The crystal structure of nylon 56 fibers post extended by drawing process was investigated by synchrotron x-ray scattering measurement. In as-cast fiber, distinct (004) and (020) diffraction peaks were observed and they were related to initial metastable alignment of nylon molecules. With increase in the drawing ratio, (110) peak intensity was increased in vertical direction with decreasing (020) peak. At the same time, (004)' peak evolved position tilted to 29 degrees from the (004) peak. This evolution is directly related to stable crystalline phase of nylon 56 originated from additional formation of hydrogen bondings between N-H and C=O by post drawing process. We also compared density variation, stress-strain curves of the fiber as a function of drawing ratio and strain. The variations of density and tanacity also supported the increase of stable structure of nylon 56.
The cyclic undecapeptide cyclosporin A (CyA), one of the most valuable immunosuppressive drugs, is produced nonribosomally by a multifunctional cyclosporin synthetase enzyme complex by the filamentous fungus Tolypocladium niveum. To increase CyA productivity by wild-type T. niveum (ATCC 34921), random mutagenesis was first performed using an antifungal agar-plug colony assay (APCA) selection approach. This generated a mutant strain producing more than 9-fold greater CyA than the wild-type strain. Additionally, a foreign bacterial gene, Vitreoscilla hemoglobin gene (VHb), was transformed via protoplast regeneration and its transcription was confirmed by RT-PCR in the UV-irradiated mutant cell. This led to an additional 33.5% increase of CyA production. Although most protoplast-regenerated T. niveum transformants tend to lose CyA productivity, the optimized combination of random mutagenesis and protoplast transformation described here should be an efficient strategy to generate a commercially valuable, yet metabolite low-producing, fungal species, such as CyA-producing T. niveum.

Earthquakes not only produce additional load on the structures and underlying soil, but also change the strength characteristics of the soil. Therefore, in order to analyze soil structures for stability, the behaviour after earthquake must be considered. In this paper, a series of cyclic triaxial tests and monotonic triaxial tests were carried out to investigate the undrained shear strength and liquefaction strength characteristics of Nak-Dong River sand soils which were subjected to cyclic loading. The sample was consolidated in the first stage and then subjected to stress controlled cyclic loading with 0.1Hz. After the cyclic loading, the cyclic-induced excess pore water pressure was dissipated by opening the drainage valve and the sample was reconsolidated to the initial effective mean principal stress(p/sub c/'). After reconsolidation, the monotonic loading or cyclic loading were applied to the specimen. In the results, the undrained shear strength and liquefaction strength characteristics depended on the pore pressure ratio(Ur=U/p/sub c/'). The volume change following reconsolidation can be a function of cyclic-induced excess pore water pressure and the maximum double amplitude of axial strain.

Cytotoxic effects of extracts from Tremella fuciformis strain FB001 were evaluated on the DLD-1 human colon adenocarcinoma cell line and the content of polyphenolic compounds in the extracts were analyzed. Hexane, chloroform, and ethyl acetate subfractions (experimental setting I) exhibited cytotoxic effects on the human colon adenocarcinoma DLD-1 cell line with $IC_{50}$ values of 350, 400, and 450 ppm, respectively. When T. fuciformis was extracted sequentially with ether, ethyl acetate, chloroform, and ethanol (experimental setting II), the ether extract demonstrated potent cytotoxicity with an $IC_{50}$ value of 150 ppm, followed by ethyl acetate and chloroform fractions. If the first extraction solvent was chloroform instead of ether (experimental setting III), exposure of the cell line to chloroform, ethyl acetate, and ether extracts at 1,000 ppm led to cell death. High levels of phenolic compounds were estimated for all hydrophobic extracts, which exhibited cytotoxic effects. We propose that this useful information gives additional support to our understanding of the biology and utility of this particular mushroom.
The Glycoprotein D (gD) gene of the HSV-1 strain F was cloned, sequenced, recombinated into the HcNPV (Hyphantria cunea nuclear polyhedrosis virus) expression vector and expressed in insect cells. The gD gene was located in the 6.43 kb BamHI fragment of the strainF. The open reading frame (ORF) of the gD gene was 1,185 by and codes 394 amino acid residues. Recombinant baculoviruses, GD-HcNPVs, expressing the gD protein were constructed. Spodoptera frugiperda cells, infected with the recombinant virus, synthesized a matured gX-gD fusion protein with an approximate molecular weight of 54 kDa and secreted the gD proteins into the culture media by an immunoprecipitation assay The fusion gD protein was localized on the membrane of the insect cells, seen by using an immunofluorescence assay The deduced amino acid sequence presents additional characteristics compatible with the structure of a viral glycoprotein: signal peptide, putative glycosylation sites and a long C-terminal transmembrane sequence. These results indicate the utility of the HcNPV-insect cell system for producing and characterizing eukaryotic proteins.
Ring forging process is more appropriate for high-length and thin walled ring, because it utilizes the forging press and hence does not require heavy-duty ring rolling mill. Although ring forging process is very simple and economic for facilities, the process is not efficient because of multi-forging-step and low material utilization. An effective ring forging process is developed using a hollow ingot. When a hollow ingot is used with a workpiece, the ingot can be forged into a final ring without multi-stage pre-forging process, such as, cogging, upsetting, and piercing, etc.. Finally it has advantages of the material utilization and process improvement because a few reheating and forging process are not necessary to make workpiece for ring forging. The important design variables are the applied plastic deformation energy to eliminate cast structure and make uniform properties. In this study, the mechanical properties after forging of hollow cast ingot were investigated from the experiment using circumferential sectional model. Also, the effects of process variables were studied by FEM simulation on the basis of thermo-visco-plastic constitutive equation. Applied strain is different at each position in length direction because diameter of hollow ingot is different in length direction. The different strain distribution become into a narrow gap by additional plastic deformation during diameter extension process.

