Inhibitory effects of Tinospora cordifolia and Rubia cordifolia Linn. on egg albumin-induced experimental allergic conjunctivitis in rats

  • Allergic conjunctivitis is the most common allergic disease. These diseases are severe & frequent which requires search of new treatments. The aim of the study was to investigate the effects of Tinospora cordifolia (TC), Rubia cordifolia Linn. (RC) on experimentally induced allergic conjunctivitis in rats. In this study, dried water soluble extracts of TC and RC. (250 and 500 mg/kg, p.o. for 7 days) were evaluated for their antiallergic activity in Wistar rats. They were tested for inhibition of egg albumin-induced vascular permeability, inhibition of histamine release from the rat conjunctiva as well as in histamine content in tears. TC and RC showed significant (P < 0.05) inhibition in vascular permeability, inhibition in histamine release from the rat conjunctiva which is reflected by reduced level of histamine content in tears. The activities were found to be comparable to azelastine hydrochloride. These results suggest that the inhibitory effect on egg albumin-induced experimental allergic conjunctivitis in rat may be due to the antihistaminic activity of TC and RC. Our studies provide evidence that TC and RC may be beneficial in the treatment of allergic conjunctivitis.

Inhibitory Effects on the Type 1 Hypersensitivity and Inflammatory Reaction of Herba Patriniae Aqua-acupuncture (패장약침(敗醬藥鍼)이 면역반응(免疫反應)에 미치는 영향)

  • Objective : Experimental studies were done to research the clinical effects of Herba Patriniae(HP) aqua-acupuncture ($BL_{13},\;BL_{17},\;BL_{13}{\cdot}BL_{17}$ and free points) on the anti-allergic inflammatory response. Methods : We measured active systemic anaphylatic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. And we measured total IgE and plasma WBC level, serum total protein, albumin, immunoglobulin and NO levels induced by egg albumin. Results : HP aqua-acupuncture pretreatments at all acupoints inhibited active systemic anaphylatic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. Total IgE and plasma WBC level inhibited by HP aqua-acupuncture pretreatment at $BL_{13}\;BL_{17}$ and free points. However, HP aqua-acupuncture didn't effect serum total protein, albumin, immunoglobulin and NO levels. Conclusion : These results suggest that HP aqua-acupuncture may be beneficial in the regulation of type Ⅰ allergic reaction, but is further required immunological studies on the allergic reaction.

  • pH-sensitive P(MAA-co-PEGMA) hydrogel particles were prepared and their feasibility as smart delivery carriers for cosmetic ingredients was evaluated. P(MAA-co-PEGMA) hydrogel particles having an average size of approx. $2{\mu}m$ were synthesized via dispersion photopolymerization. There was a drastic change in the swelling ratio of P(MAA-co-PEGMA) particles at a pH of around 5 due to the ionization of MAA in the hydrogel and as the amount of MAA in the hydrogel increased, the swelling ratio increased at a pH above 5. The P(MAA-co-PEGMA) hydrogel particles showed a pH-sensitive release behavior. Thus, at pH 4 almost none of the albumin permeated through the skin while at pH 6 relatively high skin permeability was obtained. The albumin loaded in the P(MAA-co-PEGMA) hydrogel particles was hardly degraded in the presence of pepsin and its stability was maintained.

  • Purpose : Minimal Change Disease(MCD) is the most common primary nephrotic syndrome in children. Some suggested that tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) are involved in the pathogenesis of MCD. This study was done to see the changes of plasma and urinary $TNF-{\alpha}$, and their effects on the permeability of glomerular basement membrane. Methods : Study patients consisted of 19 biopsy-proven MCD children aged 2-15 years old. Both plasma and urinary $TNF-{\alpha}$ were measured. Employing the Millicell system, $TNF-{\alpha}$ were screened for the permeability factors. Results : Urinary $TNF-{\alpha}$ during relapse was significantly increased(P<0.01). No significant change was seen in the plasma $TNF-{\alpha}$ during relapse when compared to those in remission and the healthy controls. Furthermore, in the in vitro Millicell system, $TNF-{\alpha}$ did not produce a significant change in albumin permeability. Conclusion : Therefore, it seems that $TNF-{\alpha}$ may not play a disease-specific role in the pathogenesis of MCD.

  • Experimental studies were done to research the Clinical effects of Insambakhab-tang on the Anti-allergic effect and pulmonary function of O₃ intoxicated Mice. Anti-allergic effect experiment consisted of vascular permeability responses to intradermal histamine and serotonin, 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, and delayed type hypersensitivity responses to Picryl Chloride and SRBC. Pulmonary function of O₃ intoxicated Mice experimental consisted of pulmonary thromboembolism (Sodium Arachidonate-induced and ADP-induced), lung TBA value, and serum Na/sup +/, K/sup +/, Cl/sup +/ level. The results obtained as follows; 1. In the effects of Insambakhab-tang on the pulmonary thromboembolism by Sodium Arachidonic acid and ADP, Insambakhab-tang group revealed significant effect. 2. In the effects of Insambakhab-tang on the vascular permeability responses to intradermal histamine, Insambakhab-tang group revealed significant effect. 3. In the effects of Insambakhab-tang on the vascular permeability responses to intradermal serotonine, Insambakhab-tang group revealed significant effect. 4. In the 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, Insambakhab-tang group revealed significant effect. 5. In the delayed type hypersensitivity responses to Picryl Chloride, Insambakhab-tang group revealed significant effect. 6. Insambakhab-tang group revealed significant effect on decrease of the lung TBA value of lung. 7. In the effects of Insambakhab-tang on Serum Na/sup +/, K/sup +/ Level in O₃-intoxicated Mice. Insambakhab-tang group revealed none significant effect, but In the effects of Insambakhab-tang on Serum Cl/sup +/ Level in O₃-intoxicated Mice, Insambakhab-tang group revealed significant effect.

