  • The chromosomal DNA of Agrobacterium tumefaciens contains the genes required for bacterial attachment to plant cell which is an essential atage in crown gall tumorigenesis by Ti-plasmid. In order to clone the genes, Agrobacterium tumefaciens strain A5512 was mutagenized by transposon Tn5 and two Agrobacterium tumefaciens mutants which are attachment-defective and nontumorigenic were isolated. From one of the two mutants, a chromosomal virulence region which was required for attachment to the plant cells was cloned.

  • These studies were carried out to obtain the basic information about transformation of ginseng plant by potential vector system, utilization of opine compound by Agrobacterium sap. , and initiation of crown gall tumor callus. Crown gall tumors were induced from stem of Panax ginseng C.A. Meyer by infection of Agrobacterium tumefaciens. Therefore, it was clarified that transformation of ginseng by Ti plasmid was possible. The crown gall tumors induced by Agrobacterium tumefaciens isolated. from the soil were different in a shape, size, and growth rate. Especially, infection of ginseng by Agrobacterium tumefaciens Y104 led to the amorphic tumor, Tumor tissue derived from stem crown gall could not be continuously cultured on the medium which did not contain phytohormone, and did not form the callus even on the medium supplemented with 2,4-D. On the other hand, the root crown gall tumors formed the calli but the formation rate of callus was quite low. As for the utilization of octopine and nopaline, it was found that 3 strains of Agrobacterium app., Y104, Y110 and C58, utilized nopaline only, Y109 utilized octopine, and Y101 failed to utilize either compound.

  • This study was carried out to research antimicrobial agents from medicinal plants, Glycyrrhiza uralensis, Dryopteris crassirhizoma, Dictamnus albus, Paeonia lactiflora, Angelica dahurica, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum falcatum, Magnolia kobus, Artemisia princeps, Arctium lappa, Aster tataricus, Hovenia dulcis, Citrus unshiu, Asparagus cochinchinensis, Gardenia jasminoides, Smilax china, Hovenia dulcis, Prunus sargentii, Scutellaria baicalensis. The ethanol extracts of 20 medicinal plants were tested for the antimicrobial activity against Agrobacterium tumefaciens. The extracts of Glycyrrhiza uralensis, Dryopteris crassirhizoma, Cimicifuga heracleifolia, Bupleurum falcatum showed antimicrobial activities against Agrobacterium tumefaciens.

  • Genetic transformation in dandelion(Taraxacum mongolicum Hand). was studied. We used for transformation by Agrobacterium tumefaciens strian LBA4404 harboring a binary vector pBI121 carrying the CaMV 35S promoter-GUS gene fusion used as a reporter gene and NOS promoter-NPTII gene as a positive selection marker. To obtain transformed plants, leaf explants of dandelion were cocultured with Agrobacterium tumefaciens LBA4404 for 10 mins, then transferred to MS medium containing 1 $\mu$M IAA, 1$\mu$M BA, 100$\mu$g/ML carbenicillin and 50 $\mu$g/ML kanarmycin sulfate. After two weeks of subculture of the explants, Kanamycin-resistant shoots were formed on explants survived. When subjected to GUS histochemical assay, all of the regenerants showed the GUS-positive responses. Plantlets were be be transformed to soil for further growth.

Isolation of Hypervirulent Agrobacterium spp from Korea and Application for Transformation of Tobacco (한국산 고감염 Agrobacterium spp의 분리 및 연초의 형질전환에 이용)

  • Total of 78 strains were characterized based on the morphological characteristics of colonies isolated on Schroth, and New & Kerr's media for selection of hypervirulent wild-type Agrobacterium spp from galls, hairy root-like process and soil of Populus, Malus, Salix and Diopyros in Korea. Among them, 48 strains were able to induce tumors in carrot disc. Hypervirulent A. tumefaciens SP101 and SM042 were identified as biotype 1 and biotype 2, respectively, These strains formed fast growing, larger tumors as compared to those induced by other strains. The binary vector pGA643 with kanamycin resistant gene was mobilized from E. coli MC100 into A. tumefaciens strain SM042 isolated from soil, and/or disarmed vector PC2760 using a triparental mating method with E. coli HB101/pRK2013, and transconjugants, A. tumefaciens SM643 and PC643 were obtained in minimal media containing kanamycin and tetracycline. Tobacco tissues were cocultivated with conjugant Agrobacterium and then transferred to selective medium with 2,4-D and kanamycin to induce the transformants. Calli were formed more efficiently in cocultivation with A. tumefaciens SM643 than that with A. tumefaciens PC643. Most of calli transformed with A. tumefaciens PC643 were friable and regenerated into normal plantlets, while the calli transformed with A. tumefaciens SM643 were compact, hard, and mixed with friable calli. The friable calli formed normal shoots, while compact calli did not form shoots but only grew to typical compact tumor calli. When the shoots formed directly from tobacco stems without callus induction after transformation by A. tumefaciens SM643 with wild-type Ti-plasmid, normal transformed plants can be induced without using disarmed Ti-plasmid.

