A novel method that utilizes poly(5-methyl-2-thiozyl methacrylamide-co-2-acrylamido-2-methyl-1-propanesulfonic acid-co-divinylbenzene) [MTMAAm/AMPS/DVB] as a solid-phase extractant was developed for simultaneous preconcentration of trace Cd(II), Co(II), Cr(III), Cu(II), Fe(III), Mn(II), Ni(II), Pb(II), and Zn(II) prior to the measurement by flame atomic absorpiton spectrometry (FAAS). Experimental conditions for effective adsorption of the metal ions were optimized using column procedures. The optimum pH value for the simultaneously separation of the metal ions on the new adsorbent was 2.5. Effects of concentration and volume of elution solution, sample flow rate, sample volume and interfering ions on the recovery of the analytes were investigated. A high preconcentration factor, 100, and low relative standard deviation values, $\leq$1.5% (n = 10), were obtained. The detection limits (${\mu}gL^{-1}$) based on the 3s criterion were 0.18 for Cd(II), 0.11 for Co(II), 0.07 for Cr(III), 0.12 for Cu(II), 0.18 for Fe(III), 0.67 for Mn(II), 0.13 for Ni(II), 0.06 for Pb(II), and 0.09 for Zn(II). The validation of the procedure was performed by the analysis of two certified reference materials. The presented method was applied to the determination of the analytes in various environmental samples with satisfactory results.
SMB 크로마토그래피를 위한 예비 실험으로서 L-ribose와 L-arabinose의 혼합물의 분리 특성을 $NH_2$ 컬럼을 사용하여 확인하였고 두 성분 분리 최적 조건으로 Acetonitrile과 증류수의 조성이 85/15 (v/v)에서 실험을 수행하였다. PIM(Pulse input method)과 Aspen chromatography 전산모사를 통해 L-ribose와 L-arabinose가 선형 흡착등온식을 따르는 것을 알 수 있었으며 각 성분의 Henry상수를 추산하였고 다음과 같다. $$C_{S,Ribose}=3.51{\cdot}C_{M.Ribose}$$$$C_{S,Arabinose}=5.07{\cdot}C_{M.Arabinose}$$ 이를 바탕으로 SMB 크로마토그래피의 최적 운전조건을 수립하기 위하여 Triangle 이론에 의한 운전 조작변수를 추산함으로서 $m_2\;=\;3.51$, $m_3\;=\;5.07$일 때 즉, 삼각형에서 꼭지점의 조건에서 L-ribose와 L-arabinose의 순도가 각각 85, 80% 정도로 두 성분의 분리에 있어서 가장 효과적인 운전 조건임을 확인 할 수 있었다.
An investigation was conducted on the enhancement of production and purification of an oxidant and SDS-stable alkaline protease (BHAP) secreted by an alkalophilic Bacillus horikoshii, which was screened from the body fluid of a unique Korean polychaeta (Periserrula leucophryna) living in the tidal mud flats of Kwangwha Island in the Korean West Sea. A prominent effect on BHAP production was obtained by adding 2% maltose, 1% sodium citrate, 0.8% NaCl, and 0.6% sodium carbonate to the culturing medium. The optimal medium for BHAP production contained (g/l) SBM, 15; casein, 10; $K_2HPO_4$, 2; $KH_2PO_4$, 2; maltose, 20; sodium citrate, 10; $MgSO_4$, 0.06; NaCl, 8; and $Na_2CO_3$, 6. A protease yield of approximately 56,000 U/ml was achieved using the optimized medium, which is an increase of approximately 5.5-fold compared with the previous optimization (10,050 U/ml). The BHAP was homogenously purified 34-fold with an overall recovery of 34% and a specific activity of 223,090 U/mg protein using adsorption with Diaion HPA75, hydrophobic interaction chromatography (HIC) on Phenyl-Sepharose, and ion-exchange chromatography on a DEAE- and CM-Sepharose column. The purified BHAP was determined a homogeneous by SDS-PAGE, with an apparent molecular mass of 28 kDa, and it showed extreme stability towards organic solvents, SDS, and oxidizing agents. The $K_m$ and $k_{cat}$ values were 78.7 ${\mu}M$ and $217.4s^{-1}$ for N-succinyl-Ala-Ala-Pro-Phe-pNA at $37^{\circ}C$ and pH 9, respectively. The inhibition profile exhibited by PMSF suggested that the protease from B. horikoshii belongs to the family of serine proteases. The BHAP, which showed high stability against SDS and $H_2O_2$, has significance for industrial application, such as additives in detergent and feed industries.
