Phylogenetic Relationships among Allium subg. Rhizirideum Species Based on the Molecular Variation of 5S rRNA Genes

  • This study has demonstrated the molecular variation of 5S rRNA genes in 15 Allium subgenus Rhizirideum and 1 Allium subg. Allium. For cloning of the 5S rRNA genes, PCR products were obtained from amplification with oligonucleotide primers which were derived from the conserved coding region of 5S rRNA genes. These amplified PCR products were cloned and identified by FISH and sequence analysis. The 5S rRNA loci were primarily located on chromosomes 5 and/or 7 in diploid species and various chromosomes in alloploid species. The size of the coding region of 5S rRNA genes was 120 bp in all the species and the sequences were highly conserved within Allium species. The sizes of nontranscribed spacer (NTS) region were varied from 194 bp (A. dektiude-fustykisum, 2n=16) to 483 bp (A. sativum). Two kinds of NTS regions were observed in A. victorialis var. platyphyllum a diploid, A. wakegi an amphihaploid, A. sacculiferum, A. grayi, A. deltoide-fistulosum and A. wenescens all allotetraploids, while most diploid species showed only one NTS region. The species containing two components of NTS region were grouped with different diploid species in a phylogenetic tree analysis using the sequences of 5S rRNA genes and adjacent non-coding regions.

Chromosome Doubling of Allium wakegi Araki by In Vitro Cultures (기내배양에 의한 쪽파의 체세포 염색체 배가)

  • Induction of embryogenic callus from Allium wakegi Araki explants was promoted on medium containing 2,4-D, and production of abnormal embryos from the embryogenic callus increased as 2,4-D concentration was raised. Shoot tip was found to be the best explant source for embryogenic callus formation followed in the order by bulb scale and leaf section. Medium containing 0.09 M sucrose was effective for embryogenic callus production. The regenerants from embryogenic callus on medium containing 2,4-D and BA at different concentrations was consisted of diploids, tetraploids and a few mixaploids of 2n+4n, and their chromosomal aberration rate ranged from 8.0 to 33.3%. Frequency of chromosomal aberrants was the highest (18.7%) in the regenerants obtained from bulb scale-derived embryogenic callus among others. Plant regeneration rate was high (33.5%) from the shoot tip-derived embryogenic callus and the frequency of chromosomal aberrants was very low (7.0%). The plantlets regenerated on medium containing 0.26 M sucrose resulted in high chromosomal aberrants. The regenerants on medium containing sucrose at 0.09∼0.20 M produced chromosomal aberrants at around 15.2∼16.6%.

Antibacterial Activities against Pathogenic Bacteria of Lactic Acid Bacteria Isolated from Allium wakegi (쪽파로부터 분리된 유산균의 병원성균에 대한 항균활성 )

  • In this study, we isolated lactic acid bacteria from Allium wakegi and examined the usability of culture supernatants obtained from these lactic acid bacteria. The antibacterial activity of the culture supernatant obtained from the isolated lactic acid bacteria against the pathogens Escherichia and Salmonella spp. was measured. The obtained lactic acid bacteria culture medium showed significant antibacterial activity against pathogenic bacteria in a dose-dependent manner. The effects of pH and heat denaturation on the observed anti-pathogenic bacterial activity was also investigated. Adjusting the culture supernatant to pH 7 resulted in loss of all antibacterial activity against pathogenic bacteria, suggesting that the antibacterial activity of the obtained culture supernatant against pathogenic bacteria is influenced by organic acids. Assessment of the heat stability of the anti-pathogenic bacterial activity revealed that heat treatment did not diminish activity. The obtained lactic acid bacteria culture medium is thus stable against heat.

  • Growth status, plant nutrition and soil chemical properties were investigated for lettuce, spring onion and red pepper grown under vinyl house near Yesan. Low Ca and Mg with high K in soil resulted in Ca deficiency with slight Mg insufficiency in Korean lettuce. In this soil pH was low and EC was extremely low. Spring onion(Allium wakegi) can not emerge or showed poor growth(50%) due to high EC(above 0.5m mho/cm) and low pH (below 6.2). Red pepper plant showed wilt disease probably due to low soil pH.

Application of Practical Immobilizing Agents for Declining Heavy Metal (loid)s Accumulation by Agricultural Crop (Allium wakegi Araki)

  • In order to reduce the accumulation of toxic metals (As, Cd and Pb) in the chives, various immobilizing agents such as a soil pH change-inducing immobilizing agent (lime), sorption agent (compost, spent mushroom compost), soil pH change and sorption agent (biochar) and, dissolved organic carbon (DOC) coagulator (gypsum) and uncontaminated soil were applied to the contaminated soils in isolation and in combination. Then chives were grown and determined for As, Cd and Pb concentrations accumulated in the edible part at harvest. The Cd and Pb concentrations of the chive plant grown in the contaminated soil (no treatment) exceeded the legislated Korean guideline values (Cd: $0.05mg\;kg^{-1}$, Pb $0.1mg\;kg^{-1}$) and As concentration ($21mg\;kg^{-1}$) was 1,000 times higher than chives plant grown in uncontaminated environment in Korea. Application of lime and gypsum significantly reduced As, Cd and Pb concentrations in all chives examined, due to the increased soil pH and decreased soil DOC. Also, application of combination treatments involving DOC coagulator such as gypsum together with lime decreased As, Cd and Pb concentrations from 21, 1.3 and $9.7mg\;kg^{-1}$ to 2.1, 0.1 and $1.1mg\;kg^{-1}$, respectively. Consequently, it was concluded that pH change-inducing immobilizing agent (lime) which was already well known and DOC coagulator such as gypsum could be used as a promising immobilizing agent for safer chives plant production.
