적조에 관한 조사는 육안과 샘플링에 의한 현지조사에 의해 일반적으로 진행되고 있다. 이와 같은 기존의 조사와 분석방법은 적조의 발생과 소멸까지의 경로를 정확하게 예측하기 곤란하였다. 이에 좀더 객관적이고 과학적인 방법으로 적조의 발생과 그 이동경로 등 시·공간적인 환경인자를 분석해 볼 필요성이 대두되고 있다. 본 연구에서는 적조발생에 대한 피해를 최소화하기 위해 원격탐측기법을 적용하였다. 적조의 주요 원인이 되는 해수 온도의 변화, 조류의 변화 등을 자료와 탐측데이터를 활용하였으며, 육지로부터 유입되는 부하 물질 등의 적조원인 데이터를 해석함으로서 남해연안에 발생하는 적조의 현상을 사전에 예측하는 기법을 연구하였으며 그 활용가능성을 제시하고자 하였다.
For the test organism of algal growth potential (AGP), the diatom in the genus Stephanodiscus which cause blooms in the Nakdong River was used instead of generally used strains of Selenastrum, Microcystis, or Anabaena. AGP results indicated that all the samples in the Nakdong River except for that from the Nakdan Bridge site were eutrophic state. Furthermore, the sample from Kumho River site was hypertrophic state. In the main stream Nakdong River, the value of AGP was lowest at the upstream Nakdan Bridge site and was highest at Koryoung Bridge site which is just downstream of Kumho River confluent point indicating the seriousness of pollution contributed by the Kumho River to the Nakdong River. Changes in the concentration of nutrients before and after the AGP tests and inter-relationship among the nutrients indicated that the growth of the Stephanodiscus in the AGP tests were mostly affected by the nitrate, silicate and phosphate. The limiting nutrient was identified by the nutrient addition experiments and the results showed that phosphate was the limiting nutrient for the growth of Stephanodiscus in the tested samples.
The purpose of this study is to analyze the characteristics and correlations of the Yeongcheon Lake in order to reduce the occurrence of harmful cyanobacteria. In this study, we investigated the water quality and phytoplankton of the lake from May to November in 2017. Correlation and data mining analyses were performed to analyze the relationship between the two factors. The water temperature was lowest at the point where the Yeongcheon Lake inflow occurs at Imha Lake. It was highest at the point where the outflow occurs to Angye Lake. The pH was also highest at the outflow point, but in the case of DO, it was highest at the midpoint between the inflow and outflow. The main cyanobacteria that emerged during the study period were Oscillatorialimosa, Microcysti saeruginosa and Aphanizomenon flos-aquae. As a result of correlation analysis, the water temperature, inflow, COD loading, TOC loading at the inflow point of the Yeongcheon Lake were the items that were related to the harmful cyanobacteria. The data mining analysis indicated that the TP loading and harmful cyanobacteria in the inflow point of the Yeongcheon Lake were influential on the detrimental cyanobacteria in the Yeongcheon Lake outflow point. When the TP loading was less than 39.0 kg/day at the inflow site, it was expected that the amount of harmful cyanobacteria could be maintained below 10,000 cells/mL.
The present study assessed the efficient removal of nutrients and Chlorophyll-a (Chl-a) by using methyl esterified sericite (MES) and pine needle extracts (PNE), a low cost and abundant green hybrid material from nature. For this purpose, the optimal conditions were investigated, such as the pH, temperature, MES and PNE ratio, and MES-PNE dose. In addition, a Microcystis aeruginosa control using MES-PNE was also analyzed with various inhibition models. The removal of the nutrient and Chl-a onto MES-PNE was optimized for over 95% removal as follows: 2-2.5 for the MES-PNE ratio, 7-8 pH and a $22-25^{\circ}C$ temperature. In this respect, approximately 1.52-2.20 g/L of MES-PNE was required to remove each 1 g of dry weight/L of Chl-a. Total phosphorus (TP) has a greater influence on the increase in Chl-a than total nitrogen (TN) according to the correlation between TN, TP and Chl-a. Moreover, the Luong model was the best model for fitting the biodegradation kinetics data from Chl-a on MES-PNE from lake water. The novel hybrid material MES-PNE was very effective at removing TN, TP and Chl-a from the lake and can be applied in the field.
