Characterization of Differentiation of the Supernumerary Dental Pulp Stem Cells toward the Odontoblast by Application Period of Additives (과잉치 치수유래 줄기세포의 분화제 처리 기간에 따른 상아모세포 발현 특성)

  • The aim of this study was to investigate the possibility of the supernumerary teeth for the stem cell source in dentistry. The Real Time Quantitative Reverse Transcription Polymerase Chain Reaction (Real Time qRT-PCR) method was used to evaluate the differentiation toward the odontoblast of the supernumerary dental pulp stem cells (sDPSCs). Supernumerary dental pulp stem cells were obtained from 3 children (2 males and 1 female, age 7 to 9) diagnosed that the eruption of permanent teeth was disturbed by supernumerary teeth. The common genes for odontoblasts are alkaline phosphatase (ALP), osteocalcin (OC), osteonectin (ON), dentin matrix acidic phosphoprotein 1 (DMP-1), dentin sialophosphoprotein (DSPP). The sDPSCs were treated for 0 days, 8 days and 14 days with additives and then Real Time qRT-PCR was performed in intervals of 0 days, 8 days and 14 days. The alizarin-red solution staining was performed to visualize the stained color for the degree of calcification at 7 days, 14 days, 21 days and 28 days after treating additives to the sDPSCs. From the result of the Real Time qRT-PCR, the manifestation exhibit maximum value at 8 days after additive treatment and shifted to a decrease trend at 14 days. Alizarin-red solution staining exhibit light results at 7 days after staining and generalized dark result at 14 days. Consequently, in studies with sDPSCs, appropriate treatment time of additives for Real Time qRT-PCR is 8 days. Also, a suitable period of Alizarin-red solution staining is 14 days.

Analysis of Degradation Products in Madder Dyed Fabrics in Selective Degradation Conditions (퇴화조건에 따른 꼭두서니 염색물의 퇴화물 연구)

  • The purpose of this investigation was to investigate the degradation products of the dye component extracted from madder dyed fabrics using the GC-MS analysis and to evaluate the change of color due to degradation treatment. Four different degradation protocols were used in this study,; refrigeration at $7^{\circ}C$ (LT), room temperature (RT), oven treatment at $100^{\circ}C$ (OV), and $H_2O_2/UV(PER)$ method. Degradation times for each thermal system were 6 hour, 24 hour, 48 hour, 1 week, 2 week, 4 week. Alizarin was detected from the control and degraded samples of both alizarin dyed and madder dyed fabrics. Benzoic acid, 2, 4-di-tert-butylphenol, phthalic anhydride were detected as the degradation products for both alizarin dyed and madder dyed fabrics. The result suggest that these products can be used as the fingerprints of GC-MS analysis for the identification of madder dye in archaeological textiles. Both alizarin dyed and madder dyed samples became less red and less yellow after degradation. In the PER degradation system madder dyed sample showed the greatest color difference even after 1 week of degradation treatment. Further research is necessary for investigating the color change in the exhumed textiles, which is caused by the dual action of dye fading and the staining of organic matters in the soil.

Solvent Extraction of Trace Mo(VI) in Natural Water Samples by Chelation and Ion-pairing (킬레이트 및 이온쌍 형성을 이용한 자연수 중 극미량 Mo(VI)의 용매추출)

  • The formation of Mo(VI)-alizarin red S chelate ion and its extraction into an organic solvent by ion-pairing were studied for the separative determination of trace Mo(VI) in natural water samples. Natural water 100mL was sampled in 250mL separatory funnel. After Mo(VI)-ARS chelate ion was formed by adding 0.01M alizarin red S solution 0.5mL to the water sample of pH 4.0, 0.2% aliquat-336 chloroform solution 10mL was added and the solution was vigorously shaked for about 30 seconds to from the ion-pair between Mo(VI)-ARS and aliquat-336, completely. The solution was stood for about 90 minutes. And the organic phase was taken for the absorbance measurement of the ion-pair at 520 nm. The content of Mo(VI) in sample was obtained from the standard calibration curve. Several extraction conditions such as pH, adding amounts of alizarin red S and aliquat-336, and shaking and standing times were optimized. This procedure was applied to the analysis of river and tap waters. It could be confirmed from the recoveries of over 99% in samples spiked with a given amount of Mo(VI) that this method was quantitiative in the determination of trace Mo(VI) in a natural water.

