Epidemiological aspects of Salmonella spp infections of domestic animals in Gyeongbuk province (경북지역 가축에서 Salmonella속 균 감염증에 대한 역학적 특성)

  • The result of studying the epidemiological characteristics of Salmonella strains which have been isolated from the domestic animals in Gyeongbuk province from February 1998 to August 2000 were summarized as follows. The isolation rates of Salmonella strains were 2.0% from cattle feces, 6.3% from cattle lymph node, 9.5% from pig feces, and 25.1% from pig lymph node. In poultry, the isolation rates were 30.3%. The isolates of Salmonella showed positive reaction for MUCAP test, methyl red test, but showed negative reaction for urea test, indole test, Voges Proskauer test. On TSI agar, the isolates showed acid butt, alkaline slant. Also, the isolates were identified as Salmonella strain by API 20E kit. Non H$_2$S Production Salmonella strains isolated from poultry were identified as S gallinarum. As a result of serotyping, B group were the most common in cattle and pig, Dl in chickens. 21 serovars were found. the common serovar from the domestic animals was S typhimurium, S derby, S agona, S schwarzenground, S enteritidis and S gallinarum. The most commonly encountered serovars in cattle were S agona and S typhirimurium in pig, S gallinarum in chicken. As a result of antimicrobial susceptibility test, all Salmonella isolates were susceptible to amikacin, ciprofloxacin, norfloxacin; cefotaxime and polumcin B. The resistance rates to tetracycline and streptomycin was 58% and 56%, respectively. 69.3% of all isolates were resistant to more than one antimicrobial agent. Out of the resistant isolates, the isolates resistant to streptomycin and tetracycline was 36%. There were 24 strains of multiresistant isolates resistant to more than 5 antimicrobial agents. S typhimurium were resistant to all antimicrobial agents, also had a lot of multiresistant strains. Therefore, S typhimurium was considered as a major agent of antimicrobial resistance.

Antimicrobial-resistant Escherichia coli isolated from dogs and cats at animal hospitals in Daegu (대구지역 동물병원에서 입원중인 개와 고양이로부터 분리된 항생제 내성 대장균)

  • This study was carried out to investigate the antimicrobial resistance profiles and resistance genes in 62 Escherichia coli isolated from dogs and cats hospitalized at animal hospitals in Daegu. E. coli isolates showed high resistance to nalidixic acid (46.8%) and ampicillin (45.2%). Resistance to the other antimicrobial agents was less than 30%, and no resistant isolates were detected for imipenem and amikacin. Of the 28 ampicillin-resistant isolates, TEM and CTX-M genes were detected in 16 (57.1%) and 11 (39.3%), respectively. The aadA gene was found in 4 (26.7%) of 15 gentamicin-resistant isolates, and strA-strB gene was found in 10 (66.7%) isolates. The sul I and sul II genes were detected in 11 (61.1%) and 14 (77.8%) of 18 trimethoprim/sulfamethoxazole-resistant isolates, and tetB gene in 9 (81.8%) of 11 minocycline-resistant isolates, and cmlA gene in 2 (22.2%) of 8 chloramphenicol-resistant isolates. The qnrB and qnrS genes were found in 3 (10.3%) and 1 (3.4%) of 28 nalidixic acid-resistant isolates, respectively. Whereas, none of the SHV, CMY-2, tetA, dfr Ia and dfr VII, and qnrA genes were found. Our results show a wide variety of resistance genes in E. coli isolates from dogs and cats. This study also represents the first report of qnrB and qnrS gene producing E. coli isolates from dogs in republic of Korea.

Biochemical characteristics and serotypes of Actinobacillus pleuropneumoniae isolated from pneumonic lungs of pigs (돼지 폐렴병소에서 분리한 Actinobacillus pleuropneumoniae의 특성에 관한 연구)

