We have introduced polyelectrolyte micro-patterning technique employing agarose plane stamp and a hard substrate having microscale features on its surface. With this method, chemically micropatterned surfaces with both positive and negative functionalities were successfully embedded in well-defined microstructures, and selective impartment of charge functionalities was confirmed by patterning bead bearing surface charge. Furthermore, this technique allows highly sensitive immobilization of protein onto targeted surface simply by endowing functionalities, which extends the potential of its use as a tool for high-throughput protein microarray and proteomics. Because plane agarose stamp is free of structures on its surface, there is no concern for pattern collapse, and the combination of agarose plane stamp with patterned substrate is more suited for selective protein patterning compared with adopting surface-patterned agarose stamp with flat substrate. Our technique using agarose plane stamp and a substrate having microscale features on its surface suggests a range of possible applications, including the micropatterning of biofunctionalized copolymer having polyelectrolyte block, immobilization of micro- and nanoparticle with biofunctionalities such as biotin and streptavidine, and establishing optoelectronic microstructures with micro-beads on various surfaces.
For deciphering the cyclic guanosine monophosphate (cGMP) signaling pathway, we employed chemical proteomics to identify the novel target molecules of cGMP. We used cGMP that was immobilized onto agarose beads with linkers directed at three different positions of cGMP. We performed a pull-down assay using the beads as baits on tissue lysates and identified 9 proteins by MALDI-TOF (Matrix-Assisted Laser Desorption/Ionization Time-of-Flight) mass spectrometry. Some of the identified proteins were previously known cGMP targets, including cGMP-dependent protein kinase and cGMP-stimulated phosphodiesterase. Surprisingly, some of the co-precipitated proteins were never formerly reported to associate with the cGMP signaling pathway. The competition binding assays showed that the interactions are not by nonspecific binding to either the linker or bead itself, but by specific binding to cGMP. Furthermore, we observed that the interactions are highly specific to cGMP against other nucleotides, such as cyclic adenosine monophosphate (cAMP) and 5'-GMP, which are structurally similar to cGMP. As one of the identified targets, MAPK1 was confirmed by immunoblotting with an anti-MAPK1 antibody. For further proof, we observed that the membrane-permeable cGMP (8-bromo cyclic GMP) stimulated mitogen-activated protein kinase 1 signaling in the treated cells. Our present study suggests that chemical proteomics can be a very useful and powerful technique for identifying the target proteins of small bioactive molecules.
인체 혈액 응고 9인자는 간에서 생성되며 461개의 아미노산으로 구성된 당 단백질이다. 따라서 인체 혈액 응고 9인자 cDNA를 찾기 위해 태아의 간(fetal liver) cDNA library를 PCR(Polymerase Chain reaction) 방법으로 screening하였으며, 그 결과 ATG개시 코돈으로부터 TAA종료 코돈까지 포함하는 1.4 kb의 9인자 cDNA를 찾았다. 또한 클론된 9인자 cDNA를 박테리아에서 발현시키기 위해 박테리아 발현 벡터인 pGEX-2T 플라스미드에 클로닝하므로써 pGEX-F9 플라스미드를 제조하였다. pGEX-F9로 형질전환된 E. coli에서 PGEX-F9의 발현을 유도하면 73 kDa 크기의 GST-factor9 융합 단백질이 다량생성되며 , 이 단백질이 혈액 응고 9인자 단백질을 함유하는 융합 단잭질임을 혈액 응고 9인자 항체를 이용한 Western blot으로 입증하였다. E. coli에서 발현된 GST-factor 9 융합 단백질은 전체 단백질의 약 20%를 차지하며 GST agarose bead를 이용한 one step purificarion 방법을 통해 GST-factor9 융합 단백질을 쉽게 분리 할 수 있다.

