대전 대덕구 출토 복식 유물에 부착된 세균의 종류 및 섬유소분해효소 활성 (Bacterial Strains and Their Cellulase Activity from the Excavated Clothes at Daedeok-gu, Daejeon)

  • This study was aimed to isolation and identification of attached bacteria on the clothes excavated from Deajeon area dating on the 16th century. From the observation with colony shape, 17 bacterial strains were isolated, and then 7 bacterial strains were identified with morphological and biochemical characteristics. Streptococcus sp., Alcaligenes faecalis, Gemella sp., Acinetobacter sp., Pseudomonas vesicularis, Aeromonas sain. salmonicida, Moraxella spp. In observation of the bacterial strains by the sort of textile, more bacterial strains were found in silk, cotton, and cotton batt than in ramie and hemp. It is suggest that hemp has antibacterial characteristics due to the presence of lignin. In the comparison washed samples with unwashed ones, there were more kinds of bacterial strains in washed samples. In the cellulase activity tests, all isolated bacteria had low level cellulase activity.

정수처리용 생물활성탄(BAC) 부착 박테리아를 이용한 회분식 반응기에서의 Geosmin 생분해 특성 (Characteristics of Biodegradation of Geosmin using BAC Attached Bacteria in Batch Bioreactor)

  • 3가지 재질의 생물활성탄 부착 박테리아들을 부리 동정한 결과 총 9종류의 부착 박테리아가 동정되었다. Pseudomonas 속이 차지하는 비율이 평균 56.5%로 나타나 가장 높은 우점비율을 나타내었고, 다음으로 Pasteurella속 18.9%, Chryseomonas 속 4.0%, Agrobacterium속 3.5%, Aeromonas속 2.0% 순으로 검출되었다. 순수 분리된 9종의 박테리아들의 성장곡선을 조사한 결과 24~96시간 내에 최대의 생체량을 나타내어 geosmin을 유기탄소원으로 활용하는 능력이 뛰어난 것으로 조사되었다. $4^{\circ}C$$25^{\circ}C$의 운전조건에서 geosmin에 대한 생분해능을 조사한 결과 Pseudomonas vesicularis, Pseudomonas fluorescens, Agrobacterium radiobacter 및 Stenotrophomonas maltophilia 등이 뛰어난 생분해율을 나타낸 반면 Chryseomonas luteola, Spingomonas paucimobilis, Spirillum spp. 등은 비교적 낮은 geosmin 생분해능을 나타내었다. Geosmin의 생분해능은 수온이 $4^{\circ}C$일 경우 생분해율 속도상수가 $0.00006{\sim}0.00020\;hr^{-1}$의 범위에서 $25^{\circ}C$에서는 $0.0043{\sim}0.0046\;hr^{-1}$의 범위로 나타나 수온 상승에 따라 큰 폭으로 증가하였으며, 또한 투입된 geosmin의 농도가 10~10,000 ng/L로 증가할수록 생분해율 속도상수도 $0.0003{\sim}0.0882\;hr{-1}$로 증가하였다.

Inhibition of Clinical Nosocomial Bacteria by Chlorine Dioxide

  • Chlorine dioxide is an effective chemical to inhibit the growth of bacteria and viruses or to disinfect infected areas. In this study, the effects of chlorine dioxide on several bacteria in hospitals were analyzed. Alloiococcus otitis, Kocuria rosea, Leuconostoc mesenteroides spp. and Staphylococcus lentus as gram-positive bacteria and Acinetobacter lwoffii, Aeromonas salmonicida, Brucella melitensis, Oligella ureolytica as gram-negative bacteria were done for the inhibitory analysis. The growth and morphology of the bacteria were analyzed by placing a plastic stick which was called "FarmeTok (medistick/Puristic)" provided by Purgofarm, co, Ltd. to release ClO2 (13 ppmv/hr) next to the plate where the bacteria were incubated for 24 hours. Less than 10 bacterial colonies were evaluated as having 99% inhibitory effect. The initial bacterial culture concentration of 0.5 McFaland turbidity was good for analyzing the chlorine dioxide inhibitory effect. All bacteria could be easily counted post 24 hr co-incubation with ClO2, but A. otitis and A. lwoffii without ClO2 gas were not countable due to very dispersed colony types which were not affected for result analysis. As shown in this study, the FarmeTok plastic stick, which discharges chlorine dioxide at 13 ppmv / hour, was evaluated to be sufficient to suppress the above bacteria in the hospital. Bacteria existing in the clinic such as this hospital will be used as a data to inhibit the growth of bacteria by using ClO2, and molecular biology analysis using the gene of bacteria will be possible in the future rather than inhibiting the growth of bacteria itself.

Experimental Analysis of Interactions Among Saprotrophic Fungi from A Phosphorous-Poor Desert Oasis in the Chihuahuan Desert

  • Fungal ecological interactions play a key role in structuring community assemblages. These associations may involve both antagonistic and synergistic relationships, which are commonly influenced by abiotic factors such as nutrient conditions. However, information for extreme, oligotrophic systems remain poor. Herein, interactions among key members of the aquatic transient fungal community (Aspergillus niger, Cladosporium sp., and Coprinellus micaceus) of a low-nutrient freshwater system in the Cuatro Ci enegas Basin, Mexico were studied. Pairwise interaction bioassays were explored in vitro under different nutrient conditions, including carbohydrates-rich, carbohydrates and amino peptides-rich, and low nutrients. Our results indicated that antagonistic patterns prevail among the studied taxa. However, nutrient-dependent changes were observed in Cladosporium sp. shifting to synergy under carbohydrates-rich conditions, suggesting changes in the fungal community composition as a result of nutrient enrichment. Remarkably, our findings contrast with previous work demonstrating mainly synergistic interactions between our tested fungal isolates and co-occurring autochthonous bacteria (Aeromonas spp. and Vibrio sp.) under low-nutrient conditions. This observation may indicate that bacteria and fungi exhibit distinct community-level responses, driven by nutrient conditions. This contributes to the knowledge of fungal community dynamics and interspecific interactions in an oligotrophic ecosystem, highlighting the relevance of nutrient-based shifts and antagonistic interactions in ecosystem dynamics.
