Expression of Nutritionally Well-balanced Protein, AmA1, in Saccharomyces cerevisiae

  • Food yeast, Saccharomyces cerevisiae, is a safe organism with a long history of use for the production of biomass rich in high quality proteins and vitamins. AmA1, a seed storage albumin from Amaranthus hypochondriacus, has a well-balanced amino acid composition and high levels of essential amino acids and offers the possibility of further improving food animal feed additives. In order to find an effective means of expressing AmA1 in yeast, the gene was cloned into an episomal shuttle vector. Four different promoters were tested: the glyceraldehyde-3-phosphate dehydrogenase promoter, galactose dehydrogenase 10 promoter, alcohol dehydrogenase II promoter, and a hybrid ADH2-GPD promoter. The recombinant AmA1 genes were then introduced into the yeast Saccharomyces cerevisiae 2805. Northern and Western blot analyses of the yeast under appropriate conditions revealed that AmA1 was expressed by all four promoters at varying levels. An enzyme-linked immunosorbent assay demonstrated that the amount of AmA1 protein in the recombinant yeast was 1.3-4.3% of the total soluble proteins. The highest expression level was obtained from the hybrid ADH2-GPD promoter.

Microstructures and Mechanical Properties of AM80-xSn Magnesium Alloys with Semi-Solid casting and Hot Extrusion Process (AM80-xSn 마그네슘 합금의 반응고 주조 및 압출에 따른 미세조직 및 기계적 특성)

  • In a recent study, the microstructures and mechanical properties of AM80-xSn magnesium alloys with semi solid casting and hot extrusion process were investigated. With increasing Sn content, the amount of ${\beta}$(Mg2Sn) phase increased, while the ${\alpha}-Mg$ dendritic size decreased. The hardness was increased by the Mg2Sn as the Sn content increased. With increasing Sn content, permanent mold cast and semi solid cast AM80 Mg alloy showed less reduction of hardness and also of extruded AM80 Mg alloy after annealing. In the case of the mechanical properties, the extruded semi solid casting AM80 Mg alloy showed higher tensile strength and yield strength with increasing Sn content compared to the extruded permanent mold cast AM80 Mg alloy at room temperature.

Multi-step Metals Additive Manufacturing Technologies

  • Metal additive manufacturing (AM) technologies are classified into two groups according to the consolidation mechanisms and densification degrees of the as-built parts. Densified parts are obtained via a single-step process such as powder bed fusion, directed energy deposition, and sheet lamination AM technologies. Conversely, green bodies are consolidated with the aid of binder phases in multi-step processes such as binder jetting and material extrusion AM. Green-body part shapes are sustained by binder phases, which are removed for the debinding process. Chemical and/or thermal debinding processes are usually devised to enhance debinding kinetics. The pathways to final densification of the green parts are sintering and/or molten metal infiltration. With respect to innovation types, the multi-step metal AM process allows conventional powder metallurgy manufacturing to be innovated continuously. Eliminating cost/time-consuming molds, enlarged 3D design freedom, and wide material selectivity create opportunities for the industrial adoption of multi-step AM technologies. In addition, knowledge of powders and powder metallurgy fuel advances of multi-step AM technologies. In the present study, multi-step AM technologies are briefly introduced from the viewpoint of the entire manufacturing lifecycle.

Structure Elucidation of a Potent Anti-MRSA Antibiotic, AM3, Produced by Streptomyces sp. (방선균에 의해 생산된 항 MRSA 항생물질 AM3의 구조 연구)

  • In order to find a potent anti-methicillin resistant Staphylococcus aureus (MRSA) antibiotic, actinomycetes isolated from the samples collected in Korean marine silt were screened. From the culture broth of the isolated Streptamyces strain AM045, a substance showing excellent biological activity against MRSA was found, isolated and named AM3. The compound showed strong activities against MRSA, S. epidermidis, E. faecium and E. faecalis, which were better than those of vancomycin and teicoplanin. Unfortunately, AM3 was identified as Actinomycin V. However, this paper reports the three dimensional study of AM3 based on high resolution nmr and Computer Aided Molecular Modeling(CAMM), and the fact that the structure of the pentapeptide lactone ring with oxo-proline in chloroform solution does not have 'C conformation' any more.

Functional Characteristics from the Barley Leaves and its antioxidant mixture - Study on the Nitrite Scavenging Effect - (맥엽 및 항산화 조성물의 생리 기능적 특성에 관한 연구 - 아질산염 소거능을 중심으로 -)

  • This study was conducted to estimate the biofunctional characteristics, namely nitrite scavenging effect of barley leaves (BL) and their mixture (AM) with other antioxidants. Aqueous or methanol extracts were obtained from BL and AM. Aqueous and methanol extracts from barley leaves had relatively higher nitrite scavenging effects and its activities and contents increased in a concentration-dependent manner. The activities and contents of methanol extracts obtained from BL and AM were higher than those of aqueous extracts. Especially, AM containing BL and other antioxidant mixture had the highest activities and contents increased in a concentration-dependent manner. BL or AM were added to macrophage cells of RAW 264.7. Survival rates of the cells treated with BL and AM were measured to be different. $IC_{50}$ value decreased in a concentration-dependent manner by the addition of aqueous or methanol extracts from BL and AM. Especially, methanol extracts of AM had the highest nitrite scavenging effects. Thus, BL and AM may have protective effect against carcinogen and immune ability against reactive molecules through NO (nitric oxide) signal pathway. From the above results, barley leaves appear to contain natural anticancer and immune-related agents and may have potentiality to be used as functional food ingredients.

