Effect of Cardiopulmonary Bypass on Platelet (체외순환이 혈소판에 미치는 영향)

  • The effect of cardiopulmonary bypass on platelet count, platelet function, and bleeding time was studied in 60 patients. Platelet count was significantly reduced during and after cardiopulmonary bypass. Platelet function also had a reduced aggregation response to adenosine diphosphate. Bleeding time was prolonged to over 30 minutes during cardiopulmonary bypass and not returned to normal level until postbypass 1 hour. The amount of postoperative bleeding was proportional to the degree of decrease in platelet count and function, degree of decrease in platelet count and function. There was no significant correlation between duration of cardiopulmonary bypass and platelet count, platelet function, bleeding time, or amount of postoperative bleeding. Patients with cyanotic congenital heart disease showed a larger amount of postoperative bleeding than patients with acyanotic congenital heart disease [P<0.01], and this difference was due to the fact that platelet function was more significantly affected by cardiopulmonary bypass in cyanotic group. Patients using membrane oxygenator showed a less amount of postoperative bleeding than patients using bubble oxygenator [p<0.005] reflecting better preservation of platelet count and function by membrane oxygenator.

The Effect of Fat Diet on Inflammatory Markers and Blood Coagulation System in Rats

  • This study was undertaken to know the effect of fat diet (for eight weeks) on changes of inflammatory markers [tumor necrosis factor (TNF-${\alpha}$) and prostaglandin $E_2$ ($PGE_2$)] and blood coagulation system [platelet aggregation function (PAF), prothrombin time (PT), activated partial thromboplastin time (aPTT)] in rats. Serum TNF-${\alpha}$, $PGE_2$, biochemical markers, PAF, PT, aPTT, and body weight were measured and compared between the control (normal diet-rats) and the fat group (fat diet-rats). The weights in the fat group were higher than those of the control group. TNF-${\alpha}$, $PGE_2$, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), and creatinine levels were greater in the fat group compared with the control group. The degree of platelet aggregation was lower, whereas PT and aPTT levels were longer in the fat group than in the control group. These findings have shown that fat diet may cause inflammatory response, diabetes, liver and renal dysfunction, and disturbances of fibrinolysis and coagulation system.

Salsolinol, a tetrahydroisoquinoline-derived neurotoxin, induces oxidative modification of neurofilament-L: protection by histidyl dipeptides

  • Salsolinol (1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline) is a compound derived from dopamine metabolism and is capable of causing dopaminergic neurodegeneration. Oxidative modification of neurofilament proteins has been implicated in the pathogenesis of neurodegenerative disorders. In this study, oxidative modification of neurofilament-L (NF-L) by salsolinol and the inhibitory effects of histidyl dipeptides on NF-L modification were investigated. When NF-L was incubated with 0.5 mM salsolinol, the aggregation of protein was increased in a time-dependent manner. We also found that the generation of hydroxyl radicals (${\bullet}OH$) was linear with respect to the concentrations of salsolinol as a function of incubation time. NF-L exposure to salsolinol produced losses of glutamate, lysine and proline residues. These results suggest that the aggregation of NF-L by salsolinol may be due to oxidative damage resulting from free radicals. Carnosine, histidyl dipeptide, is involved in many cellular defense processes, including free radical detoxification. Carnosine, and anserine were shown to significantly prevent salsolinol-mediated NF-L aggregation. Both compounds also inhibited the generation of ${\bullet}OH$ induced by salsolinol. The results indicated that carnosine and related compounds may prevent salsolinol-mediated NF-L modification via free radical scavenging.

  • This paper proposes the design of protocol management system for aggregationg messages based on certification between vehicles which not only prevents the messages between vehicles from being forged and altered by Sybil attack by authenticating the them, and but also provides the efficient communication by aggregating the redundant vehicle messages which frequently happens when communicating. For this, the proposed system proposes the SKLC(Session Key Local Certificate) design which is a local certificate based on a session key, and the MAP(Message Aggregation Protocol) design which aggregates the redundant vehicle messages. Therefore, when the proposed system checks the certificate of vehicle, it provides the reliable information securely by verifying the integrity of vehicle with a hash function operation, and improves communication efficiency by reducing the processing time.

  • The quality of satellite imagery should be improved and stabilized to satisfy numerous users. The radiometric characteristics of an optical sensor can be a measure of data quality. In this study, a band aggregation technique and spectral response function of hyperspectral images are used to simulate multispectral images. EO-1 Hyperion and Landsat-8 OLI images acquired with about 30 minutes difference in overpass time were exploited to evaluate radiometric coefficients of OLI. Radiance values of the OLI and the simulated OLI were compared over three subsets covered by different land types. As a result, the index of agreement shows over 0.99 for all VNIR bands although there are errors caused by space/time and sensors.

