농산 폐기물인 Allium속 뿌리를 이용한 Ni와 Pb 이온 제거 (Removal of Ni and Pb Ion from Aqueous Solution by the Agricultural Wastes, Allium Roots)

  • A batch experiment was conducted to evaluate the removal capacity of welsh onion(Allium fistulosum L.), shallot (Allium ascalonicum L.), garlic (Allium sativum L.) roots as an adsorbent for Ni and Pb in aqueous solution. One gram of the dried Allium root powder was reacted in 100ml of solution containing 10mg of each heavy metal and effects of metal concentration, pH, temperature, and size of adsorbent on the removal efficiency were evaluated. The results were as follows ; The amount of adsorption of heavy metal ions were higher with the smaller particles size of adsorbent. Garlic root was high adsorption capacity of Pb, especially. The higher concentration of heavy metal solution was, the more amount of adsorption of heavy metals was. The adsorption ratio was differed from a kind of heavy metal. As the temperature increased, the amount of adsorption of Ni and Pb by shallot and welsh onion were decreased. The amount of adsorption of Ni was high under alkali conditions but the amount of adsorption of Pb was high under neutral and acidity condition.

Allium속 향신채 첨가가 찐어묵의 저장성과 품질에 미치는 영향 (The Effect of Herbs of Alium Species on Quality and Storage Characteristics of Kamaboko)

  • The effects of the addition of herbs belonging to Allium species on the quality and storage characteristics of kamaboko were studied. The herbs employed in the study were garlic (Allium sativum for.Pekinenese Makino), leek(.Allium tuberosum Roth), onion(Allium cepa Linnaeus), and onion skin. water activity(Aw) , pH, TBA, VBN, microbial load, textural characteristics and sensory evaluation were tested. 1. Aw of all the samples decreased on storage, the Aw on lower day ranged from 0.937~o.950. All thesamples containing herbs retained the pH 6 ~7 during the entire storage period even though it decreased gradually on storage. 2. In general, TBA and VBN of the samples containing herbs were lower than the control. The sample containing 3% onion skin showed the lowest TBA and VBN value. 3. The total plate count of the samples containing herbs was low compared to the control even though the total count increased during the storage. The samples containing 3% garlic and 3% onion skin showed the lowest total plate count on 20 days of storage. 4. The sample with garlic showed remarkably low value in sensory evaluation. The samples containing onion and onion skin, however, reached to the high sensory points as storage period increased. They received high points in taste as well as overall acceptance.

  • 본 연구에서는 몇 가지 암세포를 대상으로 마늘 열수추출물(WEAS)의 항암활성을 조사하였으며, 비교적 높은 감수성을 보인 백혈병세포를 대상으로 세포증식억제가 apoptosis 유도와 연관성이 있음을 제시하였다. WEAS에 의한 U937 세포의 apoptosis 유도는 세포주기 G2/M arrest와 연관성이 있었으며, 다양한 apoptosis조절 유전자들의 발현 변화를 동반하였음을 확인하였다. 이러한 apoptosis조절 인자들의 발현변화는 미토콘드리아 막 전위의 소실과 연관성이 있었으며, WEAS에 의한 암세포의 G2/M arrest에 의한 apoptosis 유발에 ROS가 중요한 조절자로 작용함을 알 수 있었다.

  • The health benefits of garlic (Allium sativum L.) are derived from a wide variety of components and from the different ways it is administered. The known health benefits of garlic include cardiovascular protective effects, stimulation of immune function, reduction of blood glucose level, protection against microbial, viral and fungal infections, as well as anticancer effects. In the present study, it was examined the effects of water extract of A. sativum (WEAS) on the growth of cultured human tumor cells in order to investigate its anti-proliferative mechanism. Treatment of WEAS to tumor cells resulted in the growth inhibition, especially in leukemia cells, which was associated with induction of G2/M arrest of the cell cycle and apoptosis. In order to further explore the critical events leading to apoptosis in WEAS-treated U937 human leukemia cells, the following effects of WEAS on components of the mitochondrial apoptotic pathway were examined: generation of reactive oxygen species (ROS), alteration of the mitochondrial membrane potential (MMP), and the expression changes of Bcl-2 and IAP family proteins. The cytotoxic effect of WEAS was mediated by its induction of apoptosis as characterized by the occurrence of DNA ladders, apoptotic bodies and chromosome condensation in U937 cells. The WEAS-induced apoptosis in U937 cells was correlated with the generation of intracellular ROS, collapse of MMP, activation of caspase-3 and down-regulation of anti-apoptotic proteins. The quenching of ROS generation with antioxidant N-acetyl-L-cysteine conferred significant protection against WEAS-elicited ROS generation, caspase-3 activation, G2/M arrest and apoptosis. In conclusion, the present study reveals that the cellular ROS generation plays a pivotal role in the initiation of WEAS-triggered apoptotic death in U937 cells.