복분자 열매에 함유된 항산화활성 물질을 탐색하기 위하여 MeOH로 추출하고, 이 추출물을 용매분획하여 EtOAc가용 중성획분을 얻었다. 이 획분에 함유된 항산화활성 물질을 흡착성과 분자체 효과를 이요한 Sephadex LH-20 column chromatography로 정제하고, HPLC에 의해 1종의 항산화활성물질을 단리하였다. 분리된 활성물질의 구조 해석을 위해 EI-MS와 $^1H-NMR$ 분석을 실시하여, 활성물질은 구조식이 $C_{15}H_{10}O_7$이며 분자량이 302인 quercetin으로 동정되었다. 또한 항산화활성 물질 quercetin은 HPLC에 의해 복분자 열매 100g 중에 $0.25{\pm}0.02\;mg$이 함유되어 있는 것으로 분석되었다.
The effect of water activity on degradation of pigments in dried lavers, Porphyra tenera Kjellm. was examined when stored at room temperature for fifty days. Chlorophyll pigment was extracted with methanol-petroleum ether mixture solvent(2:1 v/v), partitioned in ether, and analysed spectrophotometrically at 662 nm as chlorophyll a. The degradation products of chlorophyll were isolated on sugar-starch column(85:15 w/w) with n-propanol-petroleum ether solution(1:200 v/v) as a developing solvent. The isolated green colored zones were analysed individually at the wavelengths of 650, 662, and 667 nm as allomerized product, chlorophyll a retained, and pheophytin formed respectively. Carotenoida were also extracted with the methanol mixture solvent, partitioned in ether, and finally redissolved in acetone after the evaporation of ether in a rotary vacuum evaporator. The total carotenoid content was measured as lutein at 450 nm. From the results, it is noted that the rate of chlorophyll degradation reached a minimum at 0.11 to 0.33 water activity while progressively increased at higher moisture levels resulting in rapid conversion of chlorophyll to pheophytin. At lower activity, autocatalysed oxidizing reaction like allomerization seemed prevailing the acid catalysed conversion reaction. The loss of carotenoid pigment was also greatly reduced at the range of 0.22 to 0.34 water activity with much faster oxidative degradation at both higher and extremely lower moisture levels. These two moisture levels indicated above at which the both pigments exhibited maximum stability are considerably higher than the BET monolayer moisture which appeared 7.91 percent on dry basis at Aw=0.10 calculated from the adsorption isothermal data of the sample at $20^{\circ}C$. The rate of pigment loss in heat treated samples at 60 and $100^{\circ}C$ for 2 hours prior to storage somewhat decreased, particularly at higher moisture levels although the final pigment retention was not much stabilized.
심해와 연안의 서로 다른 해양퇴적토 환경에서 ATP농도를 이용한 총 미생물 생체량을 측정하였다. 표층 ATP의 분포는 연안역에서 가장 높았으며, 육지로부터 멀어질수록 감소하였다. 또한 수직적으로는 깊이에 따라 급격히 감소하는 양상을 나타내, 저층의 생태계가 수층으로 부터의 영양원 공급에 의해 일차적으로 조절되고 있는 것으로 나타났다. 한편, 유기물은 퇴적물로의 흡착으로 인해 환경 내 체류시간이 길어짐으로 인해, 유기탄소의 함량 변화가 ATP에 비해 급격히 일어나지 않았으며, 특히, 연안퇴적토의 경우 유기탄소의 함량 변화에 비해 ATP의 농도변화가 일정하게 나타나 두 변수간의 유의성 이 없는 것으로 나타났다. 이러한 결과는 분해가능한 유기물의 공급이 많은 연안퇴적토의 경우, 영양원에 의한 조절보다는 상위영양단계의 포식활동이 미생물의 생체량을 최종적으로 조절하는 요인으로 작용하기 때문 인 것으로 사료된다. 지역분포 특성 및 수직적 분포 특성으로부터 ATP는 서로 다른 저서 생태환경의 생체량 분석을 위한 일차적 인 도구로 유용하게 쓰일 수 있는 것으로 나타났다.
