Effect of irradiation on the oxidation kinetics of TODGA-based extraction mixtures at atmospheric pressure

  • The gas evolution from mixtures consisting of 0.2 M solution of N,N,N',N'-tetra-n-octyldiglycolamide (TODGA) in n-alcohol (n-decanol or n-nonanol) with Isopar-M diluent was investigated during thermal oxidation. The effect of ionizing radiation on their thermal stability has been studied. It has been determined that the volume of gaseous thermolysis products increases by 260% in the case of n-nonanol and 80% in the case of n-decanol compared to non-irradiated solutions. It has been shown that the gas evolution rate and gas volume increase when the irradiated mixture saturated with nitric acid is heated. However, there are no prerequisites for the development of autocatalytic oxidation.

  • During the synthesis of 4-fluorobenzaldehyde via the SNAr reaction of 4-nitrobenzaldehyde with TBAF, it was found that an equivalent amount of TBAF could oxidize benzaldehyde to benzoic acid. The reaction of 4-nitrobenzaldehyde with tetrabutylammonium fluoride (TBAF) gave 4-nitrobenzoic acid in high yield. Depending on the reaction conditions, other aromatic aldehydes produced acids with fewer amounts of alcohols. However, this type of oxidation has limited practical applications. Nevertheless, the mechanism is quite different from the Cannizzaro reaction because the amounts of the acid salt and alcohol formed were different.

  • Methanol electro oxidation on the surface of carbon paste modified by $NiCl_2/6H_2O$ was studied in 1M NaOH by potentiostatic and potentiodynamic methods. Ni/C catalyst by the concentration of 5% Ni showed about twice higher electro catalytic activity than Ni metal. The amount of monolayer's on the surface of electrode is almost one order higher for Ni/C than Ni electrode. The kinetic parameters and the diffusion coefficient of methanol were derived from chronoamperometry (CA) and electrochemical impedance spectroscopy (EIS) measurements.

  • The removal of 2,4-dichlorophenoxyacetic acid (2,4-D) in aqueous solution by coupled electro-oxidation and Fe(II) activated persulfate oxidation process was investigated. The electrochemical oxidation was performed using carbon sheet electrode and persulfate using Fe(II) ion as an activator. The oxidation efficiency was investigated by varying current density (2 - 10 mA/㎠), electrolyte (Na2SO4) concentration (10 - 100 mM), persulfate concentration (5 - 20 mM), and Fe(II) concentration (10 - 20 mM). The 2,4-D removal efficiency was in the order of Fe(II) activated persulfate-assisted electrochemical oxidation (Fe(II)/PS/ECO, 91%) > persulfate-electrochemical oxidation (PS/ECO, 51%) > electro-oxidation (EO, 36%). The persulfate can be activated by electron transfer in PS/ECO system, however, the addition of Fe(II) as an activator enhanced 2,4-D degradation in the Fe(II)/PS/ECO system. The 2,4-D removal efficiency was not affected by the initial pHs (3 - 9). The presence of anions (Cl- and HCO3-) inhibited the 2,4-D removal in Fe(II)/PS/ECO system due to scavenging of sulfate radical. Scavenger experiment using tert-butyl alcohol (TBA) and methanol (MeOH) confirmed that although both sulfate (SO4•-) and hydroxyl (•OH) radicals existed in Fe(II)/PS/ECO system, hydroxyl radical (SO4•-) was the predominant radical.

대장균에서 사람 ALDH2 유전자의 발현 (Expression of Human ALDH2 Gene in escherichia coli)

  • 사람의 미토콘드리아에 있는 aldehyde dehydrogenase(ALDH2)는 체내에서 알코올 대사 과정 중에 생성되는 아세트알데히드를 산화시키는 주된 역할을 담당하고 있다. 이 ALDH2가 알코올 대사에 미치는 영향을 연구하기 위하여 가용화된 효소가 필요하다. 알려져 있는 유전자의 염기서열 데이터를 바탕으로 ALDH2의 cDNA는 cDNA 라이브러리에서 선별하였으며, 이를 여러 가지 대장균 발현벡터에 연결하였다. 제조한 발현벡터를 형질전환시킨 대장균을 사용하여 단백질의 발현을 확인한 결과 대부분의 계에서 ALDH가 과발현되고 있었다. 그러나 발현된 단백질의 대부분은 inclusion body로 형성되어, 실제로 가용화된 효소의 양은 전체 발현된 양의 5% 이하 였고 이들 몇 가지 발현 system으로 재조합 미오2DML 발현을 확인하였다.

