Isolation and Characterization of Saccharomyces cerevisiae from nuruk for Production of Ethanol from Maltose (누룩으로부터 맥아당 이용능과 에탄올 생산성이 우수한 효모의 분리와 특성)

  • Wild-type yeast strains were isolated from nuruk, a type of microbial starter culture used for fermenting grains to produce alcoholic products, that was collected from different areas in Korea. Strains were identified based on the analysis of 18S rRNA sequences. Fifty strains shared the highest sequence similarity with Saccharomyces cerevisiae and were designated MBYK1-MBYK50. Among these S. cerevisiae isolates, MBYK45 produced $44.0{\pm}0.3g$ of ethanol from 200 g maltose after incubation at $30^{\circ}C$ for 48 h. Maximum ethanol production of $110.80{\pm}0.81g/l$ with productivity of $3.79{\pm}0.14g^{-1}l^{-1}h^{-1}$ was obtained at optimum culture conditions of pH (6.0), maltose (200 g/l), and temperature ($35^{\circ}C$). This study indicates that the MBYK45 strain of S. cerevisiae, isolated from nuruk, might be suitable for traditional liquor production from malts.

  • To produce inulase from Kluyveromyces fragilis No.351 using Jerusalem artichoke tuber extract, optimization of the condition was conducted. As results, the optimal concentration of artichoke tuber extract was 3.5 % and bactocasitone showed better production than yeast extract. The optimal temperature and pH were $30^{\circ}C$ and 5.5, respectively. The addition of $NH_4H_2PO_4$ increased the enzyme production. Inulase synthesis was growth-associated and the enzyme production increased as concentration of dissolved oxygen increased. A higher quantity of industrial grade inulase was prepared by the combination of ultrafiltration and ethanol precipitation with 72% recovery.

Soil physio-chemical properties of Mt. NamSan on Kyungju in Korea (경주 남산의 토양 이화학적 특성)

  • This study was carried out in order to produce useful material for the forest multiple use and forest protection management by soil physio-chemical analysis of studied area in Mt. Namsan. The result of soil physio-chemical analysis and statical analysis represented as following. In side of physical properties of forest soil in Mt. Namsan, Soil depth was average 31.4cm and available soil depth was average 20.0cm. Soil type was Brown forest soil that representative soil type in Korea. Soil texture was sandy loom(SL) except valley area. In side of chemical properties, the range of soil acidity was 4.29 ~ 5.19 (average 4.76), organic matter content was 3.17% that compared the lowest value to organic matter content of Korea forest soil. Available phosphorus was 3.64ppm that was lower than others forest soil. Exchangeable cation content was similar to the Korea brown forest soil. Cation exchange capacity(CEC) was $8.22cmol_c/kg$ in Mt. Narnsan.

Production of Functional Whey Protein Concentrate by Monitoring the Process of Ultrafilteration

  • This investigation was undertaken in order to elicit the relationship between the extent of ultrafiltration processing of whey and its effect on composition and yield of resultant whey protein concentrate (WPC). Cheddar cheese whey was fractionated through ultrafiltration to an extent of 70, 80, 90, 95, 97.5% and 97.5% volume reduction followed by I stage and II stage diafiltration. After each level of ultrafiltration, the composition of WPC was monitored. Similarly, the initial whey was adjusted to 3.0, 6.2 and 7.0 pH levels and ultrafiltration was carried out to elicit the effect of pH of ultrafiltration on the composition. Further, initial whey was adjusted to different levels of whey protein content ranging from 0.5 to 1.0 per cent and subjected to ultrafiltration to different levels. The various range of retentate obtained were further condensed and spray dried in order to assess the yield of WPC per unit volume of whey used and the quantity of whey required to produce unit weight of product. With the progress of ultrafiltration, there was a progressive increase in protein content and decrease in lactose and ash content. The regression study led to good relationships with $R^2$ values of more than 0.95 between the extents of permeate removed and the resultant changes in composition of each of the constituents. Whey processed at pH 3.0 had significantly a very low ash content and high protein content as compared to processing at 6.2 and 7.0. The yield of WPC per unit volume of whey varied significantly with the initial protein content. Higher initial protein content led to higher yield of all ranges of WPC and the quantity of whey required per unit weight of spray dried WPC significantly reduced. Regression equations establishing the relationship between initial protein content of whey and the yield of various types of WPC have been derived with very high $R^2$ values of 0.99. This study revealed that, the yield and composition of whey can be monitored strictly by controlling the processing parameters and WPC can be produced depending on the food formulation requirement.

Physical, Chemical and Biomethanation Characteristics of Stratified Cattle-Manure Slurry

  • In the quiescent state, cattle-manure slurry stratifies into three discernible layers, namely a floating scum layer, a bottom sludge layer and a watery middle layer. The proportions of top (scum), middle and bottom (sludge) layers were approximately 20, 60 and 20% respectively of the volume of the whole slurry. Particulate matter from the different stratified layers was characterised for particle size distribution and cellulose, hemicellulose and lignin composition. Total solids concentrations of top, middle and bottom layers were 12.7, 2.8 and 7.4% respectively. Larger particles were found in the top layer compared with the bottom. The top layer contained the highest amounts of Neutral Detergent Fibre (NDF), Acid Detergent Fibre (ADF), cellulose and hemicellulose, but the lowest amount of Total Kjeldahl Nitrogen (TKN). The bottom layer contained the highest amounts of Acid Detergent Lignin (ADL) and TKN. With increase in particle size, there were increases in NDF, ADF, cellulose and hemicellulose, accompanied by decreases in ADL and TKN. Biochemical methane potential of the three layers was also measured. The top layer was found to produce the most methane with the middle layer producing the least. Biomethanation rate from the top layer was also the highest. Differences in biomethanation rates and biochemical methane potential were attributed to differences in chemical composition of the particulate matter. About 48%, 23% and 30% of the total chemical oxygen demand (COD) in the top, middle and bottom layers respectively of the slurry was found to be degradable.

