The hairy root cultures of Centella asiatica were established by infection leaf explants with Agrobacterium rhizogenes A4, 15834 in 1/2 Murashing and skoog liquid medium supplemented with 50 $\mu$M acetosyringone. The induced hairy roots were subjected to paper electrophoresis for the detection of opine and opine-positive clones which were considered to have been transformed. Five hairy root clones were selected according to the different bacterial strains used, growth rate and pattern. Among media tested, MS basal medium substituted phosphate concentration by 2.5mM K2HPO4 showed the highest growth rate in the dark condition.
Hairy roots of Korean ballon flower (Platycodon grandiflorum A. DC) were induced from the root tissues infected with Agrobacterium rhizogenes ATCC 15834. Growth and polyacetylene [lobetyol (1), lobetyolin (2) and lobetyolinin (3)] production fo ten hairy root clones cultured in 1/4 Gamborg B5 (B5) liquid medium were determined. One selected hairy root clone (D6) grew well in hormone free-B5 liquid medium and showed maximum content of polyacetylenes at week 6 for 1 (0.375% dry wt) and at week 7 for 2 and 3 (3.030% and 0.206% dry wt, respectively) whose levels were much higher than those of the intact plant root (1:0.019%, 2:0.077% dry wt, 3 was not detected).
In order to solve the shortage of natural Tripterygium wilfordii Hook. f. plant resource for the production of the important secondary metabolites triptolide and wilforine, hairy roots were induced from its root calli by Agrobacterium rhizogenes. Induced hairy roots not only could be maintained and grown well in hormone-free half-strength Murashige and Skoog medium but also could produce sufficient amounts of both triptolide and wilforine. Although hairy roots produced approximately 15% less triptolide than adventitious roots and 10% less wilforine than naturally grown roots, they could grow fast and could be a suitable system for producing both secondary metabolites compared with other tissues. Addition of $50{\mu}M$ methyl jasmonate (MeJA) could slightly affect hairy root growth, but dramatically stimulated the production of both triptolide and wilforine, whereas $50{\mu}M$ salicylic acid had no apparent effect on hairy root growth with slightly stimulatory effects on the production of both secondary metabolites. Addition of precursor nicotinic acid, isoleucine, or aspartic acid at the concentration of $500{\mu}M$ had varying effects on hairy root growth, but none of them had stimulatory effects on triptolide production, and only the former two had slightly beneficial effects on wilforine production. The majority of triptolide produced was secreted into the medium, whereas most of the produced wilforine was retained inside of hairy roots. Our studies provide a promising way to produce triptolide and wilforine in T. wilfordii hairy root cultures combined with MeJA treatment.

Hairy root cultures of Rheum undulatum L. induced by a co-culture with Agrobacterium rhizogenes ATCC15834 were established and the production of tannin in hairy roots was investigated. The growth of hairy roots was maximized in WPM liquid medium supplemented with 2 mg/L IAA, and 3% sucrose (pH 5.7). The highest yields of tannin were obtained from WPM medium containing 0.5 mg/L ABA, and 5% sucrose (pH 5.5). The growth and tannin production in hairy root cultures were influenced by the addition of elicitors to the medium. The addition of 50 mg/L chitosan enhanced the production of tannin with about 1.7 fold.

The hairy roots of Crotalaria sessiliflora were induced from the tissue segments infected with Agrobacterium rhizogenes ATCC 15834. The induced hairy roots were subjected to paper electrophoresis fro the detection of opine-positive clones which were considered to have been transformed. Mannopine and agropine were presented in hairy root clones while mannopine was presented in two hairy root clones. Eight hairy root clones were selected and cultured in MS, B5 and WP media. Each of hairy root clones was showed a difference in branch pattern and growth rate. The best culture medium and culture conditions of hairy roots were in $\frac{1}{2}$MS(3% sucrose, pH 5.7) liquid medium at 25$^\circ C$, 70 rpm under dark, the growth rate in $\frac{1}{2}$MS liquid medium was increased with 210-fold more than that of inoculated hairy roots and with 2-fold more than that in MS liquid medium. Also, the adequate condition for hairy root growth was such that concentration of KH$_2PO$_4 was 1.25mM and the ratio of NH${_4}{^+}$ : NO${_3}{^-}$ was 1 to 3 in MS medium. The presence of pyrrolizidine alkaloids, monocrotaline, in the hairy roots was detected by TLC.
Transformed hairy roots were induced from in vitro grown plantlets of Trichosanthes kirilowii by infection with Agrobacterium rhizogenes strain ATCC15834. Transformed hairy roots exhibited active growth with high branching of roots on plant growth regulators-free medium. Cloned line (TR-03) of hairy root was tested for its growth and extracellular protein accumulation in medium under various culture conditions. Among the culture media tested, a full-strength MS medium had a pronounced effect on root biomass and extracelluar protein accumulation in medium. The maximum root biomass (2.4 g DRW/flask) and extracellular total protein contents $(28.3ug/m\ell)$ in medium was obtained at inoculum size of 2 g (FRW) and in MS medium supplemented with 4% sucrose. In addition, the optimal shaking speed for root growth and extracellular protein accumulation in medium were 100 rpm. The total extracellualr protein concentration reached a maximum of $28.3ug/m\ell$ at 4 weeks and decreased thereafter. Protein translation inhibitory activity was observed in culture broths and reached levels of 21.3 unit. These studies demonstrate that the transformed hairy roots can be utilized for the in vitro production of ribosome-inactivating proteins.