최근 금속자원 재활용과 환경보호에 대한 기술적 필요성에 의해 구리(Cu)를 주성분으로 하는 동합금으로부터 니켈, 주석 등의 유가금속 분리/회수에 대한 관심이 높아지고 있다. 본 연구에서는 동합금에 포함된 유가금속들을 분리/회수하는 기초연구중 하나로서, 킬레이트 수지를 이용하여 동합금 제련시 발생하는 부산물에 포함되어 있는 대표적인 유가금속인 니켈(Ni)을 분리/추출할 수 있는 농축 공정의 가능성에 대해 검토하였다. 킬레이트 수지는 원자량이 높은 $Cu^{2+}$ 이온을 $Ni^{2+}$ 이온보다 선택적으로 흡수하였으며, 이러한 과정을 수차례 반복함으로써 실제 동합금 부산물 샘플의 $Ni^{2+}$ 및 $Cu^{2+}$ 이온을 농축할 수 있었으며, XRF 분석을 통하여 각각 70 % 및 99 %의 농도로 분리할 수 있음을 확인하였다.
현재 암모늄과 질산이온에 의한 수질 오염은 생태계에 있어 심각한 문제로 떠오르고 있다. 미생물에 의한 생물학적인 제거 공정은 질화 과정과 탈질 과정으로 구분되는데 암모늄이온은 질화 과정에 의해 질산염 이온으로 산화되고 질산염 이온은 다음 단계인 탈질 과정에서 질소 기체로 되어 제거 된다. 천연 제올라이트는 양이온 교환능이 뛰어나 암모늄이온($NH_4{^+}$)의 제거에 우수한 것으로 알려져 있지만 흡착만으로 암모늄과 질산이온($NO_3{^-}$)을 충분히 제거할 수 없다. 이러한 문제를 해결하기 위해 제올라이트와 미생물을 이용해 생물학적인 방법으로 암모늄과 질산이온을 동시에 제거하기 위한 실험을 수행하였다. 암모늄이온의 제거는 분말형 Beneficial bacteria(Savio, USA)의 종배양 단계를 거쳐 제올라이트가 충진된 컬럼과 진탕배양을 동시에 하였을 경우에 14시간 후에 완전히 제거되었고 질산이온은 제올라이트에 미생물을 자연 흡착시켜 컬럼 처리시 4시간 후에 100% 제거됨을 확인하였다.
어유의 탈취장치 대신 컬럼에 활성탄, 활성 알루미나, 실리카겔, 대나무 숯 등 흡착제를 단독 혹은 적당한 비율로 혼합하여 충진시켜 탈색과 동시에 탈취시킴으로서 효율적인 탈취효과를 검토하였다. 대멸치유의 탈검효과는 수산 30%를 어유 100 ml에 대하여 18 ml첨가한 것이, 탈산은 수산화 나트륨을 2.5%로 하여 $40^{\circ}C$에서 30분간 처리한 것이 효과적이었다. 탈취의 경우 흡착제(활성탄, 활성알루미나, 실리카겔)를 농도별로 단독 첨가할 경우는 실리카겔30% 첨가가, 그리고 흡착제를 혼합하여 첨가할 경우는 활성알루미나 30%와 실리카겔 10% 혼합 첨가가 가장 우수하였으나 흡착제에 녹차분말 2%를 첨가할 경우 큰 효과가 없었다. 대멸치유의 탈검, 탈산 및 탈취 후의 주요지방산 조성은 16:0, 20:5n-3, 18:1n, 16:1n-7, 22:6n-3 및 18:0이었다. 대멸치 정제유에 $\alpha$-tocopherol를 첨가하여 $25^{\circ}C$에서 40일간 저장시 산화 안정성은 대조구는 20일까지 항산화제를 첨가한 경우는 30일까지라 생각되며 $\alpha$-tocopherol 0.02%첨가보다 0.01% 첨가가 더 효과적이었다.
