Traditional culture technology of medicinal plants mainly depends on the field culture, which has many problems. With progress of modern culture technology, it has become possible to produce valuable secondary metabolites from medicinal plants. In this paper, we discuss about the pigment and saikosaponin production from too medicinal plants, Carthamus tinctorius and Bupleurum falcatum, through bioreactor culture system. A two-stage bioreactor culture system was established for the production of yellow and red pigments and saikosaponins by cell suspension cultures of Carthamus tinctorius and Bupleurum falcatum. In Carthamus tinctorius, balloon type airlift bioreactors and column type airlift bioreactors were employed for the tell culture and for the pigment production, respectively. The greatest pigment production was obtained on White medium supplemented with 4 mg/L kinetin, high levels of sucrose concentration and photosynthetic photon flux. In Bupleurum falcatum, adventitious roots were cultured in balloon type airlift bioreactors and the root growth was greatest on SH medium containing 5 mg/L IBA and 0.2 mg/L kinetin. HPLC analysis showed that the contents of main active saikosaponins a, c, and d in adventitious roots were almost the same as those in field cultured root.
A bench-scale submerged-type membrane filtration system (SMFS) was constructed to study a feasibility of membrane filtration for solid-liquid separation in water and wastewater treatment processes. In the case of applying the SMFS to a biological wastewater treatment process, so-called membrane bioreactor, aeration underneath membrane modules is usually employed in order to provide oxygen demand for microbial growth as well as to control membrane fouling. A study was investigated the effects of operation parameters by aeration intensity, feed concentration, foulant type and airlift pore size on critical flux. Critical flux tends to increase with aeration rate. Optimal aeration flow rate was found to be 10 L/min/module. Feed concentration and foulant type has a significant effect on membrane fouling and filtration performance. But downward position and pore size of airlift has no a significant effects on membrane fouling and filtration performance.
For the optimization of operating conditions for plant cell suspension culture in bioreactors, effects of bioreactor types, various kinds of impellers, and aeration rates were examined using Nicotiana tabacum cells as a model system. Stirred tank bioreactor and airlift bioreactor were used for the comparison of bioreactor type. Growth rates in both bioreactors were lower than in shake flasks. In terms of final cell concentration, stirred tank bioreactor supported a little bit better growth compared to airlift bioreactor. Impeller type did not affect cell growth significantly, but it was apparent that cell size index decreased in the case of using hollowed paddle impeller. When the aeration rate was maintained at 0.3 vvm, cell growth was the best. At above 1.0 vvm, growth inhibition as well a browning was noticed. In addition, it was found that cell size index reduced proportionally to the increased of aeration rate.
A series of studies were carried out to establish micropropagation system, using airlift bioreactors (ebb $\varepsilon$ flood type, 5 L), of Lilium oriental hybrid 'Casa Blanca'. The bulblets with swollen basal plate were formed from bulb scales, then proliferated to bulblet clusters with swollen basal plate. Finally normal bulblets were formed from the sections. Bulblet formation and proliferation with swollen basal plate were not accomplished entirely in liquid culture of 5 L airlift bioreactors, but leafy bulb scales grew vigorously. Bulblet clusters with swollen basal plate were proliferated by periodic immersion culture. Bulblet proliferation was not affected by light, but scale leaves grew under light. MS medium containing 2.0 mg/L benzyl adenine (BA) and 0.3 mg/L indole acetic acid (IAA) was favorable to the bulblet proliferation with swollen basal plate. In liquid culture of 5 L bioreactors, bulblets from bulblet sections with swollen basal plate grew vigorously on MS medium with 70 g/L sucrose. It was effective for bulblet growth to replace the new medium after 8 weeks in culture during 16 weeks of cultural period. 15 g injection of bulblet sections as a cultural material was suitable for bulblet growth in 5 L bioreactors.
Suspension culture of Schisandra chinensis for production of gomisin J was perfomed in bioreactor. The inoculum size and initial sucrose concentration had significant effect on the cell growth and gomisin J accumulation. The maximum dry cell weight $(DCW;\;43.5\;g/{\ell})$ and gomisin J content $(0.71\;{\times}\;10^{-3}\;{\mu}g/g\;DCW)$ were obtained at inoculum size of 100 g fresh cell weight (FCW) per liter and MB5 medium containing 6% sucrose after 8 weeks of culture. The effect of oxygen supply on the cell growth and gomisin J accumulation was also investigated in an airlift-type bioreactor. The optimal cell growth and gomisin J content was obtained under 0.5 vvm. The productivity of gomisin J was 0.7 fold in bioreactor culture lower than that obtained in a flask cultivation.

