Nutritional regulation of gene expression associated with growth and feeding behavior in avian species can become an important technique to improve poultry production according to the supply of nutrients in the diet. Insulin-like growth factor-I (IGF-I) found in chickens has been characterized to be a 70 amino acid polypeptide and plays an important role in growth and metabolism. Although it is been well known that IGF-I is highly associated with embryonic development and post-hatching growth, changes in the distribution of IGF-I gene expression throughout early- to late-embryogenesis have not been studied so far. We revealed that the developmental pattern of IGF-I gene expression during embryogenesis differed among various tissues. No bands of IGF-I mRNA were detected in embryonic liver at 7 days of incubation, and thereafter the amount of hepatic IGF-I mRNA was increased from 14 to 20 days of incubation. In eyes, a peak in IGF-I mRNA levels occurred at mid-embryogenesis, but by contrast, IGF-I mRNA was barely detectable in the heart throughout all incubation periods. In the muscle, no significant difference in IGF-I gene expression was observed during different stages of embryogenesis. After hatching, hepatic IGF-I gene expression as well as plasma IGF-I concentration increases rapidly with age, reaches a peak before sexual maturity, and then declines. The IGF-I gene expression is very sensitive to changes in nutritional conditions. Food-restriction and fasting decreased hepatic IGF-I gene expression and refeeding restored IGF-I gene expression to the level of fed chickens. Dietary protein is also a very strong factor in changing hepatic IGF-I gene expression. Refeeding with dietary protein alone successfully restored hepatic IGF-I gene expression of fasted chickens to the level of fed controls. In most circumstances, IGF-I makes a complex with specific high-affinity IGF-binding proteins (IGFBPs). So far, four different IGFBPs have been identified in avian species and the major IGFBP in chicken plasma has been reported to be IGFBP-2. We studied the relationship between nutritional status and IGFBP-2 gene expression in various tissues of young chickens. In the liver of fed chickens, almost no IGFBP-2 mRNA was detected. However, fasting markedly increased hepatic IGFBP-2 gene expression, and the level was reduced after refeeding. In the gizzard of well-fed young chickens, IGFBP-2 gene expression was detected and fasting significantly elevated gizzard IGFBP-2 mRNA levels to about double that of fed controls. After refeeding, gizzard IGFBP-2 gene expression decreased similar to hepatic IGFBP-2 gene expression. In the brain, IGFBP-2 mRNA was observed in fed chickens and had significantly decreased by fasting. In the kidney, IGFBP-2 gene expression was observed but not influenced by fasting and refeeding. Recently, we have demonstrated in vivo that gizzard and hepatic IGFBP-2 gene expression in fasted chickens was rapidly reduced by intravenous administration of insulin, as indicated that in young chickens the reduction in gizzard and hepatic IGFBP-2 gene expression in vivo stimulated by malnutrition may be, in part, regulated by means of the increase in plasma insulin concentration via an insulin-response element. The influence of dietary protein source (isolated soybean protein vs. casein) and the supplementation of essential amino acids on gizzard IGFBP-2 gene expression was examined. In both soybean protein and casein diet groups, the deficiency of essential amino acids stimulated chickens to increase gizzard IGFBP-2 gene expression. Although amino acid supplementation of a soybean protein diet significantly decreased gizzard IGFBP-2 mRNA levels, a similar reduction was not observed in chickens fed a casein diet supplemented with amino acids. This overview of nutritional regulation of IGF-I and IGFBP-2 gene expression in young chickens would serve for the establishment of the supply of nutrients to diets to improve poultry production.
