Effect on Inhibition of Matrix Metalloproteinase-1 in Human Dermal Fibroblasts by Production of Exopolysaccharide from Mycelial Culture of Grifola frondosa (잎새버섯이 생산하는 세포외 다당체의 사람 섬유아세포에서 Matrix Metalloproteinase-1 발현저해 효과)

  • We investigated the effect on inhibition of matrix metalloproteinase (MMP) in human dermal fibroblast (HDF) by production of exopolysaccharide (GF-glucan) from mycelial culture of Grifola frondosa HB0071. The photoprotective potential of GF-glucan was tested in HDF exposed to ultraviolet-A (UVA) light. It was revealed that GF-glucan had an inhibitory effect on MMP-1 expression in UVA-irradiated HDF without any significant cytotoxicity. The treatment of UVA-irradiated HDF with GF-glucan resulted in a dose-dependent degrease in the expression level of MMP-1 protein and mRNA (by maximum $54.4\%$ at an $0.5\%$ GF-glucan). These results suggest that GF-glucan obtained from mycelial culture of G. frondosa HB0071 may contribute to inhibitory action in photoaging by reducing the MMP-1 related matrix degradation system.

Effect of red ginseng NaturalGEL on skin aging

  • Background: In aged skin, degradation of collagen fibers, which occupy the majority of the extracellular matrix in the dermis, and changes of aquaporin 3 (AQP3) and skin constituents, such as hyaluronic acid and ceramide, cause wrinkles and decrease skin moisturization to contribute to dryness and lower elasticity skin. Red ginseng (RG) is used as a cosmetic and food material and is known to protect from UVB-induced cell death, increase skin hydration, prevent wrinkles, and have an antioxidative effect. But, in general, RG used as a material is the soluble liquid portion in the solvent, and the part that is not soluble in the solvent is discarded. Thus, we made the whole RG into microgranulation and dispersed in water to produce gel form for using entire RG, and it was named red ginseng NaturalGEL (RG NGEL). Methods: RG NGEL was investigated for matrix metalloproteinases inhibitory activity, induction of Type I collagen, AQP3, hyaluronan synthetase 2, serine palmitoyl transferase, ceramide synthase 3, and filaggrin expression and compared with RG water extract. Results: RG NGEL reduced the levels of UV-induced matrix metalloproteinases and increased Type I collagen in human fibroblast cells and upregulated AQP3, hyaluronan synthetase 2, serine palmitoyl transferase, ceramide synthase 3, and filaggrin expressions in human keratinocytes compared with RG water extract. Conclusion: RG NGEL has the potential as an effective reagent for antiaging cosmetics to improve wrinkle formation and skin hydration.

  • The performance of ultrasonic testing systems for industrial or medical purpose largely depends on the performance of ultrasonic transducers. Generally, the information about an ultrasonic transducer performance characteristics are expressed by the ultrasonic R/F signal back from a reflector and its frequency characteristics in the data sheet provided by manufacturers. In case of focused ultrasonic transducers, the two pieces of information can, however, hardly assure that the focused ultrasonic transducer would produce well-focused C-scan images. Therefore, we propose the measured size of focal spot and the reconstructed shape of effective focal zone in the focused sound field as novel measures for the performance evaluation of the focused ultrasonic transducers. The process of getting the both measures of the transducers is conducted by the implemented software including sound field scanning and virtual 3D reconstruction functions which requires the echo of a point reflector. The proposed method could, otherwise impossible in the existing method, effectively and simply distinguish superior ones among many transducers made in the same specification and be also used to detect the performance degradation due to the aging of the transducers. Eventually, the quality of performance of the ultrasonic testing systems for industrial or medical purpose is secured.

