Classification and Comparative Analysis of the Contents of Acorus species and Anemone altaica by UPLC-PDA Analysis (UPLC-PDA를 이용한 창포류의 분류 및 함량 분석)

  • A quantitative method using ultra performance liquid chromatography with a photodiode array detector (UPLCPDA) was established for the analysis of 2 major plant metabolites: ${\beta}$-asarone and ${\alpha}$-asarone from Acorus gramineus, A. tatarinowii, A. calamus and Anemone altaica, and their contents are compared with other herbs of Acorus species. The method was validated according to the International Conference on harmonization (ICH) guideline for validation of analytical procedures with respect to precision, accuracy, and linearity. The average content of ${\beta}$-asarone in Acorus gramineus was significantly higher than that in others, with the second highest concentration observed in A. tatarinowii, and only a trace amounts found in A. calamus and Anemone altaica. In contrast, the average content of ${\alpha}$-asarone in A. calamus was the highest, followed by that in Acorus gramineus and A. tatarinowii. principle component analysis (PCA) confirmed that ${\beta}$-asarone and ${\alpha}$-asarone content differed among the species. These results suggest that this UPLC-PDA method can be considered as good quality control criteria for Acorus gramineus.

  • The purpose of this study was to introduce the differential standard of the types of Acori Graminei Rhizoma. We established the classificatory standard according to the external and internal morphology for the several types of Acori Graminei Rhizoma. 1. External shape: Acorus gramineus has smaller size and fibroidal section. A. calamus has bigger size and cancellate section. A. tatarinowii has middle size and fibroidal section. Anemone altaica has very small long-spindle shape and flat section that has 6-12 row of vascular bundle. 2. Internal shape: Acorus gramineus has some bundle sheath fiber, but A. calamus has not. A. tatarinowii has many calcium oxalate crystalloid around bundle sheath. Anemone altaica has obscure cambium. All sample except A. altaica, in the mass, has scattered bundle, thin cell wall, broad cortex and well-developed endodermis. For the future, such results will be used as the basic source of additional research, and a far-reaching comparative study is needed to distinguish between many kinds of same genus-degree of relatedness.

Comparative Study of Changpo(Ch$\={a}$ngp$\'{u}$) species on Antioxidant Activity and HPLC Pattern Analysis (4종 창포류의 성분 패턴 및 항산화 효능 비교)

  • Objective : This study was to compare antioxidant activity and HPLC pattern analysis from 4 species of changpo(ch$\={a}$ngp$\'{u}$). Methods : To compare the antioxidant activity and HPLC pattern analysis from the 4 species of changpo, we performed the in vitro anti-oxidative activity assays and HPLC analysis from 70% ethanol extracts of Acorus gramineus Sol. (=AG), A. tatarinowii Schott (=AT), A. calamus L. (=AC) and Anemone altaica Fisch. ex C.A.Mey (=AA) taken in the herbal medicine market of Korea. Results : AG has the most effective anti-oxidative activity among 4 species of changpo. As the HPLC pattern analysis, AT was detected the unknown peak at retention time 14.9 min whereas AG was not showed any peak at the same retention time. These results suggest that AG could be used rather than AT when it need to be prescribed as anti-oxidative medicine. Conclusions : This result can be used as the basic data contributing to the stability of AG according to an appropriate clinical application.
