LC-MS/MS Analysis of Surface Layer Proteins as a Useful Method for the Identification of Lactobacilli from the Lactobacillus acidophilus Group

  • For precise identification of a Lactobacillus K1 isolate, LC-MS/MS analysis of the putative surface layer protein was performed. The results obtained from LTQ-FT-ICR mass spectrometry confirmed that the analyzed protein spot is the surface layer protein originating from Lb. helveticus species. Moreover, the identified protein has the highest similarity with the surface layer protein from Lb. helveticus R0052. To evaluate the proteomic study, multilocus sequence analysis of selected housekeeping gene sequences was performed. Combination of 16S rRNA sequencing with partial sequences for the genes encoding the RNA polymerase alpha subunit (rpoA), phenylalanyl-tRNA synthase alpha subunit (pheS), translational elongation factor Tu (tuf), and Hsp60 chaperonins (groEL) also allowed to classify the analyzed isolate as Lb. helveticus. Further classification at the strain level was achieved by sequencing of the slp gene. This gene showed 99.8% identity with the corresponding slp gene of Lb. helveticus R0052, which is in good agreement with data obtained by nano-HPLC coupled to an LTQ-FT-ICR mass spectrometer. Finally, LC-MS/MS analysis of surface layer proteins extracted from three other Lactobacillus strains proved that the proposed method is the appropriate molecular tool for the identification of S-layer-possessing lactobacilli at the species and even strain levels.

Changes in terpenes of three kinds of pine needles during litter decomposition

  • This study was designed to evaluate changes in the terpene composition of 3 types of pines (Pinus densiflora, Pinus thunbergii and Pinus rigida), while decomposing their leaf litter. Needle litters were placed at two different organic layer depths, one on the surface and the other beneath the litter layer. Changes in the terpene composition of this litter were detected using a gas chromatograph-mass spectrometer. Among the monoterpenes acquired from the fresh needles of P. densiflora and P. rigida, $\alpha$-pinene (12.05% and 19.87%, respectively) was the major one, followed by $\beta$-pinene (2.90% and 14.07%). However, from the needles of P. thunbergii, $\beta$-pinene (20.77%) was the major one, followed by $\alpha$-pinene (10.79%). Among the sesquiterpenes detected in P. densiflora, trans-caryophyllene (3.12%) was the highest composition compound, whereas germacrene-D (6.09%) for P. thunbergii and 1,6-cyclodecadiene (7.41%) and endo-1-bourbonanol (7.41%) for P. rigida were the highest content compounds. However, the total amounts of terpenes decreased sharply by 40-85.4% in all three types of pine needle after 90-120 days of the experiment. The concentration of each terpene differed during decomposition, and the majority of compounds disappeared from beneath the litter layer. It was determined that three types of reducing patterns of each compound appeared on the rate of loss of concentration during decomposition; one pattern decreasing sharply during the initial period, another pattern steadily or slowly decreasing, and a newly detected pattern at low concentration occurring during decomposition.

  • 할미송이버섯으로부터 혈전용해효소 (FE-2)를 DEAE-cellulose, Mono-S column으로 분리 정제하였다. 이 효소는 분자량이 18.2 kDa 이었으며, ICP-MS (inductively coupled plasma mass spectrometer) 분석 결과 $Zn^{2+}$을 포함하고 있었다. 15번째까지의 N-terminal amino acid 서열은 A-L-Y-V-G-X-S-P-X-Q-Q-S-L-L-V이고, pH 7.5에서 활성이 가장 큰 염기성 단백질 분해효소로, EDTA와 1,10-phenanthroline에 의해 활성이 감소되는 metalloprotease였다. $Mg^{2+}$, $Zn^{2+}$, Fe$^{2+}$, Co$^{2+}$의 첨가 시 활성이 증가하였으나 Hg$^{2+}$의 경우 활성이 완전히 소멸되었다. 이 효소 (FE-2)는 단백질 분해효소 저해제인 E-64 (trans-epoxysuccinyl-L-leucylamido-(4-guanidino)-butane), PMSF (phenylmethylsulfonyl fluoride), pepstatin 과 2-mercaptoethanol의 영향을 받지 않으며, 섬유소원 (fibrinogen)과 반응 시 $A\alpha$ chain과 B$\beta$ chain은 분해시키지만 $\gamma$ chain과는 반응하지 않았다.

