Early Healing Responses of the Skin Wounds in the Korean Fire-Bellied Toad, Bombina orientalis (무당개구리 (Bombina orientalis) 피부의 초기 상처치유반응)

  • Early wound healing responses in the skin of the Korean fire bellied toad, Bombina orientalis, were examined using histochemical , scanning and transmission electron microscopical techniques. Cutaneous wounds were induced by sharp razor blade , and maintained in special cages for up to 3 days. By this treatment whole parts of epithelium and dermis were profoundly affected. After injury, retraction of damaged surface of the skin was observed. At 3 hours after injury, poly-band was form by mucous substance. The wound skin consisted of mucous gland secreted mucous substance which flows into wound cavity and the wound area was sealed by mucous substance which form thick layer at 6 hours. It appeared that natural disposition showed acid and neutral mucous substance by reaction of PAS and Alcian blue (pH 2.5) staining. After formation of the poly-band, epithelial cells were formed by migration of regenerating epithelial cells. These cells moved over the wound surface by migration at 12 hours after wounding. Poly-band was much increased in thickness and migrating cells were completely formed wound closure at 24 hours post-wounding. After 72 hours regenerated epithelial layer consists of four or five cell layers which are similar to clear, granular, and spinous layer.

Antianaphylactic Effect of Sinpo-tang by Anal Therapy (肛腸療法에 의한 辛蒲湯의 抗아나필락시 效果)

  • Objective : Anal Therapy is another way of taking medicine and applies to each field of clinical treatment extensively. Sinpo-tang(SPT) has been used for the treatment of a allergic rhinitis. In this study, the auther investigated the anti anaphylactic action of Sinpo-tang by anal therapy was investigated on cutaneous allergic reaction models. Methods : Results : 1. Sinpo-tang (0,001-0.1 g/kg) dose-dependently inhibited the compound 48/80-induced ear swelling response in mice. Inhibitory effect of Sinpo-tang was significant (P < 0.05) at the doses of 0.01, 0.1 g/kg. 2. Sinpo-tang (0.001-0.1 g/kg) inhibited the cutaneous allergic reaction activated by anti-dinitrophenyl IgE in rats. Of special note, Sinpo-tang (0.1 g/kg) inhibited the cutaneous allergic reaction by $68\%$. 3. This inhibitory effect of Sinpo-tang was confirmed by observation of alcian blue/nuclear fast red stained-mast cells in the cutaneous tissue. 4. Sinpo-tang (0.01-1 g/L) dose-dependently inhibited the compound 48/80-induced histamine release from the peritoneal mast cells. Conclusions :These results indicate that anal therapy of Sinpo-tang may be beneficial in the treatment of mast cell-mediated anaphylaxis by inhibition of histamine release from mast cells in vivo and in vitro.

Study on the Expectorant Activity of Ethanol Extracts from Some Crude Drugs Containing Saponins (Saponin을 함유한 생약추출물의 거담작용에 관한 연구)

  • The effect of ethanol extracts of Gilkyung(Platicodi Radix, Platicodongrandiflorum A, DC), Onji(Polygalae Radix, Polygala tennuifolia Willdenow) and Deoduk(Codonopsis lanceolate Radix, Codonopsis lanceolata) on expectorant activity of rat trachea was investigated. Following treatment of 50% ethanol extract of these medicinal plants (25 mg/rat), the content of neurotransmitters such as acetylcholine and histamine in tissue was significantly increased. The secretions of acid glycoproteins and the artificially injected phenol red were also increased. However, there was no significant difference except Onji From the histological study through periodic acid Schiff and alcian blue stain, the thickness of inner membrane of acinar glands and the stained glycoproteins on surface of epithelium and on the glands were observed in all the rats trachea treated with extract of medicinal plants. In vitro, the viscosity of mucin solution was slightly decreased with an addition of the extracts. Onji showed the most effective expectorant activity among them at the identical conditions. The mechanism of expectorant activity of these medicinal plants seems to be due to stimulation of secretion and changes of rheological properties of mucus.

The Toxicological Effects of Ahnjon-Yichun-Tang in Pregnant Rats and Fetuses (안존이천탕 추출물이 흰쥐의 모체 및 태자에 미치는 영향)

  • The objective of this study was to characterize the adverse effects of Ahnjon-Yichun-Tang during early pregnancy. Following successful mating, female Sprague-Dawley rats were given Ahnjon-Yichun-Tang(AYT) extract by oral administration daily with dose of 150mg (n=10), 300mg(n=10), 450mg(n=10) for 20 days of pregnancy. The rats in Control group(n=10) were orally administrated with Saline. All pregnant rats of Ahnjon-Yichun-Tang-treated and Control groups were sacrificed on day 20 of pregnancy. The pregnancy outcome was determined and the internal and reproductive organs of pregnant rat were observed. The fetuses were examined for the presence of various developmental toxic endpoints and stained with alcian blue and alizarin red S, and observed skeletal malformations. The results obtained in this study represent that there is no significant changes between Control and Ahnjon-Yichun-Tang-treated groups in body weight, organ weight, blood chemistry values, hematological values and pregnancy indexes of pregnant rat. The skeletal malformation of fetus was not observed as well. These results suggest that oral administration of Ahnjon-Yichun-Tang does not produce either maternal or developmental toxicity.

