  • In this work, two types of copolymers are manufactured by a radical polymerization to develop a material for humid membrane. Each copolymer contains three monomers that have functions to improve humid-resistance, membrane stability, flexibility, impedance, and adhesion to the electrode. MDBAB (N,N'-dimethyl-2-methacryloxyethyl-3-bromopropyl ammonium bromide) having a salt form decreases the impedance of the humid membrane and reacts with amines to produce a cross-linking structure. HEMA (2-hydroxyethyl methacrylate) has an important role which reduces the impedance and increases the adhesion strength to the electrode. The other monomers are DAEMA (N,N'-dimethylamino ethyl methacrylate), 4-VP(4-vinyl pyridine), and 2-EHA(2-ethylhexyl acrylate) and all the monomers are formulated with several compositions to make a humid membrane. At specific composition, we could attain a satisfactory results having good performance and long term durability.

Effect of Benzalkonium Chloride on Percutaneous Absoption of Antisense Phosphorothioate Oligonucleotides

  • The effect of benzalkonium chloride on skin permeability of partially modified antisense phosphorothioate oligonucleotides (PS-ODN), which are designed as scar formation inhibitor, was investigated using Franz Diffusion Cell. When the concentration ratio of PS-ODN-quarternary ammonium salt complex is more than 1:100, the apparent partition coefficient (APC) of each complex was increased in the following order; tetraphenyl phosphonium chloride (TPP) < cetyltrimethyl ammonium bromide(CTAB) < benzalkonium chloride (BZ). The permeability of PS-ODN through the rat skin increased in the presence of BZ. The fluxs of PS-ODN with BZ were increased by addition of Pluronic F 68 or Triton X-100 to phosphate buffered saline (PBS), respectively. When the mole ratio of PS-ODN to BZ is 1:10, the fluxs penetrated of PS-ODN with BZ was greatest. The increase of the permeability in the presence of BZ might be due to the formation of lipophilic ion-pair complex between PS-ODN and BZ. By regulation of mole ratio of PS-ODN to BZ, the development of topical dosage forms using PS-ODN as scar formation inhibitor will be possible with minimal systemic exposure.

The Role of Excipients in Iontophoretic Drug Delivery: In vitro Iontophoresis of Isopropamide and Pyridostigmine through Rat Skin and Effect of Ion-pair Formation with Organic Anions

  • The iontophoretic delivery across rat skin of quaternary ammonium salts (isopropamide: ISP, pyridostigmine: PS), which are positively charged over a wide pH range, was measured ill vitro. The study showed that: (a) iontophoresis significantly enhanced delivery of ISP and PS compared to respective passive transport; (b) delivery of ISP and PS was directly proportional to the applied continuous direct current density over the range of $0-0.69\;mA/cm^2;$ (c) delivery of ISP and PS was also proportional to the drug concentration in the donor compartment over the range of $0-2{\time}l0^{-2}M:$ (d) sodium ion in the donor compartment inhibited the drug transport possibly due to decreasing the electric transference number of the drug; (e) delivery of ISP and PS increased as the pH of the donor solution increased over the pH range 2-7 suggesting permselective nature of the epidermis, and inhibition of the transference number of the drugs by hydronium ion; (f) some organic anions such as taurodeoxycholate, salicylate and benzoate which form lipophilic ion-pair complexes with ISP inhibited the delivery of ISP. The degree of inhibition by the organic anions was linearly proportional to the extraction coefficient $(K_e)$ of ISP from the partition system with each counteranion between phosphate buffer (pH 7.4) and n-octanol. For PS, however, taurodeoxycholate, but not salicylate and benzoate inhibited the iontophoretic delivery. It suggests that not only sodium ion and hydronium ion but also the counteranions which form lipophilic ion-pairs with quaternary ammonium drugs are not favorable components in formulating the donor solution of the drugs to achieve an effective iontophoretic delivery.

  • The carboxylation of the fluorenyl epoxide with a spiro framework, 9,9'-Bis(4-oxiranylmethoxyphenyl) fluorine (2) was catalyzed by some onium salts such as quaternary ammonium and phosphonium salts to produce the corresponding five-membered cyclic carbonate (3) in an efficient and environmentally benign fashion. The coupling reactions depend greatly on the kind of the halide anions and alkyl chain length of the onium salts. While the reaction was sensitive to the reaction temperature, the reaction trends suggest that the catalytic efficiency of the quaternary ammonium halides may correlate strongly with the melting points of the halides. The reactions using a catalytic amount (2 mol %) of quaternary ammonium bromide with an n-butyl chain at 75.9 bar of $CO_2$ and 393 K give the highest yield of the cyclic carbonate (92%).

  • The 2.4-kb cryptic plasmid (pKH8) of multidrug-resistant Staphylococcus aureus SA2 was characterized by complete nucleotide sequencing and homology comparison. pKH8 was found to contain three open reading frames. Protein analysis of pKH8 showed that pKH8 was a multidrug resistance plasmid and mediated resistance to ethidium bromide and quaternary ammonium compounds.