  • The present study was conducted to investigate the biochemical and serologic characteristic of Actinobacillus pleuropneumoniae isolated from pneumonic lungs of pigs during the period from January 1992 to April 1993. A pleuropneumoniae was isolated from 17(27.0%) of 63 growing pigs with respiratory signs and 21(6.4%) of 330 pneumonic lungs of slaughtered pigs. The seasonal isolation frequency of A pleuropneumoniae was higher in winter and spring than that in summer or fall. The biochemical and cultural properties of A pleluropneumoniae isolated from the pneumonic lungs of pigs were identical to those of the reference strains used. The isolates were highly susceptible to ampicillin, cephalothin, ceftiofur, ciprofloxacin(MIC : ${\leq}0.39{\mu}g/ml$) and moderately susceptible to amikacin, chloramphenicol, erythromycin, kanamycin, methicillin, penicillin-G, streptomycin(MIC : 0.78~25IU or ${\mu}g/ml$), respectively. Sulfadimethoxine, sulfamerazine, tylosine showed no response to the isolates(MIC : ${\geq}100{\mu}g/ml$). Among the 38 isolates, 21(55.3%) and 13(34.2%) were resistant to oxytetracycline aid lincomydn, respectively(MIC : ${\geq}50IU$ or ${\mu}g/ml$). The majority of 38 A pleuropneumoniae isolates were turned out as serotype 2(47.4%) or serotype 5(54.7%) and the remaining 3 isolates were evenly classified to serotype 7, 10 or 12. It was noted A pleuropneumonine serotype 5 isolates were more resistant to oxytetracycline than serotype 2 isolates.

L-glutamine:D-fructose-6-phosphate Aminotransferase as a Key Protein Linked to Multidrug Resistance in E. coli KD43162

  • A microarray study has been employed to understand changes of gene expression in E. coli KD43162 resistant to ampicillin, ampicillin-sulbactam, piperacillin, piperacillin-tazobactam, cefazolin, cefepime, aztreonam, imipenem, meropenem, gentamicin, tobramycin, ciprofloxacin, levofloxacin, moxifloxacin, fosfomycin, and trimethoprim-sulfamethoxazole except for amikacin using disk diffusion assay. Using Sodium dodecyl sulphate-polyacrylamide gel electrophoresis and MALDI-TOF MS analyses, 36 kDa of outer membrane proteins (OMPs) was found to be deleted in the multidrug resistant E. coli KD 43162. Microarray analysis was used to determine up- and down-regulated genes in relation to multidrug resistant E. coli KD43162. Among the up-regulated genes, these genes were corresponded to express the proteins as penicillin-binding proteins (PBPs), tartronate semialdehyde reductase, ethanolamine utilization protein, shikimate kinase I, allantoinase, predicted SAM-dependent methyltransferase, L-glutamine: D-fructose-6-phosphate aminotransferase (GFAT), phospho-glucosamine mutase, predicted N-acetylmannosamine kinase, and predicted N-acetylmannosamine-6-P epimerase. Up-regulation of PBPs, one of primary target sites of antibiotics, might be responsible for the multidrug resistance in E. coli with increasing amount of target sites. Up-regulation of GFAT enzyme may be related to the up-regulation of PBPs because GFAT produces N-acetylglucosamine, a precursor of peptidoglycans. One of GFAT inhibitors, azaserine, showed a potent inhibition on the growth of E. coli KD43162. In conclusion, up-regulation of PBPs and GFATs with the loss of 36 kDa OMP refers the multidrug resistance in E. coli KD 43162.

A Case of Ochrobactrum anthropi Infection after Using Medicinal Plants (약초 복용 후 Ochrobactrum anthropi에 감염 1예)

  • Ochrobactrum anthropi, previously known as Achromobacter species biotypes 1 and 2 (CDC groups Vd-1, Vd-2), belong to the groups of non-Enterobacteriaceae- nonfermentative Gram negative bacilli. Achromobacter is not presently a recognized genus. Achromobacter xylosoxidans has been transferred to genus Alcaligenes as A. xylosoxidans subsp. xylosoxidans, and "Achromobacter" sp. group Vd has been named Ochrobactrum anthropi. O. anthropi was isolated from a blood culture. Organisms were identified as O. anthropi by use of the biochemical test and the VITEK 2(bioMerieux, USA). The Organism was susceptible only to colistin, imipenem, meropenem, and tetracycline, but were resistant to amikacin, aztreonam, cefepime, ceftazidime, cefpirome, ciprofloxacin, gentamicin, isepamcin, netilmicin, pefloxacin, piperacillin, piperacillin/tazobactam, ticarcillin, ticarcillin/clavulanic acid, tobramycin, and trimethoprim/sulfamethoxazole. We report the clinical and microbiologic characteristics of O. anthropi infection in the patient. This is the first case of O. anthropi infection after using a plant as medicine at Chosun University Hospital.

