Antioxidant capacity in seedling of colored-grain wheat under water deficit condition

  • Nutritious and functional foods from crop have received great attention in recent years. Colored-grain wheat contains high phenolic compound and a large number of flavonoid. The anthocyanin and polyphenolic synthesis and accumulation is generally stimulated in response to biotic or abiotic stresses. Here, we analyzed genome wide transcripts in seedling of colored-grain wheat response to ABA and PEG treatment. About 900 and 1500 transcripts (p-value < 0.05) from ABA and PEG treatment were aligned to IWGSC1+popseq DB which is composed of over 110,000 transcripts including 100,934 coding genes. NR protein sequences of Poaceae from NCBI and protein sequence of transcription factors originated from 83 species in plant transcription factor database v3.0 were used for annotation of putative transcripts. Gene ontology analysis were conducted and KEGG mapping was performed to show expression pattern of biosynthesis genes related in flavonoid, isoflavonoid, flavons and anthocyanin biopathway. DroughtDB (http://pgsb.helmholtz-muenchen.de/droughtdb/) was used for detection of DEGs to explain that physiological and molecular drought avoidance by drought tolerance mechanisms. Drought response pathway, such as ABA signaling, water and ion channels, detoxification signaling, enzymes of osmolyte biosynthesis, phospholipid metabolism, signal transduction, and transcription factors related DEGs were selected to explain response mechanism under water deficit condition. Anthocyanin, phenol compound, and DPPH radical scavenging activity were measured and antioxidant activity enzyme assays were conducted to show biochemical adaptation under water deficit condition. Several MYB and bHLH transcription factors were up-regulated in both ABA and PEG treated condition, which means highly expressed MYB and bHLH transcription factors enhanced the expression of genes related in the biosynthesis pathways of flavonoids, such as anthocyanin and dihydroflavonols in colored wheat seedlings. Subsequently, the accumulation of total anthocyanin and phenol contents were observed in colored wheat seedlings, and antioxidant capacity was promoted by upregulation of genes involved in maintaining redox state and activation of antioxidant scavengers, such as CAT, APX, POD, and SOD in colored wheat seedlings under water deficit condition. This work may provide valuable and basic information for further investigation of the molecular responses of colored-grain wheat to water deficit stress and for further gene-based studies.

Effect of a new phosphorus source, magnesium hydrogen phosphate (MHP) on growth, utilization of phosphorus, and physiological responses in carp Cyprinus carpio

  • Magnesium hydrogen phosphate (MHP, $MgHPO_4$) recovered from swine manure was prepared as an alternative phosphorus (P) source. Conventional P additives, monocalcium phosphate (MCP), dicalcium phosphate (DCP), and tricalcium phosphate (TCP) were compared with the MHP in terms of growth and P availability by juvenile carp Cyprinus carpio. A basal diet as a negative control was prepared using practical feed ingredients without P supplementation to which four supplemental P sources were added at the level of 2%. Five groups of 450 fish having mean body weight of 6.5 g following 24 h fasting after 2 weeks of adaptation period were randomly distributed into each of 15 tanks (30 fish/tank). Fish were hand-fed to apparent satiety twice a day for 9 weeks. Fish fed the MHP had weight gain (WG), feed conversion ratio (FCR), protein efficiency ratio (PER), and specific growth rate (SGR) comparable to those fed the MCP. Those values of both the MHP and MCP groups were significantly different (p < 0.05) from the other groups. Fish groups fed control and the TCP showed the lowest WG, PER, and SGR and the highest FCR among treatments. No fish were died among treatments during the experimental period. Fish fed control and the TCP showed hematocrit and hemoglobin significantly lower (p < 0.05) than fish fed the MHP. The lowest inorganic P (Pi) in plasma was found in the control group. Even though Pi was not significantly different (p > 0.05) from other phosphate groups, fish fed the MCP and MHP retained higher P in whole body than the other groups. P availability was determined to be 93.2, 62.4, 6.1, and 98.0% for MCP, DCP, TCP, and MHP, respectively. The present results suggested that the MHP recovered from wastewater stream could be used as an alternative P source in carp diet.

