High Cell Density Culture of Micro-algal Dunaliella bardawil (미세조류 Dunaliella bardawil의 고농도 세포배양)

  • High cell density cultivation of microalga Dunaliella bardawil using nitrogen fed-batch cultures was studied in batch flask. Optimum environmental conditions include concentrated nutrients except NaCl and carbon sources, carbon sources, pH, light, agitation, nitrate and phosphate ions. Cell growth, consumption rates of nitrate and phosphate ions were monitored. Optimal conditions for higher cell density were found to be(in the range tested): 5 times concentrated media(1 times-10 times concentrated media) pH 8.0 (7.0-9.0) white light(blue and red light) 15mM of nitrate (0.94-15mM) 250mM $NaHCO_3$ and $CO_2$ gas. However, the addition of phosphate ions did not enhance the algal maximum cell density and specific growth rate. Nitrate was found to be effective for the cell growth. The maximum cell density of fed-batch culture using nitrate ions in $8.955{\times}106$cells/ml after 189hr incubation.

Effect of Agitation and Aeration Rate on Nicotiana tabacum Suspension Cell Culture in Bioreactors (Bioreactor를 이용한 담배세포 현탁배양에서 교반형태와 통기량이 미치는 영향)

  • For the optimization of operating conditions for plant cell suspension culture in bioreactors, effects of bioreactor types, various kinds of impellers, and aeration rates were examined using Nicotiana tabacum cells as a model system. Stirred tank bioreactor and airlift bioreactor were used for the comparison of bioreactor type. Growth rates in both bioreactors were lower than in shake flasks. In terms of final cell concentration, stirred tank bioreactor supported a little bit better growth compared to airlift bioreactor. Impeller type did not affect cell growth significantly, but it was apparent that cell size index decreased in the case of using hollowed paddle impeller. When the aeration rate was maintained at 0.3 vvm, cell growth was the best. At above 1.0 vvm, growth inhibition as well a browning was noticed. In addition, it was found that cell size index reduced proportionally to the increased of aeration rate.

Effects of Mixing Performance and Conditioned Medium on hCTLA4Ig Production in Transgenic Rice Cell Suspension Cultures (형질전환 벼 현탁세포 배양에서 혼합효율과 조정배지가 hCTLA4Ig 생산에 미치는 영향)

  • Transgenic rice cells using RAmy3D promoter can provide high productivity, and the production of recombinant protein is induced by sugar starvation. In this system, productivity was reduced during the scale-up processes. To ensure the influences of shear stress and oxygen transfer rate, working volume and mixing performances were investigated under various agitation speeds and working volumes. In addition, inoculation methods including suspended cells and filtered cells were compared. Working volumes and shaking speeds were 300, 450 mL and 80, 120 rpm, respectively. Hydrodynamic environment of each condition was measured numerically like mixing time and $k_La$. Good mixing performance and high shear stress were measured at high agitation speed and low volume. The highest level of hCTLA4Ig was 30.7 mg/L at 120 rpm, 300 mL. When conditioned medium was used for inoculation, increased cell growth was noticed during the day 0~4 and decreased slower than filtered cells. Compared with filtered cells, the maximum hCTLA4Ig level reached 37.8 mg/L at 120 rpm, 300 mL and lower protease activity level was observed. In conclusion mixing performance is critical factor for productivity and conditioned medium can have a positive effect on damaged cells caused by hydrodynamic shear stress.

  • This study was conducted to determine the influence of cultural conditions such as temperature, time, water content, koji-thickness and agitation on the production of saccharifying amylase by Rhizopus sp. ZB9 isolated from Korean Nuruk during the preparation of rice koji, which is used in brewing Korean rice wines, Takju and Yakju. Rice kojies were made under different cultural conditions, and the saccharifying activities of each koji were tested. The temperature range suitable for the production of saccharifying amylase was $28{\sim}36^{\circ}C$. Based on the saccharifying activity and color, 60 hours of cultivation at $28^{\circ}C$ was believed to produce the optimum results. The water contents of steamed rice suitable for the production of saccharifying amylase were 35~40%. An increase in koji-thickness induced no adverse effects on the production of saccharifying amylase, but agitation-work during cultivation had a harmful effect.

Production of a Fibrinolytic Enzyme in Bioreactor Culture by Bacillus subtilis BK-17

  • Bacillus subtilis BK-17 which produces a novel protease with fibrinolytic activity was isolated from soybean paste. Bioreactor production of the enzyme was studied in order to optimize fermentation conditions such as medium concentration, pH, agitation speed, and temperature. Under most cultural conditions, enzyme production initially began when the cell growth stopped. The onset of the enzyme production was indicated by rapid increase in both dissolved oxygen (DO) and pH. Two- to three-times more concentrated medium than the flask optimum medium yielded higher enzyme production in the bioreactor fermentation. When the medium pH was controlled constant, pH 6.5 exhibited the highest activity in the range of 6.0 to 7.5, but the activity was similar to the case when the pH was initially adjusted to 7.5 and subsequently maintained within a relatively wide range of 6.4 to 7.8. Agitation speed did not affect the enzyme production with an exception of DO reaching zero. Fermentation time was reduced when temperature increased within the range of $25^{\circ}C$ to$37^{\circ}C$. However, the highest activity, along with the slow decrease of the enzymatic activity after reaching the maximum value, was observed at $25^{\circ}C$. By shifting the temperature from $37^{\circ}C$ to $25^{\circ}C$immediately after DO reached the minimum level, the high enzyme production of 1,100 U/ml along with the short fermentation period of 13 h could be obtained.

