Soy isoflavones could be recovered with adsorption resin column chromatography from soybean cooking water produced during soymilk or tofu manufacturing process. The main isoflavones in the soybean cooking water were genistin and daidzin, and their concentration was $0.083{\pm}0.019$ and $0.11{\pm}0.017\;mM$, respectively. Their aglycones were not detected. pH of soybean cooking water was critical in this chromatographic process and the recovery of isoflavones, both genistin and daidzin, was maximum at pH 4.0. Adsorption of genistin on the resin was stronger than that of daidzin. Elution rate and height/diameter ratio also affected the recovery yield. Under the optimal conditions, about 85% of genistin and 70% of daidzin could be isolated from soybean cooking water. Soy saponins were also recovered with isoflavones.
Urokinase (UK), a thrombolytic enzyme used to clear catheters obstructed by blood clots, can be also used industrially in the recombinant protein purification system to cleave a fusion protein linked with a certain fragment of GST. We have immobilized UK by covalent attachment to activated Sepharose 6B-Cl gels and evaluated its performance to cleave a fusion protein of hGH and GST. The Sepharose gels were activated by etherification with glycidol (2,3-epoxypropanol) and further oxidized with periodate resulting in glyceryl-Sepharose gels. After the activation treatment, surface density of the aldehyde groups was 7-30 $\mu$mol-aldehde/mL-gel. Immobilization yield was higher than 99% at high pH (10.5), and the immobilized UK maintained ca. 80% specific activity of the soluble UK. In a column reaction the cleavage yield heavily depended on the feed rate, and it was nearly 86% of that from soluble UK. And the immobilized UK was successfully regenerated by unfolding and refolding with 6M GuHCl. After cleavaging reaction, the monomeric hGH was purified by using expanded bed adsorption chromatography.

Recent publications have paid attention on the utilization of solid reagents for the removal of substances causing eutrophication, in particular $PO_4^{3-}$ ions. The adsorption of dissolved inorganic phosphate on slag produced by the refining process of iron ore was fundamentally studied for suppressing the liberation of phosphate from wastewater. This study has been conducted in order to find a possibility to improve the phosphate removal and to evaluate the phosphate removal variation to form hydroxyapatite, when the converter slag is used for phosphate removal. The result shows that the converter slag can be applied to remove phosphate using Freundlich isotherm. The size of converter slag, $2{\sim}0.425 mm$ was more efficient than $2{\sim}4.75mm$ to remove phosphate. In particular, 1 mg/L of phosphate can be removed up to 80% of the initial concentration for the continuous column experiment.
In most of studies on solidification/stabilization with waste, small columns have been used to examinate adsorption or leachate. However, these experiments using small apparatus have been limited to apply with the field. In this study, considering an application to the field, a large Lysimeter(100cm$\times$100cm$\times$100cm) used for the simulation. Then, the open dumping waste was mixed directly with bentonite to simulate the stabilization of waste, environmental aspects and several basic tests. As a result, the concentration of heavy metals and contaminated substances changed with bentonite rate. Most of contaminated substances were decreased in leaching with increased mixing rate of bentonite. Especially, the concentration of CODcr removed 25~30%. Also, the residual soil in dumping waste produced approximately 80% of total volume by 40$\times$40mm screen.
The anticariogenic effects and inhibition of glucosyltransferase activity of Chrysanthemun indicum L. extracts were investigated against 5 strains of microorganisms. The ethanol extracts of Chrysanthemun indicum L. showed the growth inhibition effects on the Streptococcus mutans and oral bacteria. The inhibition rate of glucosyltransferase activity of Chrysanthemun indicum L. extracts showed 78.4 to 92.3% range. The extracts were analysed by using solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 column chromatography, TLC, HPLC techniques. Three components whose molecular weights ranged from 200 to 400 were confirmed to have the anticariogenic activity.
Background: Ginsenosides with less sugar moieties may exhibit the better adsorptive capacity and more pharmacological activities. Methods: An efficient method for the separation of four minor saponins, including gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, from Panax notoginseng leaves (PNL) was established using biotransformation, macroporous resins, and subsequent preparative high-performance liquid chromatography. Results: The dried PNL powder was immersed in the distilled water at $50^{\circ}C$ for 30 min for converting the major saponins, ginsenosides Rb1, Rc, Rb2, and Rb3, to minor saponins, gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, respectively, by the enzymes present in PNL. The adsorption characteristics of these minor saponins on five types of macroporous resins, D-101, DA-201, DM-301, X-5, and S-8, were evaluated and compared. Among them, D-101 was selected due to the best adsorption and desorption properties. Under the optimized conditions, the fraction containing the four target saponins was separated by D-101 resin. Subsequently, the target minor saponins were individually separated and purified by preparative high-performance liquid chromatography with a reversed-phase column. Conclusion: Our study provides a simple and efficient method for the preparation of these four minor saponins from PNL, which will be potential for industrial applications.