E. coli의 PheA 단백질은 chorismate mutase and prephenate dehydratase (CMPD) 활성을 가지며 마지막 산물인 페닐알라닌에 의하여 되먹임제어가 되는 생합성 경로의 주요 조절 효소 중의 하나이다. 그러므로, 이 PheA 단백질은 필수 아미노산 중의 하나인 페닐알라닌의 대량 생산에 이용하기 위한 단백질 공학의 타겟이 될 수 있다. 이러한 목적으로 PheA 단백질의 마지막 생산물인 페닐알라닌에 의한 되먹임저해 저항성 유전자원을 선별하였다. 이 유전자의 산물인 $PheA^{FBR}$은 118번째 류신이 페닐알라닌으로 치환되었고, 기질인 prephenate에 대한 친화도가 야생주단백질과 비교하여 약 3.5배 정도 높았다. $PheA^{FBR}$은 세포내에서 축척되어져 되먹임저해를 하는 페닐알라닌 농도에서(약1 mM와 10 mM)에서도 50%와 40%의 활성을 유지 하고 있었고, 페닐알라닌 존재하에서 기질의 결합 성향이 협동적(cooperative) 모드에서 단독적(hyperbolic) 모드로 전환되었다. 이는 기존 연구와 비교해 볼 때, 이 돌연변이 부위는 이 융합기능 효소인 PheA 단백질의 새로운 조절 부위의 존재를 암시 한다. 효소 동력학적 결과는 PheA 단백질의 되먹임저해 저항성 획득이 아미노산 돌연변이에 의한 단백질 구조의 변화 유도에 의한 것으로 생각된다. 더 나아가, 본 연구에서 선별된 돌연변이 유전자는 생물전환법을 이용한 필수아미노산 생산에 산업적으로 응용 가능성이 있다.
본 연구에서는 (E)-2-(substituted benzylidene)-2,3-dihydro-1H-cyclopenta[a]naphthalen-1-one 유도체들을 합성했으며, 합성된 유도체들이 멜라닌 생성과정의 주요 효소인 tyrosinase 활성을 저해할 수 있는지에 대하여 확인하였다. 19종류의 유도체들의 tyrosinase 저해활성을 측정해본 결과, MHY3655 ($IC_{50}=0.1456{\mu}M$)가 가장 큰 저해활성을 나타냈으며 이는 대조군인 코직산($IC_{50}=17.2{\mu}M$) 보다 큰 효능을 보였다. 게다가, Lineweaver-Burk 분석법을 이용하여 MHY3655가 경쟁적 저해 기전으로 tyrosinase 활성을 저해하였고, docking simulation으로 MHY 3655가 tyrosinase에 직접 결합함을 재확인하였다. 마지막으로, B16F10 melanoma 세포에서 MHY3655의 세포독성을 평가한 결과 $1-20{\mu}M$ 사이의 MHY3655는 세포독성을 나타내지 않았다. 이상의 결과에서, 신물질 MHY3655은 우수한 tyrosinase 저해활성을 나타내며, 이는 미백 화장품 소재로서 활용할 가치가 있으리라 사료된다.
본 연구는 황금찰수수의 에탄올 추출물을 이용하여 항산화 활성 및 뉴라니미데이즈 억제 효과를 조사하였다. 헥산, 에틸아세테이트, 메탄올과 70% 에탄올 조추출물에서 항산화력의 IC50값을 비교한 결과 70% 에탄올 추출물이 DPPH 라디칼 소거능, ABTS 라디칼 소거능에서 각각 83.2±2.7, 85.6±2.4 μg/ml으로 가장 높았고, 뉴라니미데이즈 억제활성 IC50값을 비교한 결과 메탄올 추출물에서 1.8±0.1 μg/ml로 나타났다. 또한 황금찰수수 추출물에서 컬럼크로마토그래피와 UPLC-PDA-MS/MS 분광기 분석을 통해, Gallic acid (1), Protocatecuic acid (2), p-Hydroxy benzoic acid (3), Vanillic acid (4), Caffeic acid (5), Ferulic acid (6), Luteolinidin (7), Apigeninidin (8), Luteolin (9), 총 9종의 폴리페놀 화합물을 확인하였다. 또한 각각의 화합물에 대한 항산화력의 IC50값을 비교한 결과 Luteolinidin이 DPPH 라디칼 소거능, ABTS 라디칼 소거능에서 각각 10.9±0.5, 8.6±3.0 μM로 가장 우수 하였고, 뉴라니미데이즈 억제활성은 Luteolin이 12.9±3.8의 IC50값과 비경쟁적 저해모델을 보였다. 결과적으로 황금찰수수는 높은 항산화 효과와 뉴라미니데이즈 억제활성을 보여 식품, 사료 등의 새로운 기능성소재로 다양하게 활용될 수 있음을 시사한다.
