Mild and Efficient Silylcyanation of Aldehydes Catalyzed by Zirconium Complex

  • An efficient method of addition of trimethylsilyl cyanide to aldehydes by employing a zirconium complex as the catalyst has been described. A variety of aromatic, aliphatic, cyclic and heterocyclic aldehydes has been converted into corresponding trimethylsilyl ethers in excellent yield within short reaction time.

Volatile Flavor Components of Scent, Colored, and Common Rice Cultivars in Korea

  • To compare the composition of volatile flavor components of three different cultivars of rice, Hyangnambyeo (aromatic cultivar), Heugjinjubyeo (pigmented cultivar) and Dongjinbyeo (normal cultivar), the volatile flavor components of brown rice were isolated by Likens-Nickerson simultaneous steam distillation and extraction apparatus. The flavor concentrates obtained were analyzed by gas chromatography and gas chromatography-mass spectrometry. A total of 65 components, including 14 aliphatic aldehydes and ketones, 7 aliphatic alcohols, 8 aromatic alcohols, 13 hydrocarbons, 9 esters, 7 aliphatic acids, and 7 miscellaneous components were identified. The aliphatic aldehydes, which are known as contributors to the overall flavor of cooked rice, were present in larger amounts in Hyangnambyeo than in Heugjinjubyeo and Dongjinbyeo, while the difference in quantity of these components between Heugjinjubyeo and Dongjinbyeo was not remarkable. Hyangnambyeo and Heugjinjubeyo contained 562 ng and 259 ng of 2-acetyl-1-pyrroline per gram of brown rice based on dry weight, respectively, which is a key compound contributing to the popcorn-like aroma in aromatic rice. Dongjinbeyo contained about 6 ng.

  • A systematic study of the partial reduction of nitriles to the corresponding aldehydes with thexylbromoborane-methyl sulfide $(ThxBHBr{\cdot}SMe_2)$ under practical conditions has been carried out. The yields of aldehydes are good in the aliphatic series. However, the yields of aromatic series vary with substituents and ring itself.

  • A systematic study of the partial reduction of carboxylic acids and their derivatives to the corresponding aldehydes with lithium tris(dialkylamino)aluminum hydrides under practical conditions has been carried out. The diethylaminosubstituted derivative of lithium aluminum hydride, lithium tris(diethylamino)aluminum hydride (LTDEA), shows quite general applicability in the conversion of carboxylic acids, carboxylic esters, and primary carboxamides to the corresponding aldehydes. Lithium tripiperidinoaluminum hydride (LTPDA) also appears to be a reagent of choice for such partial transformation of primary carboxamides. In additioin, both LTDEA and LTPDA reduce tertiary carboxyamides to aldehydes in high yields. Finally, lithium tris(dihexylamino)aluminum hydride (LTDHA) is capable of achieving the chemoselective reduction of aromatic nitriles to aldehydes in the presence of aliphatic nitriles under practical conditions.

  • Catecholalane (1,3,2-Benzodioxaluminole, CA) prepared from catechol and aluminum hydride in tetrahydrofuran (THF) is found to be a selective reducing agent. The systematic study in order to characterize the reducing properties of the reagent under practical conditions (THF, 0 or 25 ℃, the quantitative amount of reagent to compound) has been done. The reagent reduces aldehydes, ketones, esters and acid chlorides to the corresponding alcohols, and primary amides to the corresponding amines. Especially noteworthy is that the reagent can convert both aromatic and aliphatic nitriles to the corresponding aldehydes in very high yields.

  • Potential of acetates and related compounds in glacial acetic acid as a catalyst for aldol-type condensation reactions was examined. Reactions of cycloalkanones or selected heteroaromatics with aldehydes in presence of 10 mol% of various acetates in acetic acid afforded ${\alpha},{\alpha}$'-bis(substituted-benzylidene)cycloalkanones and substituted-benzylidene-mackinazolinones, respectively, in good yields. Among the compounds tested, ammonium acetate is the best and effective especially towards the reactions of mackinazolinone and aliphatic aldehydes to afford 6-alkylidenemackinazolinones.

Purification and Reaction Mechanism of Rat Brain Succinic Semialdehyde Dehydrogenase

  • Rat brain succinic semialdehyde dehydrogenase (EC 1.2.1.24 SSADH) activity was detected in mitochondrial, cytosolic and microsomal fractions. Brain mitochondrial soluble SSADH was purified by ammonium sulfate precipitation, DEAE Sephacel, and 5'-AMP Sepharose 4B affinity chromatography. The purified enzyme was shown to consist of four identical subunits, and the molecular weight of a subunit was 55 kD. The $K_m$ for short chain aliphatic aldehydes and aromatic aldehydes were at the $10^{-3}M$ level but that for succinic semialdehyde was 2.2 ${\mu}M$. Either $NAD^+$ or $NADP^+$ can be used as a cofactor but the affinity for $NAD^+$ was 10 times higher than that for $NADP^+$. The brain cytosolic SSADH was also purified by ammonium sulfate precipitation, DEAE Sephacel, Blue Sepharose CL-6B and 5'-AMP Sepharose 4B affinity chromatography and its Km for short chain aliphatic aldehydes was at the $10^{-3}$ level but that for succinic semialdehyde was 3.3 ${\mu}M$. $NAD^+$ can be used as a cofactor for this enzyme. We suppose that both enzyme might participate in the oxidation of succinic semialdehyde, which is produced during GABA metabolism. The activity of both cytosolic and mitochondrial SSADH was markedly inhibited when the concentration of succinic semialdehyde was high. The reciprocal plot pattern of product inhibition and initial velocity indicated a sequential ordered mechanism for mitochondrial matrix SSADH. Chemical modification data suggested that amino acid residues such as cysteine, serine and lysine might participate in the SSADH reaction.

Analysis of the Terpenoids from Syneilesis palmata Essential Oil and the Variation of the Sesquiterpene Compounds by Harvest Year (우산나물 정유의 테르펜 화합물 분석 및 수확 연도에 따른 Sesquiterpene 화합물 변화 조사)

  • This study was investigated the chemical composition from Syneilesis palmata essential oil and the tendency of variation of the sesquiterpene compounds according to the harvesting time. The essential oils obtained by hydro distillation from the aerial parts of Syneilesis palmata were analyzed by GC and GC-MS. Ninety-eight compounds consisting of 9 aliphatic hydrocarbons, 17 sesquiterpene hydrocarbons, 11 aliphatic aldehydes, 1 terpene aldehyde, 8 aliphatic alcohols, 4 monoterpene alcohols, 16 sesquiterpene alcohols, 3 diterpene alcohols, 6 ketones, 11 esters, 8 oxides and epoxides, 3 acids and 1 miscellaneous one were identified from the oil. Spathulenol (22.33%) was the most abundant compound, followed by ${\beta}$- caryophyllene (6.23%), germacrene D (5.57%), longipinane (4.10%), and epiglobulol (3.65%). The volatile composition of Syneilesis palmata was characterized by higher contents of sesquiterpene compounds, especially sesquiterpene alcohols. The total content of 13 sesquiterpene compounds was decreased significantly from 2010 to 2012. ${\alpha}$-Caryophyllene, ${\beta}$-bisabolene, elemol, germacrene D, ${\beta}$-zingiberene, longipinane, and ${\beta}$-caryophyllene alcohol contents decreased, while ${\beta}$-bisabolol content increased during 3 years. The ecological responses to recent climate change may be influenced in the chemical components of natural plant terpenoids.
