High Occurrence Conditions of Hollow Heart and Internal Brown Spot in Potatoes (Solanum tuberosum L.) (내부갈색반점(內部褐色斑點)과 내부공동(內部空洞) 저항성(抵抗性) 감자 계통 선발을 위한 다발성(多發生) 조건 구명(究明))

  • To screen potato clones with high resistance to hollow heart (HH) and internal brown spot (IBS), field conditions were set up to induce high frequencies of physiological disorders in 'Atlantic' potato through various treatments of mulching, periodic shadings, and plant growth regulators such as trinexapac-ethyl (Tr-E) and dicloprop-triethanol mine (DTA). IBS occurrence was as high as 67.5% in the field plot mulched with transparent film and shaded for 10 days beginning at 80 days after planting. The highest level of HH was 22.9% in the plot mulched with transparent film and shaded for 10 days beginning at 60 days after planting. Very high level of IBS (66.3%) also occurred in the plot treated with 1500 mg/L of Tr-E 40 days after planting, while HH occurred by 21.3% in the plot treated with 1000 mg/L of DTA 70 days after planting. In the plots which were treated with 1,500 mg/L of Tr-E after 40 days of planting and 1000 mg/L of DTA after 70 days of planting, 'Superior' (moderately highly resistant) and 'Atlantic' (very susceptible) could be clearly distinguished to be resistant and susceptible. High occurrence condition set up in this study could be applied for the potato breeding program to screen potato clones with high resistance to HH and IBS.

Effect of Planting Date, Plant Spacing, and Harvest Time on the Production of Small-sized Sweetpotato in the Alpine Zone of Korea (재배조건이 소형 간식용 고구마 생산에 미치는 영향)

  • The optimum planting date, plant spacing, and harvest time for production of small-sized sweetpotato was evaluated in 2003 to 2004 at the Mokpo Experiment Station. Small-sized sweetpotato is important as snack food in Korea. Optimum planting date and plant spacing were with plastic mulch, and mid-April and $75{\times}15{\sim}20cm$ respectively, mid-June and $75{\times}15cm$ respectively without plastic mulch. Storage root yield decreased with early harvest, but the ratio of small-sized sweetpotato was higher in plots harvested after 100 days of planting (DAP) than that of 80 or 120 DAP. Considerable differences in storage root yield was noted with planting dates and plant spacings, but the use of plastic mulch resulted to smaller yield variations in plots, harvested in mid-April. Planting in mid-April and harvesting after 100 DAP increased income by about $32{\sim}61%$ compared with plantings mid-May.

Survey on pesticide use by chinese cabbage growers in gangwon alpine farmland (강원도 고냉지대 배추 경작자들의 농약 사용 실태)

  • The objective was to know if chinese cabbage growers in Gangwon alpine farmland control agricultural pests including weeds effectively and use pesticide properly. Examiners visited 185 farmers at Taebaek, Pyongchang, and Jeongseon and surveyed 33 questions on pest control methods pesticide use. Chinese cabbage farmers have noxious plant diseases such as clubroot, bacterial soft rot, downy mildew, anthracnose, and mosaic disease, and also noxious insects such as diamondback moth, aphid, beet armyworm, common cabbage worm, and Japanese native slug. In addition, farmers have noxious weeds such as common chickweed, marsh pepper, hairy crabgrass, common purslane, and horseweed. To control diseases and insects, 51.3% of farmers used many chemical agents, while 20.7% of farmers used chemical agents with too much emphasis on paraquat and glyphosate to control weeds: 87.2% of the answered farmers have a preference of the both non-selective herbicides. Farmers in the survey area selected pesticides on the basis of their own experience and sales managers' recommendation (84.2%) which results in the use of inappropriate pesticides such diniconazole. Many farmers have experienced phytotoxicities (46.7%) and pesticide poisoning (51.2%). We conclude that a systematic educational program for the proper selection and use of pesticides should be conducted for chinese cabbage growers in Gangwon alpine farmland.

Development of a Nutrient Solution for Potato (Solanum tuberosum L.) Seed Tuber Production in a Closed Hydroponic System (수경재배용 감자 배양액 개발)

  • To develop a nutrient solution for a closed hydroponic system in potato (Solanum tuberosum L.) 'Atlantic' and 'Superior' potatoes were grown with the nutrient solutions whose strengths were 0.25, 0.5, 1.0, and 1.5 of the concentration of the nutrient solution developed by the National Horticultural Experiment Station in Japan. The best results in potato growth and yield were obtained with 0.5 and 1.0 strength nutrient solutions, and nutrient compositions for potato were determined based on the 1.0 strength nutrient solution; $14.4me{\cdot}L^{-1}\;N,\;4.2me{\cdot}L^{-1}\;P,\;7.5me{\cdot}L^{-1}\;K,\;5.5me{\cdot}L^{-1}\;Ca$, and $3.5me{\cdot}L^{-1}\;Mg$ for stolon growth stage and $14.8me{\cdot}L^{-1}\;N,\;4.0me{\cdot}L^{-1}\;P,\;8.5me{\cdot}L^{-1}\;K,\;6.5me{\cdot}L^{-1}\;Ca$, and $3.0me{\cdot}L^{-1}\;Mg$ for tuber growth stage. To examine the suitability of the nutrient solutions developed for potato, the strengths of 1.0 (PS 1.0S), 0.75 (PS 0.75S), or 0.5 (PS 0.5S) were compared with half-strength of Japanese Horticultural Experiment Station' solution (JH 0.5S). Changes in pH, EC, and mineral concentrations in nutrient solutions depended more on solution strength and growth stage than on the type of nutrient solution. However, most elements in solution remained constant with plant age in PS 0.75S solution during stolon growth stage, and in PS 0.5S solution during tuber growth stage. The greatest growth and tuber yield was obtained in the standard strength solution (PS 1.0S), and potato solution developed in this experiment was recommended for hydroponic culture of potato in a closed system.