A Study on Changing Patterns of Shigella spp. by Years (Shigella spp.의 연차적 추이에 관한 조사연구)

  • A total of 3,879 cases of feces and rectal swab from patient was collected in H. hospital from January 1974 to December 1986 in Seoul. Among the materials, the number of cases of Shigella spp. isolated were 197 strains of 139 patients. Infectous pattern and antibiotic sensitivity of Shigella spp. were as follows; The range of percentage of an identified Shigella spp. among total feces and rectal swabs was $1.5{\sim}12.5%$ yearly. The isolation ratios of Shigella spp. per each patient 1.35 for male and 1.19 for female. The isolation ratio of male to female was 1.28:1 in whole group. The isolated Shigella species was 81.0% in S. flexneri, 1.1% in S. boydii and 17% in S. sonnei. The highest number of Shigella spp. was found in August and September according to monthly isolation, on the other hand the lowest number of Shigella spp. was obserbed in March. The seasonal isolation rate of Shigella spp. was 31.7% in Fall, 27.3% in Summer, 21.6% in Winter and 19.3% in Spring. The age specific frequency of Shigellosis was 46.8% in $0{\sim}9$ year group, 8.6% in $10{\sim}19$, 7.2% in $40{\sim}49$ and 6.5% in $50{\sim}59$. The antibiotics showing over 80% susceptibility against Shigella spp. were gentamicin, kanamycin, amikacin, tobramycin, cefoperazone, cefoxitin, cefamandole nafate, cefotaxine and sisomycin.

Antimicrobial Susceptibility of Vibrio parahaemolyticus and Vibrio alginolyticus from Fish Farms on the Southern Coast of Korea (남해안 어류양식장에서 분리된 Vibrio parahaemolyticus와 Vibrio aiginolyticus의 항균제 감수성)

  • The antimicrobial resistance patterns to the 10 antimicrobial agents of potential pathogenic vibrios isolated from seawater and a variety of farmed fishes, including oliver flounder (Payalichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagyus major) and sea bass (Lateolabyax japonicus), were investigated from May to October, 2004. A total of 314 strains of the genus vibrios were isolated from 126 collected samples, and the number of isolated strains of Vibrio payahaemolyticus and Vibrio alginolyticus were 194 and 120, respectively. Apparently $98.5\%$ of V. parahaemolyticus and $100\%$ of V. alginolyticus isolates demonstrated antimicrobial resistance against at least one antimicrobial agent. The resistance of V. parahaernolyticus isolates to ampicillin ($97.9\%$) was highest, followed by oxolinic acid ($26.8\%$), amikacin ($19.1\%$) and tetracycline and amoxicillin/clavulanic acid ($6.7\%$). V. alginolyticus isolates were resistant to ampicillin ($100\%$), solfamethoxazoleit,imethopenem ($25\%$), amikacln ($21.7\%$), amoxicillin/clavulanic acid ($15.8\%$), ciprofloxacin ($13.3\%$), and tetracycline and doxycycline ($11.7\%$). The rate of multiple antimicrobial resistance to at least four antimicrobials was higher in the V. alginolyticus isolates ($20.8\%$) than in the V. parahaemolyticus ($6.7\%$).

Character of Listeria spp. isolated from livestock products and their related environmental areas

  • This study was carried out to investigate the characters of Listeria monocytogenes isolated from food, animal feces, dry cattle food, and the environment in Seoul and Kyonggi province during the period from 1998 to 2003. Serotyping of 70 L. monocytogenes isolates was performed according to the manufacturer's instruction. Minimum inhibitory concentrations (MICs) were determined by the microdilution method according to the guidelines of the Clinical and Laboratory Standards Institute. All the isolates were tested against 20 antimicrobial agents. The serotypes of the 70 L. monocytogenes isolates were 1/2c (62.8%), 1/2a (20%) and 1/2b (17.2%). Of the 70 L. monocytogenes isolates, 67.1%, 57.1%, 11.4%, 5.7%, 2.8%, 1.4% and 1.4% were resistant to tetracycline (Te), minocycline (Mi), norfloxacin (Nor), ciprofloxacin (Cip), neomycin (N), chloramphenicol (C) and cephalothin (Cf), respectively. However, all isolates were 100% sensitive to antibiotics such as amikacin, ampicillin, erythromycin, gentamycin, imipenem, kanamycin, ofloxcin, streptomycin, penicillin, trimethoprim, trimethoprim/sulfamethoxazole, tobramycin, and vancomycin. Multiple resistance patterns of the isolates were observed in TeMiNor Cip (1.4%), TeMiNor (7.1%), TeMiCip (2.9%), TeMiN (1.4%) and TeMi (44.3%). The results of this study indicate that many L. monocytogenes isolates are resistant to antimicrobial agents including Te and Mi. The possibility that the isolates could increasingly acquire multiple antimicrobial resistant properties cannot be precluded.