질병과 밀접한 관계가 있는 hCGL 단백질의 경우 대량 배양 시 유도체를 사용하지 않아도 발현이 되는 점과 유전자 측면에서 조작이 쉬운 E.coli를 이용하여도 발현이 된다는 점에 있어서 중요한 이점을 가지고 있다. 본 연구에서는 배양되는 온도와 발현에 관련 있는 유도체의 농도, 600 nm에서의 균 성장 정도에 따른 유도체의 첨가 그리고 배지의 양을 조절하면서 유입되는 aeration의 조건으로 hCGL 단백질 발현의 최적의 조건 확립을 목적으로 하였다. 또 각 발생되는 inclusion body의 양을 측정하면서 보다 많은 가용성 단백질을 발현시키는 조건을 확립하고자 하였다. hCGL 단백질은 저온에서 보다 많은 양의 단백질이 발현되며 inhibitor의 억제를 담당하는 유도체의 농도와는 상관없이 발현이 되었다. 또한 균의 성장 정도에 따라 유도체의 첨가시기를 달리 하였을 때, 발현 비율에 차이는 있었으나 전체적인 단백질 양과 비교해 보면, 이는 hCGL 발현에 큰 영향을 미치지 않는다. 배지의 양을 달리하여 살펴본 aeration에 따른 hCGL 발현 정도는 배지의 부피가 15%일 때 높은 aeration으로 균의 양은 많았으나 목적 단백질인 hCGL의 발현은 aeration이 되지 않는 조건에서 더 잘되는 것을 확인하였다. 그리고 His-TEV-hCGL의 활성은 야생형 hCGL의 활성을 기준으로 하였을 때, L-cystathionine을 기질로 하였을 경우 76%, L-cysteine을 기질로 하였을 경우 88% 수준으로 유사한 활성을 나타내었고, 이는 손쉽게 정제 가능한 His-TEV-hCGL을 야생형을 대신하여 사용할 수 있음을 시사한다. 또한 His-TEV-hCGL이 야생형 hCGL과 같이, 427 nm에서 흡광을 가지는 것으로 보아 보효소PLP를 포함하고 있음을 알 수 있었다. 이로써 homocysteine 대사연구에 필수적인 hCGL 효소를 다량 얻는 방법을 확립하고, 관련 연구에 기여하리라 사료된다.
멜라닌 색소는 포유동물의 피부, 머리카락, 눈, 신경계에 풍부하게 존재한다. 멜라닌은 다양한 환경적 스트레스로부터 피부를 보호하며, 생리학적 산화-환원 완충 작용을 통해 항상성을 유지한다. 그러나, 과도한 멜라닌 축적은 간반, 주근깨, 노인성 흑자, 염증성 색소침착을 일으킬 수 있다. 티로시나아제는 멜라닌의 생합성 경로 조절에 아주 중요한 역할을 하는 것으로 알려져 있다. 티로시나아제의 활성을 저해하는 다양한 미백제가 개발되었지만 알러지, DNA손상, 세포독성, 돌연변이 유발 등을 야기하는 부작용으로 인해 임상적 적용이 제한되었다. 본 논문에서 여러 4-크로마논 유도체를 합성하여 티로시나아제 억제 활성을 조사하였다. 이들 화합물 중 MHY1294는 IC50가 5.1±0.86 μM으로 양성 대조군인 코직산(14.3±1.43 μM) 보다 나은 티로시나아제 효소 억제 활성을 나타냈다. 또한 MHY1294는 티로시나아제의 촉매 부위에서 경쟁적인 억제 작용을 보였으며 코직산보다 더 큰 기질 결합 친화성을 가지는 것으로 확인되었다. 뿐만 아니라, MHY1294는 B16F10 흑색종 세포에서 멜라닌 세포 자극 호르몬 (α-MSH)에 의해 유도되는 멜라닌 합성과 세포 내 티로시나아제 활성을 유의적으로 억제하였다. 결론적으로 본 연구는 MHY1294가 과도한 멜라닌 축적에 대한 약물 제제 및 미백제로서의 개발 가능성이 있음을 시사한다.