Quantifying the abundance of Chattonella species is necessary to effectively manage the threats from ichthyotoxic raphidophytes, which can cause large-scale mortality of aquacultured fish in temperate waters. The identification and cell counting of Chattonella species have been conducted primarily on living cells without fixation by light microscopy because routine fixatives do not retain their morphological features. Species belonging to the Chattonella marina complex, including C. marina and C. marina var. ovata, had high genetic similarities and the lack of clear morphological delimitations between the species. To estimate the abundance of C. marina complex in marine plankton samples, we developed a protocol based on the droplet digital polymerase chain reaction (ddPCR) assay, with C. marina complex-specific primers targeting the internal transcribed spacer (ITS) region of the rDNA. Cell abundance of the C. marina complex can be determined using the ITS copy number per cell, ranging from 25 ± 1 for C. marina to 112 ± 7 for C. marina var. ovata. There were no significant differences in ITS copies estimated by the ddPCR assay between environmental DNA samples from various localities spiked with the same number of cells of culture strains. This approach can be employed to improve the monitoring efficiency of various marine protists and to support the implementation of management for harmful algal blooms, which are difficult to analyze using microscopy alone.
The occurrence of harmful algae on the coast of Korea has been a cause of damage to the aquaculture industry and deterioration of the coastal ecosystem environment. A method is required to predict their outbreak in real-time at the site. Therefore, this study attempted to develop a small hybrid optical sensor and real-time monitoring system based on LiDAR that can be used in the field and laboratory and can be applied to various platforms. FMS-L specifically suggested the amount of Chlorophyll a (Chl a) in the sample by measuring and analyzing the fluorescence emitted by the irradiating light. The accuracy of FMS-L was verified by measuring the concentrations of standard Chlorophyll a substances and Margalfidinium polykirkoids. In addition, the precision was verified by comparing the measurement results of FMS-L using commercial equipment Phyto-PAM-II. This equipment is compact and easy to move. Therefore, it can be easily applied to field surveys, allows short time measurements (10 s), and can be applied at a distance of 10 m from the measurement site.
본 연구는 2013년 4월 하순~5월 동안에 영산강의 승촌보 상류에서 군체형 녹조류 Eudorina elegans의 단일 종에 의해 유례없이 대발생한 녹조현상의 전개 과정을 모니터링하였다. 영산강은 전형적인 조절 하천으로서 외적 또는 내적 요인에 의한 부영양화가 극도로 심각한 실정이었다. 하수처리수의 과잉 영양염을 기반으로 유속, 일사량 및 수온의 구조적 또는 비구조적 복합 영향에 의한 조류(규조류(겨울), 녹조류(봄~여름) 및 남조류(여름))의 대발생은 뚜렷한 계절적 잠재력으로 상존하였다. 이 중에서 봄철 녹조현상은 E. elegans에 의한 것으로서 그 수준은 최대 $1,000mg\;m^{-3}$(>$50{\times}10^4cells\;mL^{-1}$)을 초과하였고, 발생 초기에 폭증하였다가 시간이 경과하면서 점진적으로 감소하는 양상이 현저하였다. 또한, 간헐적인 강우에 의해 하류로 이송되면서 분포 범위가 급속도로 확대되는 특성을 보였다. 보 구조물의 조작으로 펄스유량을 시험 적용하였으나 근본적인 문제를 해결하는 대응책은 아니었고, 하류 하천에 대한 영향을 고려해야 하는 문제점을 포함하고 있었다. E. elegans 녹조현상은 군체형 운동성 조류의 전형적 특성을 나타내었고, 최종적으로 후속되는 강우사상 (>45 mm)에 의해 소멸되었다.