A Study on Experiments the Environmental Conditions and the Adaptation of the Human Body in the Vinyl House (Vinyl House 내의 환경조건과 인체적응에 관한 실험연구)

  • The purpose of this study is to experiments the environmental conditions and the adaption of the human body in the vinyl house. The study was done in spring and winter and experimental clothes were used working clothes in the vinyl house. The results are as follows. 1. Environmental Conditions In the spring season, the indoor air temperature was $27.4{\pm}3.7^{\circ}C$ and the outdoor air temperature was $14.4{\pm}2.7^{\circ}C$. In the winter season, the indoor air temperature was $18.3{\pm}4.8^{\circ}C$ and the outdoor air temperature was $7.6{\pm}2.5^{\circ}C$ on the average. 2. Skin Temperature In the spring season, the mean skin temperatures indoor and outdoor were $33.81{\pm}0.7^{\circ}C\;and\;31.57{\pm}0.8^{\circ}C$ respectively, a difference of $2.24^{\circ}C$. In the winter season, they were $31.95{\pm}1.93^{\circ}C\;and\;29.86{\pm}0.55^{\circ}C$ respectively, a difference of $2.09^{\circ}C$. 3. Clothing Climate In the spring season, the temperature and humidity in the inner layer of clothing were $34.77{\pm}0.80^{\circ}C\;and\;70.75{\pm}1.65%$ indoor, $31.9{\pm}0.52^{\circ}C\;and\;51.9{\pm}3.70%$ outdoor respectively. In the winter season, those were $32.52{\pm}1.04^{\circ}C\;and\;64.65{\pm}3.68%$ indoor, $30.27{\pm}0.96^{\circ}C\;and\;45.07{\pm}2.68%$ outdoor respectively. 4. Physiological Factors Body temperature increased slightly and the pulse rate also rises, but blood pressure decreased a little with the rise of environmental temperature both in the spring and winter seasons. 5. Psychological Factors Thermal sensation in the spring season was expressed as 'slightly warm' or 'warm' indoor and as 'neutral' in the open air, while in the winter it was expressed as 'neutral' or 'slightly warm' outdoor the house and as 'cold' in the open air. Comfort sensation was characterized as 'uncomfortable' or 'slightly uncomfortable' indoor both in the spring and winter seasons, but in the open air it was characterized as 'comfortable' in the spring and as 'slightly uncomfortable' in the winter.

Comparison of Thermal Stress Induced Heat Shock Factor 1 (HSF1) in Goldfish and Mouse Hepatocyte Cultures (붕어와 마우스의 간세포 배양에서 열 스트레스에 의해 유도되는 heat shock factor1 (HSF1)의 비교)

  • Heat shock proteins (HSPs) are induced in response to various physiological or environmental stressors. However, the transcriptional activation of HSPs is regulated by a family of heat shock factors (HSFs). Fish models provide an ideal system for examining the biochemical and molecular mechanisms of adaptation to various temperatures and water environments. In this study, we examined the pattern differentials of heat shock factor 1 (HSF1) and expression of heat shock protein 70 (HSP70) in response to thermal stress in goldfish and mouse hepatocyte cultures by immune-blot analysis. Goldfish HSF1 (gfHSF1) changed from a monomer to a trimer at $33^{\circ}C$ and showed slightly at $37^{\circ}C$, whereas mouse HSF1 (mHSF1) did so at $42^{\circ}C$. This experiment showed similar results to a previous study, indicating that gfHSF1 and mHSF1 play different temperature in the stress response. We also examined the activation conditions of the purified recombinant proteins in human HSF1 (hmHSF1) and gfHSF1 using CD spectroscopy and immune-blot analysis. The purified recombinant HSF1s were treated from $25^{\circ}C$ to $42^{\circ}C$. Structural changes were observed in hmHSF1 and gfHSF1 according to the heat-treatment conditions. These results revealed that both mammal HSF1 (human and mouse HSF1) and fish HSF1 exhibited temperature-dependent changes; however, their optimal activation temperatures differed.

Contractile Function of a Flight Muscle Over the Range of Hibernation Temperature in Bats (박쥐의 동면온도 범위에서 작용하는 비행근의 수축기능)

  • Studies on thermal adaptation postulate that optimal temperature of physiological properties matches a usually experienced body temperature (Tb) of organisms. Hibernating bats maintain Tb's that are slighdy higher than ambient temperatures (9$^{\circ}$-12$^{\circ}$C) of their wintering sites. To test the hypothesis that muscle function is adjusted to the Tb range of the hibernating animals, we examined contractile function of the biceps brachil muscle of Korean greater horseshoe bats, Rhinolophus ferrumequlnum korai (n = 5) at tissue temperatures between 1O$^{\circ}$ and 35$^{\circ}$C. Relative tetanic force (% of maximum force) was highest at temperatures of 1O$^{\circ}$-15$^{\circ}$C, which match well their Tb's during hibernation. Because non-hibernating endotherms with Tb of around 37$^{\circ}$C show the optimal temperature for muscle force over 30$^{\circ}$-40$^{\circ}$C, our results strongly suggest that the flight muscle of the bats may exhibit thermal adjustments according to their seasonal Tb's. The capacity to generate strong force at such low body temperatures may be adaptive, because bats must have muscles functioning to fly for occasional watering or excretion, or to move away from potential predators during hibernation.