A Study on Correlation of Microstructural Degradation and Mechanical Properties of 9-12%Cr-Steel for Ultra-Super Critical Power Generation (초초임계압 발전용 소재의 장시간 열처리에 따른 미세조직 변화와 기계적 특성의 상관관계 연구)

  • For the good combination of high-temperature strength, toughness and creep property, $9-12\%$ chromium steels are often used for gas turbine compressors, steam turbine rotors, blade and casing. In this study, the correlation of microstructural evolution and mechanical properties was investigated fur the specimens heat-treated at 600, 650 and $700^{\circ}C$ for 1000, 3000 and 5000 hrs. The microstructure of as-received specimen was tempered martensite with a high dislocation density, small sub-grains and fine secondary phase such as $M_23C_6$. Aging for long-time at high temperature caused the growth of martensite lath and the decrease of dislocation density resulting in the decrease in strength. However, the evolution of secondary phases had influence on hardness, yield strength and impact property. In the group A specimen aged at $600^{\circ}C\;and\;650^{\circ}C$, Laves phase was observed. The Laves phase caused the increase of the hardness and the decrease of the impact property. In addition, the abrupt growth of secondary phases caused decrease of the impact property in both A and B group specimens.

  • Conductive adhesives have been investigated for use in microelectronics packaging as a lead-free solder substitute due to their advantages, such as low bonding temperature. However, high resistivity and poor mechanical behavior may be the limiting factors for the development of conductive adhesives. The metal fillers and the polymer resins provide electrical and mechanical interconnections between surface mount device components and a substrate. As metal fillers used in conductive adhesives, silver is the most commonly used due to its high conductivity and the stability. However the cost of conductive adhesives with silver fillers is much higher than usual lead-free solders and silver has poor electro-migration performance. So, copper can be a promising candidate for conductive filler metal due to its low resistivity and low cost, but oxidation causes this metal to lose its conductivity. In this study, electrically conductive adhesives (ECAs) using surface modified copper fillers were developed. Especially, in order to overcome the problem associated with the oxidation of copper, copper particles were coated with silver, and the silver-coated copper was tested as a filler metal. Especially the effect of silver coating on the electrical resistance just after curing and after aging was investigated. As a result, it was found that the electrical resistance of ECA with silver-coated copper filler was clearly lower and more stable than that of ECA with pure copper filler after curing process. And, during high temperature storage test, the degradation rate of electrical resistance for ECA with silver coated copper filler was quite slower than that for ECA with pure copper filler.

Agastache rugosa Kuntze Attenuates UVB-Induced Photoaging in Hairless Mice through the Regulation of MAPK/AP-1 and TGF-β/Smad Pathways

  • Chronic exposure to ultraviolet (UV) radiation, regarded as a major cause of extrinsic aging or photoaging characterized by wrinkle formation and skin dehydration, exerts adverse effects on skin by causing the overproduction of reactive oxygen species. Agastache rugosa Kuntze, known as Korean mint, possesses a wide spectrum of biological properties including anti-oxidation, anti-inflammation, and anti-atherosclerosis. Previous studies have reported that A. rugosa protected human keratinocytes against UVB irradiation by restoring the anti-oxidant defense system. However, the anti-photoaging effect of A. rugosa extract (ARE) in animal models has not yet been evaluated. ARE was orally administered to hairless mice at doses of 100 or 250 mg/kg/day along with UVB exposure for 12 weeks. ARE histologically improved UVB-induced wrinkle formation, epidermal thickening, erythema, and hyperpigmentation. In addition, ARE recovered skin moisture by improving skin hydration and transepidermal water loss (TEWL). Along with this, ARE increased hyaluronic acid levels by upregulating HA synthase genes. ARE markedly increased the density of collagen and the amounts of hydroxypoline via two pathways. First, ARE significantly downregulated the mRNA expression of matrix metalloproteinases responsible for collagen degradation by inactivating the mitogen-activated protein kinase/activator protein 1 pathway. Second, ARE stimulated the transforming growth factor beta/Smad signaling, consequently raising the mRNA levels of collagen-related genes. In addition, ARE not only increased the mRNA expression of anti-oxidant enzymes but also decreased inflammatory cytokines by blocking the protein expression of nuclear factor kappa B. Collectively, our findings suggest that A. rugosa may be a potential preventive and therapeutic agent for photoaging.