A dense pellicular solid matrix has been fabricated by coating 4% agarose gel on to dense zironia-silica(ZS) spheres by watr-in-oil emulsification . The agarose evenly laminated the ZS bead to a depth of 30㎛, and the resultin gpellicular assembly was characterised by densities up to 2.39g/mL and a mean particle dimeter of 136 ㎛. In comparative fluidisation tests, the pellicular solid phase exhibited a two-fold greater flow velocity than commercial benchmark ad-sorbents necessary to achieve common values of bed expansion. Furthermore, the perlicular parti-cles were characterised by improved qualities of chromatographic behaviour, particularly with re-spect to a three-fold increase in the apparent effective diffusivity of lysozyme within a pellicular assembly modified with Cibacron Blue 3GA. The properties of rapid protein adsorption/desorp-tion were attributed to the physical design and pellicular deployment of the reactive surface in the solid phase. When combined with enhanced feedstock throughput, such practical advantages recommend the pellicular assembly as a base matrix for the selective recovery of protein products from complex, particulate feedstocks(whole fermentation broths, cell disruptates and biological extracts).
Glutathione S-transferase는 식물의 해독작용에 중추적인 역할을 하는 화학 효소이다. 본 연구에서는 제초제 검출 키트 개발에 응용을 위하여 식물 해독작용에 중추적인 역할을 하는 glutathione Stransferase의 고정화 방법을 연구하였다. Chloroacetanilide계 제초제에 높은 효소 활성을 보이는 벼 유래 OsGSTF3에 공유결합을 통한 polystyrene-alkylamine 비드와 리간드결합을 통한 agarose-aminoalkyl 비드,포괄법을 통한 Na-alginate 비드를 이용하여 고정화를 실시하였다. 정제된 OsGSTF3 10 mg을 사용하여 고정화 하였을 때 0.62 mg/g 비드로 polystyrene-alkylamine에 가장 효율적으로 고정화 되었다. 고정화 된 OsGSTF3의 효소 활성은 야생형의 30%를 나타내었으며, 재사용에 의한 효소활성 측정시 3회 까지 처음 활성의 80% 이상을 유지하였다.
국내에서 시판되는 감자와 수입 감자스낵류로부터 상용 DNA 추출 kit 및 CTAB-phenol/chloroform 추출법등을 이용하여 시료특성에 따른 genomic DNA를 추출방법을 선정하고 PCR 정성검사를 실시하였다. 생감자의 경우 STE 용액으로 과량의 전분을 제거한 다음 DNA를 추출한 경우 순도 높은 DNA를 추출할 수 있었으며 상용 추출 kit를 이용한 경우 lysis buffer와 함께 ${\alpha}-/{\beta}$-amylase를 각각 또는 혼합으로 처리하거나 추출된 DNA 용액에 마지막 단계에서 효소를 처리한 시료군에서 고순도의 DNA를 추출할 수 있었으며, 효소 처리군에서는 ${\alpha}-/{\beta}$-amylase를 혼합으로 처리한 경우에 DNA 추출수율이 높았다. 냉동가공감자의 경우 silica-coated bead법을 이용하여 효소를 처리한 경우와 CTAB-페놀 클로로포름 처리군에서 DNA가 추출되었다. 또한, 각 방법으로 추출한 DNA에 대하여 감자의 내인성 유전자인 Pss 프라이머를 사용하여 PCR을 한 결과 모든 시료에서 추출된 DNA에 대하여 내부표준유전자 증폭산물이 검출되었다. 고도의 가공처리를 거친 수입 감자스낵(fabricated potato chips)과 냉동가공 감자(frozen fried potato) 등은 계면활성제인 CTAB(cetyl trimethyl ammonium bromide)과 페놀-클로로포름 혼합액을 이용하여 추출하고 이를 template로 하여 PCR 증폭을 실시하였다. 그 결과, Fig. 8에 제시한 바대로 감자의 내인성 유전자인 Pss 특이적 산물인 216bp의 산물이 냉동감자가공품과 감자칩에서 검출되었으며 재조합유전자인 New leaf plus 유래의 증폭산물(234bp)와 New lear Y유래의 증폭산물(225bp)는 검출되지 않았다. 본 실험의 결과 시료의 가공특성과 적용한 추출 kit 및 방법에 따라 genomic DNA 순도 및 추출수율이 크게 차이가 났으며 이것이 결국 PCR 결과에 의음성 혹은 의양성 등에 영향을 미치게 될 것으로 판단된다. 또한, 동일한 DNA추출방법에 의해서도 DNA가 추출되지 않은 경우가 있어서 동일한 시료에서 2회 반복 추출하는 것이 의음성결과를 피할 수 있는 방법으로 판단된다.
