Background: A leaf cuticle has different structures and functions as a barrier to water loss and as protection from various environmental stressors. Methods: Leaves of Panax ginseng were examined by scanning electron microscopy and transmission electron microscopy to investigate the characteristics and development of the epicuticular structure. Results: Along the epidermal wall surface, the uniformly protuberant fine structure was on the adaxial surface of the cuticle. This epicuticular structure was highly wrinkled and radially extended to the marginal region of epidermal cells. The cuticle at the protuberant positions maintained the same thickness. The density of the wall matrix under the structures was also similar to that of the other wall region. By contrast, none of this structure was distributed on the abaxial surface, except in the region of the stoma. During the early developmental phase of the epicuticular structure, small vesicles appeared on wallecuticle interface in the peripheral wall of epidermal cells. Some electron-opaque vesicles adjacent to the cuticle were fused and formed the cuticle layer, whereas electron-translucent vesicles contacted each other and progressively increased in size within the epidermal wall. Conclusion: The outwardly projected cuticle and epidermal cell wall (i.e., an epicuticular wrinkle) acts as a major barrier to block out sunlight in ginseng leaves. The small vesicles in the peripheral region of epidermal cells may suppress the cuticle and parts of epidermal wall, push it upward, and consequently contribute to the formation of the epicuticular structure.

Background: Both Panax ginseng Meyer and Panax quinquefolius are obligate shade-loving plants whose natural habitats are broadleaved forests of Eastern Asia and North America. Panax species are easily damaged by photoinhibition when they are exposed to high temperatures or insufficient shade. In this study, a cytohistological study of the leaf structures of two of the most well-known Panax species was performed to better understand the physiological processes that limit photosynthesis. Methods: Leaves of ginseng plants grown in soil and hydroponic culture were sectioned for analysis. Leaf structures of both Panax species were observed using a light microscope, scanning electron microscope, and transmission electron microscope. Results: The mesostructure of both P. ginseng and P. quinquefolius frequently had one layer of non-cylindrical palisade cells and three or four layers of spongy parenchymal cells. P. quinquefolius contained a similar number of stomata in the abaxial leaf surface but more tightly appressed enlarged grana stacks than P. ginseng contained. The adaxial surface of the epidermis in P. quinquefolius showed cuticle ridges with a pattern similar to that of P. ginseng. Conclusion: The anatomical leaf structure of both P. ginseng and P. quinquefolius shows that they are typical shade-loving sciophytes. Slight differences in chloroplast structure suggests that the two different species can be authenticated using transmission electron microscopy images, and light-resistant cultivar breeding can be performed via controlling photosynthesis efficiency.
Aiming to determine the taxonomic identity of Gang-hwa-yak-ssuk as a cultivated plant, this study analyzes ITS sequences and compares their external morphologies with the genus Artemisia, which have a similar external morphology. Thus far, we have considered A. indica as the original plant of Gang-hwa-yak-ssuk, but Gang-hwa-yak-ssuk is better grouped with A. argyi, not A. indica, for the following reasons: Gang-hwa-yak-ssuk has the same characteristics as A. argyi in terms of the natural habitat, in that it is 1-bipinnately cleft or parted in the external morphology of its leaves, and because the white glandular trichome is distributed on the adaxial surface. This is in addition to the result of ITS sequence analysis. Therefore, we can define Gang-hwa-yak-ssuk as a cultivated plant that originates from A. argyi and not A. indica.
The adult of the Cotton caterpillar, Palpita indica (Saunder) laid eggs more on mature leaves than on aged and developing leaves. They laid more on leaves than on petiole and stem, and more on the adaxial than on the abaxial surface side of the leaves. Larvae of the Cotton caterpillar showed their preference in the order of the cucumber (Cucumis sativus), gourd (Lagernaria siceraria), watermelon (Citrullus lanatus) > oriental melon (Cucumis melon L. var makuwa), wax gourd (Benincasa hispida), melon (Cucumis melo), star cucumber (Sicyos angulatus) > sponge cucumber (Lufa cylindrica), cotton (Gossypium indicum). There were no distinct differences among varieties and lines that were collected and hybridized at the Gochang Watermelon Experiment Station in Korea. There was a positive relationship between the leaf area and the degree of damage by the Cotton caterpillar. The feeding amounts of the Cotton caterpillar excrement were gradually increased to 16-18 days after hatching, after that the amounts of excrement were rapidly decreased.
