Anti-arthritic effect of radix Paeoniae rubra herb-acupuncture: a behavioral evaluation on adjuvant-induced arthritis rats

  • Radix Paeoniae rubra (PR) has been widely used for recovering acute inflammation with swelling and pain in Far East Asian countries. In order to investigate the anti-arthritic effect of PR herbacupuncture, several behavioral parameters such as body weight, knee circumference, squeaking threshold, and weight distribution ratio were investigated in an adjuvant-induced mono-arthritis rat model. We performed PR herb-acupuncture on Zusanli (ST36) of the right hind-leg once every second day for a total of 6 times from the second day of initiating arthritis with adjuvant injection, while the control group was treated with saline in the same way. In the experimental group, body weight significantly increased, knee circumference and squeaking threshold significantly decreased indicating the reduction of edema, and both the pain and inflammatory swelling of the hind paws measured by incapacitance meter decreased. Conclusively, PR herb-acupuncture has more promising effect in alleviating arthritic pain and inflammation than saline-acupuncture in rat model, hence further clinical study is required.

  • Aerial parts of Sida rhombifolia Linn. (Malvaceae) were extracted successively using various solvents and screened for various parameters of antiarthritic activity such as adjuvant-induced arthritis, motor performance, and histopathological study. The ethanol and the aqueous extracts showed potent activity; further these extracts were fractionated by using column chromatography. The fraction ET1 isolated from ethanol extract showed the most potent antiarthritic activity.

Effects of Achyranthis Radix Herbal-Acupuncture on Adjuvant Athritis in Rats (우슬약침(牛膝藥鍼)이 Rat의 Adjuvant 관절염(關節炎)에 미치는 영향(影響))

  • To investigate effects of Achyranthis Radix herbal-acupuncture on adjuvant arthritis in rats, the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum and histological test of the muscular tissue were measured in the arthritis part. 1. After elicitating arthritis of Sprague dawley(SD) rats by injection of Freund's complete adjuvant for 2 weeks, normal saline was injected for the Exp. I group and Achyranthis Radix herbal-acupuncture was injected for the Exp. II group during 30days. Selected point was $D\acute{u}b\acute{i}(ST_{35})$ in both the groups. And then the volume of the paw were checked. The volume of the paw was $0.84{\pm}0.14mm$ in the Exp. I group and $0.47{\pm}0.11mm$ in the Exp.II group, the swelling of the paw was restricted significantly in the Exp. II group(p<0.05). 2. The number of WBC was $10.34{\pm}0.14(10^3/ml)$ in the normal group and $37.47{\pm}5.46(10^3/ml)$ in the Exp. I group. It was $21.24{\pm}2.58(10^3/ml)$ in the Exp. II group. This fact showed that the group Exp. II with Achyranthis Radix herbal-acupuncture was more effective than the Exp. II group in the treatment of arthritis(p<0.05). 3. The content of the total protein in the blood serum was $6.14{\pm}0.43g/dl$ in the normal group, $7.95{\pm}0.94g/dl$ in the Exp. I group, and $6.41{\pm}0.68g/dl$ in the Exp. II group. There was no significance in total protein between the Exp. II group and the Exp. I group from the statistical analysis. 4. The content of albumin in the blood serum was $2.94{\pm}0.13g/dl$ in the normal group, $2.01{\pm}0.48g/dl$ in Chang Tong-young the Exp. I group and $3.15{\pm}0.27g/dl$ in the Exp. II group. This fact showed that the Exp. II group had significant increase in the serum albumin from the statistical analysis compared with the Exp. I group. 5. The content of the globulin in the blood serum was $3.19{\pm}0.48g/dl$ in the normal group, $4.70{\pm}1.26g/dl$ in the Exp. I group and $3.26{\pm}0.57g/dl$ in the Exp. II group. There was no significance in the serum globulin between the Exp. II group and Exp. I group from the statistical analysis. 6. In histological finding, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in the Exp. I group. But the Exp. II group showed small amount of inflammatory cells, the refrained inflammatory state and even recovering state.

