Operational Strategy for Increasing Ethanol Production in Repeated Fed-batch Ethanol Fermentation Using Saccharomyces cerevisiae (Saccharomyces cerevisiae 를 이용한 반복 유가식 ethanol 발효에서 ethanol 생산량을 증가를 위한 운전 전략)

  • We designed the optimal operational strategy in repeated fed-batch ethanol fermentation using Sacchromyces cerevisiae ATCC 24858 in views of ethanol yield, specific ethanol production rate, and ethanol productivity, when the aeration rate were controlled at 0.0 and 0.33 vvm. Coincidentally, the time intervals of withdrawal-fill of culture medium (24 and 36 h) were investigated. Ethanol yield and ethanol productivity when the aeration was carried out at 0.33 vvm were superior to those when the aeration was not carried out. Additionally, those parameters when the time interval of withdrawal-fill of culture medium was 24 h were superior to those when time interval of withdrawal-fill of culture medium was 36 h. The total ethanol production reached at the greatest value, 703.8 g-ethanol, when the aeration was carried out at 0.33 vvm and the time interval of withdrawal-fill of culture medium was 24 h. In this study, we verified experimentally the necessity of designing the operational strategy for increasing ethanol production in terms of aeration rate and time interval of withdrawal-fill of culture medium in the repeated fed-batch ethanol fermentation.

Effects of mixing/aeration ratio and SRT on nutrient removal in SBR process (연속회분식반응조 공정에서 교반/폭기비와 SRT가 영양염류제거에 미치는 영향)

  • In this study, nutrients treatment by sequencing batch reactors(SBR) was performed. Nitrogen and phosphorus removal efficiencies were evaluated by changing SRT and mixing/aeration ratio. Not only nitrogen but also phosphorus removal patterns were investigated through track studies on 1 cycle. As SRT was fixed and mixing/aeration ratio was changed, maximum nitrogen removal efficiency was 87.6% at mixing/aeration ratio 0.67. Phosphorus removal efficiencies were more than 85.5% except no mixing condition. As mixing/aeration ratio was fixed and SRT was changed, nitrogen removal efficiencies were 70.5~79.8%, which represented slight changes, while phosphorus removal efficiencies were 49.0~97.3%, which represented sharply decreasing tendency at less than 20 day. Both phosphorus release rate k and maximum phosphorus release rate $P_{max}/M$ were are decreased as SRT was decreased, but they were not affected by mixing/aeration ratio. It was found that there is a linear relationship between ortho-phosphate uptake and maximum ortho-phosphate release.

Effects and Batch Kinetics of Agitation and Aeration on Submerged Cultivation of Ganoderma Iucidum (영지의 액체배양에 미치는 통기.교반의 효과와 동력학적 특성)

  • The effects of agitaion and aeration on mycelial growth, exo-polysaccharide (EPS) production, and substrate consumption upon the submerged cultivation of G. lucidum were investigated, and the batch kinetics of the EPS fermentation of G. lucidum were interpreted as function of agitation speed and aeration rate. In a 2.6 L jar fermenter system, the optimum agitation speed and aeration rate for EPS production were determined to be 400 rpm and 1.0 vvm, respectively. The maximum production of EPS obtained was 15 g/L. The logistic model for mycelial growth fitted the experimental data better than that determined by the Monod and the two-thirds power models. The Luedeking-Piret equation adequately modelled the kinetic data obtained for product and substrate.

  • The effects of agitation and aeration for exopolysaccharide(EPS) production through batch cultivation of an Enterobacter sp. isolated from the composter were investigated. During the EPS fermentation under conditions of constant agitation speed from 200 to 900 rpm and constant aeration rate of 0.5-2.5 vvm, the low yields of EPS(4.8-5.2g/L) was observed as the viscosity increase of culture broth. With the stepwise increases in agitation speed and aeration rate, the EPS production and the viscosity of EPS were increased 1.3~1.4 times and 2.3~3.6 times higher than those of the fixed conditions, respectively. Therefore, these stepwise increases were considered as the key operating parameters for enhancing EPS production. The max. EPS(6.8g/L) and viscosity(14,000cp) were obtained when the agitation speed was increased from 300 to 900 rpm for 54hrs at 1.5 vvm.