Length and Weight Relationship for Two Dominant Antarctic Notothenioid Fishes Caught in the Coastal Water off King Sejong Station, King George Island, Antarctica (남극, 킹조지섬, 세종과학기지 연안에 우점하는 남극암치아속 어류 두 종의 길이와 무게의 관계)

  • Length and weight relationship (LWR) for dominant Antarctic fishes was determined in two species of the family Nototheniidae; black rockcod (Notothenia coriiceps) and marbled rockcod (Notothenia rossii). Samples were caught in the offshore sea around King Sejong station located on King George Island, Antarctica in January, 2017. A total of 30 N. coriiceps and 7 N. rossii were caught by fishing rod and hook. Average total length was 266.0 mm for N. coriiceps and 275.4 mm for N. rossii. Average total weight was 283.1g for N. coriiceps and 290 g for N. rossii. In terms of LWR and b value, the results showed that both two species had positive allometries (b>3) in good health. This size information of two dominant Antarctic fishes would be useful for future physiological studies to understand of adaptation mechanism and biological pathway of Antarctic marine organisms.

Projecting the Spatio-Temporal Change in Yield Potential of Kimchi Cabbage (Brassica campestris L. ssp. pekinensis) under Intentional Shift of Planting Date (정식일 이동에 따른 배추 잠재수량성의 시공간적 변화 전망)

  • Planting date shift is one of the means of adapting to climate change in Kimchi Cabbage growers in major production areas in Korea. This study suggests a method to estimate the potential yield of Kimchi Cabbage based on daily temperature accumulation during the growth period from planting to maturity which is determined by a plant phenology model tuned to Kimchi Cabbage. The phenology model converts any changes in the thermal condition caused by the planting date shift into the heat unit accumulation during the growth period, which can be calculated from daily temperatures. The physiological maturity is estimated by applying this model to a variable development rate function depending either on growth or heading stage. The cabbage yield prediction model (Ahn et al., 2014) calculates the potential yield of summer cabbage by accumulating daily heat units for the growth period. We combined these two models and applied to the 1km resolution climate scenario (2000-2100) based on RCP8.5 for South Korea. Potential yields in the current normal year (2001-2010) and the future normal year (2011-2040, 2041-2070, and 2071-2100) were estimated for each grid cell with the planting dates of July 1, August 1, September 1, and October 1. Based on the results, we divided the whole South Korea into 810 watersheds, and devised a three - dimensional evaluation chart of the time - space - yield that enables the user to easily find the optimal planting date for a given watershed. This method is expected to be useful not only for exploring future new cultivation sites but also for developing cropping systems capable of adaptation to climate change without changing varieties in existing production areas.

A Study on the Circadian Rhythm of Opiate Receptor in Rat Brain (백서 뇌내 Opiate 수용체의 일주기 변동에 대한 연구)

  • To investigate diurnal variations of opiate receptor binding and its modification by experimental condition or treatment of various centrally-acting drugs, the amount of maximum $^3H-morphine$ binding in rat midbrain homogenates was measured at 4 hour intervals for 24 hours. Animals were conditioned under the controlled L : D, 12 : 12 cycle or D: D, 12 : 12 cycle, for 3 weeks and treated with 0.5 ml of physiological saline or drugs for 2 weeks. A highly significant diurnal rhythm with peak at 22 hour of early dark phase with an amplitude$(0.68{\pm}0.06\;pmole/mg\;protein)$ of +51.1% and nadir $(0.33{\pm}0.03\;mole/mg\;prtein)$ at 18 hour of late light phase with an amplitude of -26.6% was found in control group. 24 tour mean of $^3H-morphine$ binding was $0.45{\pm}0.03\;pmole/mg$ protein respectively. Constant dark adaptation or treatment of reserpine, pargyline, imipramine, amphetamine and chlorpromazine modified the diurnal rhythm in the time of peak and nadir binding shape, phase, amplitude of the diurnal curve and 24 hour mean of $^3H-morphine$ binding. However, Kd values were not changed in all experimental groups : Statistical analysis at times of least and great binding indicates that the differences in $^3H-morphine$ binding were due to changes not in the affinity, but in the number of binding sites. The results are interpreted with regard to the diurnal rhythm of opiate receptor finding. The modes of action of psychoactive drugs are closely related to postulated changes of receptor sensitivity in neuropharmacological aspects.

