Inhibitory Effects of Plant Extracts on Adjuvant-induced Arthritis

  • Twenty seven plant extracts were selected on the basis of ancient literature search for rheurnatoid arthritis or similar syndrome. Methanol extract of each plant was prepared and administered orally to rats everyday at a dose of 200 mg/kg/day. Experimental arthritis was induced by subplantar injection of heat-killed Mycobacterium butyricum to right hind paw of rats. This treatment provoked swelling of the treated paw in two phases, acute primary swelling and secondary arthritic swelling. An inhibition of secondary swelling was considered to be antiarthritic activity. Several plant methanol extracts such as Akebia quinata (caulis), Ephedla sinica (herba) and 5ophorae subprostratd (radix) were found to show significant inhibitory activity against secondary swelling at the dose tested. Our results strongly suggested an antiarthritic potential of these plant extracts.

Effects of Akebia quinata extract on alcohol-induced damage of liver, spleen and thymus in rats (목통(木通)이 알코올 투여로 유발된 흰쥐의 간, 비장 및 가슴샘의 손상에 미치는 영향)

  • Objective : This study was conducted to investigate the effects of Effects of Akebia quinata (AQ) extract on alcohol-induced damage of liver, spleen and thymus in rats. Method : Experimental animals were divided in to 4 groups; Normal group, Alcohol group, AQ50 group and AQ200 group. All rats, except for Normal group, were fed 25 % ethanol for 55 days. During experimental period, Normal group and Alcohol group were administrated saline, and AQ50 group and AQ200 group were administrated AQ extract at dose of 50 and 200 mg/kg/day, respectively. We measured organ weight, liver triglyceride contents, and alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and triglycerid levels in serum. Also, we conducted histomorphometry and histopathological observation of liver, spleen and thymus. Results : AQ significantly decreased the level of AST, ALT and triglyceride in serum and the liver triglyceride contents induced by ethanol. Also, AQ significantly inhibited lipid droplets accumulation in hepatocytes. The decreased relative organ weight of spleen and thymus by ethanol were increased by AQ administration. In histopathological analysis of spleen, the rats administrated AQ 200 mg/kg presented significantly increased mean diameters of white pulps, numbers of white pulps and splenic thicknesses. The administration of 200 mg/kg AQ improved decreased lobular thickness and cortex thickness of thymus, which were decreased by ethanol. Conclusions : The results of present study indicated that AQ has an ameliorating effect for fatty degeneration of liver and damage of spleen and thymus.

Anti-obesity Effects of Extracts from Young Akebia quinata D. Leaves (어린 으름잎 추출물(Akebia quinata D. Leaves)의 항비만 효과)

  • We investigated the in vitro and in vivo anti-obesity effects of extracts from young Akebia quinata D. leaves, including hot water (AQH) and 80% ethanol (AQE) extracts. The inhibitory effects of AQH and AQE on lipid accumulation in 3T3-L1 cells were examined by Oil Red O staining. Compared to control, lipid accumulation was significantly reduced by 18.3% with the treatment upon AQE at a concentration of $5{\mu}g/mL$. The levels of intracellular triglycerides and free glycerol were also reduced by 52.8% and 9.1% at the same concentration of AQE. The in vivo anti-obesity effect of AQE was evaluated in terms of body and white adipose tissue weights in ICR mice. Experimental groups were divided into the following five groups: normal diet (ND), high fat diet (HFD), high fat diet with 60 mg/kg/day of Orlistat (HFD-RF), high fat diet with 200 mg/kg/day of AQE (HFD-AL), and high fat diet with 600 mg/kg/day of AQE (HFD-AH). Feeding of HFD for eight weeks resulted in significant increases in body weight as well as weight gain compared to the ND group. HFD-AH group showed reduced body weight, weight gain, epididymal white adipose tissue weight, and perirenal white adipose weight as compared to the HFD group. These results indicate that AQE supplementation might have beneficial effects on anti-obesity by inhibiting lipid accumulation.