Fundamental materialistic characterization and adsorption/neutralization behavior of waste egg shell for heavy metal ion have been studied for its application to wastewater treatment. To investigate the structural change and thermal decomposition characteristics of egg shell. X-ray diffraction and FT-IR analysis were conducted for egg shell treated at $105^{\circ}C$ and $700^{\circ}C$, respectively. For the result of FT-IR analysis, the sample treated at $700^{\circ}C$ showed a reduced C-O absorption band compared with that of egg shell treated at $105^{\circ}C$, which may be due to the $CO_2$ release. Unlike to the result of FT-IR analysis, the XRD patterns of egg shell were almost similar for the cases of $105^{\circ}C$ and $700^{\circ}C$ treatment. however, characteristic diffraction pattern of CaO was observed for $850^{\circ}C$ treatment, at which $CaCO_3$ is known to be completely converted to CaO. TGA/DTA analysis showed a slow decline in weight loss up to $600^{\circ}C$ and, for $600{\sim}800^{\circ}C$ range, the weight loss became drastic by reason of $CO_2$ discharge, which was accompanied by an appearance of major endothermic peak. The ratio of practical breakthrough time to ideal one, total transfer unit, and mass transfer coefficient were observed to be increased as the adsorption was progressed in a multiple-column fixed-bed reactor using egg shell as an adsorbent, which signified the distribution effect of mass transfer for continuous adsorption reaction. The neutralization effect of egg shell for several types of acidic wastewater made of different mineral acids was not much different from each other except for the case of $H_2SO_4$, for which the neutralization reaction was thought to be retarded by the formation of gypsum.
A new method developed for simultaneous purification of enterotoxin A and C from Staphylococcus aureus strain L 350/1 consisted of chromatography on carboxymethyl (CM)-cellulose using a buffer of variable pH, gel filtration on Ultro gel, and fast protein liquid chromatography(FPLC) using a buffer of variable pH. The enterotoxin A and C were purified by three steps: batchwise adsorption from culture supernatant on Amberlite CG-50; chromatography on CM-cellulose using a buffer of constant pH and molarity; and gel filtration on Sephadex G-75. The purified enterotoxin appeared homogeneous by gel diffusion and polyacrylamide gel electrophoresis. Upon treatment with CM-cellulose using a elution of variable pH, enterotoxin A and C were so close that they were not separated completely. After elution from gels, the enterotoxins appeared as a single peak at the same position. Gel filtration gave a reaction of complete identity to enterotoxin A and C in Ouchterlony immunodiffusion. In FPLC using a CM-cellulose, enterotoxin A and C were simultaneously separated at pH 8.6 and 6.8. When each fraction was performed to gel immunodiffusion, at peak of enterotoxin A and C were not detected each other. In a method of elution by pH-gradient was to be more efficient as a simultaneous separation method in terms of speed, yields and simplicity. The purified toxin A and C were identical to type A and C reference enterotoxin on both disc electrophoresis and Ouchterlony gel diffusion.

In this paper, the removal efficiency of THMs (Trihalomethanes) and DOC (Disolved organic carbon) was compared under different GAC (Granual activated carbon) filling methods. One method is "full filling method" in which column is fully filled with GAC at once and the other is "periodic cumulative method" in which column is partially filled with GAC (10, 20, 33 and 50% of total column volume) and added each ratio during 300 days. The effluent concentration of both THMs and DOC under full filling method was low during the initial period, however, steadily increased with operating time. In the contrast, with periodic cumulative method, it maintained (relatively) evenly during the operating period. Periodic cumulative method was more efficient for removing THMs than full filling method. However, when the ratio of chlorodibromomethane or bromoform among THMs was significantly higher than chloroform and bromodichloromethane, full filling method was more efficient than periodic cumulative method. Full filling method had benefit to total DOC removal and control of average DOC concentration in effluent. Overall, periodic cumulative method is more efficient to equalize the removal efficiency of THMs and DOC, so the more frequent refilling of column with small amount of GAC is more advantageous.
A specific ammino auid in a mixture can be crystallized inside an ion exchange column when displacer concentration is high enough to concentrate the amino acid in a pure band beyond its solubility limit. Glutamic acid formpd a discrete crystal layer in a cation exchanger column by operating displacement development mode and using a high concentration of displacer NaOH. The glutamic acid crystal formed was eluded from the column with the effluent stream and collected in a fraction collector. When 1.0 M of NaOH was used as a displacer, more than 60% of the loaded glutamic acid was recovered as crystal. The continuous crystallization and dissolution of crystal occurred, resulting in apparent movement of the crystal along the column without clogging or pressure increase. NaOH was proved a better displacer than NaCl because hydroxide ions neutralized hydrogen ions released from the resin and thus reduced the number of hydrogen ion competing with sodium ion for re-adsorption. The displacement development process coupled with crystallization provided higher concentration and recovery of glutamic acrid than conventional chromatography.