연구배경 : 결핵은 아직도 한국에서는 중요한 질병의 하나이지만 결핵을 진단하기 위한 보편적인 방법들이 단점인 예민도와 검사기간등이 문제가 되고 있다. 최근 분자유전학에 PCR 기법이 도입된 이래 결핵균의 DNA를 추출하여 짧은 시간내에 결핵을 진단하고자 하는 많은 보고들이 있어 왔다. 그러나 보고된 방법들의 다양성과 각 방법에 따른 소요시간등이 매우 다른 것이 문제가 되고 있고 임상에서 빠른 시간내에 비교적 간단한 방법으로 결핵율의 DNA를 분리한다면 보다 빨리 결핵을 진단하여 치료를 할 수 있다고 생각되어 결핵균의 DNA를 분리하는 방법중 가장 좋은 방법을 확인하고자 하였다. 방법 : 결핵율의 DNA를 추출하는 방법은 총 6가지의 방법으로서 SDS-microwave oven method, NaOH lysis method, Triton X-100-proteinase K method, Lysis buffer method, SDS-proteinase K method 및 Bead beater method를 사용하여 비교하였으며 사용된 균주는 Mycobacterium tuberculosis $H_{37}Rv$ strain 이었고 임상가검물중 도말양성 객담 5예 및 결핵성 뇌막염이 의심되고 도말음성이었으나 항결핵제 투여로 완치판정을 받은 환자의 뇌척수액 5예를 대상으로 하였다. 사용된 primer는 IS6110의 123bp를 target DNA를 하였고 PCR반응은 각각 $94^{\circ}C,\;68^{\circ}C$ 및 $72^{\circ}C$ 로 각각 2분씩 총 30주기를 시행하였다. 결과 : $H_{37}Rv$ strain에서 각 방법으로 추출된 DNA를 10배씩 serial dilution하여 PCR을 시행한 결과 SDS-microwave oven법과 NaOH lysis법은 50 fg까지 양성이었으며 나머지 4가지 방법에서는 5fg까지 양성이었다. 또한 각각의 방법으로 도말양성 환자의 객담에서 PCR을 시행한 결과 전예에서 양성이었고 결핵성 뇌막염이 의심스러운 환자의 뇌척수액에서는 SDS-microwave법과 NaOH lysis법으로는 5예중 3예에서 양성이었고 나머지 4가지 방법으로는 5예중 4예에서 양성이었다. 결론 : 이상의 실험으로 경제적이고 간편한 측면에서 볼 때 SDS-proteinase K법이 가장 좋은 방법임을 알 수 있었다.
IFN-${\gamma}$의 대량생산을 위한 기초연구로서 IFN-${\gamma}$의 아미노 말단에 glucagon과 ferritin을 융합파트너로 각각 결합시켜 재조합 IFN-${\gamma}$의 발현을 유도하였다. 대장균 내에서 발현되는 IFN-${\gamma}$는 그 자체로 매우 강한 소수성 결합의 양상을 나타내어 inclusion body 형태로 발현된다고 알려져 있으나 OrigamiTM(DE3) 균주로부터 50% 이상의 수용성 형태로 발현시켰다. IFN-${\gamma}$로부터 융합파트너를 제거할 수 있는 system을 개발하기 위해 융합파트너와 IFN-${\gamma}$ 사이에 enterokinase cleavage site를 도입하였으며, enterokinase에 의해 IFN-${\gamma}$에는 영향을 미치지 않고 효과적으로 융합파트너를 제거할 수 있었다. 재조합 IFN-${\gamma}$의 분리 및 정제를 위해 발현벡터상의 융합파트너와 IFN-${\gamma}$사이에 6X His-tag을 도입하였고 융합파트너의 N-말단에도 6X His-tag을 추가적으로 도입함으로써 융합파트너와 더불어 enterokinase에 의해 분해되지 않은 융합단백질을 Ni-NTA agarose column으로 제거함으로서 IFN-${\gamma}$를 완전 정제할 수 있었다. IFN-${\gamma}$의 발현을 유도하는 발현유도체로서 15 mM lactose를 이용하여 5 L 발효조에서 IFN-${\gamma}$의 발현을 검토한 결과, 재조합 균체의 단위 건조질량(dry cell weight, g)으로 약 11 g DCW/L 수준의 재조합 융합단백질을 얻을 수 있었다.