  • There are the farmer's needs to develop early-maturing cultivar adaptable to mid-northern inland plain and alpine area. Furthermore, it is required to develop a rice variety to produce new rice before concentrated marketing dates, even in the years of early Chuseok. 'Joun' is a new extremely early-maturing japonica rice cultivar developed in 2009 from the cross of SR14880-173-3-3-2-2-2/Unbong20 at Cheolwon Substation, National Institute of Crop Science (NICS), Rural Development Administration (RDA). The heading date of 'Joun' is July 23 in mid-northern alpine area, which is 7 days earlier than that of Odaebyeo. It has about 61 cm in culm length with semi-erect plant type. Panicle has a few awns and its exertion is good. The number of spikelets per panicle is smaller than that of Odaebyeo and 1,000 grain-weight of brown rice is 21.2 g which is less than 26.3 g of Odaebyeo, but the complete grain ratio is higher. Milled kernels are translucent with non-glutinous endosperm and palatability of cooked rice is good. It shows strong resistance to cold treatment, lodging, premature heading, wilting and viviparous germination during ripening stage. This cultivar shows resistance to leaf blast disease but susceptible to bacterial blight, virus disease and insect pests. The milled rice yield performance of 'Joun' is about 5.18 MT/ha by ordinary culture in local adaptability test for three years. This cultivar may be highly adaptable to the mid-northern inland plain and alpine area, north-eastern coastal area and middle plain area.

Characterization and Partial Nucleotide Sequence Analysis of Alfalfa Mosaic Alfamoviruses Isolated from Potato and Azuki Bean in Korea

  • Alfalfa mosaic alfamoviruses(AIMV) were isolated from infected potato (Solanum tuberosum) and azuki bean (Paseolus angularis) in Korea. Two AIMV isolated from potatoes were named as strain KR (AIMV-KR1 and KR2) and AIMV isolated from azuki bean was named as strain Az (AIMV-Az). Each isolated AIMV strain was characterized by using their host ranges, symptom developments, serological relations and nucleotide sequence analysis of coat protein (CP) gene. Strains KR1, KR2, and Az were readily transmitted to 20 of 22 inoculated plant species including bean, cowpea, tomato, tobacco, and potato. AIMV-KR1 and KR2 produced the typical symptoms like chlorotic or necrotic spots in Chenopodium quinoa and Solanum tuberosum cv. Superior. AIMV-Az caused bright yellow mosaic symptom and leaf malformation in Nicotiana glauca, which were different from the common mosaic symptom caused by AIMV-KR1 and KR2. Electron microscope observation of purified virus showed bacilliform virions containing a single-stranded plus-strand RNAs of 3.6, 2.6, 2.0 and 0.9 kbp in length, respectively, similar in size and appearance to those of Alfamovirus. In SDS-PAGE, the coat protein of the two viruses formed a consistent band that estimated to be about 24kDa. The CP genes of the AIMV strains, KR1, KR2, and Az have been amplified by RT-PCR using the specific primers designed to amplify CP gene from viral RNA-3, cloned and sequenced. Computer aided analysis of the amplified cDNA fragment sequence revealed the presence of a single open reading frame capable of encoding 221 amino acids. The nucleotide and peptide sequence of viral CP gene showed that strain KR1, KR2, and Az shared highest nucleotide sequence identities with AIMV strain 425-M at 97.7%, 98.2%, and 97.2%, respectively. CP gene sequences of two strains were almost identical compared with each other. Altogether, physical, serological, biological and molecular properties of the purified virus.

Effects of Selenate Ion Concentration in Nutrient Solution on the Growth and Essential Oil Content of Wormwood( Artemisia absinthium L.) (배양액 내의 Selenate 이온농도가 웜우드(Artemisia absinthium L.)의 생육 및 정유함량에 미치는 영향)

  • This study was conducted to evaluate appropriate selenate ion concentration for the production of high functional vegetables. Sodium selenate was treated 0, 2, 4, 6 and 8mg/$\ell$ using herb nutrient solution developed by European R & D Vegetable Center in Belgium. Low level of Na2se04 concentration increased the growth of wormwood, but high selenate concentration decreased the growth. Total chlorophyll content was increased by sodium selenate. The higher selenate ion concentration in the nutrient solution, the more total chlorophyll content was. The vitamin C content in wormwood was high at 2 and 4mg/$\ell$ treatment, showing good growth, at higher concentration, however, the vitamin C content decreased. At 4mg/$\ell$ selenate ion concentration, essential oil content of wormwood was best. But higher selenate ion concentration decreased essential oil content. The uptake of Se by the plant increased with the increase of selenate ion concentration.