Post-earthquake observations revealed that seismic performance of beam-column connections in precast concrete structures affect the overall response extensively. Seismic design of precast reinforced concrete structures requires improved beam-column connections to transfer reversed load effects between structural elements. In Turkey, hybrid beam-column connections with welded components have been applied extensively in precast concrete industry for decades. Beam bottom longitudinal rebars are welded to beam end plates while top longitudinal rebars are placed to designated gaps in joint panels before casting of topping concrete in this type of connections. The paper presents the major findings of an experimental test programme including one monolithic and five precast hybrid half scale specimens representing interior beam-column connections of a moment frame of high ductility level. The required welding area between beam bottom longitudinal rebars and beam-end plates were calculated based on welding coefficients considered as a test parameter. It is observed that the maximum strain developed in the beam bottom flexural reinforcement plays an important role in the overall behavior of the connections. Two additional specimens which include unbonded lengths on the longitudinal rebars to reduce that strain demands were also tested. Strength, stiffness and energy dissipation characteristics of test specimens were investigated with respect to test variables. Seismic performances of test specimens were evaluated by obtaining damage indices.
The methods of design available for geocell-supported embankments are very few. Two of the earlier methods are considered in this paper and a third method is proposed and compared with them. In the first method called slip line method, plastic bearing failure of the soil was assumed and the additional resistance due to geocell layer is calculated using a non-symmetric slip line field in the soft foundation soil. In the second method based on slope stability analysis, general-purpose slope stability program was used to design the geocell mattress of required strength for embankment. In the third method proposed in this paper, geocell reinforcement is designed based on the plane strain finite element analysis of embankments. The geocell layer is modelled as an equivalent composite layer with modified strength and stiffness values. The strength and dimensions of geocell layer is estimated for the required bearing capacity or permissible deformations. These three design methods are compared through a design example. It is observed that the design method based on finite element simulations is most comprehensive because it addresses the issue of permissible deformations and also gives complete stress, deformation and strain behaviour of the embankment under given loading conditions.
The extrinsic Fabry-Perot interferometric (EFPI) optical fiber sensor shows good sensitivity and resolution, and has many advantages over optical fiber sensors of other types. However, this EFPI optical fiber sensor has a disadvantage that the distinction of measuring directions is difficult due to the measurement method by using only fringe counting. In this paper, the transmission-type extrinsic Fabry-Perot interferometric (TEFPI) optical fiber sensor was developed, which has been improved by the additional function and whose measuring system is different from that of the conventional EFPI optical fiber sensor. Then the application result of the TEFPI optical fiber sensor to the strain and temperature measurement was explained in detail.

In this paper, the time series based damage detection algorithms developed by Nair, et al. (2006) and Nair and Kiremidjian (2007) are applied to the benchmark experimental data from the National Center for Research on Earthquake Engineering (NCREE) in Taipei, Taiwan. Both acceleration and strain data are analyzed. The data are modeled as autoregressive (AR) processes, and damage sensitive features (DSF) and feature vectors are defined in terms of the first three AR coefficients. In the first algorithm developed by Nair, et al. (2006), hypothesis tests using the t-statistic are applied to evaluate the damaged state. A damage measure (DM) is defined to measure the damage extent. The results show that the DSF's from the acceleration data can detect damage while the DSF from the strain data can be used to localize the damage. The DM can be used for damage quantification. In the second algorithm developed by Nair and Kiremidjian (2007) a Gaussian Mixture Model (GMM) is used to model the feature vector, and the Mahalanobis distance is defined to measure damage extent. Additional distance measures are defined and applied in this paper to quantify damage. The results show that damage measures can be used to detect, quantify, and localize the damage for the high intensity and the bidirectional loading cases.