Liquid Crystal Based Optical Sensor for Imaging Trypsin Activity at Interfaces Between Aqueous Phases and Thermotropic Liquid Crystals

  • In this study, we developed a liquid crystal (LC)-based optical sensor for monitoring enzymatic activity through orientational changes in liquid crystals (LCs) coupled to the properties of a poly-${\small{L}}$-lysine (PLL)-based polymeric membrane. We prepared a PLL-based polymeric membrane at the planar interface between the thermotropic liquid crystal and aqueous phases. The PLL-based polymeric membrane was obtained by contacting the PLL solution with water immiscible LCs, 4-cyano-4'-pentyl-biphenyl (5CB) doped with adipoyl chloride. We then investigated the membrane properties by examining the permeability of the membrane to phospholipids, 1,2-didodecanoyl-rac-glycero-3-phosphocholine (DLPC). The permeability of the membrane to transport phospholipids was monitored through the orientational transition of 5CB in contact with the dispersions of DLPC. Since trypsin can enzymatically catalyze the hydrolysis of PLL, we incubated an aqueous trypsin solution with the membrane for 2 h at room temperature to cause an increase in the permeability of the polymeric membrane to DLPC. As a result, a bright to dark optical shift of LCs was observed, which implied that an enzymatic reaction between trypsin and PLL-based membrane occurred. Two control experiments using chymotrypsin and bovine serum albumin (BSA) revealed no sign of improved permeability based on the orientational transition of LCs.

In the effect of Hyunggaeyeungyotang on vascular permeability responses to intradermal serotonin in rats, Hyunggaeyeungyotang revealed significant effect. 3. In the 48hr homologous passive cutaneous anaphylaxis in rats provoked by the IgE-like antibody against egg albumin, Hyunggaeyeungyotang revealed significant effect. 4. In the effect of Hyunggaeyeungyotang on Delayed-type hypersensitivity responses to picryl chloride in mice, Hyunggaeyeungyotang revealed none of significant. 5. In the effect of Hyunggaeyeungyotang on Delayed-type hypersensitivity responses to SRBC in mice, Hyunggaeyeungyotang revealed none of significant. According to above-stated results, Hyunggaeyeungyotang is concluded to be effective as anti-allergic regimen and recommended to be used for treatment of allergic disease.

  • Experimental studies were done to research the clinical effects of Sopoongyangjetang on the Anti-allergic effect, Analgesic action and Anti-inflammatory action. The results obtained as follows ; 1. In effect of Sopoongyangjetang on vascular permeability responses to intradermal serotonin, it had tendency to decrease, but was none significant effect. 2. In effect of Sopoongyangjetang on vascular permeability responses to intradermal histamine, it revealed significant effect. 3. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Sopoonyangjetang showed significant effect. 4. In the delayed type hypersensitivity responses to picryl chloride, Sopoongyangjetang was proved significant effect. 5. In the delayed type hypersensitivity responses to SRBC, Sopoongyangjetang revealed significant effect. 6. In analgesic action by acetic acid method, Sopoongyangjetang was recognized significantly. 7. In Anti-inflammatory action by carrageenine method, Sopoongyangjetang showed significant effect.

  • Chitosan-alginate capsules were formed by the electrostatic interactions and had appropriate mechanical strength, permeability to albumin and stability to hepatocyte. Pig hepatocytes were isolated and immobilized in chitosan-alginate capsules. Encapsulation in 3 minutes and spheroid formation period of 24 hours were optimum condition for the high liver function. Pig hepatocytes density of $90.{\times}10^6$ cells/mL in capsules was suitable for the application to bioartificial liver support system.

We studied the vacular permeability response induced by histamine and serotonin, homologous PCA provoked by IgE-like antibody against EWA, contact dermatitis induced by picryl chloride and delayed type hypersensitivity response to SRBC. The results were obtained as follows: 1. In effects of the Leejungtang and Leejungtanggamibang on the vascular permeability responses to intradermal serotonin, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 2, In effects of the Leejungtang and Leejungtanggamibang on the vascular permeability responses to intradermal histamine, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 3. In the homologous PCA provoked by IgE-like antibody against white albumin, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 4. In the delayed type hypersensitivity responses to picryl chloride, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 5. In the delayed type hypersensitivity responses to sheep red blood cell the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect.
