서양삼 추출물의 초단파 및 식초 처리에 의한 인삼 사포닌 성분 변화 (The Change of Ginsenoside Composition in American Ginseng (Panax quinquefolium) Extract by the Microwave and Vinegar Process)

  • The purpose of this study is to develop a new preparation process of American ginseng (Panax quinquefolium) extract featuring high concentration of ginsenoside $Rg_3$, $Rg_5$, and $Rk_1$, Red ginseng special components. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of American ginseng were processed under several treatment conditions of microwave and vinegar (about 14% acidity). The results showed that the quantity of ginsenoside $Rg_3$ increased by over 0.9% at the 20 minutes of the pH 2~4 vinegar and microwave American ginseng ethanol extract compared with other process times. The result of MAG-20 indicates that the American ginseng microwave and vinegar-processed American ginseng extracts (about 14% acidity) treated for 20 minutes produced the highest amount of ginsenoside $Rg_3$ (0.969%), $Rg_5$ (1.071%), and $Rk_1$ (0.247%). Besides, MAG-15 indicates that the microwave - and vinegar-processed American ginseng extracts (about 14% acidity) treated for 15 minutes produced the highest amount of ginsenoside $Rg_3$ (0.772%), $Rg_5$ (1.330%), and $Rk_1$ (0.386%). This indicates that American ginseng treated with microwave and vinegar had the quantity of the ginsenoside $Rg_3$ over 32 times the amount of the ginsenoside $Rg_3$ (which was not found in raw and American ginsengs) in the average commercial Red ginseng.

고려홍삼 성분이 암독소 호르몬-L의 체지방 분해작용에 미치는 영향 (Effect of Korean Red Ginseng Component on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid)

  • This study was devised to observe the inhibitory effects of 3 kinds of petroleum ether extracts (percolation by petroleum ether) from Korean red ginseng, Chinese red ginseng and American white ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been Inoculated with sarcoma-180. The yields of petroleum ether extract from Korean red ginseng, Chinese red ginseng and American white ginseng were 0.64, 0.47 and 0.58 and respectively, indicating that the yield of Korean red ginseng was the highest. In vitro, at the concentration of 2 mg /ml, the inhibition rate of lipolysis by the petroleum ether extract of Korean red ginseng, Chinese red ginseng and American white ginseng were 55.1, 50.0 and 44.9% respectively, and the total inhibitory activity per gram of ginseng material were 18, 12 and 13 unit respectively, indicating that the Korean red ginseng was the most effective in the inhibition of the lipolysis.

Phytochemical Characteristics of Coffee Bean Treated by Coating of Ginseng Extract

  • The principal objective of this study was to assess the instrumental and sensory characteristics of ginseng coffee with different ratios of the ingredients: type of coffee bean (Colombia, Brazil, and Indonesia), type of ginseng extract (white ginseng, red ginseng, and America ginseng) and concentration of ginseng extract (3, 6, and 9 w/v %). The sensory optimal condition of white ginseng coffee, red ginseng coffee and America ginseng coffee were as follows: 3% Indonesian coffee bean coated with 3% white ginseng extract, Colombian coffee bean coated with 6% red ginseng extract and Colombian coffee bean coated with 3% American ginseng extract, respectively. In particular, the Colombian coffee bean coated with 6% red ginseng extract had significantly higher scores than other samples in terms of flavor, taste, and overall preference. Additionally, the contents of total ginsenoside and total sugar and total phenolic compounds were also highest in the Colombian coffee bean coated with 6% red ginseng extract.

