Effect of Salinity Stress on Growth, Yield, and Proline Accumulation of Cultivated Potatoes (Solanum tuberosum L.) (염 스트레스에 따른 감자 품종 (Solanum tuberosum L.) 간 생육, 수량 및 proline 함량 변이)

  • This study evaluated the responses of 18 potato cultivars to three levels of salinity stress (electrical conductivity, EC: 1.0, 4.0, and $8.0dS{\cdot}m^{-1}$). Stem, leaf, root, chlorophyll, tuber yield, and proline content were investigated and statistically analyzed using analysis of variance (ANOVA) and correlations. Stem number and stem diameter were not affected by salinity, but stem length and aerial weight showed highly significant responses to salinity. Aerial weight decreased with increasing salinity levels in most cultivars, while it increased in some the cultivars 'Daejima', 'Goun', 'Haryeong', and 'LT-8'. Leaf number, leaf area index, and leaf weight were most significantly affected by salinity and the cultivar ${\times}$ salinity interaction. Root length, root weight, total chlorophyll and chlorophyll a were affected by salinity, but not by the cultivar ${\times}$ salinity interaction. The opposite trend was shown in chlorophyll b. Although there was great variability among cultivars, tuber yield decreased in all cultivars, and was most significantly influenced by salinity and the cultivar ${\times}$ salinity interaction. 'Superior', 'Kroda', 'Romana', and 'Duback' gave better tuber yields under salinity at EC 4.0 and $8.0dS{\cdot}m^{-1}$ than the cultivars with better aerial weights. Proline content was increased by salinity in all cultivars, and was more remarkable in the cultivars with better aerial weights than in cultivars such as 'Superior' and 'Kroda' with better tuber yields. Leaf number, leaf area index, leaf weight, and root length parameters were considered to be useful criteria in the evaluation of salt tolerance because of their high positive correlation with tuber yield; however, given its negative correlation with tuber yield under high salinity, proline content was not. Salinity tolerances varied greatly among potato cultivars. The low correlation between growth and yields of aerial parts under high salinity suggests that, in commercial agriculture, it might be more practical to compare relative yields to controls. Additionally, 'Superior', 'Kroda', 'Romana', and 'Duback' might be very useful cultivars to use in breeding programs to develop salinity-tolerant potatoes, as well as for sustainable potato production in saline areas.

Effects of Mulching Materials on Growth and Yield of Atractylodes macrocephala Koidz (피복재료(被覆材料)가 백출(白朮)의 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • This study was conducted to investigate the soil mulching effect on growth and yield of Atractylodes macrocephala Koidz. The number of stem, stem diameter, and fresh weight of aerial part in black poly-ethylene film and transparent poly-ethylene film mulched condition were higher than those of non-mulching and rice-straw mulching. Number of flowers were high in the order of non-mulching, rice-straw, trans-P. E. film, and black P. E. film. Yield of underground part was higher in mulching condition than that of non-mulching. Dry rhizome yield in black P. E. film mulching was significantly high to compare with the other treatments. There was lower disease incidence of Phytophthora blight in trans-P. E and black P. E. film compare with the other treatments but any difference was not observed in essential oil content of rhizome. Significant positive correlation was found between rhizome weight and fresh weight of aerial part. It was also shown that the fresh weight of aerial part was greatly dependent on the number of stems and stem diameter.