Antioxidant Effect of some Phenolics on Soybean Oil (식용대두유에 대한 Phenolics의 항산화 효과)

  • Some phenolics were examined as an antioxidant for the autoxidation of soybean oil. Soybean oil was autoxidized under a mild condition (the flow rate of 67ml $O_{2}/min$ and $50^{\circ}C$). The antioxidant effect was estimated by active oxygen method. Phenolics show distinctive antioxidant effect, and the effect is prominent when cupper or iron was added. Phenolics showed a tendency to increase antioxidant effect with an increase of the number of hydroxyl group, and the increasing order was ferulic acid, quinalizarin, sesamol, alizarin, fisetin and purpurogallin. However, the effect was remarkably low in ferulic acid, alizarin and quinalizarin. It was found that the antioxidant effect was dependent on the functional group and geometric molecular structure of phenolics.

  • Electrochemical behavior of $Ln^{3+},$-ALC complexes($Gd^{3+},$ $Tb^{3+},$ $Dy^{3+},$ $Ho^{3+},$ $Er^{3+},$ $Yb^{3+}$ and $Lu^{3+}$-alizarin complex-one) has been investgated by d.c polarography, differential pulse polarography and cyclic voltammetry. The reduction mechanism of ALC comes to the conclusion that the two electron make one step of reversible processes, and that there is few adsorption in the electrode reaction. The new complex is made from one lanthanide ion and one ALC. This complex is proven to make an adsorptive complex wave, by the experiments of differential pulse polarography and cyclic voltammetry. The reduction potential of complex wave($P_2$)turns up more negatively than ligand wave($P_1$) does. Linear calibration curves of the decreasing P1 and increasing $P_2$ is obtained when the lanthanide concentration varys from $2.5X10^5$M to $1X10^4M$.

Method for Marking on Scales of Juvenile Black Rockfish by Immersion in Three Chemicals (세 화학 물질, Oxytetracycline hydrochloride, Alizarin red S 및 Calcein의 침지 처리에 의한 조피볼락 치어의 비늘 표지)

  • In this study, we evaluated the efficiency of chemical marking of black rockfish scales by immersion in oxytetracycline hydrochloride (OTC, 500 ppm), alizarin red S (AR, 250 ppm) and calcein (CARL, 250 ppm) diluted rearing water. Immersion treatment of chemicals had no effects on both mortality and growth of black rockfish. Marking sucess was 100% in all treatment durations (24, 48 and 72 hours) with three chemicals and marking quality was higher in 48 and 72 hours than 24 hours treatment. Marking retention rates at 24 weeks after treatment were 100% in OTC and CAL treated group, but marking quality was higher in CAL treated group (brilliant 92%, bright 8% and dim 0%) than in OTC treated group (brilliant 4%, bright 70% and dim 26%). AR treated group had lower marking retention rates and marking quality than OTC and CAL treated group. As a results, immersion treatment with OTC and CAL was effective in marking scales of black rockfish and practical in releasing program and other studies requires same rearing environment.

  • The intact dental pulps which were free of their tooth bud from adult rat incisors, and oral mucosa were transplanted subcutaneously in homologous rats to study the formation of calcified tissue. The rat were sacrificed after 1,2,3 and 4 weeks following transplantation of dental pulp and oral mucosa. The samples which contained the transplanted and surrounding tissue were fixed in 10% NBF, stained with hematoxylin and eosin, alizarin red S, von Kossa, and alcian blue. Microscopic examinstins revealed as follows: 1. The transplanted oral mucosas were not calcified but tended to form the epithelial cysts. 2. At 1 week after transplantation of dental pulp the calcified structures were appeared at the periphery of the transplantation of dental pulp but weakly reacted to alizarin red S, von Kossa, and alcian blue. 3. At 2 weeks after transplantation of dental pulp the calcified structures began to expand from the periphery to the center of the transplanted dental pulp and occupied the large areas comparatively, and strongly reacted to alizarin red S, and von Kossa stains. 4. At 3 weeks after transplantation of pulp tissue the fibrous components were grown at the periphery of the transplanted pulp tissuesand at 4 weeks a large amount of fibrous tissues were observed. The transplanted pulp tissue tended to form foreign bodies gradually.