North American ginseng influences adipocyte-macrophage crosstalk regulation of inflammatory gene expression

  • Background: Adipocyte-macrophage communication plays a critical role regulating white adipose tissue (WAT) inflammatory gene expression. Because WAT inflammation contributes to the development of metabolic diseases, there is significant interest in understanding how exogenous compounds regulate the adipocyte-macrophage crosstalk. An aqueous (AQ) extract of North American (NA) ginseng (Panax quinquefolius) was previously shown to have strong inflammo-regulatory properties in adipocytes. This study examined whether different ginseng extracts influence adipocyte-macrophage crosstalk, as well as WAT inflammatory gene expression. Methods: The effects of AQ and ethanol (EtOH) ginseng extracts ($5{\mu}g/mL$) on adipocyte and macrophage inflammatory gene expression were studied in 3T3-L1 and RAW264.7 cells, respectively, using real-time reverse transcription polymerase chain reaction. Adipose tissue organ culture was also used to examine the effects of ginseng extracts on epididymal WAT (EWAT) and inguinal subcutaneous WAT (SWAT) inflammatory gene expression. Results: The AQ extract caused significant increases in the expression of common inflammatory genes (e.g., Mcp1, Ccl5, Tnf-${\alpha}$, Nos2) in both cell types. Culturing adipocytes in media from macrophages treated with the AQ extract, and vice versa, also induced inflammatory gene expression. Adipocyte Ppar-${\gamma}$ expression was reduced with the AQ extract. The AQ extract strongly induced inflammatory gene expression in EWAT, but not in SWAT. The EtOH extract had no effect on inflammatory gene expression in either both cell types or WAT. Conclusion: These findings provide important new insights into the inflammo-regulatory role of NA ginseng in WAT.

고려인삼과 미국삼 뿌리 중심부의 단백질 패턴 비교 (Comparison of Protein Patterns of the Root Pith from Panax ginseng and Panax quirnquefolium)

  • The purpose of this study was to analyze the electrophoretic patterns of soluble proteins from ginseng roots and to compare the protein patterns from Korean ginseng and American quinquefolium. The size difference was found in the major protein bands of a molecular weight of about 27,000 between Korean ginseng and American quinquefolium. The protein band of a molecular weight of 22,000 showed a quantitative difference in its amount. The major 27 K proteins appeared to form a complex heterodimer of 66,000 and to have internal bisulfide bonds, from band shifting studies under non-denaturing conditions. Three peaks appeared when the protein extract from root homogenates was purified using gel filtration and DEAE ion exchange chromatography. The examination of physiological activity and further purification of these fractions are underway.

Increased Production of Ginsenoside Compound K by Optimizing the Feeding of American Ginseng Extract during Fermentation by Aspergillus tubingensis

  • The ginsenoside compound K (C-K) is widely used in traditional medicines, nutritional supplements, and cosmetics owing to its diverse pharmacological activities. Although many studies on C-K production have been conducted, fermentation is reported to produce C-K with low concentration and productivity. In the present study, addition of an inducer and optimization of the carbon and nitrogen sources in the medium were performed using response surface methodology to increase the C-K production via fermentation by Aspergillus tubingensis, a generally recognized as safe fungus. The optimized inducer and carbon and nitrogen sources were 2 g/l rice straw, 10 g/l sucrose, and 10 g/l soy protein concentrate, respectively, and they resulted in a 3.1-fold increase in the concentration and productivity of C-K (0.22 g/l and 1.52 mg/l/h, respectively) compared to those used before optimization without inducer (0.071 g/l and 0.49 mg/l/h, respectively). The feeding methods of American ginseng extract (AGE), including feeding timing, feeding concentration, and feeding frequency, were also optimized. Under the optimized conditions, A. tubingensis produced 3.96 mM (2.47 g/l) C-K at 144 h by feeding two times with 8 g/l AGE at 48 and 60 h, with a productivity of 17.1 mg/l/h. The concentration and productivity of C-K after optimization of feeding methods were 11-fold higher than those before the optimization (0.22 g/l and 1.52 mg/l/h, respectively). Thus, the optimization for the feeding methods of ginseng extract is an efficient strategy to increase C-K production. To our knowledge, this is the highest reported C-K concentration and productivity via fermentation reported so far.

