The Arabidopsis female gametophyte contains seven cells with eight haploid nuclei buried within layers of sporophytic tissue. Following double fertilization, the egg and central cells of the gametophyte develop into the embryo and endosperm of the seed, respectively. The epigenetic status of the central cell has long presented an enigma due both to its inaccessibility, and the fascinating epigenome of the endosperm, thought to have been inherited from the central cell following activity of the DEMETER demethylase enzyme, prior to fertilization. Here, we present for the first time, a method to isolate pure populations of Arabidopsis central cell nuclei. Utilizing a protocol designed to isolate leaf mesophyll protoplasts, we systematically optimized each step in order to efficiently separate central cells from the female gametophyte. We use initial manual pistil dissection followed by the derivation of central cell protoplasts, during which process the central cell emerges from the micropylar pole of the embryo sac. Then, we use a modified version of the Isolation of Nuclei TAgged in specific Cell Types (INTACT) protocol to purify central cell nuclei, resulting in a purity of 75-90% and a yield sufficient to undertake downstream molecular analyses. We find that the process is highly dependent on the health of the original plant tissue used, and the efficiency of protoplasting solution infiltration into the gametophyte. By isolating pure central cell populations, we have enabled elucidation of the physiology of this rare cell type, which in the future will provide novel insights into Arabidopsis reproduction.

This paper discusses an energy system that can convert wave energy into electrical energy. This wave energy generation system is movable and has 12 arms and one generator. A multibody dynamic model for this system is established by using kinematic constraints. A gear mechanism, several kinematic constraints, and force elements are included in the model. Wave forces are obtained numerically from the time domain formulation based on the Morison equation. The MSC/ADAMS program is employed to carry out dynamic analysis of the wave energy generation system. The dynamic behavior responses of this system are analyzed for design verification. According to the results of the dynamic analysis, the yaw motion is relatively stable and kinetic energy sufficient to generate electrical energy is obtained when the wave height exceeds 1m.
What van Fraassen calls 'Reflection Principle' is claimed to meet several counterexamples, one of which stands out in the form of the Sleeping Beauty problem. Adam Elga argues that what he believes is the correct answer to the Sleeping Beauty problem shows that Reflection is subject to counterexamples. David Lewis proposes a different answer which preserves Reflection intact. Recently, Nick Bostrom presents a hybrid view which is supposed to allow us to keep Reflection. In proposing his hybrid view Bostrom criticizes both Elga and Lewis while taking some 'good' parts from each. He claims that Elga's view is not entirely acceptable because it presupposes the 'Self-Indication Assumption'. I shall claim, however, that Elga could avoid Bostrom's criticisms by introducing Bostrom's notion of agent-part. I believe that several probability-related puzzles including the Sleeping Beauty problem indicate a promising view concerning the way we should regard our future selves' opinions. According to this view, whether one takes the outsider stance or insider stance makes a difference in an important way that one and the same proposition is associated with different degrees of belief by one agent.
In this study, we developed a machine learning-based model for predicting the production stage temperature of distillation process. It is necessary to predict an accurate temperature for control because the control of the distillation process is done through the production stage temperature. The temperature in distillation process has a nonlinear complex relationship with other variables and time series data, so we used the recurrent neural network algorithms to predict temperature. In the model development process, by adjusting three recurrent neural network based algorithms, and batch size, we selected the most appropriate model for predicting the production stage temperature. LSTM128 was selected as the most appropriate model for predicting the production stage temperature. The prediction performance of selected model for the actual temperature is RMSE of 0.0791 and R2 of 0.924.
This paper investigated the influence of simple support girds on flow, irrespective of having mixing vanes, in a 1 × 3 array rod bundle by using CFD methodology and the most accurate turbulence model which could reflect the actual physics of the flow was determined. In this context, a CFD model was created simulating the experimental studies on a single-phase flow [1] and the results were compared with the experimental data. In the first part of the study, influence of mesh was examined. Tetra, hybrid and poly type meshes were analyzed and convergence study was carried out on each in order to determine the most appropriate type and density. k - ε Standard and RSM LPS turbulence models were used in this section. In the second part of the study, the most appropriate turbulence model that could reflect the physics of the actual flow was investigated. RANS based turbulence models were examined using the mesh that was determined in the first part. Velocity and turbulence intensity results obtained on the upstream and downstream of the spacer grid at -3dh, +3dh and +40dh locations were compared with the experimental data. In the last section of the study, the behavior of flow through the spacer grid was examined and its prominent aspects were highlighted on the most appropriate turbulence model determined in the second part. Results of the study revealed the importance of mesh type. Hybrid mesh having the largest number of structured elements performed remarkably better than the other two on results. While comparisons of numerical and experimental results showed an overall agreement within all turbulence models, RSM LPS presented better results than the others. Lastly, physical appearance of the flow through spacer grids revealed that springs has more influence on flow than dimples and induces transient flow behaviors. As a result, flow through a simple support grid was examined and the most appropriate turbulence model reflecting the actual physics of the flow was determined.
AWS (all wheel steering) is applied to improve the stability and the turning performance. Most automotive cars are mainly controlled by FWS (front wheel steering) system except some cars which are made to improve their stability by using AWS. Articulated vehicles with a pivoting joint for easy turn are difficult to make a sharp turn because of the long body and long wheelbase. Therefore applying AWS to the articulated vehicles is effective to reduce the turning radius. The AWS control method for the articulated vehicles is currently applied to only Phileas vehicles which were developed by APTS. The paper on the design of a controller to guide an articulated vehicle along the path was published but control algorithm for manual driving has not been reported. In the present paper, steering, characteristics of the Phileas vehicles have been analyzed and then new algorithm has been proposed. To verify the AWS algorithm, Commercial S/W, ADAMS was used for validity of the dynamic model and algorithm.