지렁이 Peroxidase의 특성 및 페녹시계 제초제의 분해 (characteristics of Peroxidase from the Earthworm, Lumbricus rubellus and Degradation of Phenoxyherbicides)

  • Peroxidase has been isolated to apparent homogeneity from earthworm, Lumbricus rubellus, using ammonium sulfate fractionation, Sephacryl S-2000 gel filtration, CM-cellulose cation exchange chromatography and native-PAGE elution. Some of its enzymatic characteristics were examined. The optimum pH for gruaiacol oxidation of earthworm peroxidase was determined to be 6.0, and the $K_{m}$ values against guaiacol and $H_2O_2$ were 1.25 mM and 3.4mM, respectively. When various compounds were tested as the possible substrates of the enzyme, o-dianisidine was used as the substrate. However, earthworm peroxidase could not oxidize esculetin and ferulic acid as substrates, suggesting the different characteristics of the enzyme from plant peroxidases. The optimum pH for veratryl alcohol and $H_2O_2$ oxidation was determined to be 2.5 when lignin peroxidation activity was examined. The $K_{m}$ values for veratryl alcohol and $H_2O_2$ were 0.02 mM and 0.13 mM, respectively. Furthermore, the earthworm peroxidase could oxidize phenoxyherbicides such as 2,4-D, 2,4-DP and MCPA as substrates. The optimum pHs for 2,4-D, 2,4-DP and MCPA were determined to be 4.0, 2.0 and 2.0, respectively. The most available substrate was 2,4-DP, followed by MCPA and 2,4-D when their peroxidation activities were compared.

Effect of Herbal Extracts Mixtures on Antioxidant System in Chronic Enthanol-treated Rats

  • Disturbance of antioxidant system is very common in chronic alcoholics and herbal or natural products with antioxidant activity have been used for its treatment. This study was to investigate the effect of Vitis vinifera extract(V), Schisandra chinensis extract(S), Taraxacum officinale extract(T), Gardenia jasminoides extract(G), Angelica acutiloba extract(A) and Paeonia japonica extract(P), and their combinations on the antioxidant and ethanol oxidation system. Male Sprague-Dawley rats were subjected to Lieber-DeCarli ethanol liquid diet(ED) and were then given different herbal extract mixtures for 6 weeks including VST(V 100+S 150+T 150mg/kg/day), VSG(V 100+S 150+G 150mg/kg/day), VTG(V 100+T 150+G 150mg/kg/day), and VAP(V 100+A 150+P 150mg/kg/day). When the activity of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) were compared between ED only group and herbal extracts treatment group, the differences were statistically significant. Phase I and II(glutathione-S-transferase, phenol sulfatransferase) enzyme activities were found to be significantly higher in the VAT treatment group compared to the ED group. Herbal extracts not only repressed the ethanol-induced elevation of malondialdehyde level, but also protected against ethanol-induced decrease in glutathione content, glutathione reductase, glutathione peroxidase, catalase and superoxide dismutase activities. The administration of the herbal extracts was found to be effective in eliminating lipid-peroxides induced by long-term consumption of alcohol by activating various enzyme systems and physiological active compound formation system. After a chronic consumption of alcohol, Angelica Radix protected the liver via activating the ethanol-metabolism enzyme system, and Paeoniae Radix via activating the ethanol-metabolism enzyme and the phase I, II-metabolism enzyme system. Taraxaci Herba was also effective in liver protection via activating the ethanol-metabolism enzyme system and the phase I, II-metabolism enzyme system, Gardeniae Fructus via activating the phase II-metabolism enzyme system and the anti-oxidation system enzyme, and Schisandra Fructus and a grapestone via activating the anti-oxidation system. Our data suggest that these herbal extracts may be useful as a health functional food or new drug candidate for fatty liver and hepatotoxicity induced by chronic alcohol consumption.
