Isolation, Purification and enzymatic characterization of the Cellulase produced by Aspergillus Phoenicis (Aspergillus phoenices K.U. 175이 생성하는 셀루라아제의 분리, 정제 및 효소학적 성질)

  • Avicelase, CMCase and salicinase of A.phoenicis K.U. 175 were purified from wheat bran culture by salting out with ammonium sulfate, dialysis and successive column chromatography Sephadex G-100. Optimum pH and temperature of avicelase were pH 3.8-4.8, $35-55^{\circ}C$ and that of CMCase, salicinase were pH4.5-5.5, $45-60^{\circ}C$ and pH 4.5-6.0, $45-60^{\circ}C$ respectively. These enzymes were relatively thermostable, alkali unstable and inhibited by $Ca^{++},\;Mn^{++},\;Cu^{++},\;and\;Hg^{++}$. Km values of avicelase, CMCase and salicinase were calculated to be $1.5{\times}10^{-4}M,\;5.5{\times}10^{-4}M\;and\;2.75{\times}10^{-5}M$ and Vmax values $1.66{\times}10^{-4}mM/min,\;3.33{\times}10^{-4}mM/min\;and\;1.14{\times}10^{-4}mM/min$, respectively.

  • This study aims to manufacture non-sintering cement(NSC) by adding phosphogypsum(PG) and waste lime(WL) to granulated blast furnace slag(GBFS) as sulfate and alkali activators. This study also investigates the pore structure of NSC Matrix. The result of experiment of pore structure properties, showed no considerable difference for total pore volume by cement mixing ratio but shows a large distinction in distribution of pore diameter. On the whole, pore-diameter of paste of NSC show that occupation ratio of pore diameter below 10mm is larger and is smaller than OPC and BFSC at pore diameter of over 10nm. Such a reason is that the hydrate like CSH gel and ettringite formed dense pore structure of NSC matrix.

  • This research investigates the characteristics of chloride fixation in non-sintering cement(NSC) matrix. NSC was manufactured by adding phosphogypsum and slack lime to granulated blast furnace slag as sulfate and alkali activators. As a result, the concentration of chloride ion in pore solution of NSC-solidified matrix is more low than that of OPC-solidified matrix containing the same chloride content in cement paste. Also, the concentration of chloride ion in pore solution of NSC-solidified matrix is similar with that of BSC-solidified matrix containing the same chloride content in cement paste.

Degradation mechanisms of concrete subjected to combined environmental and mechanical actions: a review and perspective

  • In-service reinforced concrete structures are simultaneously subjected to a combination of multi-deterioration environmental actions and mechanical loads. The combination of two or more deteriorative actions in environments can potentially accelerate the degradation and aging of concrete materials and structures. This paper reviews the coupling and synergistic mechanisms among various deteriorative driving forces (e.g. chloride salts- and carbonation-induced reinforcement corrosion, cyclic freeze-thaw action, alkali-silica reaction, and sulfate attack). In addition, the effects of mechanical loads on detrimental environmental factors are discussed, focusing on the transport properties and damage evolution in concrete. Recommendations for advancing current testing methods and predictive modeling on assessing the long-term durability of concrete with consideration of the coupling effects are provided.