  • We examined the effects of various phenolic compounds, Ti plasmids(octopine, nopaline) and A. tumefaciens on the vir gene expression. Nine phenolic compounds were tested using 3 types of Ti plasmid and 3 strains of A. tumefaciens on the vir gene expression. It was also investigated how the levels of vir gene expression could be related to specific phenolic compounds. Six phenolic compounds as 4-hydroxyacetophenone, phenol, catechol, resorcinol, acetosyringone and vanillin had exhibited a strong effect on the vir gene expression of A. tumefaciens MW107 containing nopaline Ti plasmid. The vir genes of A. tumefaciens MW105 and MW108 containing octopine Ti plasmids were remarkably stimulated only by acetosyringone. Thus, it appeared that the vir gene inducing abilities were differed by the kinds of phenolic compounds, A. tumefaciens and Ti plasmids.

Host Range of pTi12 Contained Agrobacterium tumefaciens KU12 Isolated from Korea (pTi-12를 함유한 한국산 Agrobacterium tumefaciens KU12의 숙주범위)

  • In order to investigate the host range of Agrobacterium tumefaciens KU12 containing pTi-12, 28 species of dicotyledonous plants were infected with KU12, A136 without Ti plasmid and A348 containing pTi A6, respectively. KU12 and A348 induced tumor in 20 species and 14 species, respectively. This results showed that KU12 has a wide host range. Therefore, it was confirmed that KU12 and pTi-12 are very useful for developing plant vector system having a broad host range.

Influence of Phenolic Compounds on vir Gene Expression in Various Agrobacterium tumefaciens (여러 종류의 Agrobacterium tumefaciens에서 vir 유전자의 발현에 영향을 미치는 페놀화합물)

  • The virulence(vir) genes in Ti plasmid at Agrobacterium tumefaciens are expressed by a phenolic compound synthesized at plant wound site. The vir genes inducing abilities of 8 phenolic compounds were tested using three wild type strains of A. tumefaciens. It was also investigated how the levels of vir gene expression among the strains tested could be related to the kinds of specific phenolic compounds. Five phenolic compounds like as 4-hydroxyacetophenone, phenol, catechol, resorcinol, and vanillin had exhibited a strong effect on the vir gene expression of A. tumefaciens MW102 whereas they did not be either non-functional or weakly inducible to the vir gene expression of other strains i.e. A. tumefaciens MW105 and MW108. Furthermore, the vir gene of A. tumefaciens MW102 was lowly expressed by acetosyringone that exposed an strong effect on the vir gene induction of other two strains. Thus, it appeared that the vir gene inducing abilities were differed by the kinds of phenolic compounds and Ti plasmids. In conclusion, we suppose that a change in vir gene inducing ability could be resulted from a difference of sensor protein expressed by vir A gene.

Epidemiology of the Crown Gall (Agrobacterium tumefaciens) of Persimmon Tree (감나무 근두암종병(Agrobacterium tumefaciens)의 발생실태)

  • Occurrence and epidemiology of crown gall caused by A. tumefaciens in persimmon tree was investigated. Persimmon tree was more sensitive to crown gall disease than peach and jujube tree. Over 80% persimmon trees were infected to crown gall in all investigated region with no relation to planted years. Yield of infected tree was severely decreased to 54%. Incidence of crown gall in nursery field of persimmon tree was also severe and the infection rate of persimmon seedlings collected from market was 34%. It was guessed that the primary inoculum of crown gall caused by A. tumefaciens is infected persimmon seedlings.

Inducers for the vir Gene Expression of Ti-Plasmid in Korean Agrobacterium tumefaciens KU12 (한국산 Agrobacterium tumefaciens KU12so Ti-plasmid의 vir 유전자 발현 유도 물질)

  • In order to investigate the phenolic compounds inducing the expression of Ti-plasmid vir genes of Agrobacterium tumefaciens KU12, we tested whether the eighteen phenolic compounds known as vir gene inducer have the activity to induce vir operon of the Ti-plasmid in A. tumefaciens KU12 transformed with pSM358cd. Also, the phenolic compounds in some tumor-uninduced dicotyledons and the attachment ability of a. tumefaciens KU12 on such dicotyledonous plants were investigated in an effort to analyze the reason why no tumor is formed on the plants. As results, fifteen among eighteen phenolic compounds known as vir gene inducer induced the expression of the Ti-plasmid vir genes. Some dicotyledonous plants which do not form the tumor have the phenolic compounds inducing the vir genes, but have less ability of the attachment than the dicotyledonous plants forming tumor.