  • Structural Health Monitoring (SHM) is used to provide reliable information about the structure's integrity in near realtime following extreme incidents such as earthquakes, considering the inevitable aging and degradation that occurs in operating environments. This paper experimentally investigates an integrated wireless sensor network (Wi-SN) based monitoring technique for damage detection in concrete structures. An effective SHM technique can be used to detect potential structural damage based on post-earthquake data. Two novel methods are proposed for damage detection in reinforced concrete (RC) building structures including: (i) Jerk Energy Method (JEM), which is based on time-domain analysis, and (ii) Modal Contributing Parameter (MCP), which is based on frequency-domain analysis. Wireless accelerometer sensors are installed at each story level to monitor the dynamic responses from the building structure. Prior knowledge of the initial state (immediately after construction) of the structure is not required in these methods. Proposed methods only use responses recorded during ambient vibration state (i.e., operational state) to estimate the damage index. Herein, the experimental studies serve as an illustration of the procedures. In particular, (i) a 3-story shear-type steel frame model is analyzed for several damage scenarios and (ii) 2-story RC scaled down (at 1/6th) building models, simulated and verified under experimental tests on a shaking table. As a result, in addition to the usual benefits like system adaptability, and cost-effectiveness, the proposed sensing system does not require a cluster of sensors. The spatial information in the real-time recorded data is used in global damage identification stage of SHM. Whereas in next stage of SHM, the damage is detected at the story level. Experimental results also show the efficiency and superior performance of the proposed measuring techniques.

  • As the aging of large domestic SOC facilities accelerates, facility maintenance is also changing from safety evaluation based on the current condition to performance-oriented preventive and preemptive maintenance based on the prediction of the level of future obsolescence. In particular, in the case of bridges, class 1 and 2 bridges are systematically managed along with many studies, but for small and medium-sized class 3 bridges there is no collection and utilization of historical data presenting performance degradation during their service life. Therefore, in this study, 3D model-based demonstration DB system was designed and developed to intuitively check the damage change rate at the damage location by registering the maintenance history by life cycle for each member's exterior damage in the 3D bridge object and to enable API-based comprehensive performance evaluation.

Numerical analysis of the combined aging and fillet effect of the adhesive on the mechanical behavior of a single lap joint of type Aluminum/Aluminum

  • Bonded joints have proven their performance against conventional joining processes such as welding, riveting and bolting. The single-lap joint is the most widely used to characterize adhesive joints in tensile-shear loadings. However, the high stress concentrations in the adhesive joint due to the non-linearity of the applied loads generate a bending moment in the joint, resulting in high stresses at the adhesive edges. Geometric optimization of the bonded joint to reduce this high stress concentration prompted various researchers to perform geometric modifications of the adhesive and adherends at their free edges. Modifying both edges of the adhesive (spew) and the adherends (bevel) has proven to be an effective solution to reduce stresses at both edges and improve stress transfer at the inner part of the adhesive layer. The majority of research aimed at improving the geometry of the plate and adhesive edges has not considered the effect of temperature and water absorption in evaluating the strength of the joint. The objective of this work is to analyze, by the finite element method, the stress distribution in an adhesive joint between two 2024-T3 aluminum plates. The effects of the adhesive fillet and adherend bevel on the bonded joint stresses were taken into account. On the other hand, degradation of the mechanical properties of the adhesive following its exposure to moisture and temperature was found. The results clearly showed that the modification of the edges of the adhesive and of the bonding agent have an important role in the durability of the bond. Although the modification of the adhesive and bonding edges significantly improves the joint strength, the simultaneous exposure of the joint to temperature and moisture generates high stress concentrations in the adhesive joint that, in most cases, can easily reach the failure point of the material even at low applied stresses.

  • A half portion of Korean railway bridges depends on the type of steel plate girder bridge. Since these bridges have been built in the early stage of Korean economical boom, numerous maintenance effort suffers from aging and progressive degradation issues at present. In accordance with these efforts, this paper would like to address the detailed analyses of thin steel plates with bolts in order to simulate the connection regions of steel plate girder bridge. The fundamental modal analysis, transient dynamic analysis with 3D piezoelectric element in open circuit loop and signal process with aids of TOF(time of flight) and WC(wavelet coefficient) are extensively discussed.
