  • In this experiment, etching was performed on the incisor enamel surfaces of 13 Albino rats, weighted about 130gm, with both 30% and 50% concentrations of the phosphoric, citric acids, and EDTA solution for 60 seconds. Immediately after extraction of the animal teeth, laboratory procedures for scanning electron microscopic observations were made and the author derived the following conclusions: 1. On the enamel surface treated with 30% and 50% phosphoric acids, all of the periphery, center of the enamel rods, and the enamel matrices were severely etched. 2. In the etched with 30% citric acid group, only the small part of the enamel rods were etched out. In case of 50% citric acid the periphery, center of enamel rods were partially or entirely etched, and the intermatrix spaces were widened. 3. In the 30% and 50% of the EDTA etched groups, the periphery of the enamel rods explicitely were etched, and the spaces between matrices were widened.

Protective effects of Hydro-alcoholic extract of the roots of Kasondi (Cassia occidentalis L.) against gentamicin-induced nephrotoxicity in albino Wistar rats

  • Objectives: Cassia occidentalis L. is a weed belonging to the Caesalpiniaceae family. The root of this medicinal plant is used for the treatment of various ailments, including kidney diseases. The present study was aimed at evaluating the nephroprotective effects of HAE of the roots of Cassia occidentalis L. against gentamicininduced renal toxicity in albino Wistar rats. Methods: The renal toxicity was induced by subcutaneous administration of gentamicin at 100 mg/kg in the rats belonging to the disease control and treatment groups from the 4th to the 8th day. The rats in the treatment group received HAE of the roots of Cassia occidentalis L. at 67 mg/kg b. w. orally for 8 days, while no treatment was given to the rats in the disease control and plain control groups. At the end of the experiment, renal biomarkers viz; s. creatinine, b. urea, and s. uric acid, were investigated. The histopathological examination of the kidney specimens was also carried out. Results: The results of the present study revealed that renal function biomarkers such as s. creatinine, b. urea, and s. uric acid were significantly reduced in the rats of the treatment group as compared to those of the disease control group. Moreover, the histoarchitecture reports of the treatment group's kidney specimens showed significant improvements. Conclusion: The results suggested that the HAE of Cassia occidentalis L. roots promisingly prevented kidney injury in gentamicin-induced nephrotoxic rats. This effect might be due to improved clearance of gentamicin from the renal tubule and decreased generation of reactive oxygen species (ROS).

모과(木瓜)가 장운동(腸運動)에 미치는 영향 연구 (Study of the Intestinal Motility Effects of Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus)

  • Objectives : This study was conducted to compare Chaenomelis Sinensis Fructus with Chaenomelis Lagenariae Fructus, and to examine Fructus chaenomelis having an influence on Intestinal Motility. Methods : We compared Chaenomelis Sinensis Fructus with Chaenomelis Lagenariae Fructus, by observing their effects on Intestinal Motility. Oral administration of water extracts of Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus into albino rats was followed by dealing with carbachol or loperamide, injecting charcoal meal and measuring the moving length in the intestine. Results : By Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus, the Intestinal Motility of normal albino rats did not change significantly. Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus controled the accelerated Intestinal Motility of albino rats. Chaenomelis Sinensis Fructus was not different from the extracting method, Chaenomelis Lagenariae Fructus had effects of control on extracting by methanol significantly. When Chaenomelis Sinensis Fructus was compared with Chaenomelis Lagenariae Fructus, the former was proved to have control effects on Intestinal Motility in decoction extracts and extracts by ethyl ether higher than the latter. By both Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus, the declined Intestinal Motility was not changed significantly. Conclusion : Chaenomelis Fructus suppressed the exasperated Intestinal Motility by carbachol, but did not influence the dropped Intestinal Motility by loperamide. In addition, Chaenomelis Sinensis Fructus was more excellent than Chaenomelis Lagenariae Fructus in its control effects of Intestinal Motility. inhibitory effects by DKT represent a potential therapeutic approach to the treatment of inflammatory diseases.