흑마늘의 항산화, 항균 및 항혈전 활성 (Antioxidation, Antimicrobial and Antithrombosis Activities of Aged Black Garlic (Allium sativum L.))

  • 흑마늘을 이용한 식의약품 기능성 소재 개발 연구의 일환으로, 생마늘과 흑마늘 추출물을 조제하여 항산화, 항균, 항혈전 활성을 비교하였으며, 이후 흑마늘 추출물의 순차적 용매 분획물을 조제하여 활성분획을 확인하고자 하였다. 흑마늘의 열수 추출효율은 40.6%로 생마늘의 4배의 효율을 나타내었으며, 고온 숙성과정을 통해 생마늘보다 총 폴리페놀은 4배, 총 플라보노이드 함량은 1.56배, 총 당 및 환원당은 각각 3.36배 및 6.75배 증가되었다. 항산화 활성 평가 결과, 흑마늘 추출물은 DPPH 음이온에 대한 소거능은 생마늘 추출물보다 미약하였으나, ABTS 양이온 소거능 및 nitrite 소거능은 생마늘 추출물보다 우수하였다. 특히 흑마늘 추출물의 ethylacetate 분획은 매우 강력한 항산화 활성을 나타내었다. 흑마늘 추출물은 생마늘 추출물에 비해 항균 및 항혈전 활성을 상실한 반면, 흑마늘 추출물의 ethylacetate 분획은 Staphylococcus aureus 및 Bacillus subtilis에 대한 항세균 활성과 프로트롬빈, 혈액응고인자 저해, 인간 혈소판 응집저해를 나타내어 강력한 항혈전 활성을 확인하였다. 흑마늘 열수 추출물 및 분획물은 적혈구 용혈활성은 나타나지 않았다. 본 연구결과는 흑마늘 추출물의 ethylacetate 분획이 향후 항혈전 활성을 위한 기능성 식의약품 소재로 개발 가능함을 제시하고 있다.

High frequency Plant Regeneration of Garlic (Allium sativum L.) Calli Immobilized in Calcium Alginate Gel

  • Calli obtained from a shoot-tip of garlic, Allium sntivum L., were encapsulated using a calcium alginate gel. Some of the encapsulated calli were cultured on a 1/2 MS medium supplemented with 3% sucrose, 10$\^$-5/ kinetin, and 5 ${\times}$ 10$\^$-6/ M NAA whereas the remainder was stored for 40 days at 4$^{\circ}C$. All the naked calli regenerated on the solid medium, while 95% of the encapsulated calli regenerated, and 88% of the encapsulated calli regenerated after 40 days of storage at 4$^{\circ}C$. The capsule matrix delayed the germination time of the encapsulated calli, yet activated the shoot formation of the artificial garlic seeds. The shoot length of the encapsulated garlic calli was much longer than that of the naked garlic calli. The encapsulated garlic calli were dried in a laminar airflow cabinet and the conversion frequency of the dried artificial garlic seeds on a 1/2 MS medium remained at 93% with a water Loss of Less than 50%.

Allicin-induced apoptosis of gastric epithelial cells is associated with changes of caspase-independent effector and involvement of PKA

  • Garlic (Allium sativum) has been used as a general food and a remedy in Oriental for a long time. Since garlic compounds have been also shown to inhibit growth of tumors and to modulate the activity of carcinogenesis, the effects of allicin on growth and survival in human gastric epithelial cells were evaluated by cell viability, cell cycle analysis and DNA fragmentation. Protein levels of cytochrome C, Bcl-xL, Bax and AIF were detected by Western blotting. Effects of recombinant VacA on caspase proteases activity were also determined. (omitted)
