  • To gain a greater understanding of the potential for future harmful algal bloom (HAB) outbreaks and to trace the dispersal paths of dinoflagellates, sediment samples were collected from 13 stations in the southeastern Yellow Sea. 23 different types of dinoflagellate resting cysts were identified from the samples. Protoceratium reticulatum (1-391 cells/g dry weight), Gonyaulax scrippsae (0-254 cells/g dry weight), G. spinifera (0-301 cells/g dry weight) and Alexandrium spp. (ellipsoidal type) (0-76 cells/g dry weight) were the dominant species at all surveyed stations. The overall distribution pattern demonstrated that the resting cyst densities were highest in the offshore area and decreased gradually toward the Korean coast. On the other hand, the composition rate of resting cysts of the heterotrophic dinoflagellate species to the total dinoflagellates was higher in the Korean coast region than in the offshore area. We supposed that this distribution pattern of dinoflagellate resting cysts appeared to be influenced by the hydrographic features and environmental conditions of the Yellow Sea.

  • Yellow Sea surface sediment samples collected on October 15-31, 2003 were analyzed using the palynological process to investigate the spatial distribution of dinoflagellate resting cysts. The sampling areas comprised four latitudinal transects, the northernmost of which was located off the Shandong Peninsula, China and the southernmost off Jeju Island, Korea. Each transect line was composed of six to nine stations, spanning the distance between the Chinese and Korean coasts. Twenty-five different types of dinoflagellate cysts were identified. Gonyaulax scrippsae, Alexandrium spp. (ellipsoidal type), and G. spinifera were the most dominant at all stations surveyed. Dinoflagellate cysts belonging to the Gonyaulacales comprised over 50% of all cysts collected. The latitudinal distribution trend showed that cyst concentrations along the two middle transects were much higher than those along the two northern and southern transects. Cyst concentrations in the offshore central areas reached their highest values within each transect and gradually decreased toward the Chinese and Korean coasts. Overall, cyst concentrations were markedly elevated in the offshore central Yellow Sea areas and gradually decreased outward in all four directions. This concentric cyst distribution pattern was consistent with the hydrographic features of the Yellow Sea, such as circular current systems, sedimentary properties, and water depth.

  • Relative DNA contents in some harmful algae were measured using DAPI staining and image analysis system. This method was useful to identify some morphologically similar species and isolates from harmful algal blooms (HABs). In exponential phase, Prorocentrum micans had higher relative DNA content (RD) of $1.83\pm0.52$ than any other isolates, followed by Cochlodinium polykrikoides $(1.10\pm0.46)$ Alexandrium tamarense $(0.93\pm0.32)$ Gyrodinium impudicum $(0.56\pm0.17)$, Scrippsiella trochoidea $(0.41\pm0.26)$ and P. minimum$(0.05\pm0.01)$. When they were fixed with Lugol's solution, it was difficult to d,iscern C. polykrikoides from G. impudicum under the light microscope, but the DNA contents were quite different in two species. C. polykrikoides contained about twice as much RD as G. impudicum under the same culture conditions and exponential phase. DAPIstained DNA feature in C. polykrikodes showed concentrated in the peripheral part of the cell, but in G. impudicum showed a compact structure in the central part. Although A. tamarense and S. trochoidea were morphologically similar under the light microscope, nuclear DNA content of A. tamarense was twice as much as that of S. trochoidea.

  • Mortality of several species of fish and shellfish exposed to Harmful Algal Blooms (HABs) caused by Cochlodinium polykrikoides, Heterosigam akashiwo, Alexandrium tamarense, Eutreptiella gymnastica, Heterocapsa triquetra and Prorocentrum micans was studied. When fish were exposed to a cell density of 8,000 cells $mL^{-1}$ in C. polykrikoides, $35\%$ of flatfish and darkbanded rockfish died within 48 hrs. However, jacopever rockfish had mortality of higher than $85\%$. Rock bream, filefish and red sea bream showed $100\%$ mortality within 10 hrs with an exposure cell density of 8,000 cells $mL^{-1}$. The rest of HABs except for C. polykrikoides showed that there was no fish and shellfish death throughout the 48 hrs even in the maximum cell density of 100,000 cells $mL^{-1}$ These results imply that C. polykrikoides can have a serious impact on fish mortality and it is regarded as an ichthyotoxic dinoflagellate. The fish death may be attributed to anoxia caused by a combination of the production of reactive oxygen species (ROS) and polysaccharide from C. polykrikoides during blooms.