In order to investigate how the microstructure of fuel/matrix-interaction (FMI) layers change during irradiation, different U-7Mo dispersion fuel plates have been irradiated to high fission density and then characterized using scanning electron microscopy (SEM). Specifially, samples from irradiated U-7Mo dispersion fuel elements with pure Al, Al-2Si and AA4043 (~4.5 wt.%Si) matrices were SEM characterized using polished samples and samples that were prepared with a focused ion beam (FIB). Features not observable for the polished samples could be captured in SEM images taken of the FIB samples. For the Al matrix sample, a relatively large FMI layer develops, with enrichment of Xe at the FMI layer/Al matrix interface and evidence of debonding. Overall, a significant penetration of Si from the FMI layer into the U-7Mo fuel was observed for samples with Si in the Al matrix, which resulted in a change of the size (larger) and shape (round) of the fission gas bubbles. Additionally, solid fission product phases were observed to nucleate and grow within these bubbles. These changes in the localized regions of the microstructure of the U-7Mo may contribute to changes observed in the macroscopic swelling of fuel plates with Al-Si matrices.

In this paper, we studied some design methodologies of typical FIR filters based on the peak-error constrained least square criterion which was first introducedd by Adams in 1991. This method is a mixed type of the classical least squared error method(LSM) and the so-called min-max error method (MMM). And by considering both the least squared error as well as the maximum error, the solution, i.e. the impulse response of the filter, can be found only when the restrictions on maximum gain, transition bandwidth, and the squared error are satisfied simultaneously under some trade-off conditions. We used the multiple exchange algorithms for optimization procedure and applied the design methodology to the cases of the multiband filter, the differentiator, and the Hilbert transformer by taking the balance of two design criteria into account. The results show that the peak-error constrained least weighted square error design method(PLEM) is superior in performance to the existing LSM and MMM from both the squared error and the maximum error standpoints. And it is verified that PLEM can be applied to not only the case of simple low pass filter, but also to various types of FIR filters.

The performance of ultrafiltration (UF) membranes with molecular weight cut off (MWCO) of 1000 and 3500 Da in clarifying sugar cane juice was investigated, as well as the performance of a nanofiltration (NF) membrane with MWCO of 200 Da and a reverse osmosis (RO) membrane in concentrating sugar cane juice. For both cases the sugar cane juice had been limed and partially clarified. The UF membranes were found to be effective at clarifying the sugar cane juice in terms of purity rise and reduction in turbidity, colour, starch and protein. A purity rise of approximately 6 was achieved by both UF membranes at trans-membrane pressures (TMP) from 15 to 25 bar. However, Brix reduction in the permeate was between 14.5 and 41.85% and 12.11 and 26.52% for 1000 Da and 3500 Da membranes respectively. For the 200 Da and RO membranes the Brix in the concentrate was increased from 7.65 to 12.3 after 3 hours of operation for the 200 Da membrane at a TMP of 10 bar, whilst the Brix in the concentrate was increased from 15.65 to 27.6 after 3 hours of operation for the RO membrane at a TMP of 35 bar. Overall, UF membranes were found to be unsuitable for clarification of sugar cane juice since significant amount of Brix is reduced in the permeate, whilst RO membranes were found to be effective for concentration of sugar cane juice.
Objective: This study tried to examine the muscle activity of the neck and trunk according to head posture changes during use of laptops. Design: Cross-sectional study. Methods: This study included nineteen young men and women. All subjects maintained each posture while practice typing on laptops for ten minutes with a 5-minute break between each posture. For the neutral head posture, the subjects practiced typing while pulling their chins down towards the Adam's apple and were able to look at their knees while having their external auditory meatus, acromion, and greater trochanter vertical to the ground. For the natural head posture, they practiced typing while balancing their posture between extension and flexion of the cervical vertebrae by themselves without any guidelines. While a forward head posture was created by having their heads face the front horizontal to the rope hanging from the ceiling, they practiced typing with their external auditory meatus located in the place which was 5 cm in front of the vertical plane. The subjects used general word process but practiced typing with accuracy and high speed. Muscle activities were randomly measured using surface electromyography according to each postures. Results: The research result had found that muscle activity with the natural head posture was more significantly reduced than that of the forward head posture in the sternocleidomastoid muscle, upper trapezius, cervical vertebral spinae, and thoracic vertebrae spinae muscles (p<0.05) and that the neutral head posture was more significantly reduced than that of forward head posture in the upper trapezius, cervical vertebral spinae, and thoracic vertebrae spinae muscle activity (p<0.05) with significant increases in lumbar spinae muscle activity (p<0.05). Also, muscle activity with the neutral head posture significantly increased more in the sternocleidomastoid muscle and lumbar spinae than that of the natural posture (p<0.05). Conclusions: Our study results suggest that in order to prevent musculoskeletal pain, the neutral head posture with use of laptops is effective in reducing load to the shoulders and vitalizing the postural muscles.