Fresh specimens of the aquatic macrophyte, Spirodela polyrhiza, have been examined employing scanning and transmission electron microscopy. Observations revealed the occurrence of microbial colonization during development. Submerged parts of the small, free-floating S. polyrhiza body exhibited a variety of microorganisms such as bacteria, cyanobacteria, and diatoms throughout their development. However, immature and/or young plants normally demonstrated much less microbial colonization compared to mature plants. During the study, heavy colonization by the microorganisms was routinely encountered at maturity, especially in the fully developed abaxial fronds and root caps. The mucilaginous layer was shown along the root caps, and the microorganisms appeared to be either clustered or attached to this layer. In contrast, only moderate degrees of colonization were observed in the root, and little to no colonization was observable in the adaxial frond surface. Transmission electron microscopy clearly demonstrated the microbial colonization to be external in the S. polyrhiza specimen examined in the current study. The association between the microorganisms and S. polyrhiza has been considered non-harmful, as no frond senescence and almost no mechanical penetration of the plant by the microorganisms were noticed during the study.

BACKGROUND: Before generating transgenic plant using the CRISPR-Cas9 system, the efficiency test of sgRNAs is recommended to reduce the time and effort for plant transformation and regeneration process. The efficiency of the sgRNA can be measured through the transient expression of sgRNA and Cas9 gene in tomato cotyledon; however, we found that the calculated efficiency showed a large variation. It is necessary to increase the precision of the experiment to obtain reliable sgRNA efficiency data from transient expression. METHODS AND RESULTS: The cotyledon of 11th, 15th, 19th, and 23rd-day-old tomato (Solanum lycopersicum cv. Micro-Tom) were used for expressing CRISPR-Cas9 transiently. The agrobacterium harboring sgRNA for targeting ALS2 gene of tomato was injected through the stomata of leaf adaxial side and the genomic DNA was extracted in 5 days after injection. The target gene edition was identified by amplifying DNA fragment of target region and analyzing with Illumina sequencing method. The target gene editing efficiency was calculated by counting base deletion and insertion events from total target sequence read. CONCLUSION: The CRISPR-Cas9 editing efficiency varied with tomato cotyledon age. The highest efficiency was observed at the 19-day-old cotyledons. Both the median and mean were the highest at this stage and the sample variability was also minimized. We found that the transgene of CRISPR-Cas9 system was strongly correlated with plant leaf development and suggested the optimum cotyledon leaf age for Agrobacterium-mediated transfection in tomato.

This study aimed to identify and compare the characteristics of Elymus humidus on common wheat (Triticum aestivum L. cv. Chinse Spring, CS). The seed length, width, height, and weight of E. humidus were smaller than those of the CS. In particular, the germination rate of E. humidus was substantially lower than that of CS. In the anatomical dissection of the leaf, E. humidus showed a considerably different xylem diameter of the main vascular bundle in the main vein; however, there was no difference in the phloem of the main vascular bundle compared with the xylem and phloem of the main vascular bundle in the main vein of CS, although E. humidus showed a leaf structure similar to that of CS. In addition, E. humidus had a thinner epidermis than that of CS. Regarding stomatal traits, E. humidus showed a graminoid stomata type similar to that of CS. On the adaxial and abaxial sides, the density, length, and width of the stomata in E. humidus were smaller than those in CS, whereas the distance between stomata in E. humidus was greater than that in CS. The chromosomes of E. humidus were classified as long and short based on their respective lengths. Long chromosomes were classified based on the ratio of the long arm to the short arm e.g., 1:1 or 2:1. Short chromosomes showed the same trend and some short chromosomes were microsatellites. To evaluate genetic diversity, 38 barley EST markers with polymorphisms between E. humidus and CS were selected from 236 barley EST markers.