Comparison of TLC and GC-MS Method in the Analysis of Dye Extracted from Madder Plant (꼭두서니 추출염료에 대한 TLC와 GC-MS 분석법의 비교 연구)

  • This research was aimed to investigate and compare the effectiveness of TLC and GC-MS methods in the analysis of chromophoric substances extracted from madder plant. Alizarin and purpurin 0.3% solution were used as comparative standards; madder extraction was prepared by heating the solution of powdered madder at 80℃, pH 1.5, for 90 min. Best elution solvent for TLC in silica gel plate was toluene:ethyl acetate=9:1, which resulted in red and yellow spots from madder extraction each of which showed R/sub f/ values 0.32-0.43 and 0.07-0.11. Although the red spot in particular exhibited similar characteristics as standard purpurin in color, shape, and R/sub f/ values, the result was inconsistent throughout different TLC trials. GC-MS analysis showed only small amount of alizarin and no purpurin in the madder extraction. Other chromophoric substance such as 2-furancarboxaldehyde, 5-(hydroxymethyl)-, anthralin, and danthron were also detected in small amounts. The result indicated that TLC was less sensitive to detecting and identifying the natural dyestuff which is generally constituted with a number of similar but chemically different chromophoric substances.

Determination of Trace Mo(VI) in Seawater Samples by Ion Pair Formation and Solvent Extraction (이온쌍 형성-용매추출에 의한 해수 중 극미량 Mo(VI)의 정량)

Removed Fe(III) and Zn(II), and Cu(II) by precipitating with anthranilic acid at pH 4.0 and 2.0, seawater 100mL was sampled in 250mL separatory funnel. After Mo(VI)-ARS chelate ion was formed by adding 0.01M alizarin red S solution 1.0mL to the water sample of pH 4.6, 0.6% aliquat-336 chloroform solution 10mL was added and the solution was vigorously shaked for about 30 seconds to form the ion-pair between Mo(VI)-ARS and aliquat-336 perfectly. The solution was stood for about 30 minutes. And the organic phase was taken for the absorbance measurement of the ion-pair at 520nm. The content of Mo(VI) was obtained from the standard calibration curve. Several extraction conditions such as pH, adding amounts of alizarin red S and aliquat-336, and shaking and standing times were optimized and the interferences and release of concomitant ions was also studied. This procedure was applied to the analysis of Eastern and Yellow seawaters. It could be confirmed from the recoveries of over 85% in samples spiked with a given amount of Mo(VI) that this method was also quantitative in the determination of trace Mo(VI) in a seawater.

A Study on Dyeability of PEI-treated Cotton Fabric with Polychromatic Natural Dyes (PEI를 처리한 면직물의 다색성 천연염료에 대한 염색성 변화)

  • This study examined the change in the dyeability of natural dyes on cotton fabrics by a PEI (polyethyleneimine) pretreatment instead of mordants. Cotton fabrics were treated with PEI and the changes in the dyeability were shown by measuring the amount of PEI on cotton fabrics. Samples treated with PEI were dyed with two natural polychromatic dyes with a different affinity to cotton fibers: Alizarin Red S and Curcumin. The changes in dyeability by three variables (time, temperature and concentration of dyes) on cotton fabrics were analyzed by the K/S value to define optimum dyeing conditions. Subsequently, the PEI treatment improved the dyeability of cotton fabrics with both dyes of low and high affinity to cotton fibers. Thus, PEI could be a suitable heavy metal mordant replacement.