  • The spatio-temporal distribution of dinoflagellate resting cysts was investigated by palynological processing to understand the harmful algal bloom (HAB) potential in the sediment of the Saemangeun area in 2003. In total, thirty-two dinoflagellate species were identified, and their concentrations were in the range of $6-1,618\;cysts{\cdot}g^{-1}$ (dry weight). The concentrations of resting cysts were higher in the spring (26 species, $64-1,101\;cysts{\cdot}g^{-1})$ and summer (30 species, $81-1,618\;cysts{\cdot}g^{-1})$ than in the autumn (32 species, $6-1,150\;cysts{\cdot}g^{-1})$ and winter (24 species, $25-728\;cysts{\cdot}g^{-1}).$ The composition rate of the heterotrophic dinoflagellate species to the total, which is closely related to the eutrophication process, to ranged from 6 to $29\%$ in the study area. The most dominant species was toxic Alexandrium tamarense/catenella $(25\%)$ followed by Gonyaulax scrippsea $(9\%)$ and toxic Protoceratium reticulatum $(5\%).$ Given the high abundance of the toxigenic dinoflagellate species, the Saemangeun area is considered to have the great potential for HABs in the future.

  • Most previous studies on dinoflagellates in Korean coastal areas were conducted without morphological descriptions and illustrations of the observed dinoflagellates. This indicates that the species and diversity of dinoflagellates may have been respectively misidentified and underestimated in the past, probably due to cell shrinkage, distortion and loss caused by sample fixation. This study provides information on the morphological observations of four dinoflagellate orders (Prorocentrales, Dinophysiales, Gonyaulacales and Gymnodiniales) from Jangmok Harbour in Jinhae Bay, Korea. The unfixed samples were collected weekly from December 2013 to February 2015. A total of 13 genera and 30 species were identified using light and scanning electron microscopy, although some samples were not clarified at the species level. Harmful dinoflagellates, Prorocentrum donghaiense, Tripos furca, Alexandrium affine, A. fundyense, Akashiwo sanguinea and Cochlodinium polykrikoides, were identified based on the morphological observations. The results also reflect the occurrence and identification of dinoflagellates that had not been previously recorded in Jangmok Harbour.

  • Lectin binding assay was conducted on 3 A. tamarense isolates (AT-A, AT-2 and AT-6). Fatty acid composition of all 3 isolates was analyzed, and total carotenoid content and $\beta$-carotene were also determined. AT-A and AT-2 treated with different lectins in this study showed the positive response, whereas potentially toxic AT-6 did not bind DBA lectin, regardless of different growth phase, but conjugated ConA, PNA, RCA, SBA, UEA and WGA. It is possible that DBA is a desirable method for rapid and easy discrimination of highly toxic A. tamarense. AT-A, AT-2 and AT-6 comprised saturated fatty acids (49.0-61.9%), monounsaturated fatty acids (8.0-20.5%) and polyunsaturated fatty acids (23.2-30.5%). In particular, 22:6 (n-3) polyunsaturated fatty acid in AT-6 had a high abundance, compared with AT-A and AT-2. However, carotenoid content and $\beta$-carotene were not contributed to discriminate each isolate. Due to variability in biochemical composition at different isolates, possibly DBA and 22:6 (n-3) polyunsaturate fatty acid provide a good information for discrimination of AT-6.

  • Harmful micro algae isolated from Korean coastal waters, were tested with FITC-conjugated lectins and observed by epifluorescent microscopy to distinguish each other. Strain-specific sugar composition at the cell surface was suggested by the affinity of lectins to different microalgae. The micro algae Cochlodinium polykrikoides (CP-1) and Gymnodinium $A_3\;(GA_{3-1}\;1)$, are morphologically similar, but exhibited different binding activity with the lectins ECA, HPA and WGA. In Peridiniales, the micro alga Alexandrium tamarense (AT) bound HPA and WGA, but Scrippsiella trochoidea (ST-1) did not bind those lectins. Three species of Prorocentrum also exhibited different binding specificity with HPA, PHA and SBA. A nontoxic Korean isolate of Heterosigma akashiwo (HA-2) bound ConA, PEA and UEA. These results suggest that lectins are useful in discriminating morphologically similar species, as well as different species or strains within the same genus.
