Effect of Alpha Lipoic Acid on in vitro Maturation of Porcine Oocytes and Subsequent Embryonic Development after Parthenogenetic Activation

  • Alpha lipoic acid (ALA) is a biological membranes compound. As the antioxidant, it decreases the oxidized forms of other antioxidant substances such as vitamin C, vitamin E, and glutathione (GSH). To examine the effect of ALA on the in vitro maturation (IVM) of porcine oocytes, we investigated intracellular GSH and reactive oxygen species (ROS) levels, and subsequent embryonic development after parthenogenetic activation (PA). Intracellular GSH levels in oocytes treated with 50uM ALA increased significantly (P < 0.05) and exhibited a significant (P < 0.05) decrease in intracellular ROS levels compared with the control group. Oocytes matured with 50 uM of ALA during IVM displayed significantly higher cleavage rates (67.8% vs. 83.4%, respectively), and higher blastocyst formation rates and total cell number of blastocysts after PA (31.6%, 58.49 vs. 46.8%, 68.58, respectively) than the control group. In conclusion, these results suggest that treatment with ALA during IVM improves the cytoplasmic maturation of porcine oocytes by increasing the intracellular GSH levels, thereby decreasing the intracellular ROS levels and subsequent embryonic developmental potential of PA.

  • Inhibition effects of alanine (Ala), histidine (His), methionine (Met) on the corrosion of aluminum were investigated in deaerated 0.5 M HCl and NaCl solution. In HCl solution the inhibition efficiency for the aluminum corrosion depended on the cathodic inhibition, and the inhibition efficiency was increased in the order of Met$10^{-4\;}M$ the adsorption process can be explained by Langmuir isotherm, however, in the case of higher concentration by Temkin logarithmic isotherm due to the interaction between the adsorbed molecules.

  • This preliminary study examined the status of data science-related course syllabi in the American Library Association (ALA) accredited Library and Information Science (LIS) programs. The purpose of this study was to explore LIS course syllabi related to data science, such as course title, course description, learning outcomes, and weekly topics. LIS programs offer various topics in data science such as the introduction to data science, data mining, database, data analysis, data visualization, data curation and management, machine learning, metadata, and computer programming. This study contributes to helping instructors develop or revise course materials to improve course competencies related to data science in the ALA-accredited LIS programs.

  • Photodynamic therapy is one of the ways to treat cancer using light and during laser irradiation, photosensitizers react and combine with oxygen to destroy cancer cells. This treatment is in the spotlight as a treatment that minimizes side effects in cancer patients. Among them, photosensitizers differ in the treatment area, treatment effect, and degree of absorption depending on the type. Therefore, in this study, a quantitative evaluation study was conducted on the effect of inhibiting cancer cell proliferation by irradiating blue LEDs on HELA cell lines injected with 5-ALA among photosensitizers.

Identification of a Potential Gene for Elevation ω-3 Concentration and its Efficiency for Improving ω-6/ω-3 Ratio in Soybean

  • This present study was to identify a novel candidate gene that contribute to the elevated α-linolenic acid (ALA, ω-3) concentration in PE2166 from mutagenesis of Pungsannamul. Major loci qALA5_1 and qALA5_2 were detected on chromosome 5 of soybean through quantitative trait loci mapping analyses of recombinant inbred lines. With next generation sequencing of parental lines and Pungsannamul, and recombinant analyses, a potential gene, Glyma. 05g221500 (HD) controlling elevated ALA concentration was identified. HD is a homeodomain-like transcriptional regulator that may regulate the expression level of microsomal ω-3 fatty acid desaturase (FAD3) genes responsible for the conversion of linoleic acid into ALA in the fatty acid biosynthetic pathway. In addition, we hypothesized that combination of mutant alleles, HD and either of microsomal delta-12 fatty acid desaturase 2-1 (FAD2-1\ could reduce the ω-6/ω-3 ratio. In populations where HD, and FAD2-1A and FAD2-1B genes were segregated, combination of a hd allele from PE2166 and either of the variant FAD2-1 alleles were sufficient to reduce the ω-6/ω-3 ratio in seeds.

  • 펩타이드 약물의 일종인 LHRH 및 그의 analogue인 〔D-Ala$^{6}$)LHRH의 경점막 수송경로를 개발하기 위하여 이들 약물을 점막 균질액과 배양시 점막에 존재하는 효소에 의해 분해되는 것을 발견하였다. 이에 저자등은 LHRH 및 〔D-Ala$^{6}$〕 LHRH의 점막 균질액 중에서의 분해산물을 HPLC에 의해 정량하였다. 또한 PITC법에 따라 아미노산 분석을 실시하여 이들 분해산물의 아미노산 조성을 밝히고 아울러 분해위치를 규명하였다. 즉 LHRH또는 〔D-Ala$^{6}$〕 LHRH를 직장, 비강 및 질점막과 일정시간 배양 후 시료를 HPLC에 주입하여 각 분해산물의 peak time에 용리액을 분획 수집하였다. 이것을 6N-HCl로 가수분해하여 유리아미노산으로 만들고 phenylisothiocyanate 유도체화 하여 PTC 아미노산으로 한후 아미노산 분석용 컬럼을 사용하여 HPLC로 정량 하였다.

  • Aminolevulinic acid(ALA) was shown to be synthesized via active pathways of either C4 or C5 ALA biosynthesis in cells of a photosynthetic bacterium, Rhodocyclus gelatinosus KUP-74, where the C5 pathway was appeared to be preferntially expressed in the cells. It was strongly suggested that L-glutamine might be utilized more effectively than L-glutamate to synthesize ALA via C5 pathway in this bacterium from the fact of relationship between the cellular uptake rates of glutamate and its Gamma-derivaties and corresponded ALA productivities in vitro and in vivo.

Overproduction and Operator DNA-Protein Blotting of R100 Mutant MerR from Shigella flexneri

  • Wild-type and four mutant R100 merR genes were cloned and the proteins overproduced under tac promoter control of pKK223-3. His118Ala, Cys117Ser, Cys126Ser, and wild-type MerR were successfully overproduced although amino-terminal 14 amino acids deletion mutant MerR was not successful. The amount of overproduced wild-type MerR protein as well as other mutant MerR was between 15%-20% of the total protein. The protein was able to be purified up to 95% homogeneity. Specific DNA-protein blotting experiments showed that the 95 bp operator containing DNA fragment could bind to Cys126Ser, His118Ala, and wild- type MerR, but not to Cys117Ser. These results were consistent with the previously reported complementation experiment results that His118Ala, Cys126Ser, and wild-type MerR could repress the mer operon but Cys117Ser could not.