  • The present study examined pharmacokinetic profiles of KBP31705-Cl27 and KBP30603-901, new platinum coordination complexes synthesized as anticancer candidates, in comparison with two well-known platinum-containing anticancer agents, cisplatin and carboplatin in rats. Under sodium pentobarbital anesthesia of male Sprague-Dawley rats, urinary bladder, and femoral artery and vein were catheterized for urine collection, blood sampling and drug injection, respectively Following i.v. administration of cisplatin (2 mg/kg), KBP31705-C127 (2 mg/kg), carboplatin (20 mg/kg) or KBP30603-901 (20 mg/kg), blood samples were collected at 2, 4, 6, 8, 10, 15, 20, 30, 45, 60 and 120 minutes. Urine samples were collected at 1-hr interval for 4 hr. Platinum concentrations in plasma and urine were measured using an inductively coupled plasmamass spectrometer. The plasma concentration-time curves were biphasic for all drugs during the time period studied. Compared with cisplatin, KBP31705-C127 showed similar decay patters in the alpha- and betaphases with slightly lower plasma concentrations. Urinary platinum excretion for cisplatin and KBP31705-C 127 was 56 and 52% of the administered dose in 4 hr, respectively. With regard to carboplatin and KBP 30603-901, a similar decay pattern was also observed in the alpha-phase. The half life of KBP30603-901 in the beta-phase, however, was much longer than that of carboplatin, which was consistent with the urinary excretion results that 46 and 59% of the administered dose were excreted in the urine in 4hr, respectively. The results suggest that platinum coordination complexes are primarily excreted via the renal route and KBP30603-901 can elicit longer duration of action due to slower renal excretion compared to carboplatin.

  • ${\gamma}-Al_2O_3$, ${\alpha}-Al_2O_3$, $SiO_2$, $TiO_2$등에 고정화된 폴리에틸렌글리콜을 $Fe(CO)_5$에 의한 니트로벤젠의 상온 환원반응에 상이동 촉매로 사용하였다. 고정화된 PEG의 몰수는 담체의 비표면적에 따라 증가하였고 PEG/${\gamma}-Al_2O_3$가 가장 좋은 활성을 나타내었다. PEG의 사슬길이가 길고 NaOH 농도가 높을수록 반응속도가 증가하였다. 적외선분광기를 이용하여 반응기구에 대한 고찰도 함께 실시하였으며 폴리에틸렌글리콜 상이동 촉매는 본 반응의 활성물질로 알려진 $HFe(CO)_4{^{-}}$이온의 생성과 이의 유기상으로의 이동을 촉진시키는 것으로 판단되었다.

태양열 집열기에 사용될 선택흡수막의 성능 평가 (Performance Evaluation of Selective Coatings for Solar Thermal Collectors)

  • Metal-metal oxide (M-M oxide) cermet solar selective coatings with a double cermet layer film structure were deposited on the Al-deposited glass substrate by using a directed current (DC) magnetron sputtering technology. M oxide (CrO and ZrO) was used as the ceramic component in the cermets, and Cr and Zr used as the metallic components. In addition, black Cr (Cr-$Cr_2O_3$ cermet) solar selective coatings were deposited on the Ni-plated Cu substrate by using a electroplating method for comparison. The thermal stability tests were carried out for performance evaluation of solar coatings. Reflectance measurements were used to evaluate both solar absorptance(${\alpha}$) and thermal emittance (${\epsilon}$) of the solar coatings before and after thermal testing by using a spectrometer. Optical properties of optimized cermet solar coatings were ${\alpha}{\simeq}0.94-0.96$ and ${\epsilon}{\simeq}0.1$ ($100^{\circ}C$). The results of thermal stability test of M-M oxide solar coatings showed that the Cr-CrO cermet solar selective coatings were more stable than the Zr-ZrO cermet selective coatings at temperature of both $400^{\circ}C$ in air and $450^{\circ}C$ in vacuum. The black Cr solar selective coatings were degraded in air at temperature of $400^{\circ}C$. The main optical degradation modes of these coatings were diffusion of metal atoms, and oxidation.

  • 블루 안료 15 : 6이라 불리는 ${\epsilon}$ 결정상의 동프탈로시아닌(${\epsilon}$-CuPc)는 LCD 패널에서 청색화소를 제공하는 중요한 소재로써, 본 연구에서는 ${\epsilon}$-CuPc 시료를 ${\epsilon}$-CuPc 나노 입자를 seed로 이용하는 방법으로 여러 조건에서 합성하여 적합한 용매 및 온도조건 등을 규명하였고, 이를 미리 합성한 ${\alpha}$${\beta}$ 결정상의 CuPc 시료들과 비교함으로써 ${\epsilon}$-CuPc 시료의 결정상, 결정 순도, 합성 수율 및 미세구조를 상호 비교 분석하였다. 시료들의 화학 구조 및 결정 구조는 푸레어 변환 적외선(FT-IR) 분광기 및 X선 회절 분석기(XRD)를 이용하여 각각 비교 분석하였다. 또한, 입자의 형상 및 크기는 주사 전자현미경(FE-SEM)으로 비교 분석하였고, 열적 특성은 열중량분석기(TGA)를 이용하여 비교 분석하였다.

