Purification and Characterization of Branching Specificity of a Novel Extracellular Amylolytic Enzyme from Marine Hyperthermophilic Rhodothermus marinus

  • An extracellular enzyme (RMEBE) possessing ${\alpha}-(1{\rightarrow}4)-(1{\rightarrow}6)$-transferring activity was purified to homogeneity from Rhodothermus marin us by combination of ammonium sulfate precipitation, Q-Sepharose ion-exchange, and Superdex-200 gel filtration chromatographies, and preparative native polyacrylamide gel electrophoresis. The purified enzyme had an optimum pH of 6.0 and was highly thermostable with a maximal activity at $80^{\circ}C$. Its half-life was determined to be 73.7 and 16.7 min at 80 and $85^{\circ}C$, respectively. The enzyme was also halophilic and highly halotolerant up to about 2M NaCl, with a maximal activity at 0.5M. The substrate specificity of RMEBE suggested that it possesses partial characteristics of both glucan branching enzyme and neopullulanase. RMEBE clearly produced branched glucans from amylose, with partial ${\alpha}-(1{\rightarrow}4)$-hydrolysis of amylose and starch. At the same time, it hydrolyzed pullulan partly to panose, and exhibited ${\alpha}-(1{\rightarrow}4)-(1{\rightarrow}6)$-transferase activity for small maltooligosaccharides, producing disproportionated ${\alpha}-(1{\rightarrow}6)$-branched maltooligosaccharides. The enzyme preferred maltopentaose and maltohexaose to smaller maltooligosaccharides for production of longer branched products. Thus, the results suggest that RMEBE might be applied for production of branched oligosaccharides from small maltodextrins at high temperature or even at high salinity.

  • The home meal replacement (HMR) market has attracted significant attention due to COVID-19 pandemic and products that utilize the combination of different processed ingredients are now being developed. In this study, Broughton's ribbed ark Scapharca broughtonii was soaked in seasoned soy sauce with the incorporation of mustard leaf Brassica juncea (M-BRA), which is known to have a unique texture as well as excellent functional properties such as antioxidation, to develop a regional specialty product. The optimal conditions for manufacturing M-BRA from the seasoned soy sauce (the sauce to be added [X1] and the soaking time [X2]), were examined using response surface methodology (RSM) to analyze the significance of the salinity (Y1), amino-N content (Y2), and overall acceptance (Y3). The coefficient of determination (R2) between X1-X2 and Y1-Y3 were close to 1, thereby confirming the suitability of the RSM model. The optimal conditions were seasoned soy sauce addition of 140% and soaking time of 156 min. The M-BRA manufactured under these conditions exhibited superior overall acceptance compared to seasoned commercial soy sauce, which was used as a control. We expect that the market for M-BRA manufactured by combining marine and agricultural materials will expand owing to superior overall acceptance compared with commercial products.

  • 현재 모든 산업에서 전산화, 자동화가 이루어지고 많은 발전을 거듭하고 있다. 이런 흐름에 따라 본 논문 역시 경매 시스템을 전산화하여 데이터 저장의 용의성과 작업을 하는데 있어서 편리성을 추구하는 것이 목적이다. 여러 경매의 형태 가운데 농수산물을 경매하는 "농수산물 도매 유통센터"(예, 가락동 농산물 시장)에서 이용한 수 있는 경매 시스템에 초점을 갖추고 있다. 경매는 물품이나 형태에 따라 특성이 많이 틀리기 때문에 모든 경매 상황을 고려하는 것 보다 한가지에 초점을 맞춰서 개발하는 것이 더욱 효과적이라 할 수 있다. 현재 경매는 거의 수신호에 의존하고 있는 경매를 전산화로 전환하여 경매 운영자가 관리를 쉽고 간편하게 한뿐만 아니라 경매 참여자가 역시 경매 참여를 간편하게 한 수 있는 경매 시스템을 개발하였다.

Physiological Roles of Adipokines, Hepatokines, and Myokines in Ruminants

  • Since the discovery of leptin secreted from adipocytes, specialized tissues and cells have been found that secrete the several peptides (or cytokines) that are characterized to negatively and positively regulate the metabolic process. Different types of adipokines, hepatokines, and myokines, which act as cytokines, are secreted from adipose, liver, and muscle tissue, respectively, and have been identified and examined for their physiological roles in humans and disease in animal models. Recently, various studies of these cytokines have been conducted in ruminants, including dairy cattle, beef cattle, sheep, and goat. Interestingly, a few cytokines from these tissues in ruminants play an important role in the post-parturition, lactation, and fattening (marbling) periods. Thus, understanding these hormones is important for improving nutritional management in dairy cows and beef cattle. However, to our knowledge, there have been no reviews of the characteristics of these cytokines in beef and dairy products in ruminants. In particular, lipid and glucose metabolism in adipose tissue, liver tissue, and muscle tissue are very important for energy storage, production, and synthesis, which are regulated by these cytokines in ruminant production. In this review, we summarize the physiological roles of adipokines, hepatokines, and myokines in ruminants. This discussion provides a foundation for understanding the role of cytokines in animal production of ruminants.