  • A Parametric study based on a computer analysis of the lubrication system of a four-cylinder gasoline engine is illustrated. Through the parametric study, the effects of various aeration levels on the change of oil flow rate and pressure are investigated. Also, at high oil temperature and low engine speed, the effect of oil aeration level on oil flow characteristics in lubrication system is investigated. The illustrated results may give to designers the guide lines of oil aeration level for the safe design of engine lubrication systems in terms of minimum pressure at crank oil bore.

  • This study was performed to evaluate the effects of air flow rate and aeration mode on the treatment of bioventing for diesel-contaminated soil. Initial concentrations of diesel-contaminated soils were about 2,500 and 9,000mg/kg. Air flow rates were 30, 60, and 100mL/min, and air was injected in the continuous and the intermittent modes. TPH removal efficiency of intermittent aeration mode was higher than that of continuous aeration mode. Greater air flow rate than the value of guidance book was needed for bioventing.

  • Using a flocculating Saccharomyes cerevisiae CA-1, an air-lift reactor equipped with a modified settler was used for ethanol fermentation. The effects of conditions such as aeration rate, initial glucose concentration, and dilution rate were studied using the air-lift reactor. In batch fermentation, optimum aeration rate was 0.5 vvm. In continuous fermentation, aeration rate and initial pH were fixed 0.5 vvm and 4.5, substrate concentration and dillution rate were changed 10-15% and 0.1-1.3. The maximum ethanol productivity was shown to be 20.4 g/l$\cdot $h in 10% glucose and 0.7 h$^{-1}$ dilution rate., and optimum operation condition considering the ethanol productivity and glucose utilization ratio was 0.5 h$^{-1}$ dilution rate in 10% glucose concentration.

  • In a 20 L fermentor experiments the level of dissolved oxygen (D.O.) strongly affected growth and PHBV production of Azotobacter vinelandii UWD. A higher D.O. (5%) increased specific cell growth rate two folds but PHBV production was 17 folds higher (62.3 wt%) at a lower D.O.(1%) level. D.O. level was not a good criterion to evaluate the effect of aeration on fermentation characteristics of A. vinelandii UWD. This strain maintained an equal D.O. (5%) by decreasing its oxygen consumption rate when oxygen transfer rate (OTR) was decreased by changing agitation speed at a fixed aeration rate. OTR rather than D.O. was a criterion to explain the effect of aeration on the cell growth and PHBV production. At 5% D.O. with a lower 0TR cell growth rate decreased but PHBV production (57.3 wt%) approached to that (62.3 wt%) of the lower (1%) D.O.

  • The growth kinetics of Streptomyces noursei NRRL 5126 was investigated under different aeration and agitation combinations in a 5.0 l stirred tank fermenter. Poly-${\varepsilon}$-lysine biosynthesis, cell mass formation, and glycerol utilization rates were affected markedly by both aeration and agitation. An agitation speed of 300 rpm and aeration rate at 2.0 vvm supported better yields of 1,622.81 mg/l with highest specific productivity of 15 mg/l.h. Fermentation kinetics performed under different aeration and agitation conditions showed poly- ${\varepsilon}$-lysine fermentation to be a growth-associated production. A constant DO at 40% in the growth phase and 20% in the production phase increased the poly-${\varepsilon}$-lysine yield as well as cell mass to their maximum values of 1,992.35 mg/l and 20.73 g/l, respectively. The oxygen transfer rate (OTR), oxygen utilization rate (OUR), and specific oxygen uptake rates ($qO_2$) in the fermentation broth increased in the growth phase and remained unchanged in the stationary phase.

  • The effect of the variation of aeration time on the microorganisms was investigated in sequencing batch reactor (SBRs). The cycling time in four SBRs was adjusted to 12 hours and then included different aerobic times as 1 hr, 2.5 hr, 4 hr and 5.5 hr, respectively. Four SBR systems have been operated and investigated for over 40 days. As the increase of aeration time, the consumption of glycogen within sludge at the 1st non-aeration time a little bit was increased and the production of glycogen at the aeration time was increased. Also, the produced PHB amounts and PHB production rate at the 1st non-aeration time were increased as the decrease of aeration time, which showed the activation of the phosphorus removal. The ratios of nitrifying microorganisms' number and GAOs to the total microorganisms' number in SBRs was decreased as the decrease of the aeration time, however, the PAOs ratio was almost constant irrespective of the variation of aeration time.