A Case of Metastatic Carcinoma of the Skin (피부전이암 1예)

  • We report a case of metastatic carcinoma of the skin in 62-year-old male who showed multiple nodules on the anterior chest, scalp, face and left axilla for 2 months' duration. He suffered from lower abdominal discomfortness for 6 months. Histopathologic findings revealed atypical cell nests and some tubular or glandular formation in the dermis and subcutaneous fat tissue. Material in the lumen showed diastase resistant and PAS positive, and alcian blue positive at pH 2.4, but negative pH 0.4. So it represented sialomucin. We did not find primary site of malignant tumor. In view of clinical and histopathological findings, it was considered to be originated from the large intestine.

Mucosubstance Histochemistry of the Epidermis in Yellowtail, Striped Beakperch, Brown Spotted Grouper, Sea Chub, and Multicolorfin Rainbowfish

  • To investigate the properties of mucosubstances of the epidermis in various teleostean species, conventional histochemical stainings were used on the skin in five species of order Perciformes, i. e., yellowtail, Seriota quinqueradiat, striped beakperch, Oplegnathus fasciatus, brown spotted grouper, Epinephelus chlorostigma, sea chub, Ditrema temmincki and multicolorfin rainbowfish, Halichoeres poecilopterus. The following methods were used: periodic acid Schiff (PAS), alcian blue (AB) pH at 2.5, AB pH at 1.0, AB pH at 2.5-PAS, AB pH at 1.0-PAS, aldehyde fuchsin (AF) pH at 1.7-AB pH at 2.5 and high iron diamine (HID)-AB pH at 2.5. The epidermis of all five species consisted of three layers: superficial, middle, and basal layer. The superficial layer was comprised of rather flattened cells. In particular, the outermost layer of striped beakperch and middle layer of sea chub consisted of mucus-secreting cells. Mucous cells, the unicellular glands, were found in epidermis but varied in number in different body regions and species. Although there was a slight difference in the amount in various species and body regions, the secretory contents of the mucous cells in the five teleostean species contained acidic mucopolysaccharides. In yellowtail, striped beakperch, and multicolorfin rainbowfish, the property of mucosubstances was identified as sialomucin, while it was sulphomucin in brown spotted grouper and sea chub.

  • The structure of skin was studied in Iksookimia longicorpus based on the micro-anatomical investigation of skin fragments taken from four regions. The epidermis was distinguished by two types of skin glands, a small mucous cell and a large club cell. The mucous cell was acid sulfomucins (some sialomueins) but the club cell did not give any histochemical tests for mucosubstances. The presence of a well defined lymphatic system with small lymphocytes was established in the stratum germinativum layer of the epidermis. A large number of blood capillaries run very close to each other just below the basement membrane, and a definite area giving AB and PAS positive was present between the basement membrane and scale.

Cytochalasin D Regulates Retinoic Acid Induced COX-2 Expression but not Dedifferentiation via p38kinase Pathway in Rabbit Articular Chondrocytes

  • Cytochalasin D (CD) is known as a disruptor of actin cytoskeleton architecture in chondrocytes. We have studied the role of CD in retinoic acid (RA) caused dedifferentiation and inflammation responses in rabbit articular chondrocytes. We have examined the effect of CD on RA induced dedifferentiation of chondrocytes. CD inhibited RA induced dedifferentiation determined by Western blot analysis and Alcian blue staining in rabbit articular chondrocytes. Also, CD additionally reduced inflammation response molecules such as cyclooxygenase-2 (COX-2) and prostaglandin $E_2$ ($PGE_2$) in RA treated cells. Treatment of CD reduced phosphorylation of p38 by treatment of RA. Inhibiton of p38kinase with SB203580 reduced expression of COX-2 and production of $PGE_2$ by treatment of CD in RA treated cells. But, Inhibiton of p38kinase with SB203580 did not any relationship with effect of CD on RA caused dedifferentiation. In summary, our results indicate that CD regulates RA reduced expression of COX-2 and production of PGE2 via p38kinase pathway.

  • The current study evaluates the capacity of eugenol to regulate immediate allergic reaction by control of histamine release. Administrations of eugenol (1M/kg, i.p.) at 60 min b efore and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) were shown the mortality rates as 0, 44.4, and 77.8%, respectively. A 60 min before administered group revealed a significant inhibition of serum histamine release compared with those of 5 and 10 min after the compound 48/80 injection. Eugenol (6-48mM) was also showed a dose-dependent activity on the compound 48/80-induced histamine release from the highly purified population of Alcian Blue-positive peritonea] mast cells. These results indicate that in vitro treatment with exogenous eugenol inhibited the active response of mast cell populations and modulated its characteristics.

Effects of Longterm Acupuncture on the Endocrine Cells and Mucus of Gastric Mucosa In Rats (족삼리 장기 자극이 흰쥐 위점막의 내분비세포 및 점액에 미치는 영향)

  • To investigate the effect of acupuncture at Zushanli (ST 36) in this study, gastric endocrine cells (G cell) by avidin-biotinylated complex (ABC) technique and histological examinations (HE; periodic acid schiff, PAS; alcian blue stain) of the stomach were perfomed at 1, 3, 6 weeks in normal rats. In other groups, omeprazole were fed for 1, 3, 6 weeks to compare with acupuncture effect. Acupuncture applied to the ST 36 acupoint and the administration of omeprazole increased G cell significantly at 1, 3, 6 weeks in time dependant manner. Furthermore, acupuncture applied to the other acupoint on GB 34 did not produce significant effect. When the common peronial nerve was dissected, acupuncture of ST 36 acupoint produced change of G cell. These data suggest that acupuncture at ST 36 increased G cell in point specific way and that effect was not related with surrounding nerve.