Background: Ginsenoside compound K (CK), the main active metabolite in Panax ginseng, has shown good safety and bioavailability in clinical trials and exerts neuroprotective effects in cerebral ischemic stroke. However, its potential role in the prevention of cerebral ischemia/reperfusion (I/R) injury remains unclear. Our study aimed to investigate the molecular mechanism of ginsenoside CK against cerebral I/R injury. Methods: We used a combination of in vitro and in vivo models, including oxygen and glucose deprivation/reperfusion induced PC12 cell model and middle cerebral artery occlusion/reperfusion induced rat model, to mimic I/R injury. Intracellular oxygen consumption and extracellular acidification rate were analyzed by Seahorse multifunctional energy metabolism system; ATP production was detected by luciferase method. The number and size of mitochondria were analyzed by transmission electron microscopy and MitoTracker probe combined with confocal laser microscopy. The potential mechanisms of ginsenoside CK on mitochondrial dynamics and bioenergy were evaluated by RNA interference, pharmacological antagonism combined with co-immunoprecipitation analysis and phenotypic analysis. Results: Ginsenoside CK pretreatment could attenuate mitochondrial translocation of DRP1, mitophagy, mitochondrial apoptosis, and neuronal bioenergy imbalance against cerebral I/R injury in both in vitro and in vivo models. Our data also confirmed that ginsenoside CK administration could reduce the binding affinity of Mul1 and Mfn2 to inhibit the ubiquitination and degradation of Mfn2, thereby elevating the protein level of Mfn2 in cerebral I/R injury. Conclusion: These data provide evidence that ginsenoside CK may be a promising therapeutic agent against cerebral I/R injury via Mul1/Mfn2 mediated mitochondrial dynamics and bioenergy.
연구배경 : NF-${\kappa}B$는 많은 염증 유발성 물질들을 발현시키는데 필요한 전사 인자로서, 염증성 폐질환 발병에 관여한다는 사실이 확인되었다. 이러한 NF-${\kappa}B$의 활성화에는 여러 신호전달 체계가 관여한다는 사실이 밝혀지고 있으며 최근 PI3K/Akt 경로도 NF-${\kappa}B$ 활성화에 관여한다는 연구 결과가 보고되고 있으나, 실험 대상 세포주마다 활성화 기전이 다르고 호흡기 상피세포에 대한 결과도 알려져 있지 않아 호흡기 상피세포에서의 NF-${\kappa}B$ 활성화에 PI3K/Akt 경로가 관여하는지를 밝히기 위하여 본 연구를 시행하게 되었다. 방법 : 인체 기관지 상피세포주인 BEAS-2B와 폐암 세포주인 A549, NCI-H157을 사용하여 Akt 활성화와 $I{\kappa}B{\alpha}$ 분해 여부를 확인하기 위해 western blot을 시행하였다. Wortmannin, LY294002 및 DN-Akt를 이용하여 Akt 경로를 억제하였고, NF-${\kappa}B$ 활성화와 전사 활성을 측정하기 위하여 각각 EMSA와 luciferase assay를 시행하였다. 결과 : BEAS-2B, A549 및 NCI-H157 세포주에 TNF-$\alpha$ 및 insulin을 처리한 경우 Akt 활성화가 유도되었다. Insulin 으로 Akt 경로를 활성화시킨 경우 $I{\kappa}B{\alpha}$ 분해가 일어나지는 않았다. Wortmannin, LY294002 및 DN-Akt 를 이용하여 Akt 경로를 억제한 경우 TNF-$\alpha$에 의한 $I{\kappa}B{\alpha}$ 분해 및 IKK 활성화가 억제되지는 않았으며, NF-${\kappa}B$ 활성화도 억제되지 않았다. Wortmannin을 처리한 경우 TNF-$\alpha$에 의한 NF-${\kappa}B$ 전사 활성이 오히려 증가하는 양상을 보였으나, DN-Akt 이입시킨 경우에는 관찰되지 않았다. 결론 : 인체 호흡기 상피세포에서는 $I{\kappa}B$/NF-${\kappa}B$ 경로의 활성화는 PI3K/Akt 경로와 무관한 것으로 판단된다.