국내 이상기후현상과 기후변화로 인해 하천, 호소에 조류의 과대성장이 빈번히 발생하고 있다. 하천의 조류는 녹조류, 남조류, 규조류, 기타조류 등으로 분류되며, 남조류 중 일부 종은 악취 유발과 독성물질의 배출로 문제를 야기하고 있다. 국내에서는 조류경보제를 시행하고 있으며, 이는 유해남조류세포수에 따라 발령이 된다. 이렇듯 유해남조류 측정은 매우 중요하며, 현재 조류의 분석방법은 현장샘플을 하여 조류검경을 통해 녹조류, 남조류, 규조류 중 유해남조류를 구분하여 세포수를 산정하기 때문에 시간이 많이 소요된다. 최근 원격탐사를 통해 남조류세포수의 대체 인자(지표)인 Phycocyanin을 통해 조류농도를 분석하고 있으나, 남조류세포수 분석에 대한 연구는 미비한 실정이다. 본 연구에서는 실제 하천에서 발생한 조류를 채취하여 조류검경을 통해 남조류세포수를 검경하였으며, 광학센서로 Phycocyanin농도, 초분광센서를 통해 조류스펙트럼을 취득하고, 이를 통해 남조류세포수 추정식을 산정하여 남조류세포수를 추정하였다.
본 연구에서는 태화강 하류에서 조수 및 조사 지점에 따른 태화강 수질 특성을 알아보고, 겨울 조류 발생에 영향을 주는 환경요인을 분석하고자 하였다. 2010년 5월에서 2011년 1월까지 태화강 하류의 신삼호교, 구삼호교, 명정천, 태화교, 명촌교 등 5개 지점에서, 사리의 만조와 간조에 시료를 채취하였다. 염분도, 용존산소(DO), 생물학적 산소 요구량(BOD), 화학적 산소 요구량(COD), 엽록소 a(Chl a), 여러 영양염류 등의 환경변수를 측정하였다. 염분도는 하류 방향으로 갈수록 증가하였으며, Chl a의 지점별 평균값은 만조 $10-26mg/m^3$, 간조 $11-53mg/m^3$였다. 특히 태화교의 경우 11월 21일-12월 22일에 Chl a 농도가 $138-296mg/m^3$로 급격히 증가하였으며, Cryptophyceae의 Rhodomonas lacustris가 우점하는 식물플랑크톤이었다. 태화교에서 Chl a 농도는 염분도, BOD, DO, COD, pH, T-N와 양의 상관관계, 수온, 기온, $NO_3{^-}$-N과는 음의 상관관계가 있었다. 반면 명촌교의 경우 Chl a 농도는 8월 25일에 $55mg/m^3$로 최고값을 보였으며, Chl a 농도는 $NH_3$-N, T-N, $PO_4{^{3-}}$-P, T-P, 일반세균과 양의 상관관계를 나타내었다. 연구의 결과는 태화강 하류에서 조사 지점에 따른 수질의 변동폭이 매우 크며, 조류 발생의 양상도 태화교와 명촌교 사이에 차이가 있음을 보여준다.
전세계적으로 연안해역에서 확산되고 있는 유해유독성 식물플랑크톤의 대발생은 수산자원생물자원에 심각한 피해를 입힌다. 본 연구에서는 유해성 미세조류 대발생을 제어하기 위해 개발된 신물질 Thiazolidinedione 유도체(TD49)의 살조능을 유해성 미세조류 성장단계(초기성장기, 대수증식기, 안정기)에 따라 조사하였다. TD49는 $Heterosigma$$akashiwo$, $Chattonella$$marina$ 그리고 $Chattonella$ sp.의 세포를 사멸시켰으며, 특히 낮은 농도($0.02{\mu}M$)의 TD49는 대수증식기와 안정기보다 초기 성장기에서 우수한 살조효과를 보였다. 또한 모든 성장단계에서 유해생물을 제어 할 수 있는 TD49의 농도는 $2{\mu}M$로 측정되었다. 무각 편모조류인 $Heterosigma$$akashiwo$, $Chattonella$$marina$ 그리고 $Chattonella$ sp.은 세포벽이 약하여 TD49물질에 의해 세포가 쉽게 파괴되어 우수한 살조효과를 보였다. 결과적으로 본 연구에서 개발된 Thiazolidinedione 유도체(TD49)는 유해적조생물 $H.$$akashiwo$, $C.$$marina$ 그리고 $Chattonella$ sp.를 제어할 수 있는 우수한 물질로 판단되었으나, 추후 현장 실용을 위해 메소코즘과 같은 인공생태계를 이용한 해양생태계 위해성 평가가 체계적으로 추진되어야 한다.