An Empirical Study on Effect of Time-Varying Quality Chang on Apple Shipment Volume for Shipment Decision Making System (출하의사결정시스템에 있어 품질변화효과가 출하량에 미치는 영향에 대한 실증연구)

  • This research is one of a series of studies to develop a system to help agricultural producers and sellers determine when and how much to ship products to the wholesale market to maximize their profit. The purpose of this research is to incorporate the time-varying quality change effect, which was not used in the previous agricultural and marine product shipping model. The researchers developed four models to measure the quality change effect: quality declining steadily over time, quality declining rapidly at first and then slowly, quality declining first slowly and then rapidly, and quality rising over time and then decreasing again. According to the results of an empirical analysis of the effect of each model's quality change effect on shipments for apples traded in the Garak Wholesale Market from 2014 to 2021, statistical significance was found in the quality change effect of all four models. And there was no significant difference in explanatory power between the four models. Therefore, any of the four models should be introduced into the decision-making system for shipping time for apples.

  • Reefs are the marine structure that can give resting, inhabiting, feeding and nursing spaces for a variety of fishes. Usually artificial reefs are made of cement and steels respectively in Japan as well as in Korea. However since resources deficiency has been getting serious, other materials are taken into consideration for the basic bodies of artificial reefs. About 300 thousand tons of waste agricultural plastic films are generated every yew in Korea, but no effective recycling techniques have been developed. In this sense, artificial reefs made of waste agricultural plastic films are the most representative symbol of the recycled products in the Resource Recycling Era. In particular, since these reefs could be made of the semi-cleaned waste agricultural plastic films that still contain high portion of soil, it is very environmentally friendly not only in manufacturing process but also in using under water. Furthermore they have some evident advantages as follows; 1) high fish swarming effect 2) good initial attachment of the marine growths 3) extremely low corrosion to brine 4) easy adjustment of the gravity 5) economical manufacture, transportation and jettison 6) excellent safety to ecosystem caused by lower elution of toxic substances 7) good recyclable property after application and so on.

  • Effective storage facilities are required to stabilize the price of agricultural and marine products to preserve their qualities due to the big fluctuation of shrimp price in Korea. It is easy to make the cave because of good conditions of the land configuration, soil and convenient transportation. The cave storage can save the cost about 40% in building site and equipment, and about 50% in maintenance comparing to existing low temperature storage facilities. The cave storage provide to improve the quality of their stored products with the low heat conductivity, the constant temperature and humidity year round. Therefore, more low temperature storage facilities are required because the items are expanded from potatoes, sweet potatoes, onions, garlics, apples and chestnuts to tangerines, grapes, cabbages, radishes, and wet ginsengs. The demands of the low temperature storage facilities can be substituted into the cave storage facilities. Thus, studies are conducted to observe the changes of the components of the pure and the seasoned salted shrimps with fermentation period during stored at room temperature in cave and to establish the storing at underground facilities to produce high quality salted fish to make profit.

  • The major purpose of the present study is to syrvey vegetables, meats, seafoods, processed foods and imported foods for the presence of Listeria spp. and to prevent listeriosis caused by the contamination of Listeria monocytogenes. Listeria spp. was not found in vegetables and processed foods. The optimum growth condition of isolates indentified as Listeria monocytogenes was pH 7.0 and 37$^{\circ}C$. the antimicrobial effect of grapefruit seed extract(GFSE) was observed in the level of more than 100ppm by disk method. When 1ml(2.5$\times$106 CFU/ml) of Listeria monocytogenes was inoculated and incubated for 3 days at 3$0^{\circ}C$, the total cell number of the organism was 4.5$\times$109 in the control, 7.2$\times$103 in 100 $\mu\textrm{g}$/ml of GFSE medium, and 3.5 $\mu\textrm{g}$/ml of GFSE medium. Direct visualization of microbial cells by using both transmission electron microscope and scanning electron microscope showed microbial cell membrane the function of which was destroyed by treating with the dilute solutions of GFSE. It could be confirmed that GFSE completely inhibited the growth of the test strain of Listeria monocytogenes.

Yield Analysis of Flavonoids in Acanthopanax divaricatus and A. koreanum Grown using Different Cultivation Methods

  • High-performance liquid chromatography was performed in order to analyze the changes in the flavonoid content (rutin, hyperin, afzelin, quercetin, and kaempferol) of Acanthopanax divaricatus and A. koreanum, in response to different cultivation methods (pinching height, planting time, and top dressing). The total flavonoid content of A. divaricatus and A. koreanum ranged from 0.201 to 0.690 mg/g with different pinching heights, 0.143 to 1.001 mg/g for different planting times, and 0.156 to 1.074 mg/g depending on the rate of fertilizer application. In both A. divaricatus and A. koreanum, the total flavonoid content in the upper section of the plant was greater than that in the lower section. These results demonstrate which cultivation methods maximize the flavonoid content of A. divaricatus and A. koreanum, and thus help to optimize flavonoid yields to improve production for nutraceutical, pharmaceutical, and cosmeceutical applications.

  • The Gang-Seo wholesale market is an experimental system in that two different distribution channels co-exist in the same marketplace; the auction system and the consignment system. The government expects the consignment system offers higher and more stable prices to farmers than the auction system. This study tested if that is the case, but the results did not support the hypotheses. The consignment system needs institutional complements and more transparent operations before wide-spreading.