Can Daily-use Lipstick Make Lips More Fresh and Healthy\ulcorner - A New Lipstick Containing $\alpha$-Glucosyl-hesperidin Can Remove the Dull-color from Lips

  • It has been known that the color of skin reflects the blood flow within. In lips, the capillaries close to the skin surface are numerous; hence lips are redder than the rest of face. However, dermatological research on lips is not as advanced as research on facial or body skin, and little was known about the relationship between relatively dull-colored lips and skin blood flow. The physiological differences between colorful and dull-colored lips were studied by a two-dimensional laser Doppler blood flow analyzer, a spectrometer for the measurement of the degree of oxygen saturation, and a confocal microscope for observing inside lips non-invasively. Dull-colored lips and the corner of lips (dull-colored compared to the center) showed relatively poor blood flow and lower oxygenated hemoglobin. It was found that colorful lips (generally the young) had a blood flow that tended to run straight in parallel with the skin surface. This unique blood-capillary structure can express clear red blood. Those with dull-colored lips had lost this unique structure. Their blood ran perpendicularly from the deep of the skin and down back again into the deep part as like the blood circulation patterns of facial skin. Therefore, the lips of the latter group had fewer blood capillaries near skin surface in the lips than that of the colorful-lips group. A lipstick containing a-glucosyl-hesperidin, which is derived from certain citrus fruits and can enhance blood circulation, was applied for evaluating its effects. Blood flow was increased 30 min after the application. After two weeks of daily application, the lips' condition became noticeably less dull. These findings suggest that the decrease of blood flow in dull-colored lips is caused by the loss of the unique capillary structure and the use of the lipstick to increase blood flow can give a vivid color to lips.

작잠견피브로인 분말의 제조와 그 특성 (Characterization of Tussah (Antheraea pernyi) Silk Fibroin Powder Prepared by HCI and NaOH)

  • 염산 또는 수산화나트륨 수용액의 농도, 온도, 시간 등 처리조건에 따른 작잠견사의 가수분해율을 측정하였으며 산처리 작잠분말과 알칼리처리 작잠분말의 형태 및 구조특성을 주사전자현미경, 적외선 분광분석, X-선 회절분석 등의 방법으로 분석한 결과 다음과 같은 결과를 얻었다. 1. 작잠견사에 대한 염산 및 수산화나트륨의 처리농도와 온도가 높을수록 또한 처리시간이 길어질수록 작잠견사의 가수분해율은 증가하는 경향을 나타내었으나 고온, 고농도의 조건하에서 수산화나트륨을 작잠견사에 처리하는 경우 작잠견사의 가수분해율이 오히려 떨어지는 경향을 보였다. 2. 산 및 알칼리 처리에 의하여 제조된 작잠분말의 형태는 용해도가 30 wt% 부근에서는 섬유상의 형태를 유지하였으며 산처리 분말의 경우 용해도가 증가함에 따라 섬유축 방향으로 조각난 미세한 입자형태를 나타내었으나 알칼리처리 분말의 경우에는 부정형의 입자형태를 나타내었다. 3. 적외선 분광분석 결과 작잠분말은 처리조건과 무관하게 ${\beta}$-sheet 구조의 특성을 반영하는 1635, 1532, 1226, 960, 700 cm$^{-1}$ 흡수대와 ${\alpha}$-helix 구조의 특성을 반영하는 616 cm$^{-1}$ 흡수대가 뚜렷하게 나타나 작잠분말의 주요 conformation은 ${\beta}$-sheet 구조와 ${\alpha}$-helix 구조임을 확인하였다. 4. X-선 회절분석 결과 가수분해율이 낮을 경우 (35 wt%)에는 ${\beta}$-sheet 구조와 ${\alpha}$-helix 구조에 기인하는 회절 피크가 뚜렷하게 나타났으나 가수분해율이 80 wt% 이상으로 증가하면 2${\theta}$=17.1$^{\circ}$ 및 20.0$^{\circ}$의 회절피크만 강하게 나타났다.

Hexaferrite 쉬트자석의 개발과 자기적 성질에 관한 연구 (The development and the magnetic properties of sheet hexaferrite magnets)

  • Hexaferrite $Ba_{0.25}Sr_{0.75}Fe_{12}O_{19}$$SiO_{2}$를 첨가한 쉬트자석의 자기적 성질의 변화를 연구하기 위해 X-선 회절 실험, 뫼스바우어 분광실험, 자기모멘트 측정실험 등을 하였다. 쉬트자석은 Dr. Blade 방법에 의해 제조되었고, 결정구조는 마그네토플럼바이트 M-type 육방결정구조이다. 첨가제 농도에 따른 격자상수의 변화는 거의 없으나, $SiO_{2}$가 2.0 wt.% 이상부터 ${\alpha}-Fe_{2}O_{3}$ 의 상이 나타나기 시작했다. 이성질체 이동값은 Fe이온들이 3가 임을 나타낸다. $SiO_{2}$의 증가에 따라 Curie 온도는 감소함을 보였다. 이것은 $Si^{4+}$가 12k-site $Fe^{3+}$의 자리를 차지함으로써 Fe-O-Fe의 초교환상호작용에 의한 원자간 거리와 양이온 사이의 각이 변함에 따른 것이다. ${\alpha}-Fe_{2}O_{3}$의 상은 $Si^{4+}$가 12k-site $Fe^{3+}$의 자리를 차지함으로 치환된 $Fe^{3+}$에 의해 나타난 것이다. 자기모멘트측정으로 부터 $SiO_{2}$를 첨가한 $Ba_{0.25}Sr_{0.75}Fe_{12}O_{19}$ 쉬트자석은 보자력 $H_{c}$, 포화자화 $M_{s}$, 잔류자화 $M_{r}$이 양이온 치환보다는 미세구조변화에 더 의존함을 알았다.