For the purpose of establishing an efficient propagation through airlift bioreactor system of Zingiber of officinale Rosc. Korean native Seosanjong, the effect of different factors and bioreactor on cultured plantlets were investigated. The highest number of plantlets, fresh weight per plant was obtained from explants when cultured in MS liquid medium including 0.3 mg/L NAA and 2.0 mg/L kinetin for 40 days. A 10 L bottle type bubble bioreactor, compared with 250 mL Erlenmeyer flask, was more efficient, producing 4.7 plantlets or from 1.5 to 1.6 times more than did the Erlenmeyer flask. The results demonstrate the rapid mass propagation of airlift bioreactor to produce normal ginger.
Airlift pumps were tested to evaluate their pumping and aeration capacities. The pumps were 34.5 inch long made of 2, 3, 4 and 6 inch nominal diameter PVC pipes. An one hp air blower was used to supply the air. The air-flow rate was measured by an anemometer type air-flow meter and air pressure was level changes in a water tank from which water was pumped. Aeration by the pumps was tested by the standard aeration test method with the center of pump outlet positioned 3 inches above water surface. Oxygen concentrations in water were measured to determine aeration rate. As pumping head increased by water level draw-down in the tank water flow decreased while air flow increased. The reduction rate of water flow was higher with 4 and 6-inch pumps. Small pumps showed very minor changes in the reduction. Aeration rates were similar among 3, 4, and 6 inch pumps. With one hp air blower 6-inch pump at the minimum pumping head achieved the best performance in terms of water circulation.
A suitable culture method and bioreactor type for itaconic acid production were chosen by comparing the maximal concentration of itaconic acid produced in various systems. In batch culture, the maximal concentration of itaconic acid produced in a bubble column reactor was about 5% greater than that produced in stirred-tank or external-loop airlift reactor. These results were thought to be due to lower shear force and higher mass transfer efficiency in a bubble column reactor in comparison with other reactors. Moreover, the fed-batch mode in a bubble column was found to be a suitable one, producing about 25% higher concentration of itaconic acid compared to batch mode.
To optimize the expression and secretion of ferritin protein associated with ion storage in the mushroom, Pleurotus eryngii, a recombinant secretion vector, harboring the ferritin gene, was constructed using a pPEVPR1b vector under the control of the CaMV 35S promoter and signal sequence of pathogen related protein (PR1b). The ferritin gene was isolated from the T-Fer vector following digestion with EcoRI and HindIII. The gene was then introduced into the pPEVPR1b secretion vector, and it was then named pPEVPR1b-Fer. The recombinant vector was transferred into P. eryngii via Agrobacterium tumefaciens-mediated transformation. The transformants were selected on MCM medium supplemented with kanamycin and its expression was confirmed by SDS-PAGE and western blotting. Expression of ferritin protein was optimized by modifying the culture conditions such as incubation time and temperature in batch and 20 L airlift type fermenter. The optimal conditions for ferritin production were achieved at 25℃ and after incubating for 8 days on MCM medium. The amount of ferritin protein was 2.4 mg/g mycelia, as measured by a quantitative protein assay. However, the signal sequence of PR1b (32 amino acids) seems to be correctly processed by peptidase and ferritin protein may be targeted in the apoplast region of mycelia, and it might not be secreted in the culture medium. The iron binding activity was confirmed by Perls' staining in a 7.5% non-denaturing gel, indicating that the multimeric ferritin (composed of 24 subunits) was formed in P. eryngii mycelia. Mycelium powder containing ferritin was tested as a feed additive in broilers. The addition of ferritin powder stimulated the growth of young broilers and improved their feed efficiency and production index.

Jar fermentor and Air-sparging fermentor performed high productivity In laccase activity by F. fraxinea. Laccase activity reached 3,540 in 8 days (Jar fermentor) and 3,100 in 6 days (Air-sparging fermentor) respectively.