First Report of Potato Stem-End Rot Caused by Fusarium oxysporum in Korea

  • In this study, we identified the causative agent of stem-end rot in potatoes that were grown in Gangwon alpine areas of Korea in 2013. The disease symptoms included appearance of slightly sunken circular lesion with corky rot on the potato surface at the stem-end portion. The fungal species isolated from the infected potatoes were grown on potato dextrose agar and produced white aerial mycelia with dark violet pigments. The conidiophores were branched and monophialidic. The microconidia had ellipsoidal to cylindrical shapes and ranged from $2.6{\sim}11.4{\times}1.9{\sim}3.5{\mu}m$ in size. The macroconidia ranged from $12.7{\sim}24.7{\times}2.7{\sim}3.6{\mu}m$ in size and had slightly curved or fusiform shape with 2 to 5 septate. Chlamydospores ranged from $6.1{\sim}8.1{\times}5.7{\sim}8.3{\mu}m$ in size and were present singly or in pairs. The causal agent of potato stem-end rot was identified as Fusarium oxysporum by morphological characterization and by sequencing the internal transcribed spacer (ITS1 and ITS4) regions of rRNA. Artificial inoculation of the pathogen resulted in development of disease symptoms and the re-isolated pathogen showed characteristics of F. oxysporum. To the best of our knowledge, this is the first study to report that potato stem-end rot is caused by F. oxysporum in Korea.

Effect of the Application of Several Organic Materials on Ginseng Growth (수종유말물 시용이 인삼생육에 미치는 영향)

  • To search for substituents of greens, several organic materials such as rice straw, barley straw, corn stem and manufactured compost were applied in a ginseng cultivating field. The yields of six year old ginseng harvested in the rice straw, barley strait and corn stem treated field were similar to or higher than that of the greens treated one. The varied amount of applied substituents resulted in a yield change, but statistical linearity was not found. The growth of ginseng aerial part and soil physicochemical property in the field fortified with these substituents showed similar results to those of the greens treated one. So, it is though that these organic materials can substitute for greens.

  • The regression line was used to predict ginseng root yield from characters of aerial parts, stem diameter, leaf length and width, and degrees of missing plants per unit area. The rates of fitness of predicted yield on practical yield investigated in field were high. Especially, theoretical yield calculated by the size of stem diameter was a good fit. Therefore, a line regression appeared to be a satisfactory fit.

  • Cephalotaxus koreana Nakai is endemic species of Korea. Since old days, the fruits have been used as a parasiticide and leaves have been used for a pierced wound by insects. The essential oil components from its aerial parts and stem bark by steam distillation were investigated. Several compounds were characterized by GC-Mass spectra. It was found that the leaves, stembark and flowers contain the compounds of essential oils, 1-octene-3-ol, hexadecanoic acid; $\alpha$-pinene, $\Delta^3$-carene mainly, linalylacetate, $\beta$-cubebene, 3,4-octadine-7-methyl , ferruginol(stem bark) and $\alpha$-pinene mainly, $\beta$-pinene , cyclopropane-1,1-dimethyl-2-(3-methyl-1,3-butadienyl), etc. This Cephalotaxus spp. contains the first components of ferruginol(M.W.286.03, $C_{20}H_{30}O)$ which belong to diterpene.

Screening of Some Plant Extracts for Inhibitory Effects on HIV-1 and Its Essential Enzymes (Human Immunodeficiency Virus Type I에 대한 수종 식물 추출물의 억제활성 검색)

  • In order to elucidate the relationship between anti-HIV-1 enzyme activity and inhibition of HIV-1 replication by natural sources, extracts from some plants using the foods and oriental medicines were tested for inhibitory effects on the viral replication, reverse transcriptase (RT), protease and ${\alpha}-glucosidase$. In the anti-RT test, water extracts of Ficus carica (leaf), Houttuynia cordata (aerial part) and Ixeris tamagawaensis (aerial part) showed more than 79% inhitibion at a concentration of $100\;{\mu}g/ml$. The protease and ${\alpha}-glucosidase-inhibiting$ samples in the screening were water extract of Syringa dilatata (leaf) and methanol extract of Hibiscus syriacus (leaf and stem), which showed more than 40% inhibition at a concentration of $100\;{\mu}g/ml$. In the primary anti-HIV-1 test, water extracts of Equisetum arvense (aerial part), Hibiscus syriacus (leaf), Ixeris tamagawaensis (aerial part) and Pueraira thunbergiana (leaf) showed the potent inhibition against HIV-1 induced cytopathic effects.