Effect of Melandrii Herba, Akebia Quinata Decaisne, and Tetrapanax Papyriferus on Milk Secretion and Lactation Related Factors in Postpartum Mice (왕불유행, 목통, 통초가 산후 생쥐의 유즙분비량과 유즙분비 관련 인자에 미치는 영향)

  • Objectives: The purpose of this study is to investigate the effect of Melandrii Herba (MH), Akebia Quinata Decaisne (AQ), and Tetrapanax Papyriferus (TP) on milk secretion and aquaporin (AQP) expression in lactating mice. Methods: For the experiment, the mice were divided into three groups, which were orally administered MH (2,720 mg/kg), TP (400 mg/kg) and AQ (2,800 mg/kg) extracts respectively for 3 weeks from Day 1 after the birth, compared with the control group (C group), which was administered distilled water. A group consisted of six infantile mice per postpartum mouse. For comparison with the C group, non-pregnant SKH-1 mice were used as the virgin group. Results: 1. When it comes to the immunohistochemical staining for prolactin receptors in the mammary glands, the AQ and MH groups showed a strong immune response to the secretory epithelial cells constituting the mammary alveoli, while the TP group represented a weaker immune response. 2. In the immunohistochemical staining for AQP in the mammary glands, AQP1 showed a strong immune response in the walls of capillaries and venules around the mammary alveoli, and AQP3 in the epithelial cells constituting the mammary alveoli, and AQP5 in some tissues between the mammary alveoli. AQP1 was expressed in the order of TP group>AQ group=C group>MH group, and AQP3 was MH group and AQ group>TP group=C group, and AQP5 was MH group>C group>AQ group and TP group. 3. In the Western blot, AQP1 was expressed in the order of TP group>AQ group>C group>MH group, and AQP3 was MH group>AQ group>C group>TP roup, and AQP5 was MH group>TP Group>C group>AQ group. All of AQP1, 3, 5 expression were significantly higher in the C group than in the Virgin group. Conclusions: The administration of Akebia Quinata Decaisne, Tetrapanax Papyriferus and Melandrii Herba have the effect of improving prolactin levels in postpartum mice and increasing the expression of prolactin receptor and AQPs in the mammary glands, suggesting that lactation might be enhanced by the development of the mammary glands.

Composition of Phenolic Acids and Flavonoids and Skin Care Cosmetic Antioxidant Activity of Akebia quinata Fruit Extracts (연복자 추출물의 Phenolic acids와 Flavonoids의 조성 및 화장품 항산화 활성)

  • In order to verify the effect of using functional cosmetic ingredients for skin beauty, the composition and content of some phenolic acid and flavonoids in the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds were analyzed, and the skin care antioxidant activity was investigated. The results are as follows. Total polyphenol and flavonoid contents were found to be higher in pericarp extract than seed extract in both hot water and 80% methanol extract. In the analysis of the composition and content of phenolic acid and flavonoids, two kinds of phenolic acids were identified in the hot water extract of pericarp, 6 kinds of phenolic acids were identified in the 80% methanol extract, and one kind of flavonoid. Two types of phenolic acids were identified in the hot water and 80% methanol extract of seeds, respectively. DPPH and ABTS radical scavenging activities tended to increase in proportion to the treatment concentration in both hot water and 80% methanol extracts, and antioxidant activity was found to be high. Therefore, from the above results, it was found that the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds contained various kinds of phenolic acid and flavonoids, and the antioxidant activity was also high. It is believed to be of value as a natural antioxidant in skin care cosmetic ingredients.

Quality Characteristics and Antioxidant Activity of Vinegar Supplemented Added with Akebia quinata Fruit during Fermentation (으름 열매 식초의 품질특성 및 항산화 활성)

  • This study investigated the physicochemical properties and antioxidant activity of vinegar added with different levels (0%, 1%, 3%, 5%, and 7%) of Akebia quinata fruit during two-step fermentation. The physicochemical properties of vinegar evaluated were pH, total acidity, alcohol, and total sugar and amino acid contents. The antioxidant activities were based on ABTS radical scavenging activity, SOD-like activity, and reducing power. During alcohol fermentation, total acidity and alcohol contents of vinegar increased, but total sugar contents decreased. During acid fermentation, total acidities of vinegar increased. Vinegar added with 7% A. quinata fruit showed the highest total sensory score. Total polyphenol contents of vinegar added with 0% and 1% A. quinata fruit were not significantly different. However, vinegar added with 3, 5, and 7% A. quinata fruit showed significantly higher total polyphenol contents of 136.6, 381.59, and 415.35 mg/100 g, respectively, after 13 days of fermentation. Further, total flavonoid contents of vinegar added with 0~7% A. quinata fruit significantly increased to 21.73, 15.79, 15.15, 26.19, and 26.87 mg/100 g, respectively, after 13 days of fermentation. In addition, tannin contents of vinegar added with 0~7% A. quinata fruit significantly increased to 0.2042, 0.2004, 0.1255, 0.1384, and 0.1255 mg/100 g, respectively, after 13 days of fermentation. Moreover, ABTS radical scavenging activities of vinegar added with 0~7% A. quinata fruit significantly increased to 5.87, 12.59, 25.63, 34.02, and 35.25, respectively, after 13 days of fermentation at a concentration of 5 mg/mL. Additionally, SOD-like activities of vinegar added with 0~7% A. quinata fruit significantly increased to 8.22, 17.49, 16.86, 16.89, and 15.68%, respectively, after 13 days of fermentation. Reducing power of 7% A. quinata fruit was 0.527 after 1 day and 1.539 at the end of fermentation. Our results demonstrate that antioxidant activity significantly increased during fermentation according to the content of A. quinata. Further, the total polyphenol, flavonoid, and tannin contents were shown to be closely related with antioxidant activities. Thus, A. quinata could be effectively used as a vinegar and functional food material based on its antioxidant activity.

