Evaluation of Hepatoprotectivte Activity of Citrullus Colocynthis Roots Against $CCl_4$ induced Toxicity in Albino Rats

  • Hepatoprotective activity of different extracts of Citrullus colocynthis L. sch. (roots) (cucurbitaceae) was investigated in albino rats by inducing hepatotoxicity with carbon tetrachloride. The alcoholic extract of Citrullus colocynthis sch. 100 mg/kg b.w. has been shown to posses significant hepatoprotective effect by lowering the serum level of transaminases (GPT & GOT), alkaline phosphate (ALP) and bilirubin (P < 0.05 to P < 0.001).

Heterotrophic Bacterial Community and Alkaline Phosphatase Releasing Bacteria in Lake Soyang (소양호에서의 종속영양세균의 종구성 및 Alkaline phosphatase 분비 세균에 관한 연구)

  • The total and heterotrophic bacterial distributions, compositions and alkaline phosphatese actibities were analyzed in Lake Soyang from Sep. 1987 to Aug. 1988. The heterotrophic bacteria was small portion, 0.07-2.63% of total bacterial number which ranged from $3.2{\times}10^{5}$ to $3.2{\times}10^{6}$ cells/${\mu}\ell$. The composition of bacterial community was less diverse in summer and at the fish farm site and Peridinium blooming site. Pseudomonas and Flavo bacterium were the dominant genera in all sites. The highest proportion and activity of alkaline phsophatase was appeared in Flavobacterium, while Pseudomonas was the most predominant group.

A Study of Alkaline Phosphatase Activity on the Preimplantation Mouse Embryos (초기 흰쥐 배아의 발생단계에 있어서의 Alkaline Phosphatase의 활성에 관한 연구)

  • In order to investigate the alkaline phosphtase activities in the mouse oocytes in matuation and preimplantation embryos in developing in culture, the enzyme activities were measured by means of biochemical method. The in vitro effect of levamisole which is known as an inhibitor of the lakaline phosphatase was also observed on the oocyte in maturation and the embryos in early embryogenesis. The results obtained were as follows: The enzyme activity was not detected in the embryos unitl the stage of 4-cell, but it appeared first in the 4-cell embryos and the level of the activity was steady through up to the blastocyst. Levamisole inhibited the alkaline phosphatase activity in the blastocyst, and the activity decreased by almost 70% at 10 mM and 50% at 1 mM as compared with the control. In addition, levamisole inhibited completely the formation of polar body by the oocytes. and induced degeneration of the preimplantation embryos at the dose of 0.5 mM or higher.

Medicinal aspects of Murraya koenigii mediated silver nanoparticles

  • The present work aimed to explore green approach via aqueous leaves extract of Murraya koenigii (ALEMk) for the synthesis of silver nanoparticles (AgNPsMk) in single step. The synthesis process was visualized with a color change and monitored by employing UV/Visible spectroscopy and a clear peak attained at 420 nm confirming the synthesis of AgNPsMk. The possible functional groups present in the extract which participated in the synthesis of AgNPsMk were identified with the help of FTIR spectroscopy. Further characterization using TEM images revealed the spherical shape of AgNPsMk with average particle size of 20 nm displaying well dispersion throughout the solution. Pronounced antioxidant activities of AgNPsMk at increased concentrations observed which evidencing strong radical scavenging ability. Moreover, AgNPsMk exhibited strong antibacterial behavior when tested against bacterial strains of Escherichia coli and Bacillus subtilis. Moving ahead, in vitro cytotoxicity work revealed potent cell viability loss appearing in AU565 and HeLa cancer cell lines on exposure to AgNPsMk at increased concentration. Finally, in vivo assessment carried out inside male Wistar rats indicated non toxic effect on examined liver tissues besides biochemical analysis including bilirubin, alkaline phosphtase (ALP) and serum glutamate pyruvate transaminase (SGPT) which found within the normal range when compared with control. The prior research work profoundly apprises the potential of green synthesized AgNPsMk to play a significant role in biomedical applications and formulations.

Effect of SAL5 on chronic ethanol-induced fatty liver model (흰쥐에서 SAL5의 알코올성 지방간 형성에 미치는 영향)

  • Objective : In this study, we investigated the effect of SAL5(mixing extracts of Schisandra chinensis Baillon, Artemisia capillaris Thunb., and Aloe vera Linne) on chronic ethanol-induced fatty liver model. Methods : Sprague-Dawley male rats were fed Liber-DeCarli (normal), ethanol liquid diet (control), SAL5 (200 mg/kg). We administrated the SAL5 on chronic ethanol-induced fatty liver model for 5 weeks. We measured alkaline phosphtase (ALP), alanine transminase (ALT), aspartate transminase (AST) and ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG), superoxide dismutase (SOD), catalase, glutathione (GSH) and malondialdehyde (MDA) level in liver. Liver histopathology was examined by Hematoxylin-eosin and Oil red O staining of the fixed liver tissues. Real-time PCR was performed to measure the mRNA expression of inflammatory cytokines and MMP-2, MMP-9. Results : SAL5 administration resulted in significantly decreased liver marker enzymes activities of alanine transminase (ALT), ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG) activities in liver. The control group decreased the activities of superoxide dismutase (SOD), catalase (CAT) with the reduced level of glutathione (GSH) in liver. On the other hand, SAL5 group increased the activities of SOD, CAT and the level of GSH. SAL5 delayed the development of an alcoholic fatty liver by reversing fat accumulation in the liver, as evidenced in histological observations. The gene expression of mRNA were significantly decreased at the $IL-1{\beta}$, $TNF-{\alpha}$, NOS-II and MMP-2 by SAL5. Conclusions : These results indicate that SAL5 might have protective effect chronic ethanol-induced fatty liver models.