Studies on Major Agronomic Characteristics of Korean Artemisia annua L. (한국산 개똥쑥의 작물학적 특성)

  • In order to development medicinal Artemisia herbs of high quality, Korean A. annua L. were investigated with its ecological and morphological characteristics, and basic statistical data of agronomic characteristics. This species, which is annual herb, is mainly distributed to marginal land, riverside, roadside, grassland. Ecological niche is low species in competition of the others. Its pollination is basically anemogamous, but is frequency pollinated by insects. This species is characterized as tap root, 1~3 pinnate compound leaf of ovate or narrowly ovate, stem is green, erect and solitary. Inflorescence is paniculate, receptacle is not hair. Capitulum, consist of ray floret and disk floret, is subglobose shape. Additionally, this species could be easily discriminated from related Artemisia herbs by the capitulum size. Ray floret is female, disk floret is bisexual. Flowering season is from August to October. Seed is achene unattached hair. Stem length ranged from 179 cm to 225 cm, and stem diameter and number of branch were $17.14\;{\pm}\;1.68\;mm$, $2.43\;{\pm}\;0.51\;mm$, respectively. Length and width of leaf were $14.5\;{\pm}\;0.5\;cm$, $15.0\;{\pm}\;1.0\;cm$, and leaf number of main stem were $48.06\;{\pm}\;10.57\;cm$, respectively. Fresh weight of aerial parts and root were $364.7\;{\pm}\;14.1\;g$, $32.6\;{\pm}\;5.1\;g$, and its dry weight were $136.6\;{\pm}\;10.0\;g$, $14.9\;{\pm}\;2.34\;g$, respectively.

Characteristics of Hybrids between Jakyungjong and Hwangsukjong in Korean Ginseng (Panax ginseng C. A. Meyer)

  • A large number of individual ginseng plants have been selected in the farmer's fields to develop new ginseng varieties with desirable traits since 1970s. Among them, Hwangsukjong with green stem and yellow berry was selected as a ginseng germplasm. The phenotype of Hwangsukjong is quite different from Jakyungjong that has violet stem and red berry and has been cultivated in most of ginseng fields. Therefore, Hwangsukjong was crossed with Jakyungjong to clarify the inheritance of stem color and then the characteristics of $F_1\;and\;F_2$ hybrids were investigated. $F_1$ hybrid plants were similar to Jakyungjong in most of aerial part characters and showed hybrid vigor in fresh weight of root and weight of 100 seeds. In $F_2$ generation, the stem color was segregated in a ratio of 3 violet to 1 green. From this result, it was elucidated that violet color was controlled by single dominant gene. In another experiment, DNA was extracted from parents (Jakyungjong and Hwangsukjong) and $F_1$ hybrid. For each primer evaluated, multiple band profile was produced comprising from one to five major bands plus a varying number of minor bands and amplified bands were detected among most primers. In case of UBC primer number 13, 17, 30, 31, and 43, band patterns of parents and $F_1$ hybrid were very similar, but the others were not. Especially, in {\sharp}1$, {\sharp}4$, and {\sharp}33$, specific band was produced in Hwangsukjong and $F_1$ hybrid while in {\sharp}6$, another specific band was produced in Jakyungjong and $F_1$ hybrid. Therefore, $F_1$ hybrid had all specific bands at these primers. So, these selective markers could be used for identification of characteristics of $F_2$ hybrids

First Report of Stem Rot in Statice Caused by Rhizoctonia solani in Korea (Rhizoctonia solani에 의한 스타티스 줄기썩음병)

  • Stem rot of perennial statice (Limonium sinuatum) was observed in Un bong, Jeonbuk from 2006 to 2007. Affected plants were randomly distributed in the greenhouses and infection rate was more than 10%. Stem and leaf of statice at soil line were dried and turned brown, initially. As the disease became severe, other stem parts and crowns were turned dark brown and then sunken. The fungal isolates were showed initially white aerial mycelium and turned brown with age. They produced few sclerotia which small, irregularly shaped with pinpoint sized. Mycelia were branched at $90^{\circ}$ angles and multinucleate in one cell. The pathogenicity of causal organism was proved according to Koch's postulates. The causal fungus of stem rot was identified as Rhizoctonia solani based on the cultural and morphological characteristics. This is the first report on stem rot of statice by R. solani in Korea.