Quality Characteristics and Biological Activities of Vinegars Added with Young Leaves of Akebia quinata (으름 어린잎 식초의 품질특성과 생리활성)

  • We investigated the characteristics and biological activities of vinegars added with different levels (0%, 0.5%, 1%, 2%, and 3%) of young leaves of Akebia quinata. During alcohol fermentation, alcohol and total acidity contents of vinegars increased. During acid fermentation, total acidity and amino acid contents increased. Vinegar added with 3% A. quinata leaf showed the highest total sensory score. The contents of total polyphenols, flavonoids, and tannin significantly increased during fermentation according to the amount of A. quinata leaf. After 22 days of fermentation, total polyphenol, total flavonoid, and tannin contents of vinegar added with 3% A. quinata were 4,079.08 mg GAE/100 g, 2,927.08 mg CE/100 g, and 3,618.00 mg TAE/100 g, respectively. ABTS radical scavenging activity of vinegar added with 3% A. quinata was 79.63%. Anti-cancer activities of vinegar added with 3% A. quinata were 48.65% and 52.90% against MCF-7 and HepG2 cells, respectively. Vinegar added with 3% A. quinata showed anti-bacterial activities against Bacillus cereus, Shigella flexneri, Salmonella enterica, Bacillus subtilis, and Klebsiella pneumoniae. Our results demonstrate that the biological activities of vinegar added with 3% A. quinata leaf (22 days of fermentation) were excellent, and their enhanced total polyphenol, flavonoid, and tannin contents were associated with antioxidant, anti-cancer and anti-microbial activities. Thus, A. quinata can be used as a functional material in vinegar and other foods.

Quality Characteristics of Muffins containing Akebia quinata Leaves Powder (으름잎 분말을 첨가한 머핀의 품질특성)

  • This study investigates the quality characteristics of muffins prepared with various concentrations of akebia leaves powder. Muffins are prepared by additions of 0%, 2%, 4%, 6% and 8% powder to the flour for basic formulation. Heights decrease significantly when the amount of Akebia leaves powder increases. When the weight is being measured, there are no meaningful differences between the samples. As a result from measuring the colors, the lightness, redness and yellowness of muffins decrease as the concentrations of the powder increase. When measuring the texture of akebia leaves muffins, the hardness, cohesiveness, gumminess and chewiness decrease, but the springiness and adhesiveness increase as the concentrations of Akebia leaves powder increase. When the sensual qualities of the muffins are measured, the color, sweetness, softness and overall quality of all of the samples have shown significant differences, and the muffins containing 4% of Akebia leaves powder reveal the highest sensual quality indexes.

  • Several kinds of medicinal plants are used as diuretics in folk medicines and oriental drugs. The diuretic action of water extracts of the ten kinds of crude drugs, such as Pachyma hoelen $R_{UMPHIUS}$ Zea mays L., Akebia quinata $D_{ECAISNE}$, Alisma orientale Juzepezuk, Atractylodes koreana $N_{AKAI}$, phytolacca americana L., Achyranthes japo-nica $N_{AKAI}$, Juncus decipiens $N_{AKAI}$, Prunella asiatica $N_{AKAI}$ and Artemisia capillaris $T_{HUNBERG}$ was examined in mouse and compared with aminophylline as a control the following results were obtained. The urine volume was found to be remarkably increased by the Pachyma hoelen RUMPHIUS, Phytolacca americana L., Prunella asiatica $N_{AKAI}$ and Artemisia capillaris $T_{HUNBERG}$, moderately increased by the Akebia quinata $D_{ECAISNE}$, Achyranthes japonica $N_{AKAI}$ and Juncus decipiens $N_{AKAI}$, and slightly increased by Zea mays $L_{ENNE}$, Alisma orientale $J_{UZFPEZUK}$ and Atractylodes koreana $N_{AKAI}$.