  • Larch ($\underline{Larix}$ $\underline{leptolepis}$ GORDON), one of the major afforestation species in Korea in view of its growing stock and rate of growth, is not favored as a raw material for pulp due to its low yield of pulp and difficulties with bleaching arising from the high content of extractives in wood, and the high heartwood ratio and the active phenolics, respectively. The purpose of this study is to investigate the characteristics of firstly pulping with various additives of cellulose protector for the yield of pulp, and secondly bleaching with oxygen for chlotination-alkali extraction of five stage-sequence to reduce chlorine compounds in bleaching effluents. The kraft cooking liquor for five age groups of larchwood was 18 percent active alkali with 25 percent sulfidity and 5 : 1 liquor-to-wood ratio, and each soda liquor for sap-and heart-wood of the 15-year-old larchwood was 18 percent alkali having one of the following cellulose protectors as the additive; magnesium sulfate ($MgSO_4$, 2.5%), zinc sulfate ($ZnSO_4$, 2.5%), aluminium sulfate ($Al_2(SO_4)_3$, 2.5%), potasium iodide (KI, 2.5%), hydroquinone (HQ, 2.5%), anthraquinone (AQ, 0.1%) and ethylene diamine (EDA, 2.5%). Then each anthraquinone-soda liquor for the determination of suitable cooking condition was the active alkali level of 15, 17 and 19 percent with 1.0, 0.5 and 0.1 percent anthraquinone, respectively. The cooking procedure for the pulps was scheduled to heat to 170$^{\circ}C$ in 90 minutes and to cook 90 minutes at the maximum temperature. The anthraquinone-soda pulps from both heartwood and sapwood of 15-year-old larchwood prepared with 0.5 percent anthraquinone and 18 percent active alkali were bleached in a four-stage sequency of OCED. (O: oxygen bleaching, D: chlorine dioxide bleaching and E: alkali extraction). In the first stage oxygen in atmospheric pressure was applied to a 30 percent consistency of pulp with 0.1 percent magnesium oxide (MgO) and 3, 6, and 9 percent sodium hydroxide on oven dry base, and the bleached results were compared pulps bleached under the conventional CEDED (C: chlorination). The results in the study were summarized as follows: 1. The screened yield of larch kraft pulp did not differ from particular ages to age group, but heartwood ratio, basic density, fiber length and water-extractives contents of wood and the tear factor of the pulp increased with increasing the tree age. The total yield of the pulp decreased. 2. The yield of soda pulp with various chemicals for cellulose protection of the 15-year-old larchwood increased slightly more than that of pure soda pulp and was slightly lower than that of kraft pulp. The influence of cellulose protectors was similar to the yield of pulps from both sapwood and heartwood. The effective protectors among seven additives were KI, $MgSO_4$ and AQ, for which the yields of screened pulp was as high as that of kraft pulp. Considering the additive level of protector, the AQ was the most effective in improving the yield and the quality of pulp. 3. When the amount of AQ increased in soda cooking, the yield and the quality of the pulp increased but rejects in total yield increased with decreasing the amount of active alkali from 19 to 15 percent. The best proportion of the AQ seemed to be 0.5 percent at 17 percent active alkali in anthraquinone-soda pulping. 4. On the bleaching of the AQ-soda pulp at 30 percent consistency with oxygen of atomospheric pressure in the first stage of the ODED sequence, the more caustic soda added, the brighter bleached pulp was obtained, but more lignin-selective bleaching reagent in proportion to the oxygen was necessary to maintain the increased yield with the addition of anthraquinone. 5. In conclusion, the suitable pulping condition for larchwood to improve the yield and quality of the chemical pulp to the level for kraft pulp from conventional process seemed to be. A) the selection of young larchwood to prevent decreasing in yield and quality due to the accumulation extractives in old wood, B) the application of 0.5 percent anthraquinone to the conventional soda cooking of 18 percent active alkali, and followed, C) the bleaching of oxygen in atmospheric pressure on high consistency (30%) with 0.1 percent magnesium oxide in the first stage of the ODED sequence to reduce the content of chlorine compounds in effluent.

Alkalinity Supplement using Sea Shell for Sulfur-utilizing Autotrophic Denitrification (황-이용 독립영양 탈질에서의 패각을 이용한 알칼리도 공급)

  • The purpose of this study was to select an effective and economical alkali source for sulfur-utilizing autotrophic denitrification. Tests on acid neutralization and denitrification at various alkali/sulfur mixing ratios were performed for charcoal, briquette ashes, sea shell, and limestone. The results of the experiments showed that sea shell was the most effective alkali source because it could provide more surface area than limestone, and the optimal alkali/sulfur mixing ratio was 1/1(V/V). In a sulfur/sea shell packed bed reactor, the denitrification efficiency was above 90% up to a loading rate of 116 g $NO_3{^-}-N/m^3-day$. but the denitrification efficiency deteriorated to 48% at the loading rate of 145 g $NO_3{^-}-N/m^3-day$. The average $SO_4{^{2-}}$ generation per g of $NO_3{^-}-N$ removed was 7.02 g, which is lower than the theoretical value of 7.54 g. Denitrification and sulfate generation appeared to be a first-order and a zero-order reaction with a reaction rate constant of 0.146 /hr and -53.1 mg/L-hr, respectively. According to nitrogen mass balance, 71~109%, with an average of 90%, of the removed nitrogen was recovered as $N_2$ gas.

Properties of Porphyran and Hemicellulose extracted with Different extract Solutions and Enzymatic Pretreatments from Porphyra (추출 용매 및 효소 전처리 방법에 따른 포피란과 헤미셀루로오즈의 특성)

  • Laver, Porphyra, is distinctive for its high content of proteins and polysaccharides such as porphyran and hemicellulose. The chemical properties of the polysaccharides extracted with different extraction methods such as hot water, dilute acid(pH 4.0) or alkali solution(2N NaOH) were examined to investigate the suitable extraction conditions for porphyran and hemicellulose from laver. For porphyran extraction, dilute acid solution was more preferable to hot water and alkali solution because of its higher 3,6-anhydrogalactose content and lower protein content. However, alkali solution was more suitable to extract the hemicellulose because of higher mannose content indicating the extraction of mannan. To decrease contamination of the polysaccharides with protein, the dried lavers were pretreated with enzymes (Protamex, Flavourzyme, Alcalase, Viscozyme) before hot water extraction. All enzyme pretreatments increased the yield of polysaccharides by compared with control (enzyme unpretreated) and Flavourzyme pretreatment was most effective to decrease protein contamination in the polysaccharide. All viscosities of porphyran solutions pretreated by enzymes were lower compared to the control porphyran solution and showed pseudoplastic behavior with yield stress. In case of alkali extraction of residues obtained after enzyme hydrolysis and hot water extraction, protease pretreatment increased the mannose contents in the polysaccharide while the xylose content was increased by Viscozyme pretreatment.