고려인삼이 Fusarium oxysporum에 의한 중금속 수은의 해독작용에 미치는 영향 (Effect of Korea Ginseng Root on Detoxification of Heavy Metal, Mercury by Fusarium oxysporum)

  • Extracts of Panax ginseng root significantly induced tolerance of Fusarium oxysporum to heavy metal, mecury, as the fungal mycelial growth was less inhibited by mercury chloride on potato dextrose medium(PDA) amended with ginseng root than on the PDA with no ginseng amendment. The most favorable concentration of ginseng root powder in detoxification of mercury chloride was 1%. The induced tolerance of F. oxysporum to mercury chloride appeared to be rather due to absorption of ginseng components, and was not related to stimulation of mycelial growth of the fungus per so by ginseng treatment. Ginseng components responsible for inducing tolerance of the fungus to mercury were involved in the water fraction of the ginseng root extract, although the water fraction had no effect on enhancement of the mycelial growth on the medium without mercury chloride. The hexane fraction of ginseng root extract, by which the mycelial growth was stimulated, was not related to the inducement of the tolerance to mercury chloride. However, more tolerance to mercury chloride was noted in PDA with both the water and hexane fractions combined than with either of the two fractions. Six-year-old ginseng roots were more effective in detoxification of mercury chloride than 4-year-old ginsng roots, and American ginseng (P quinquifolium) had no or little effect on inducing tolerance of the fungus to mercury chloride. This method may be used to screen other natural materials for test in the detoxification of mercury chloride.

American ginseng attenuates azoxymethane/dextran sodium sulfate-induced colon carcinogenesis in mice

  • Background: Colorectal cancer is a leading cause of cancer-related death, and inflammatory bowel disease is a risk factor for this malignancy. We previously reported colon cancer chemoprevention potential using American ginseng (AG) in a xenograft mice model. However, the nude mouse model is not a gut-specific colon carcinogenesis animal model. Methods: In this study, an experimental colitis and colitis-associated colorectal carcinogenesis mouse model, chemically induced by azoxymethane/dextran sodium sulfate (DSS) was established and the effects of oral AG were evaluated. The contents of representative ginseng saponins in the extract were determined. Results: AG significantly reduced experimental colitis measured by the disease activity index scores. This suppression of the experimental colitis was not only evident during DSS treatment, but also very obvious after the cessation of DSS, suggesting that the ginseng significantly promoted recovery from the colitis. Consistent with the anti-inflammation data, we showed that ginseng very significantly attenuated azoxymethane/DSS-induced colon carcinogenesis by reducing the colon tumor number and tumor load. The ginseng also effectively suppressed DSS-induced proinflammatory cytokines activation using an enzyme-linked immunosorbent assay array, in which 12 proinflammatory cytokine levels were assessed, and this effect was supported subsequently by real-time polymerase chain reaction data. Conclusion: AG, as a candidate of botanical-based colon cancer chemoprevention, should be further investigated for its potential clinical utility.

Korean Red Ginseng protects endothelial cells from serum-deprived apoptosis by regulating Bcl-2 family protein dynamics and caspase S-nitrosylation