Analgesic Effect of Daebangpung-tang on a Rat Model of Adjuvant-induced Arthritis (Adjuvant에 의해 유발된 관절염 백서모델에 있어서 대방풍탕의 진통효과)

  • Daebangpung-tang(DBPT) is one of the prescriptions used for the treatment of rheumatoid arthritis(RA) in oriental medicine. The present study aimed to examine the analgesic effect of DBPT on a rat model of CFA-induced arthritis, which is not identical to human auto-immune arthritis although it does have many features in common with RA, and the relation between DBPT-induced analgesia and steroid hormones. CFA-induced arthritis rat model used to test the effect of DBPT was chronic pain model. After the induction of arthritis, rats subsequently showed a reduced stepping force of the affected limb for at least the next 18 days. The reduced stepping force of the limb was presumably due to a painful knee, since oral injection of indomethacin produced temporary improvement of weight bearing. DBPT dissolved in water was orally administrated. After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 4 hours. DBPT produce significant improvement of stepping force of the hindlimb affected by the arthritis lasting at least 3 hours. The magnitude of this improvement was equivalent to that obtained after an oral injection of 3mg/kg of indomethacin and this improvement of stepping force was interpreted as an analgesic effect. The reduced stepping force was divided into three stages(10-30g, 30-50g, and 50-70g). All experiments was performed at 50-70g of stepping force, since both DBPT and indomethacin showed the most excellent analgesic effect at 50-70g of stepping force. DBPT produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner and showed analgesic effect on neuropathic pain as well. DBPT-induced analgesic effect could not be blocked by systemic injection of steroid antagonist mifepristone. The present study suggest 1) that DBPT produces a potent analgesic effect on the chronic knee arthritis pain model in the rat and 2) that steroids system does not mediate DBPT-induced analgesia.

The Effect of Long Chain n-3 Polyunsaturated Fatty Acids on Development of Collagen-induced Arthritis in Rats (흰쥐에서 다족쇄 n-3 다불포화지방산이 콜라젠-유도 관절염의 발생에 미치는 영향)

  • Background: The treatment of rheumatoid arthritis still depend on conserve therapy in major. Recent studies report that n-3 polyunsaturated fatty acids(PUFA) could modulate the incidence and progress of arthritis. The purpose of this study was to investigate the effects of n-3 PUFA on the development of collagen-induced arthritis in rats. Materials and Methods: Female Louvain rats were used for this experiment. Rats were randomly assigned into either normal (n=8) or collagen-immunized groups, and collagen immunized groups were divided into control(n=8, normal diet) and n-3 PUFA(n=8, 5% n-3 PUFA in diet) groups. One week after feeding n-3 PUFA to rats, they were immunized with type II collagen emulsified in incomplete Freund's adjuvant into tail and back. Development of arthritis was confirmed by x-ray and microscopic examination. Results: Incidence of arthritis at the 5th week after immunization was 38% in control and 0% in n-3 PUFA. Rats with arthritis showed edema in hind paws and inflammation in synovial membrane of the knee joint. Plasma glucose and insulin were not changed by both of immunization and diet. Plasma triglycerides and cholesterol concentrations were decreased by n-3 PUFA. Conclusion: n-3 PUFA may prevent or treat collagen-induced arthritis m rats. Further studies are needed for action mechanism of it.

Anti-inflammatory Effect of Gyulpidaehwangbakcho-tang (Jupidahuangpoxiao-tang) in the Collagen-induced Arthritis Mouse Model

  • Objectives: To investigate anti-inflammatory and anti-arthritic effects of Gyulpidaehwangbakcho-tang (GDBT) extract in a murine model of rheumatoid arthritis. Methods: The mice received $100{\mu}g$ of bovine type II collagen in Freund's complete adjuvant by intradermal injection at the base of the tail on day 0 and a booster injection on day 21. The mice were orally administered with GDBT (200 or 50mg/kg dissolved in distilled water) daily from day 1 to day 21 after arthritis incidence, and monitored for disease incidence and the severity of arthritis up to day 21. In order to evaluate the effect of GDBT on disease progression, we examined pro-inflammatory cytokines including IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II. Results: GDBT produced a significant and dose dependent inhibition of arthritis and inflammation during the entire duration of the study. This action was characterized by the decreased production of IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2, and NOS-II in vivo. Conclusion: We believe that the anti-arthritic activity of GDBT is due to its modulatory effect on the expression of pro-inflammatory cytokine in the synovium. Our results contribute towards validation of the traditional use of GDBT in the treatment of RA and other inflammatory joint disorders.