In vitro antibacterial activity, postantibiotic effects of norfloxacin and its interaction effects in combination with other antibiotics (시험관내에서 norfloxacin의 항균력과 다른 항생제와 병용투여시 상호작용)

  • 국내에서 많이 사용되고 있는 제2세대 quinolone 항생제인 norfloxacin(NFX)에 대한 약역학적인 특성을 구명하기 위하여 국내에서 분리된 동물유래 병원성 세균에 대하여 시험관내에서 실험을 수행하여 다음과 같은 결과를 얻었다. 즉, E coli(n=89) 대한 NFX의 $MIC_{50}$$MIC_{90}$는 공히 0.02g/ml이었으며, Streptococcus spp.(n=36)에 대한 NFX의 $MIC_{50}$는 2g/ml 그리고 $MIC_{90}$는 4g/ml로 나타났다. Salmonella spp.(n=56)에 대한 NFX의 $MIC_{50}$$MIC_{90}$ 모두 0.2g/ml로 강한 항균력을 보였으며, Streptococcus spp.(n=24)에 대한 NFX의 $MIC_{50}$는 2g/ml 그리고 $MIC_{90}$가 4g/ml로 나타났다. Bacillus spp.(n=34)는 NFX의 $MIC_{50}$$MIC_{90}$는 모두 0.4g/ml으로서 대부분의 병원성 세균에 대해서 $MIC_{50}$$MIC_{90}$치가 동일하든지 또는 매우 비슷한 수치를 보여주었다. 그러나 NFX는 혐기성세균인 Clostridium spp.(n=34)에 대해서는 항균력이 매우 낮았다. 현재 수의임상에서 항균제 병용요법이 많이 응용되고 있는 것을 고려하여 NFX와 다른 항생물질간의 분획억제농도 (FICs)를 E coli 88ac을 시험균주로 하여 실험한 결과, NFX와 colistin과 병용할 때 FIC 값이 0.38로서 상승작용을 그리고 gentamicin, trimethoprim, amikacin, penicillin 및 tylosin과의 병용시 FIC 값이 각각 0.52, 0.56, 0.63, 1.00 및 1.02로서 상가작용을 보여주었으며, tetracyclin과의 병용시의 FIC값은 1.49로서 길항작용을 나타냄을 알 수 있었다. 한편 실제 항균제의 임상적용시 매우 주요한 요소인 항균활성후 저농도유효성(PAE)을 알아보기 위하여 E coli AB1157을 시험균주로 측정한 결과 PAE은 0.90~1.02 시간 그리고 S aureus R-209에 대해서는 PAE가 1.58~1.99 시간으로서 그람음성균 및 그람양성균 모두에 대해서 긴 PAE를 갖고 있음을 알 수 있었다.

Identification and characterization of Shiga toxin-producing Escherichia coli isolated from diarrhea in calves (송아지 설사분변으로부터 Shiga toxin-producing Escherichia coli 의 분리 및 특성규명)

  • Shiga toxin (stx) producing Escherichia coli (STEC) causes various clinical signs in animal and human. In this study, 255 fecal samples from calves showing diarrhea were collected from cattle farms in Chonnam province during the period from January 2005 to July 2005. Twenty six STEC (10%) were isolated from 255 fecal samples by PCR. The isolates displayed three different stx combinations (stx1 [69%], stx1 and stx2 [15%], and stx2 [38%]). The isolates were further studied for virulence associated genes and antimicrobial resistance to define the virulence properties. Intimin (eaeA), enterohemolysin (hlyA), and lipopolysaccharide (rfbE) virulence genes were detected in 6 (23%), 7 (26%), and 1 (3.8%) of the isolates, respectively, by PCR. One isolate possessing rfbE gene was typed as E. coli 0157 : H7 by agglutination test with O and H antisera. All 26 isolates showed susceptibility to amikacin (100%) and the majority of isolates showed high susceptibility to gentamicin (88.5%) and chloramphenicol (73.1%). But all isolates were resistant to penicillin. These results may provide the basic knowledge to establish strategies for the treatment and prevention of enteric disease in calves.