Bisphosphonate's effect on the tongue in adult male albino rats and the possible protective role of rutin: light and scanning electron microscopic study

  • Alendronate sodium (ALS) is a nitrogen-containing bisphosphonate used for the treatment of different bone disorders. However, its adverse effect on oral soft tissue has been detected. Rutin (RUT) is natural flavonoid with antioxidant and anti-inflammatory properties. This work aimed to investigate the possible effect of ALS on the tongue of adult male albino rats and to evaluate the possible protective role of RUT. Forty adult male albino rats were equally divided into four groups: group I (control), group II (RUT): Received RUT 50 mg/kg, group III (ALS): Received ALS 1 mg/kg, group IV (ALS+RUT):(ALS+RUT): Received ALS and RUT with the same doses as pervious groups. The drugs were given once daily for 5 weeks. Tongue specimens were taken and processed for light and scanning electron microscopic inspection. ALS treated group revealed structural changes in the tongue in the form of decrease in the height of the filiform papillae with blunt ends, marked atrophy in some papillae with areas of focal loss, loss of some epithelial cells, pyknotic nuclei and cytoplasmic vacuoles in some epithelial cells. The lamina propria showed inflammatory cellular infiltration with congested blood vessels. Statistically, there were highly significant decrease in the number of proliferating cell nuclear antigen immunopositive cells, area percentage of Bcl-2 immunoexpression and highly significant increase in the collagen content compared to control group. Administration of RUT with ALS minimizes these changes. RUT protected the rat tongue against the histological and immunohistochemical changes induced by ALS through its antioxidant and anti-inflammatory properties.

Effects of solanum macrocarpon (African eggplant) on haematological parameters of wistar rats exposed to urban air pollution

  • Our study investigated the effects of Solanum macrocarpon (African eggplant) on haematological and biochemical parameters of male albino rats exposed to urban air pollution ($O_3$, $PM_{10}$, CO, $SO_2$, $NO_2$ and VOC). Male albino Wistar rats were exposed for 63 days either to urban air pollution without treatment (A); oral supplementation with Solanum macrocarpon given at 36 and 75 mg/kg body weight (BW), representing C and D respectively and exposed to air pollution; oral supplementation with Solanum macrocarpon given at 75 mg/kg BW after exposure to air pollution (B); or kept in animal house without exposure to air pollution (E). Animals exposed to air pollution showed significant alterations in haematological and biochemical parameters signaling that the blood and organs were badly injured. There were significant elevations in white blood cells (WBC) and its indices, reduction in red blood cells (RBC) and significant depletions of non-enzymic antioxidants, total protein and increase in lipid peroxidation values. Solanum macrocarpon supplementation in the feed of animals halted significantly the deleterious effects of air pollution, with co-administration during exposure given better results.

Tetracycline이 백서치질형성기(白鼠齒質形成期)에 미치는 영향(影響)에 관(關)한 실험적(實驗的) 연구(硏究) (THE EFFECT OF TETRACYCLINE ON DENTINOGENESIS OF DEVELOPING ALBINO RATS)

  • The study is about the effect of tetracycline-HCl on the amelogenesis and the dentinogenesis of the albino rats by means of histochemistry and fluorescence microscopy. Females in oestrus were mated overnight and examined the next morning for evidence of copulation. The mothers were intraperitonealy injected with a single dose of tetracycline-HCl from the eighth to tenth day of gestation. The heads of new born rats were fixed in Carnoy's solution and 10% formalin solution. The staining methods were alizarin red S stain, PAS reaction, colloidal iron reaction, Morin's stain and hematoxylin-eosin stain, The results were as follows: 1. By the single injection of tetracycline, the matrix formation of enamel and dentin were disturbed, and the shape and arrangement of ameloblast and odontoblast were distorted. 2. It seemed that, with the higher dose of tetracycline, the positive materials of PAS reaction were increased in the disturbed enamel and dentin matrix, but those of alizarin red S stain and colloidal iron reaction were decreased. 3. The fluorescence intensity in the disturbed enamel and dentin matrix were higher than the other areas and appeared to increase gradually with the higher dose of tetracycline.