Effect of Low Frequency Electroacupuncture on Nicotidamide Adenine Dinucleotide Phosphate-diaphorase(NADPH-d) Positive Neurons in the Brain Cortex of Rat with Adjuvant Induced Rheumatoid Arthritis (저빈도 전침자극이 류마토이드 관절염 유발 흰쥐 대뇌피질 Nicotidamide Adenine Dinucleotide Phosphate-diaphorase(NADPH-d) 양성세포 발현에 미치는 영향)

  • Objectives & Methods : This study was to investigate effect of low frequency electroacupuncture on NADPH-d positive neurons in the brain cortex of rat with adjuvant induced rheumatoid arthritis. Experimental groups were divided into 6 groups ; Normal, Control, $ST_{36}$, $SP_9$, $ST_{36}+SP_9$ and Non-Acupoint. Normal group, non-arthritic group, was injected normal saline, and the other groups were injected FCA. Each acupoint groups were treated by 2Hz electroacupuncture at each acupoints and NA group was treated by 2Hz electroacupuncture at non-acupoint. Each groups were evaluated by the number of NADPH-d positive neurons in primary somatosensory area(S1), secondary somatosensory area(S2), motor area and caudate putamen by using an image analyzer and a microscope. Results : 1. In S1, the number of NADPH-d positive neuron cells in the $ST_{36}$ group were significantly(p<0.05) increased compared with the control group. 2. In S2, the number of NADPH-d positive neuron cells in all electroacupuncture groups were not significantly changed compared with the control group. 3. In motor area, the number of NADPH-d positive neuron cells in $ST_{36}$ group, $SP_9$ group, NA group were significantly(p<0.05) increased compared with the control group. 4. In Caudate putamen, the number NADPH-d positive neuron cells in all electroacupuncture groups were significantly(p<0.05) decreased compared with the control group. Conclusions : Our result demonstrated that low frequency electroacupuncture on $ST_{36}$ & $SP_9$ normalized expression of NADPH-d positive neurons in the brain cortex of the rheumatoid arthritis model in rats.

Effect of Corydalidis Tuber.Carthami Flos.Paeoniae Radix Rubra on Anti-Inflamation and function of the Liver at Arthritis Rats (현호색(玄胡索).홍화(紅花).적작약(赤芍藥)이 관절염 흰쥐의 염증과 간(肝)에 미치는 영향)

  • This study is designed to find out the effect of Corydalidis Tuber Carthami Flos Paeoniae Radix Rubra mixed water extract on adjuvant arthritis, and to check the stability for Corydalidis Tuber Carthami Flos Paeoniae Radix Rubra mixed water extract about the liver. At first, rats were devided into four groups; Normal(Non-treated group with FCA), Control(Group administrated saline everyday for 1 week after treatment of FCA), Sample A(group administrated Corydalidis Tuber Paeoniae Radix Rubra Carthami Flos mixed water extract $9mg/200{\sim}220g$ everyday 1 week after treatment of FCA) and Sample B(Group administrated Corydalidis Tuber Paeoniae Radix Rubra Carthami Flos mixed water extract $18mg/200{\sim}220g$ everyday 1 week after treatment of FCA). After then, separated searches were made on the change of edema rate, the number of WBC, the contented quantities of total protein, total bilirubin, GOT, GPT in the serum. Result : In the right planter edema, both of Sample groups showed a decrease with statistical significance in comparison with control group. In the WBC count, Sample B group showed a decrease with statistical significance in comparison with control group and also sample A group showed a decrease, but did not show statistical significance. In the serum total protein, both of samples did not show statistically significant decrease in comparison with the control group. In the serum total bilirubin, GOT and GPT, both of samples did not show any harmful change about the liver in comparison with the control group. According to the above results, it is concluded that Corydalidis Tuber Paeoniae Radix Rubra Carthami Flos mixed water extract had the therapeutic effect of anti-inflammation in adjuvant arthritis with no injury to liver.