The Ethylacetate Extract of North Kangwhal(Ostericum koreanum) Attenuates the Inflammatory Responses in PMA/A23187-stimulated Mast Cells (북강활 에틸아세테이트분획의 비만세포에서의 염증반응 억제효과)

  • Objectives: In this study, the pharmacological effects of the ethylacetate extract of Ostericum koreanum(North Kangwhal; NK) on allergic inflammation were investigated in activated human mast cells. Methods: North Kangwhal was extracted with 80% methanol for 24 h, and then fractionated with ethylacetate(NK-EtOAc extract). HMC-1 cells, an human mast line, were pre-incubated with different concentrations of NK-EtOAc extract for 30 min, and then stimulated with PMA(50 nM/ml) and A23187($1{\mu}M/ml$) at indicated times. The cell toxicity was determined by MTT assay. The concentrations of prostaglandin E2(PGE2) and cytokines(TNF-${\alpha}$, IL-8) were measured by enzyme-linked immunosorbant assay. Results: NK-EtOAc extract($10{\sim}50{\mu}g/ml$) significantly inhibited the productions of $PGE_2$, TNF-${\alpha}$ and IL-8 in PMA/A23187-stimulated HMC-1 cells without cell toxicity($0{\sim}50{\mu}g/ml$). NK-EtOAc extract also inhibited PMA/A23187-induced phosphorylation of ERK1/2 MAPK and the NF-${\kappa}B$ p65 subunit translocation into the nuclear of HMC-1 cells. Conclusions: This study suggests that NK-EtOAc extract may have an anti-inflammatory property through suppressing the production of inflammatory mediators in activated mast cells and its molecular mechanism underlies the blocking of NF-${\kappa}B$ pathway.

Effects of Sulfate Ion Concentration in Nutrient Solution on the Growth and Quality of Artemisia mongolicar var. tenuifolia (배양액 내의 황산이온 농도가 참쑥의 생육과 품질에 미치는 영향)

  • This experiment was conducted to evaluate the effects of sulfate ion concentration in nutrient solution on the growth and qualify of Mongolian wormwood (Artemisia mongolica var. tenuifolia). Sulfate ion concentration was treated 0, 0.5, 1, 2 and 3mM using the modified nutrient solution composition for herb plants developed by European Vegetable R & D Center in Belgium. The growth of Mongolian wormwood was good at 3mM treatment and dry weight was best at 3mM treatment, Chlorophyll content increased with sulfate ion concentration. Mineral content did not show any significant difference among treatments. But Ca content in tissue markedly decreased at 3mM treatment. Sulfate ion uptake increased in proportion to sulfate ion concentration in nutreint solution, the higher sulfate ion concentration, the more uptake of sulfate ion by plant. At 1mM sulfate ion treatment, essential oil content was best, but the higher sulfate ion concentration resulted in decrease of essential oil content.

Attenuation of Brain Injury by Water Extract of Goat's-beard (Aruncus dioicus) and Its Ethyl Acetate Fraction in a Rat Model of Ischemia-Reperfusion

  • Ischemic stroke constitutes about 80% of all stroke incidences. It is characterized by brain cell death in a region where cerebral arteries supplying blood are occluded. Under these ischemic conditions, apoptosis is responsible for the cell death, at least in part. Goat's-beard (Aruncus dioicus var. kamtschaticus) is a perennial plant that grows naturally in the alpine regions of Korea. In the present study, we first determined whether water extract of goat's-beard (HY1646) and some of its fractions prepared by partitioning with organic solvents could improve the viability of human hepatocellular carcinoma cells (HepG2) cultured under hypoxic condition by blocking apoptotic pathways. Based on the in vitro findings, we subsequently investigated whether HY1646 and the ethyl acetate fraction (EA) selected from cell culture-based screening could attenuate brain injury in a rat middle cerebral artery occlusion (MCAO) model of ischemia (2 hr), followed by 22 hours of reperfusion. The cell number was sustained close to that initially plated in the presence of HY1646 even after 24 hr of cell culture under hypoxic condition (3% $O_2$), at which time the cell number reached almost zero in the absence of HY1646. This improvement in cell viability was attributed to the delay in apoptosis, identified by the formation of DNA ladder in gel electrophoresis. Of fractions soluble in hexane, ethyl acetate (EA) and butanol, EA was chosen for the animal experiments because EA demonstrated the best cell viability at the lowest concentration (10 ${\mu}g$/mL). HY1646 (200 mg/kg) and EA (10 and 20 mg/kg) significantly reduced infarct size, an index of brain injury, by 16.6, 40.0 and 61.0%, respectively, as assessed by 2,3,5-triphenyl tetrazolium chloride staining. The findings suggest that prophylactic intake of goat's beard might be beneficial for preventing ischemic stroke.