Preparation and Characterization of Nanoparticles Using Poly(N-isopropylacrylamide)-$Poly({\varepsilon}-caprolactone)$ and Poly(ethylene glycol)-$Poly({\varepsilon}-caprolactone)$ Block Copolymers with Thermosensitive Function

  • Thermosensitive nanoparticles were prepared via the self-assembly of two different $poly({\varepsilon}-caprolactone)$-based block copolymers of poly(N-isopropylacrylamide)-b-$poly({\varepsilon}-caprolactone)$ (PNPCL) and poly(ethylene glycol)-b-$poly({\varepsilon}-caprolactone)$ (PEGCL). The self-aggregation and thermosensitive behaviors of the mixed nanoparticles were investigated using $^1H-NMR$, turbidimetry, differential scanning microcalorimetry (micro-DSC), dynamic light scattering (DLS), and fluorescence spectroscopy. The copolymer mixtures (mixed nanoparticles, M1-M5, with different PNPCL content) formed nano-sized self-aggregates in an aqueous environment via the intra- and/or intermolecular association of hydrophobic PCL chains. The microscopic investigation of the mixed nanoparticles showed that the critical aggregation concentration (cac), the partition equilibrium constants $(K_v)$ of pyrene, and the aggregation number of PCL chains per one hydrophobic microdomain varied in accordance with the compositions of the mixed nanoparticles. Furthermore, the PNPCL harboring mixed nanoparticles evidenced phase transition behavior, originated by coil to the globule transition of PNiPAAm block upon heating, thereby resulting in the turbidity change, endothermic heat exchange, and particle size reduction upon heating. The drug release tests showed that the formation of the thermosensitive hydrogel layer enhanced the sustained drug release patterns by functioning as an additional diffusion barrier.

  • This paper proposes packet aggregation scheme for considering energy consumption of mobile devices on guaranteeing quality of voice. To our purpose, we analyze VoIP service on wireless multi-hop channel in terms of delay, packet loss and energy consumption when packet aggregation scheme is applied to VoIP service. Moreover, we induce a cost function with considering of tradeoff relation between quality of voice and energy consumption.

Anticardiovascular Diseases Effects of Fermented Garlic and Fermented Chitosan

  • Garlic is a medicinal plant used throughout the world for its anti-inflammatory, antioxidant, and antiplatelet activities. Chitosan is a natural polysaccharide obtained from chitin, and derivatives of chitosan have been shown to inhibit platelet aggregation and adhesion. We hypothesized that fermented preparations of these products may possess stronger antiplatelet effects than the non-fermented forms owing to the increased bioavailability of the bioactive compounds produced during fermentation. Therefore, we compared these compounds via in vitro and ex vivo platelet aggregation assays by using standard light transmission aggregometry and ex vivo granule secretions from rat platelets. We found that fermented preparations exerted more potent and significant inhibition of platelet aggregation both in vitro and ex vivo. Likewise, ATP release from dense granules of platelets was also significantly inhibited in fermented preparation-treated rat platelets compared to that in non-fermented preparation-treated ones. We concluded that fermented preparations exerted more potent effects on platelet function both in vitro and ex vivo, possibly as a result of the increased bioavailability of active compounds produced during fermentation. We therefore suggest that fermented products may be potent therapeutics against platelet-related CVDs and can be used as antiplatelet and antithrombotic agents.

  • Intravenous lipid emulsion is used extensively as a major component of parenteral nutrition for patients in the surgical intensive care unit. Abnormal cardiovascular function related to lipid infusion has been reported although conflicting results exist. In the present study, we investigated the effects of intravenous emulsions of long-chain triglyceride (LCT) and medium-chain triglyceride (MCT) on myocardial ischemia/ reperfusion injury and on platelet aggregation in rat. There was no difference between LCT and MCT considering the effects on left ventricular developed pressure (LVDP) and coronary flow rate (CFR) before and after ischemia/reperfusion in isolated rat heart. On the other hand, a difference was found between LCT and MCT with regard to their effects on heart rate (HR) and end diastolic pressure (EDP) after ischemia/reperfusion. After ischemia/reperfusion, HR was significantly (P<0.05) reduced and EDP significantly (P<0.05) inc.eased by LCT (18$\pm$2.0% and 42.8$\pm$8.9%, respectively), but not by MCT Ex vivo platelet aggregation induced by collagen was reduced by LCT infusion, but not by MCT These findings suggest that MCT may have slightly more favorable effect than LCT on the myocardial function after ischemia/reperfusion in rat.

Structure and Function of RGD Peptides Derived from Disintegrin Proteins

  • The Arg-Gly-Asp (RGD) sequence serves as the primary recognition site in extracellular matrix proteins, and peptides containing this sequence can mimic the biological activities of matrix proteins. We have initiated structure-function studies of two RGD containing peptides, RGD-5(AGGDD) and cyclic RGD-6(CARGDDC). Assays have shown that cyclic RGD-peptides inhibit platelet aggregation more efficiently than linear ones. NMR data revealed that RGD-5 and RGD-6 have entirely different conformation. RGD-5 has a linear extended structure and RGD-6 has a stable loop conformation. In RGD-5 the guanidinium group of Arg2 and the carboxyl group of Asp4 lie in parallel, whereas the side-chains of Arg3 and Asp5 of RGD-6 are located in different planes, supporting the idea that the stability of the cyclic form derives from the packing of the side chain of the Arg and Asp residues. The structural features of these peptides could provide a basis for designing new drugs against diseases related to platelet aggregation and as cancer antagonists.