Cure and Thermal Degradation Kinetics of Epoxy/Organoclay Nanocomposite

  • Epoxy nanocomposite was synthesized through the exfoliation of organoclay in an epoxy matrix, which was composed of diglycidyl ether of bisphenol A (DGEBA), 4,4'-methylene dianiline (MDA) and malononitrile (MN). Organoclay was prepared by treating the montmorillonite with octadecyl trimethyl ammonium bromide (ODTMA). The exfoliation of the organoclay was estimated by wide angle X-ray diffraction (WAXD) analysis. In order to measure the cure rate of DGEBA/MDA (30 phr)/MN (5 phr)/organoclay (3 phr), differential scanning calorimetry (DSC) analysis was performed at various heating rates, and the data were interpreted by Kissinger equation. Thermal degradation kinetics of the epoxy nanocomposite were studied by thermogravimetric analysis (TGA), and the data were introduced to the Ozawa equation. The activation energy for cure reaction was 45.8 kJ/mol, and the activation energy for thermal degradation was 143 kJ/mol.

  • 가스 하이드레이트는 낮은 온도와 높은 압력에서 물과 천연가스 분자가 물리적으로 안정한 결합을 하고 있는 결정질의 화합물이다. 현재 130개 이상의 가스분자들이 물분자와 결합하여서 Clathrate 하이드레이트를 형성하는 것으로 보고 되어 있다. 수소의 경우 하이드레이트를 형성하기 위해서 약 200MPa 이상의 높은 압력이 필요하다. TBAB는 semi-clathrate 하이드레이트를 형성하는 첨가제로 알려져 있으며 수소 하이드레이트의 형성 압력을 완화시키기 위한 촉진제로서 많은 연구가 수행되고 있다. Wataru Shimada는 XRD 패턴을 사용하여 semi-clathrate 하이드레이트의 특수한 결정 모델을 분석하였다. 이 모델의 경우, 대기압 하에서 TBAB와 물분자의 화학 양론적 비를 1:38로 제시하였다. 이는 처음으로 TBAB 하이드레이트 결정구조를 밝혀냈지만, 분자 동역학적 부문에서는 그 데이터가 정확히 정의 되지 않았다. 본 연구는 Wataru Shimada 모델을 수소 TBAB 하이드레이이트의 분자 동역학 연구에 적용시켰으며, 실험에서 나온 데이터를 바탕으로 RDF(Radial Distribution Function)와 MSD(Mean Square Displacement)를 측정했다.

Inhibitory Effect of {Surfactant- MnO4-} Aggregation in KMnO4 Oxidation of Proline and Methionine: A Kinetic Study

  • Anionic (sodium lauryl sulphate, NaLS) cationic (cetyl ammonium bromide, CTAB) and non-ionic (Tween-80) surfactants have been found to inhibit the rate of oxiadation L-proline and L-methionine by alkaline $KMnO_4$. A first order dependence of rate of oxidation was observed with respect to $MnO_4{^-}$. The order of reaction in substrate and alkali was found to be fractional nearby 0.65 and 0.55 in Aminoacid and $OH^-$, respectively. An aggregation/association between $MnO_4{^-}$ and surfactant has been confirmed spectrophotometrically. A mechanism, involving kinetically inactive [$MnO_4{^-}$ surfactant] aggregate and consistent with kinetic data, has been proposed. The effect of surfactants has been discussed in terms of hydrophobic and electrostatic interactions.

Encapsulation of Plasmid DNA in Pegylated Liposome

  • The purpose of the study was to prepare the pegylated liposome carrying plasmid DNA with optimal encapsulation efficiency. Plasmid DNA (pCEP4 clone 790, 10.6 kb) was entrapped in the pegylated liposome composed of neutral lipid, POPC (l-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine), cationic lipid, DDAB (dimethyl dioctadecyl ammonium bromide) and anionic lipids, DSPE-PEG 2000 (distearoyl phosphatidyl ethanolamine polyethylene glycol 2000) and DSPE-PEG 2000-maleimide by freezing/thawing method. Free plasmid DNA was separated from the encapsulated one by Sepharose CL-4B column chromatography. The DNA amount encapsulated into the pegylated liposome was increased as cationic lipid concentration, initial amount of plasmid DNA and total lipid amount were increased.

Effect of clay contents on Morphology, Thermal and Mechanical properties of Polypropylene Nanocomposites.

  • Two different sources of clay, Na-Bentonite (Thai local clay) and Na-Montmorillonite were modified with Hexadodeccyltrimethyl ammonium bromide. The nanocomposites of polypropylene were successfully prepared via melt blending in a co-rotating twin screw extruder by using PP-g-MA as a compatibilizer at various contents of organoclays. The morphology of nanocomposites was investigated by using XRD and SEM. The results showed that the intercalated and exfoliated structures were obtained. The thermal behavior was also studied by using DSC and TGA. The degradation temperature of filled PP was greater than that of unfilled PP by 20%. And, the tensile strength and modulus were improved when a small amount of organoclays were added.