  • Korean Red Ginseng extract (KRGE) is a traditional herbal medicine utilized to prevent endothelium dysfunction in the cardiovascular system; however, its underlying mechanism has not been clearly elucidated. We here examined the pharmacological effect and molecular mechanism of KRGE on apoptosis of human umbilical vein endothelial cells (HUVECs) in a serum-deprived apoptosis model. KRGE protected HUVECs from serum-deprived apoptosis by inhibiting mitochondrial cytochrome c release and caspase-9/-3 activation. This protective effect was significantly higher than that of American ginseng extract. KRGE treatment increased antiapoptotic Bcl-2 and Bcl-$X_L$ protein expression and Akt-dependent Bad phosphorylation. Moreover, KRGE prevented serum deprivation-induced subcellular redistribution of these proteins between the mitochondrion and the cytosol, resulting in suppression of mitochondrial cytochrome c release. In addition, KRGE increased nitric oxide (NO) production via Akt-dependent activation of endothelial NO synthase (eNOS), as well as inhibited caspase-9/-3 activities. These increases were reversed by co-treatment of cells with inhibitors of eNOS and phosphoinositide 3-kinase (PI3K) and pre-incubation of cell lysates in dithiothreitol, indicating KRGE induces NO-mediated caspase modification. Indeed, KRGE inhibited caspase-3 activity via S-nitrosylation. These findings suggest that KRGE prevents serum deprivation-induced HUVEC apoptosis via increased Bcl-2 and Bcl-$X_L$ protein expression, PI3K/Akt-dependent Bad phosphorylation, and eNOS/NO-mediated S-nitrosylation of caspases. The cytoprotective property of KRGE may be valuable for developing new pharmaceutical means that limit endothelial cell death induced during the pathogenesis of vascular diseases.

Gas Liquid Chromatography에 의한 각종(各種) 인삼(人蔘) 제품(製品) 및 오가피(五加皮)의 유리당(遊離糖) 조성(組成)에 관(關)한 연구(硏究) (Studies on Free Sugars in Various Ginseng Products and Acanthopanax by Gas Liquid Chromatography)

  • 한국산(韓國産) 홍삼(紅蔘), 한국(韓國), 미국(美國) 및 캐나다산(産)의 피부(皮付) 백삼(白蔘), 홍삼(紅蔘) 및 백삼(白蔘)의 추출액(抽出液), 한국(韓國) 및 쏘련산(産)의 오가피(五加皮)에 대하여 유리당(遊離糖)의 조성(組成)을 가스 크로마토그래피에 의하여, 측정(測定)한 결과(結果)는 다음과 같다. 1. 한국(韓國) 홍삼(紅蔘)과 미국(美國) 및 한국산(韓國産)의 피부(皮付) 백삼(白蔘)에서 ${\beta}-fructose,\;{\alpha}-fructose,\;{\alpha}-glucose$, galactose, ${\beta}-glucose,\;sucrose,\;{\alpha}-maltose,\;{\beta}-maltose$를 동정(同定)할 수있었고, 캐나다산(産) 피부(皮付) 백삼(白蔘)에서 위의 유리당(遊離糖) 가운데서 ${\alpha}-maltose,\;{\beta}-maltose$을 검출(檢出) 할 수 없었다. 그리고 피부(皮付) 백삼(白蔘)에는 홍삼(紅蔘)보다 유리당(遊離糖)의 함량(含量)이 훨씬 많았다. 2. 홍삼(紅蔘) 및 백삼(白蔘) 추출액(抽出液)에서 ${\beta}-fructose,\;{\alpha}-fructose,\;{\alpha}-glucose,\;galactose,\;{\beta}-glucose,\;sucrose,\;{\alpha}-maltose,\;{\beta}-maltose$를 동정(同定)할 수 있었고, 홍삼(紅蔘) 추출물(抽出物)에는 fructose, 백삼(白蔘) 추출액(抽出液)에는 sucrose의 함량(含量)이 가장 많았다. 3. 쏘련산(産) 오가피(五加皮)에서 ${\beta}-fructose,\;{\alpha}-glucose\;galactose,\;{\beta}-glucose,\;sucrose,\;{\alpha}-maltose,\;{\beta}-maltose$를 동정(同定)할 수있어서 인삼(人蔘) 유리당(遊離糖)의 패턴과 오히려 비슷하였다. 그러나 한국산(韓國産) 오가피(五加皮)에서는 ${\beta}-fructose,\;{\alpha}-$${\beta}-glucoses$, sucrose가 동정(同定) 되었을 뿐이고 함량(含量)은 쏘련산(産) 오가피(五加皮)의 1/3에 지나지 않았다.