Geochemical Evolution of CO2-rich Groundwater in the Jungwon Area (중원 지역 탄산수의 지구화학적 진화)

  • Two different types of deep groundwaters occur together in the Jungwon area: $CO_2$-rich water and alkali water. Each water shows distrinct hydrogeochemical and environmental isotopic characteristics. The $CO_2$-rich waters are characterized by lower pH(6.0~6.4), higher Eh (25~85mV) and higher TDS content (up to 3,300 mg/l), whereas the alkali type waters have higher pH (9.1~9.5), lower Eh (-136~-128mV) and lower TDS content (168~254 mg/l). The CO2-rich waters ($Pco_2$=up to 1atm) were probably evolved by the local supply of deep $CO_2$ during the deep circulation, resulting in enhanced dissolution of surrounding rocks to yield high concentrations of $Ca^{2+}, Na^+, Mg^{2+}, K^+\; and \;HCO_3\;^-$ under low pH conditions. On the other hand, the alkali type waters ($Pco_2$=about 10-4.6 atm) were evolved through lesser degrees of simple wate/rock (granite) interaction under the limited suppy of $CO_2$. The alkali waters are relatively enriched in F- (up to 14mg/l), whereas the F- concentration of$CO_2$-rich water is lower (2.2~4.8 mg/l) due to the buffering by precipitation of fluorite. The oxygen-hydrogen isotopes and tritium data indicate that compared to shaltion ($\delta$18O=-9.5~-7.8$\textperthousand$),two different types fo deep groudwaters (<1.0TU)were both derived from pre-thermonuclear (more than 40 years old) meteoric waters with lighter O-H isotopic composition ($\delta$18O=-9.5~-7.8$\textperthousand$) and have evolved through prolonged water/rock interaction. The $CO_2$-rich waters also show some degrees of isotopic re-equilibration with $CO_2$ gas. The $\delta^{34}S$ values of dissolved sulfates (+24.2~+27.6$\textperthousand$) in the $CO_2$-rich waters suggest the reduction of sulfate by organic activity at depths. The carbon isotope data show that dissolved carbon in the $CO_2$-rich waters were possibly derived either from dissolution of calcite or from deep $CO_2$ gas. However, strontium isotope data indicate Ca in the $CO_2$-rich waters were derived mainly from plagioclase in granite, not from hydrothermal calcites.

Solubility Improvement of Foodwaste by SeCAH and Potential Inhibitors for Anaerobic Co-Digestion (SeCAH를 이용한 음식물쓰레기 용해도 개선 및 혐기성 병합 소화시 저해 가능 인자)

  • Sequential quick crushing and alkali hydrolysis(SeCAH), as a pretreatment for foodwaste, incorporating mechanical crushing and alkali hydrolysis was proposed and investigated in this study. Focuses were placed on the improvement of biodegradability of foodwaste and the estimation of potential inhibitors for anaerobic digestion. The solubility of the pretreated foodwaste by SeCAH was increased 2.5 times rather than mechanical crushing. Considering solubility improvement and energy consumption, 2,000 rpm of crushing strength and 5$\sim$10 sec of crushing time are recommended. After SeCAH, the fraction of large organic particles(>2.36 mm) of foodwaste was sharply decreased from 88.0% to 29.0%, otherwise the fraction of small particles(<75 $\mu$m) was greatly increased from 10.5% to 40.7%. Ammouinum, potassium and sulfate were estimated as potential inhibitors for anaerobic digestion and their concentrations in pig slurry were 3331.3 mg/L, 4256.5 mg/L and 1017.5 mg/L, respectively.

  • Greenhouse gas reduction will be highlighted as the most pending question in the cement industry in future because the production of Portland cement not only consumes limestone, clay, coal, and electricity, but also release waste gases such as $CO_2,\;SO_3$, and NOX, which can contribute to the greenhouse effect and acid rain. To meet the increase of cement demand and simultaneously comply with the Kyoto Protocol, cement that gives less $CO_2$ discharge should be urgently developed. This study aims to manufacture non-sintering cement(NSC) by adding phosphogypsum(PG) and waste lime(WL) to granulated blast furnace slag(GBFS) as sulfate and alkali activators. This study also Investigates the hydration reaction of NSC through analysis of scanning electron microscopy(SEM), X-ray diffraction(XRD), differential thermal analysis(DTA), and pH. Results obtained from analysis of the hydrate have shown that the glassy films of GBFS are destroyed by the activation of alkali and sulfate, ions eluted from the inside of GBFS react with PG and produce ettringite, and consequently the remaining component in GBFS slowly produced C-5-H(I) gel. Here, PG is considered not only to play the role of simple activator, but also to work as a binder reacting with GBFS.