  • Changran-Jeotgal (salt fermented viscera of Alaska Pollack) produced by conventional process has been exposed to several problems, such as quality variation, salty taste and relatively short shelf-life. Therefore, Improved manufacturing process of Changran-Jeotgal was designed to resolve these problems. In this paper, we investigated the optimization of salting process. The result showed that salting stage must be maintained $11\%$ to acquire $8\%$ NaCl concentration of final products, when $12\%$ NaCl was added at $20^{\circ}C$, salting process was maintained $11\%$. Also, agitation with 10 rpm shortened salting time from 6 hours to 2 hours. To resolve the problem of water contents due to released extract from salting process, the released extract was removed at 2 hours after agitating-salting process, and it caused the water activity of products to be reduced from 0.94 to 0.88.

  • As the current heating control of the greenhouse is located in specifically designed place, there is an inevitable difference in degrees depending on the latitude in it. Even though it is necessary to maintain the proper temperature in the greenhouse producing vegetables and fruit plants, the difference between ups and downs in the facilities results in the increasing energy consumption to both warm and cool down the facilities. The newest heating method, automatic control system of vertical agitation heater, which manipulates the inner air circulation efficiently, is suggested in this paper. The proposed system utilizes both the upper temperature and the lower temperature, and controls the air circulation fan and heating independently, so that maximizes the efficiency of heating with the minimum energy and implements predictable planning of farm products.

Leaching Kinetics of Yttrium Extraction from Coal Fly Ash using Sulfuric Acid

  • Leaching kinetics for extracting yttrium from the coal fly ash was investigated in the presence of sulfuric acid during extraction. The leaching kinetics of yttrium were conducted at reactant densities of 5~1,000 g coal fly ash per L of $1.0{\sim}10.0N\;H_2SO_4$, agitation speed of 250 rpm and temperature ranging from 30 to $90^{\circ}C$. As a result, the leaching kinetic model was determined in a two-step model based on the shrinking core model with spherical particles. The first step was proceeded by chemical reaction at ash surface, and the second step was proceeded by ash layer diffusion because the leaching conversion of yttrium by the first chemical reaction increases with increased the time irrelevant to the temperature whereas it increases with increased the leaching temperature. The activation energy of the first chemical leaching step was determined to be $1.163kJmol^{-1}$. After the first chemical reaction, the activation energy of ash layer diffusion leaching was derived to be $41.540kJmol^{-1}$. The optimum conditions for leaching the yttrium metal of 60 % were found to be the slurry density of 250 g fly ash per L of $H_2SO_4$, solvent concentration of $2.0N\;H_2SO_4$, second step leaching of temperatures of $30^{\circ}C$ for 3 hours and then $90^{\circ}C$ for 3 hours at agitation rate of 250 rpm.

  • The cultural conditions of Mucor plumbeus FRI 0007 were investigated for the maximum production of felt and lipid. It was found that the lower the pH and the higher the incubation temperature, the higher accumulation of the felt and lipid. Shake culture rendered higher lipid accumulation and lower felt accumulation than static culture. Maximum production of felt and lipid content were 47.8 g/$\ell$ and 50.73%, respectively, when the organisms were static-cultured at a temperature of 37$^{\circ}C$ and pH of 3.5 for 25 days latroscan thinchrographic analysis showed that the higher amount of triglyceride was obtained when static-cultured at a low pH. Fatty acid composition of the microbial lipid was affected by the incubation temperature, types of nitrogen source and speed of agitation: lower degree of saturation was observed as the incubation temperature decreased and the speed of agitation increased. Fatty acids of monoglyceride and diglyceride were mainly palmitic and oleic acids and those of triglycerides were mainly palmitic, oleic acids.

Application of Scale-Up Criterion of Constant Oxygen Mass Transfer Coefficient ($k_La$) for Production of Itaconic Acid in a 50 L Pilot-Scale Fermentor by Fungal Cells of Aspergillus terreus

  • The scale-up criterion of constant oxygen mass transfer coefficient ($k_La$) was applied for the production of itaconic acid (IA) in a 50 L pilot-scale fermentor by the fungal cells of Aspergillus terreus. Various operating conditions were examined to collect as many $k_La$ data as possible by adjusting the stirring speed and aeration rate in both 5 L and 50 L fermentor systems. In the fermentations performed with the 5 L fermentor, the highest IA production was obtained under the operating conditions of 200 rpm and 1.5 vvm. Accordingly, we intended to find out parallel agitation and aeration rates in the 50 L fermentor system, under which the $k_La$ value measured was almost identical to that ($0.02sec^{-1}$) of the 5 L system. The conditions of 180 rpm and 0.5 vvm in the 50 L system turned out to be optimal for providing almost the same volumetric amount of dissolved oxygen (DO) into the fermentor, without causing shear damage to the producing cells due to excessive agitation. Practically identical fermentation physiologies were observed in both fermentations performed under those respective operating conditions, as demonstrated by nearly the same values of volumetric ($Q_p$) and specific ($q_p$) IA production rates, IA production yield ($Y_{p/s}$), and specific growth rate (${\mu}$). Specifically, the negligible difference of the specific growth rate (${\mu}$) between the two cultures (i.e., $0.029h^{-1}$ vs. $0.031h^{-1}$) was notable, considering the fact that ${\mu}$ normally has a significant influence on $q_p$ in the biosynthesis of secondary metabolites such as itaconic acid.