Comparison of the Marginal and Internal Gap of Metal Coping according to Processing Method of Dental CAD/CAM System (치과 캐드캠 시스템의 가공 방식에 따른 금속 코핑의 적합도 비교)

  • The purpose of this study was to evaluate the marginal and internal gap of metal coping fabricated using additive manufacturing (AM) group and subtractive manufacturing (SM) group by dental computer-aided design (CAD)/computer-aided manufacturing (CAM) systems. Twenty same cases of stone models of abutment teeth 16 by the universal numbering system were manufactured and scanned. Ten metal copings of control group were fabricated using SM and ten metal coping of experimental group were fabricated using AM. Marginal and internal gap of copings were measured using the silicone replica technique and digital microscope (${\times}140$). The data were analyzed using IBM SPSS 21.0 Statistical Software for independent samples t-test (${\alpha}=0.05$). Mean${\pm}$ standard deviation (SD) of marginal and internal gap total size of SM group was $101.00{\pm}40.33{\mu}m$ of AM group was $83.61{\pm}40.37{\mu}m$. Mean${\pm}$SD of marginal and internal gap total size of SM group was significantly greater than that of AM group (p<0.05). This study showed that AM metal copings had a better marginal and internal gap than SM metal copings.

  • LCD를 극복할 수 있는 유일한 대안이 AM OLED라는 판단에 따라 일본업체들은 소형 AM OLED의 초기 생산체제에 돌입했다. 이에 비해 국내에서는 AM OLED 사업화의 필요성은 인정하고 있으나, 여전히 시장 지배력을 확보하고 있는 TFT LCD에 무게 중심을 두고 있어 AM OLED는 일본보다 사업화가 늦어질 것으로 보인다. 현재 세계 OLED 시장은 2006년에는 10억 달러, 2009년에는 20억 달러로 성장할 것으로 예상되는 등 2011년까지 연평균 30% 이상의 안정적인 성장세를 보일 것으로 전망된다. 이들 대부분은 중소형 모바일 제품에 탑재되는 것이다.

모의 방사성용액에서 옥살산에 의한 네오디뮴과 아메리슘의 공침전

  • 알카리 및 알카리토금속, 전이금속원소, 액티나이드원소등 10개 원소로 구성된 질산매질의 모의 방사성용액에서 옥살산에 의한 Nd와 Am의 공침전 연구를 수행하였다. 모의용액중의 질산농도와 옥살산농도에 따른 Nd과 Am의 침전율과 정제도가 연구되었다. 각 원소들의 침전율은 옥살산이 증가함에 따라서 증가하고 질산농도에 따라서는 감소하였다. 각각 0.5M인 옥살산농도와 질산농도에서 Nd과 Am의 침 전율이 99%이상이 되고, 정제도 측면을 고려할 때 가장 좋은 것으로 판단되었다. Am은 Nd와 매우 잘 공침전됨을 확인하였으며, Zr이 존재할 시 Cs, Sr, Pd등과 공침됨을 알 수 있었다.

Seasonal Changes in Colonization and Spore Density of Arbuscular-Mycorrhizae in Citrus Groves (감귤뿌리에서의 Arbuscular-Mycorrhizae 형성과 감귤원 토양중 포자밀도의 계절적 변화)

  • In four citrus grow of Satsuma mandarin (rootstock of trifoliate orange) including two grove of organical management and two groves of conventional management, spores of arbuscular mycorrhizal(AM) fungi were identified and seasonal changes in spore density in soils and AM colonization of citrus roots were investigated. AM colonization in weeds found in the groves were also examined. Three species of Glomus (G.deserticola, G. vesiculiferum, G. rubiforme ) and one unknown species of Acaulospora were observed in all of the groves. Annual mean density of AM fungal spores were in the range of 10,000${\sim}$40,000 per 100g soil with more spores in the organically-managed groves. The least spores were observed in December in all groves, and the most spores in April in the organically-managed groves while in February or April in the conventionally- managed. Annual mean AM colonization more 27% of citrus root were observed in the organically-managed with the high peaks in April and October and the minimum in August, while mean colonization less than 15% in the conventionally-managed with the peak in February and the minimum in different times depending on groves and years. AM colonization corresponded to a sigmoidal curve consisting of a laf phase during winter and a subsequent increase in spring, then succeeded by a maximum, and then a decrease at the end of vegetation. Fungal spore density and AM colonization showed a parallel pattern during the sample period. The seasonality appeared to be related more to the phenology of the plant than to the soil factors. Generally more spore density and AM colonization were found in organically managed groves. AM colonization was not correlated with available P and organic matter content in soil in this field investigation. Among sixteen weed species found in the groves, Astrogalus sinicus of Leguminosae, Portulaca oleracea of Portulacaceae showed high colonization in all groves and they can be considered as a source of inoculumn and host plants for propagation of AM fungi.