A Prospect on the Changes in Short-term Cold Hardiness in "Campbell Early" Grapevine under the Future Warmer Winter in South Korea (남한의 겨울기온 상승 예측에 따른 포도 "캠벨얼리" 품종의 단기 내동성 변화 전망)

  • Warming trends during winter seasons in East Asian regions are expected to accelerate in the future according to the climate projection by the Inter-governmental Panel on Climate Change (IPCC). Warmer winters may affect short-term cold hardiness of deciduous fruit trees, and yet phenological observations are scant compared to long-term climate records in the regions. Dormancy depth, which can be estimated by daily temperature, is expected to serve as a reasonable proxy for physiological tolerance of flowering buds to low temperature in winter. In order to delineate the geographical pattern of short-term cold hardiness in grapevines, a selected dormancy depth model was parameterized for "Campbell Early", the major cultivar in South Korea. Gridded data sets of daily maximum and minimum temperature with a 270m cell spacing ("High Definition Digital Temperature Map", HDDTM) were prepared for the current climatological normal year (1971-2000) based on observations at the 56 Korea Meteorological Administration (KMA) stations and a geospatial interpolation scheme for correcting land surface effects (e.g., land use, topography, and site elevation). To generate relevant datasets for climatological normal years in the future, we combined a 25km-resolution, 2011-2100 temperature projection dataset covering South Korea (under the auspices of the IPCC-SRES A2 scenario) with the 1971-2000 HD-DTM. The dormancy depth model was run with the gridded datasets to estimate geographical pattern of change in the cold-hardiness period (the number of days between endo- and forced dormancy release) across South Korea for the normal years (1971-2000, 2011-2040, 2041-2070, and 2071-2100). Results showed that the cold-hardiness zone with 60 days or longer cold-tolerant period would diminish from 58% of the total land area of South Korea in 1971-2000 to 40% in 2011-2040, 14% in 2041-2070, and less than 3% in 2071-2100. This method can be applied to other deciduous fruit trees for delineating geographical shift of cold-hardiness zone under the projected climate change in the future, thereby providing valuable information for adaptation strategy in fruit industry.

A Study on Nutritional Management for Improvement of Exercise Capacity and Physical Fitness -For Dietary Feeding Condition- (운동 수행 능력 및 체력증진을 위한 효율적 영양관리에 관한 연구 -식이급식 조건에 대하여-)

  • This study was directed to further clarify the effect of resting time or dietary condition for the improvement of exercise capacity and physical fitness, and the changes of some body consitiuents and physiological functions which are related to the exercise metabolism. Sixteen male students(8 athletes and 8 nonathletes)were participated during 3 weeks(Aug. 20-Sep.9, 1989). Each subject performed two treadmill running trials at an absolute intensity (1 mintute in 3.4mph/15% slope and 2 minutes in 5.5mph/20% slope). In the resting time trials, general diet was fed before 1, 2, 4 and 12 hours of the treadmill exercise loaded. in the dietary condition trials, high carbohydrate(HC), high fat(HF) and high protein(HP) diet were fed before 2 hours of the treadmill exercise loaded. Control trial was that of resting time before treadmill exercise loaded after 12 hours of general diet feeding. Measurement were made to study the change of blood glucose, palmitate, lactate, blood pressure and heart rate. The results obtained are summarized as follows: Mean daily metabolizable period. In resting time trial, the blood glucose concentration of athlete and nonathlete from 2 hours group was similar to control group. The blood palmitate concentration of athlete was increased in 1 and 2 hours group but those of nonathlete was not only increased in 1 and2 hours group but was more increased in 12 hours group, compared with both control group. The blood lactate concentration was increased in all experimental group, compared with both control group and those of nonathlete was much higher than athlets. The elevation rate of blood pressure in pre-and after-exercise of athlete was lower than those of nonathlete. In dietary composition trial, the blood glucose concentration of athlete and nonathlete in HC group was higher than other diet groups. The blood lactate concentration of athlete and nonathlete in HC group was lower than other diet groups. There was no remarkable change of the blood palmitate concentration and heart rate in each dietary composition trial, but those of nonathlete was low in HC and high in HP group. In above results, it was suggested that the effective condition of resting time and dietary composition for the improvement of exercise capacity of nonathlete may be 2 hours and HC diet, respectively. But it was showed that the exercise capacity of athlete may not be affected by experimental condition of resting time, except 1 hour after feeding or of dietary composition because of well adaptation in new exercise condition.
