The Effect of Minocycline on Motor Neuron Recovery and Neuropathic Pain in a Rat Model of Spinal Cord Injury

  • Objective : Minocycline, a second-generation tetracycline-class antibiotic, has been well established to exert a neuroprotective effect in animal models and neurodegenerative disease through the inhibition of microglia. Here, we investigated the effects of minocycline on motor recovery and neuropathic pain in a rat model of spinal cord injury. Methods : To simulate spinal cord injury, the rats' spinal cords were hemisected at the 10th thoracic level (T10). Minocycline was injected intraperitoneally, and was administered 30 minutes prior surgery and every second postoperative day until sacrifice 28 days after surgery. Motor recovery was assessed via the Basso-Beattie-Bresnahan test Mechanical hyperalgesia was measured throughout the 28-day post -operative course via the von Frey test Microglial and astrocyte activation was assessed by immunohistochemical staining for ionized calcium binding adaptor molecule 1 (lba1) and glial fibrillary acidic protein (GFAP) at two sites: at the level of hemisection and at the 5th lumbar level (L5). Results : In rats, spinal cord hemisection reduced locomotor function and induced a mechanical hyperalgesia of the ipsilateral hind limb. The expression of lba1 and GFAP was also increased in the dorsal and ventral horns of the spinal cord at the site of hemisection and at the L5 level. Intraperitoneal injection of minocycline facilitated overall motor recovery and attenuated mechanical hyperalgesia. The expression of lba1 and GFAP in the spinal cord was also reduced in rats treated with minocycline. Conclusion : By inhibiting microglia and astrocyte activation, minocycline may facilitate motor recovery and attenuate mechanical hyperalgesia in individuals with spinal cord injuries.

4×4 배열 합성 혼 안테나 및 고출력 마그네트론에 의한 방사전력 측정 (Design of 4×4 Array Synthesis Horn Antenna and Radiated Power Measurement by Magnetron)

  • 본 논문에서는 배열 합성 혼 안테나를 설계하고 고출력 마그네트론을 연결하여 방사전력을 측정하였다. 제안된 안테나는 $4{\times}4$ 배열 합성 혼 안테나 특성을 바탕으로 후방 방사를 억제시키기 위해 합성 혼 개구면 상, 하단에 2단 단락구조물을 부착하여 빔폭 E-plane $8.86^{\circ}$, H-plane $7.35^{\circ}$, 전후방비 39.7 dB를 얻었다. 또한 배열된 마그네트론을 이용한 배열 혼 안테나의 방사전력을 측정하기 위하여 도파관 어댑터를 설계하여 마그네트론과 혼 안테나를 연결 시켰고 고출력의 파워를 측정하기 위해 중간단에 에어갭을 넣은 마이크로스트립 라인 커플러를 설계하였다. $4{\times}4$ 배열 합성 혼 안테나의 중앙에 4개의 마그네트론을 연결하여 측정한 결과 평균 방사 출력 0.063 W를 얻었다.

TAK1-dependent Activation of AP-1 and c-Jun N-terminal Kinase by Receptor Activator of NF-κB

  • The receptor activator of nuclear factor kappa B (RANK) is a member of the tumor necrosis factor (TNF) receptor superfamily. It plays a critical role in osteoclast differentiaion, lymph node organogenesis, and mammary gland development. The stimulation of RANK causes the activation of transcription factors NF-${\kappa}B$ and activator protein 1 (AP1), and the mitogen activated protein kinase (MAPK) c-Jun N-terminal kinase (JNK). In the signal transduction of RANK, the recruitment of the adaptor molecules, TNF receptor-associated factors (TRAFs), is and initial cytoplasmic event. Recently, the association of the MAPK kinase kinase, transforming growth factor-$\beta$-activated kinase 1 (TAK1), with TRAF6 was shown to mediate the IL-1 signaling to NF-${\kappa}B$ and JNK. We investigated whether or not TAK1 plays a role in RANK signaling. A dominant-negative form of TAK1 was discovered to abolish the RANK-induced activation of AP1 and JNK. The AP1 activation by TRAF2, TRAF5, and TRAF6 was also greatly suppressed by the dominant-negative TAK1. the inhibitory effect of the TAK1 mutant on RANK-and TRAF-induced NF-${\kappa}B$ activation was also observed, but less efficiently. Our findings indicate that TAK1 is involved in the MAPK cascade and NF-${\kappa}B$ pathway that is activated by RANK.