Isolation and Identification of Fungal Strains Producing Gibberellins from the Root of plants (식물뿌리내부에 존재하는 지베렐린 생산균 분리와 동정)

  • 249 fungal strains were isolated from the roots of 26 plants, and the production of GAs was spectrophotometric ally examined. As a result 76 fungal strains were shown to produce GAs. Bioassay of culture broth from seventy six fungal strains producing GAs was carried out with waito-c rice, that is dwarf rice. The seventy six fungi with GAs-producing activity were incubated for seven days in 40 mL of Czapek's liquid medium at $30^{\circ}C$ and 180 rpm, and the culture broth of fungi were treated on the 2-leaf rice sprout. Fifteen of these showed plant growth promoting activity and the amount of each GAs in the medium was measured by Gas chromatographymass spectrometer (GC-MS). Nine of these fungi were also identified by genetic analysis of the nucleotide sequences in the internal transcribed spacer region of the ribosomal DNA.

Roles of YehZ, a Putative Osmoprotectant Transporter, in Tempering Growth of Salmonella enterica serovar Typhimurium

  • Salmonella, a main cause of foodborne diseases, encounters a variety of environmental stresses and overcomes the stresses by multiple resistance strategies. One of the general responses to hyperosmotic stress is to import or produce compatible solutes so that cells maintain fluid balance and protect proteins and lipids from denaturation. The ProP and ProU systems are the main transport systems for compatible solutes. The OsmU system, recently identified as a third osmoprotectant transport system, debilitates excessive growth as well by reducing production of trehalose. We studied a fourth putative osmoprotectant transport system, YehZYXW, with high sequence similarity with the OsmU system. A Salmonella strain lacking YehZ, a predicted substrate-binding protein, did not suffer from hyperosmolarity but rather grew more rapidly than the wild type regardless of glycine betaine, an osmoprotectant, suggesting that the YehZYXW system controls bacterial growth irrespective of transporting glycine betaine. However, the growth advantage of ${\Delta}yehZ$ was not attributable to an increase in OtsBA-mediated trehalose production, which is responsible for the outcompetition of the ${\Delta}osmU$ strain. Overexpressed YehZ in trans was capable of deaccelerating bacterial growth vice versa, supporting a role of YehZ in dampening growth. The expression of yehZ was increased in response to nutrient starvation, acidic pH, and the presence of glycine betaine under hyperosmotic stress. Identifying substrates for YehZ will help decipher the role of the YehZYXW system in regulating bacterial growth in response to environmental cues.

  • Fresh agricultural product in Korea are currently being exported overseas through expensive air transportation, supported by the governments's farm export subsidies. However all members of the World Trade Organization(WTO) must halt government subsidies by 2023. Accordingly, it is necessary to use marine transport capable of carrying freight at low cost. Reefer containers are used for marine transportation of fresh produce but it have a problem due to the temperature difference inside the reefer container which causes of fresh cargo and drop in freshness during sea transportation. In order to solve the problem, we developed a temperature control algorithm for reefer container that maintain a constant temperature and minimizes the deviation inside reefer container. The result showed that the maintained a constant temperature within a maximum of $0.5^{\circ}C$ based on the set-point of $4.0^{\circ}C$ inside reefer container.

Effect of Water Potential on Mycelial Growth, Reproduction and Spore Germination by Fusarium moniliforme (Fusarium moniliforme의 Propagule형성(形成)과 발아(發芽)에 미치는 Water Potential의 효과(效果))

  • Hyphal growth by Fusarium moniliforme was best at -14 bars osmotic water potential. Hyphal growth was prevented at -94 bars. The production of microconidia was best at -14 bars osmotic potential and prevented at -84 bars regardless of Strain. In contrast, this fungus sporulated macroconidia best at -1.4 bars and progressively less with each increment drop in water potential below that of basal media. The rate of spore germination followed a similar pattern with all of the spores; uniformly maximal at about -1.4 bars and progressively slower as the water potential was lowered from -1.4 bars to -42 bars. Under the natural conditions, plants infected by F. moniliforme produce microconidia on the dead tissues instead of producing macroconidia. This phenomenon agrees well with the water potential experiment since the dead plant tissues have a lower water potential than the living plant.

Evaluation of L-Lactic Acid Production in Batch, Fed-batch, and Continuous Cultures of Rhizopus sp. MK-96-1196 Using an Airlift Bioreactor

  • Various processes which produce L-lactic acid using ammonia-tolerant mutant strain, Rhizopus sp. MK-96-1196, in a 3L airlift bioreactor were evaluated. When the fed-batch culture was carried out by keeping the glucose concentration at 30g/L, more than 140 g/L of L-lactic acid was produced with a product yield of 83%. In the case of the batch culture with 200g/L of initial glucose concentration, 121g/L of L-lactic acid was obtained but the low product yield based on the amount of glucose consumed. In the case of a continuous culture, 1.5g/L/h of the volumetric productivity with a product yield of 71% was achieved at dilution rate of $0.024\;h^{-1}$. Basis on these results three processes were evaluated by simple variable cost estimation including carbon source, steam, and waste treatment costs. The total variable costs of the fed-batch and continuous cultures were 88% and 140%, respectively, compared to that of batch culture. The fed-batch culture with high L-lactic acid concentration and high product yield decreased variable costs, and was the best-suited for the industrial production of L-lactic acid.