카리니주폐포자충 장염에 대한 여러 포유동물 및 흰쥐 계통에 따른 감수성 (Susceptibility of various animals to Pneumocystis carinii infection)

  • 국내에서 카리니주폐포자충(Pneumocystis carinii, 이하 Pc) 연구에 적합한 동물모델을 개발하고자, 총 7종류 의 실험동물(흰쥐, 기니픽, 햄스터, 개, 고양이, 토끼 및 돼지)에 methyl prednisolone(Depomedrol", Korea Upiohn Ltd.)을 주사하여 면역억제를 유발하고 폐를 슬라이드 글라스에 문질러 도말표본을 만들었다. 도말표본을 Diff Quik 염색 후 광학현미경으로 관찰하여 Pc의 감염전도를 음성 (0), 0.5+, 1+, 2+ 및 3+의 5등급으로 구분하였다. 실험동물 중 흰쥐에서만 Pc폐염이 유발되고 다른 동물에서는 Pc를 관찰하지 못하였다. 대부분 의 동물은 전신쇠약의 증세를 보였으며, 대체로 면역억제 6주경부터 사망하기 시작하였다. 흰쥐의 경우 실험한 세 계통 모두에서 감염을 확인하였다. Sprague-Dawley 흰쥐의 경우 총 136마리 중 86마리63.2%)가 Pc 감염 되었고, Fisher 휜쥐는 76마리 중 63마리 (82.9%)에서, Wistar 흰쥐는 106마리 중 77마리 (72.6%)에서 Pc 감염 이 양성이었다. 감염정도와 시기에서도 차이가 있어서 Wistar 흰쥐에서 더 이른 시기에 높은 감염정도로 관찰되었다. 이상의 결과에 의하면 Pc 병원체를 얻기에는 흰쥐 세 계통 중 Wistar 흰쥐가 Pc연구에 가장 적합한 실험동물이고 그외의 실험동물은 Pc 감염에 적합하지 않음을 확인하였다.

A comparative study on the hepatoprotective effect of selenium-nanoparticles and dates flesh extract on carbon tetrachloride induced liver damage in albino rats

  • Exposure to environmental pollutants such as carbon tetrachloride (CCL4) causes liver damage. This study aimed to compare the ameliorative activity of the dates flesh extract (DFE) and selenium-nanoparticles (SeNPs) on CCL4-induced hepatotoxicity and if DFE could be a useful alternative supplement. Twenty-four male albino rats were enrolled and randomly divided into four equal groups (6 rats in each group): control group received only basal diet with no medications. Group II received CCL4 in a dose of 0.5 mg/kg intraperitoneal injection twice weekly for four weeks. Group III rats were pretreated with SeNPs in a dose of 2.5 mg/kg once a day orally three times/wk for four weeks alone then combined with the previously described dose of CCL4 for another four weeks. Group IV rats were pretreated with DFE in a dose of 8 ml of the aqueous extract/kg/d orally for four weeks alone then combined with the previously described dose of CCL4 for another four weeks. The liver damage was assessed by estimation of plasma concentration of albumin and enzymes activities of alanine aminotransferase and tissue genes expression. Liver oxidation levels were assessed by measuring the tissue concentration of the malondialdehyde, superoxide dismutase, and the total glutathione. Additionally, inflammatory mediators tumour necrosis factor--α and interleukin-6 were estimated. Detecting the liver's cellular structural damage was done by histopathological and immunohistochemical examination. This study suggests that CCL4-induced liver damage in rats can be protected by administration whether the costly SeNPs or the economical DFE.