TRRAP stimulates the tumorigenic potential of ovarian cancer stem cells

  • Ovarian cancer is the most fatal gynecological malignancy in women and identification of new therapeutic targets is essential for the continued development of therapy for ovarian cancer. TRRAP (transformation/transcription domain-associated protein) is an adaptor protein and a component of histone acetyltransferase complex. The present study was undertaken to investigate the roles played by TRRAP in the proliferation and tumorigenicity of ovarian cancer stem cells. TRRAP expression was found to be up-regulated in the sphere cultures of A2780 ovarian cancer cells. Knockdown of TRRAP significantly decreased cell proliferation and the number of A2780 spheroids. In addition, TRRAP knockdown induced cell cycle arrest and increased apoptotic percentages of A2780 sphere cells. Notably, the mRNA levels of stemness-associated markers, that is, OCT4, SOX2, and NANOG, were suppressed in TRRAP-silenced A2780 sphere cells. In addition, TRRAP overexpression increased the mRNA level of NANOG and the transcriptional activity of NANOG promoter in these cells. Furthermore, TRRAP knockdown significantly reduced tumor growth in a murine xenograft transplantation model. Taken together, the findings of the present study suggest that TRRAP plays an important role in the regulation of the proliferation and stemness of ovarian cancer stem cells.

Regulation Fe65 localization to the nucleus by SGK1 phosphorylation of its Ser566 residue

  • Fe65 is characterized as an adaptor precursor (APP) through its PID2 element, as well as with the other members of the APP protein family. With the serum- and glucocorticoid-induced kinase 1 (SGK1) substrate specificity information, we found that the putative site of phosphorylation in Fe65 by SGK1 is present on its $Ser^{566}$ residue in $^{560}CRVRFLSFLA^{569}$(X60469). Thus, we demonstrated that Fe65 and the fluorescein-labeled Fe65 peptide $FITC-^{560}CRVRFLSFLA^{569}$ are phosphorylated in vitro by SGK1. Phosphorylation of the $Ser^{566}$ residue was also demonstrated using a $Ser^{566}$ phospho-specific antibody. The phospho Fe65 was found mainly in the nucleus, while Fe65 S556A mutant was localized primarily to the cytoplasm. Therefore, these data suggest that SGK1 phosphorylates the $Ser^{566}$ residue of Fe65 and that this phosphorylation promotes the migration of Fe65 to the nucleus of the cell.

Production of IL-1β and Inflammasome with Up-Regulated Expressions of NOD-Like Receptor Related Genes in Toxoplasma gondii-Infected THP-1 Macrophages

  • Toxoplasma gondii is an obligate intracellular parasite that stimulates production of high levels of proinflammatory cytokines, which are important for innate immunity. NLRs, i.e., nucleotide-binding oligomerization domain (NOD)-like receptors, play a crucial role as innate immune sensors and form multiprotein complexes called inflammasomes, which mediate caspase-1-dependent processing of $pro-IL-1{\beta}$. To elucidate the role of inflammasome components in T. gondiiinfected THP-1 macrophages, we examined inflammasome-related gene expression and mechanisms of inflammasome-regulated cytokine $IL-1{\beta}$ secretion. The results revealed a significant upregulation of $IL-1{\beta}$ after T. gondii infection. T. gondii infection also upregulated the expression of inflammasome sensors, including NLRP1, NLRP3, NLRC4, NLRP6, NLRP8, NLRP13, AIM2, and NAIP, in a time-dependent manner. The infection also upregulated inflammasome adaptor protein ASC and caspase-1 mRNA levels. From this study, we newly found that T. gondii infection regulates NLRC4, NLRP6, NLRP8, NLRP13, AIM2, and neuronal apoptosis inhibitor protein (NAIP) gene expressions in THP-1 macrophages and that the role of the inflammasome-related genes may be critical for mediating the innate immune responses to T. gondii infection.

디지털콘텐츠 식별체계 UCI (Universal Content Identifier)와 통합저작권 관리번호 ICN (Integrated Copyright Number)간의 연계와 통합에 관한 연구 (A Study on the Linkage and integration of UCI (Universal Content Identifier) between ICN (Integrated Copyright Number))

  • UCI (Universal Content Identifier) 식별체계가 국가표준식별체계로 자리를 잡으면서 다양한 저작물의 디지털 콘텐츠에 대하여 UCI 식별자가 부여되어 유통이 이루어지고 있으나, 콘텐츠의 저작권 정보 부재로 인해 의도되지 않은 저작권 침해 행위 발생 및 저작권 계약 등에 있어 많은 문제점이 발생하고 있다. 통합저작권관리번호인 ICN (Integrated Copyright Number)는 국내에서 저작권 관리를 위해 개발된 코드체계로 저작물 활용에 따른 사용내역에 대한 정산과 분배, 불법 모니터링, 필터링을 목적으로 하며, 권리자의 보호를 위해 저작물에 대한 저작권 관계 정보를 포함하고 있다. 그러나 ICN은 그 목적이 저작권 관리에 집중되어 있어 디지털콘텐츠자체의 속성이나 유통에 필요한 정보들을 포함하고 있지 않다. 따라서 생산되는 디지털저작물을 보다 효율적으로 관리하고 유통하기 위해서는 콘텐츠를 식별하기 위한 식별체계인 UCI와 디지털저작물에 대한 권리의 소유관계를 식별할 수 있는 저작권 권리관리체계인 ICN과 연계와 통합은 권리자와 사업자간의 투명하고 효과적인 저작물 유통의 기반이 될 수 있다. 본 논문에서는 UCI 식별체계와 ICN 식별체계를 연계와 통합의 관점에서 식별체계간의 정책적 요인들과 핵심적인 기술 요소를 분석하고 각 식별체계의 가치사슬간의 효율화를 기할 수 있는 연계 및 통합 인프라의 구축에 대한 모델을 제시하고자 한다.