In order to establish highly efficient gene transfer condition at early stage of soybean transformation, various experiments were performed and compared their efficiencies by transient GUS analysis; those conditions are genotype determination of Korean soybean cultivars for amenability to Agro-infection, appropriate agar and selective agent concentration, orientation of explant placement, hormone pre-culture, and liquid selection condition. In the genotype screen of Korean soybean varieties, 14 amenable genotypes were selected. For efficient Agrobacterium washing, cefotaxime was chosen and hygromycin at the concentration of 10 and 15 ppm was used as selection agent in the media. Agar concentration was slightly better in 0.6% and 0.8% for both shoot and callus formation, and explant placement with adaxial side down showed high frequency of GUS expression. For wounding treatment, oriental needle was efficient than scalpel for shoot formation and gene transfer. To increase the frequency of gene transfer, hormone pre-treatment was applied. BA at the concentration of 5 and 10 ppm resulted in better survival at the late stage of selection in shoot elongation media. Selection in liquid media after hormone pre-treatment seemed to be effective to remove the escaped non-transformants at early stage of procedure. Considering the results obtained, Eunhakong could be the first choice as a material for soybean transformation among Korean soybean genotypes.
This study was carried out to know the effect of temperature, light intensity and $CO_2$ concentration on photosynthesis and respiration in Wasabi (Wasabia japonica Matsum). The optimum temperature for photosynthesis in Wasabi was $17{\sim}20^{\circ}C$ and dark respiration rate was increased with the increasing of temperature from, $15 ^{\circ}C\;to\;30^{\circ}C$. Light compensation point was $6.0\;{\mu}E\;m^{-2}s^{-1}$ in Wasabi and $36.7\;{\mu}E\;m^{-2}s^{-1}$ in Corn, and light saturation point was $600{\mu}E\;m^{-2}s^{-1}$, similar in Wasabi and Corn. $CO_2$, compensation point was 67.3ppm in Wasabi and 11.6 ppm in Corn. Photorespiration rate in Wasabi leaf at $l000{\mu}E\;m^{-2}s^{-1}$ light intensity was 3.3 mg$CO_2$, $dm^{-2}hr^{-1}$, and then was gradually decreased as light intensity decreased. Stomatal frequency was about $76\;mm^{-2}$ on the adaxial surface and $623\;mm^{-2}$ on the abaxial surface, and the size of stomata was about 1$12{\mu}m$ on the adaxial surface and $17{\mu}m$ on the abaxial surface of the leaf.
This study was conducted to establish a systematized taxonomic problems of through the leaf morphological characters and leaf venation patterns, and stomatal cell patterns and cell characteristics of abaxial and adaxial surface of the leaflets by SEM, of 6 native species in Korea and 2 foreign species of the Genus Rhus in the Family Anacardiaceae. The results obtained from this study are summarized as followings: 1. Morphological study measured 32 characters of leaves from herbarium specimen and field-collected samples for each species. The results of cluster analysis based on the Euclidean distance showed that the species could be classified into 3 groups: R. sylvestris. R. typhina, R. succedanea: R. trichocarpa. R. chinensis. R. verniciflua: and R. ambigua. R. radicans subsp. orientale, Analysis of principal components showed 5 groups: The major factors in the first principal component group was length of petiole of the terminal leaflets, that in the second group angle of left side in the terminal leaflet bash, that in the third group area ratio between first and terminal leaflets, that in the forth group angle ratio between right and left side in the terminal leaflet base, and that in the fifth group was angle of main and secondary vein at midrib of terminal leaflet. Cumulative contribution by the first, second and third principal component group was explained with 82.6%, a large percent of all information. 2. The leaf venation pattern investigated using soft X-ray photography revealed clado-and reticulo-camptodromous types according to branching angle of the secondary vein. And three groups by the developing degree of secondary vein were R. trichocarpa, R. ambigua. R. chinensis, R. typhina; R. radicans subsp. onentale, R. succedanea, R. verniciflua: and R. sylvestris. Classification key for the Rhus of Korean-native Anacardiaceae was made by the venation pattern and devevoping degree of the secondary vein. 3. The stomatal cell patterns were greatly classified into paracytic and anomocytic types, specific among species according to stomatal and subsidiary cell patterns, and various differences among the species was determined. Microstructure of the adaxial and abaxial surfaces could be divided into synclinal and anticlinal cell wall patterns, and were specific-species. Stomatal cells of R. chinensis were surrounded with characterized villus-like cells.