Experimental study on Cervi Cornu on Adjuvant Arthritis in rats (록각(鹿角)의 Adjuvant 관절염(關節炎)에 대한 실험적(實驗的) 연구(硏究))

  • Objective: To investigate effects of Cervi Cornu on Adjuvant Athritis in rats, the edema inhibit rate, the anaJgesic effects, the number of WBC, RA facter, Platelet, the quantity of CRP, total protein, albumin and globuline in the blood serum were measured in the arthritis part. Results: The results obtained as fonows ; 1. After arthritis of Sprague dawley(SD) rats was induced by injecting Freund's complete adjuvant for 2 weeks, any treatment was not for Control group, acupunctured for Treat Ⅰ group. normal saJine was ora] administrated for the Treat Ⅱ group, Cervi Cornu Ex. was oral administrated for Treat Ⅲ, and Cervi Cornu Herbal-acupullcture was injected for Trea Ⅳ group during 2 weeks every other day. Selected point was on pressure pain point in both groups. And then the edema inhibit rate were checked. The edema inhibit rate was $46.03\%$ in Treat I group, $43.24\%$ Treat IV group, $37.44\%$ in Treat III. there was significance in the edema inhibit rate between Control group and Treat group, in order of Treat Ⅰ, Ⅳ, Ⅲ.(p<0.05) 2. The analgesic effects was $7.58{\pm}1.80$(${\times}10$gm) in Control group. $11.00{\pm}1.10$(${\times}10$gm) in Treat Ⅰ group. $99.92{\pm}1.28$(${\times}10$gm) in Treat Ⅲ group and $14.67{\pm}1.03$(${\times}10$gm) in Treat Ⅳ group. There was significance in the analgesic effects between Control group and Treat group(p<0.05) 3. The number of WBC was $14.72{\pm}1.48$(${\times}10^3$㎕) in control Group, $10.26{\pm}1.13$(${\times}10^3$㎕) in Treat Ⅰ group, $11.00{\pm}1.13$(${\times}103$㎕) in Treat Ⅱ Group and $9.63{\pm}1.75$(${\times}10^3$㎕) in Treat Ⅳ group. There was significance in the number of WBC between Control group and Treat group(p<0.05) 4. The content of total protein in the blood serum were $6.13{\pm}0.05$g/dl in control group, $5.73{\pm}0.14$g/dl in Treat I group, $5.88{\pm}0.13$g/dl in Treat Ⅲ group and $5.90{\pm}0.13$g/dl in Treat IV group. There was significance in The content of total protein in the blood serum between Control group and Treat group(p<0.05) 5. The contests of albumin in the blood serum were $2.32{\pm}0.12$g/dl in the Control group, $2.35{\pm}0.05$g/dl in Treat Ⅰ group, $2.35{\pm}0.05$g/dl in Treat Ⅱ group, $2.30{\pm}0.06$g/dl in Treat Ⅲ group, $2.42{\pm}0.08$g/dl in Treat IV group. There was no significance in The content of albumin in the blood serum between Control group and Treat group(p<0.05) 6. The contests of globulin in the blood semm were $3.68{\pm}0.08$g/dl in the Control group, $3.43{\pm}0.12$g/dl in Treat Ⅰ group, $3.55{\pm}0.10$g/dl in Treat IV group. There was significance in The content of globulin in the blood serum between Control group and Treat group(p<0.05) 7. The numbers of RA factor were $3.47{\pm}0.54$IU/ml in Control group and $2.38{\pm}0.50$IU/ml in Treat Ⅱ group. There was significance inThe numbers of RA factor between Control group and Treat group(p<0.05) 8. The numbers of platelet were $1126.33{\pm}1126.33{\pm}85.93{times}10^3$/㎕ in Control group, $1043.33{\pm}80.80{times}10^3$/㎕ in Treat Ⅰ group, $1116.82{\pm}77.93{times}10^3$/㎕ in Treat Ⅱ group, $1164.17{\pm}94.02{times}10^3$/㎕l in Treat Ⅲ group, $1076.67{\pm}54.84{times}10^3$/㎕ in Treat Ⅳ group. There was no significance in The numbers of platelet between Control group and Treat group(p<0.05) 9. The quantity of CRP were $0.05{\pm}0.01$mg/ml in Control group, $0.05{\pm}0.01$mg/ml in Treat Ⅰ group, $0.06{\pm}0.01$mg/ml in Treat Ⅱ group, $0.05{\pm}0.00$mg/ml in Treat Ⅲ group, $0.05{\pm}0.00$mg/ml in Treat Ⅳ group. There was no significance in The quantity of CRP between Control group and Treat group(p<0.05) Concluslon : From these results, it is shown Cervi Comu Herbal-acupuncture more efffective thaJJ Cervi Cornu Ex. on Adjuvant Arthritis in rats.