액티브 스너버형 플라이백 컨버터를 적용한 60W급 초소형 어댑터에 관한 연구 (A Study on 60W Class Ultra-small Adapter with Active Snubber Type Flyback Converter)

  • 본 논문에서는 액티브 스너버형 플라이백 컨버터를 60W급 어댑터에 적용하고 시험 회로를 제작하여 그 시험 결과를 보고하였다. 특히 어댑터의 시험 회로는 입력전압이 110V와 220V 겸용으로 설계하였으며 출력전압은 노트북 컴퓨터 등에서 가장 많이 사용되는 19V 60W급으로 설계하였다. 시험용 60W급 어댑터의 체적은 약 $50cm^3$, 전력밀도는 약 $1.22W/cm^3$로 설계하여 사이즈를 최소화 시켜서 휴대성을 확인 하였다. 시험 회로는 입력전압 변화에 대해 최대부하 60W에서는 최대 90.88%의 고효율을 나타내었다. 입력전압 220V에서 효율은 60W 출력에서 최대 91.04%이었다. 또한 부하 전력에 대한 출력전압 안정도는 약 0.07%이내로 관측되었다.

이중공기공급 이유체노즐의 선회각 변화에 따른 분무특성 (Effect of Swirl Angle on the Atomization Characteristics in Two-Fluid Nozzle with Dual Air Supplying System)

  • The atomization characteristics of the dual air supplying twin-fluid nozzle were investigated experimentally using PIV and PDA systems. The two-fluid nozzle is composed of three main parts: the feeding injector to supply fluid that is controlled by a PWM (pulse-width modulation) mode, the adaptor as a device with the ports for supplying the carrier and assist air and the main nozzle to produce the spray. The main nozzle has the swirl tip with four equally spaced tangential slots, which give the injecting fluid an angular momentum. The angle of the swirl tip varied with 0$^{\circ}$ 30$^{\circ}$, 60$^{\circ}$ and 90$^{\circ}$, and the ratios of carrier air to assist air and ALR(total air to liquid) were 0.55 and 1.23, respectively. The macroscopic behavior of the spray was investigated using PIV system, and the mean velocity, turbulent intensity and SMD distributions of the sprays were measured using PDA system. As the results, the mean axial velocity at the spray centerline decrease with the increase of the swirl angle. The turbulent intensities of the axial and radial velocity were increased with the increase of the swirl angle. The mean SMD (Sauter mean diameter) of the radial direction along the axial distance shows the lowest value at the swirl angle of 60$^{\circ}$.

Kaempferol이 LPS로 유도된 생쥐 중추신경계 염증에 미치는 영향 (Effects of Kaempferol on Lippolysaccharide-induced Inflammation in Mouse Brain)

  • Objectives : Brain inflammation early activates the microglia and activated microglia secrete a variety of pro-inflammatory cytokines. Kaempferol, which is a flavonoid in Cuscutae Semen, shows a wide range of physiological activities, including neurons protection and anti-inflammatory actions through inhibition of pro-inflammatory mediators. The present study examined the modulatory effect of kaempferol on cytokines [tumor necrosis factor- alpha ($TNF-{\alpha}$), interleukin-1beta ($IL-1{\beta}$) and interleukin-6 (IL-6)] and cyclooxygenase-2 (COX-2) mRNA expression and microglia activation in the brain tissue of the mouse. Methods : Kaempferol was administered orally three doses of 10, 20 and 30 mg/kg respectively, once 1 hour before the lippolysaccharide(LPS) (3 mg/kg, i.p.) injection. Brain tissue was removed at 4 hours after LPS injection. Cytokines and COX-2 mRNA expression in the brain tissue was measured by the quantitative real-time polymerase chain reaction (PCR) method. Iba1 expression was calculated by western blotting method. Microglia was observed with immunohistochemistry. Immunohistochemistry stained microglia was analyzed by using ImageJ software. Results : Kaempferol 20 and 30 mg/kg was significantly attenuated the expression of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 mRNA. Kaempfrol 10, 20 and 30 mg/kg significantly attenuated COX-2 mRNA expression in the brain tissue. Kaempferol 30 mg/kg significantly suppressed the increase of Iba1 protein expression by LPS. Kaempferol 30 mg/kg significantly decreased the number of microglia in the cerebral cortex and the number and cell size of microglia in the hypothalamic region and the area percentage of ionized calcium binding adaptor molecule 1(Iba1)-expressed microglia in the hippocampus. Conclusions : This results indicate that kaempferol plays an anti-inflammatory role in the brain.