Opium poppy (Papaver somniferum L.) is one of the most important medicinal plants, which is used as a sole commercial source of narcotic analgesic, morphine. The transformant of opium poppy we have established by infection of Rhizobium rhizogenes (formerly Agrobacterium rhizogenes) strain MAFF03-01724 showed aberrant morphology and altered opium alkaloid composition. The major alkaloid produced by this transformant was thebaine (16.3%, opium dry weight) instead of morphine. It is likely that this 'thebaine poppy' phenotype was caused by the integration of T-DNA(s) into the poppy genome DNA, and their inserted loci are of great interest. To gain an insight into the mechanism of nonnarcotic thebaine accumulation for the further approach toward the creation of 'codeine poppy' which produces codeine as a major alkaloid, the genetical and morphological analyses on the transformant was carried out. Here we report the results of the detailed analysis on the T-DNA inserted loci of T0 transfromant and the correlation between opium alkaloid composition and segregated T-DNA integration pattern in the self-pollinated T1 transformants.
The experiments were carried out to determine the optimum conditions for the induction of hairy roots in ginseng(Panax ginseng C.A. Meyer) by Agrobacterium spp. We were examined the antibiotics resistance of Agrobacterium spp and various ginseng parts, and the media for induction of hairy roots. The optimum concentration of NaOCl for sterilization of ginseng root segments without tissue damage with reduce of contamination was 7% NaOCl for 15-20 min and 9% NaOCl for 5 min, respectively. The more ginseng ages, the more contamination of ginseng root segment by sterilized in 7% NaOCl for 20 min, and especially in ginseng root segments with epidermis in six-year old roots. The growth of Agrobacterium spp were inhibited, but ginseng root segments was death in 30mg/L tetracycline. In 500mg/L cefotaxime or 500mg/L carbenicillin, the growth of Agrobacterium sup were inhibited, and root segments was grown normally. The optimum conditions for induction of hairy roots were using the root segments of three-year old ginseng cultured in 1/2MS medium supplemented with 500mg/L cefotaxime, and inoculation of Agrobacterium to root segments were better co-culture than smear method. After 2 weeks co-culture, the callus induced in cambium of root segments cultured in 1/2MS solid medium with 500mg/L cefotaxime. And then after 2 weeks, ginseng hairy roots were induced in callus of root segments. PCR analysis of rot C gene fragment confirmed that hairy roots were transgenic tissues.
This article was conducted to induce the transgenic hairy roots and determine the effect of culture conditions on optimum growth of hairy roots by Agrobacterium rhizogenes strain, 15834 in Lycium chinense Miller. Hairy roots of L. chinense Miller. were induced from leaf segments by co-cultivation with A. rhizogenes. When the hairy roots were cultured in various MS medium strength and sucrose concentrations, the highest growth of hairy roots was observed in half-strength MS media supplemented with 3% sucrose, respectively. In air lift bioreactor cultures, the liquid medium contained with 1/2 MS and 3% sucrose was also the best for optimum growth of hairy roots.
Antimicrobial activity and chemical composition of volatile flavor extracts from Agastache rugosa were investigated. The volatile flavor extracts were obtained from leaves and stems of Agastache rugosa by simultaneous distillation extraction (SDE) method. Antimicrobial activity was investigated by disc diffusion and broth dilution methods against several microorganisms of Bacillus cereus, bacillus megaterium, Bacillus subtilis, Corynebacterium xerosis, Staphylo coccus aureus, Staphylococcus epidermidis, Agrobacterium rhizogenes , Agrobacterium tumefaciences, Enterobacter cloacae, Escherichia coli, Salmonella typhi, Vibrio parahaemolyticus, Candida utilis and Saccharomyces cerevisiae. Volatile flavor extractsfrom leaves have strong antimicrobial activity against C.utilis and S.cerevisiae. When 0.12% volatile flavor extracts from fresh leaves were included in the medium, lag phase of C. utilis was extended 6 hr and that of S.utilis and S.cerevisiae was extended 2hr. Further analyses were performed to elucidatethe effective component of the extracts. The major component of volatile flavor was estragole, a phenolic compound. Minor components were determined to be terpenes , alcohols, acids , esters, ketones and aldethydes.
