CJ-11555의 안전성 약리실험 (Safety Pharmacology of CJ-11555)

  • Safety pharmacological properties of CJ-11555, an anti-cirrhotic agent, were investigated in experimental animals and in vitro test system. CJ-11555 had no effects on normal body temperature in rats, motor coordination, chemoshock induced by pentetrazol, electric shock induced by electric shocker and writhing syndromes in mice at dose levels of 100, 300 and 1,000 mg/kg. CJ-11555 inhibited intestinal activity and prolonged hexobarbital-induced sleeping time in mice at the dose level of 1,000 mg/kg. CJ-11555 affected on general activity and behaviour tests in SD rats, such as lacrimation, ptosis, piloerection, decreased body tone, abnormal dispersion within the cage, diarrhoea, red colored faeces, slight hypothermia and decreased grooming, at the dose level of 1,000 mg/kg in rats. CJ-11555 was effected on cardiovascular and respiratory system in anesthetized beagle dogs, such as tachycardia, increase of mean blood pressure and decrease of PR interval, decrease of respiratory rate and minute volume, at dose levels of 10 and 30 mg/kg. However, these effects were also observed in vehicle treated anesthetized beagle dogs. In in vitro experiments, CJ-11555 inhibited agonists (histamine, acetyl-choline or $BaCl_2$) induced contraction of isolated guinea-pig at the concentration of 30$\times$$10^6$ M. CJ-11555 was weekly inhibited hERG channel current at concentrations of 10 and 30$\times$$10^6$ M, and $IC_{50}$ was estimated to be higher than 30${\times}$$10^6$M. Based on these results, it was concluded that CJ-11555 affected on cardiovascular and respiratory system, general activity and behaviour and hexobarbital-induced sleeping time at the dose level of 1,000 mg/kg and contraction of the smooth muscle and hERG channel current at the concentration of 30$\times$$10^6$ M.

수온 및 하상 변화에 대한 참실지렁이 (Tubifex tubifex)의 행동 반응 (Behavioral Response of Tubifex tubifex to Changes of Water Temperature and Substrate Composition)

  • 담수에 서식하는 참실지렁이(Tubifex tubifex)가 수온 변화에 따라 보이는 반응과 하상의 구성에 대한 선호성을 분석하였다. 수온이 증가할 때 참실지렁이는 활동성이 증가하였다. $10{\sim}16^{\circ}C$의 낮은 온도에서 50% 이상의 개체가 몸을 수축하고 꼬여있는 상태로 관찰되었으며, $24^{\circ}C$ 이상의 온도에서는 대부분의 개체가 몸을 꼬임 없이 이완시킨 상태로 관찰되었다. 하상입자의 크기가 다른 여러 하상을 이용한 실험에서는 가장 작은 입자인 미사-진흙 (silt-clay, <0.063 mm)을 선호하였다.

Korean Red Ginseng reduces chronic social defeat stress-induced mood disorders via N-methyl-D-aspartate receptor modulation in mice

  • Background: A chronic social defeat stress (CSDS) model has been proposed as relevant to stress-induced behavioral change in humans. In this study, we examined the effect of Korean Red Ginseng (KRG) on CSDS-induced mood disorders and protein expression in an animal model. Methods: To evaluate the effect of KRG on social defeat stress, test mice were exposed in the resident aggressor's home cage compartment for 14 days beginning 1 h after KRG treatment (10, 20, and 40 mg/kg, per oral (p.o.)). After the exposure, behavioral tests to measure anxiety, social interaction, and depression-like behavior were performed. To investigate the underlying mechanism, N-methyl-D-aspartate receptor expression levels in CSDS-induced mice were evaluated using Western blot analysis. Results: CSDS induced anxiety-like behaviors by decreasing central activity in the open-field test and open-arm approach in the elevated plus maze test and led to social avoidance behavior in the social interaction test. CSDS mice showed upregulated NR1, NR2A, and NR2B expression in the hippocampus. KRG 20 and 40 mg/kg ameliorated anxiety-like activities and KRG 20 mg/kg alleviated social avoidance by decreasing time in the corner zone. KRG treatment recovered CSDS-induced NR1, NR2A, and NR2B protein levels in the hippocampus. Conclusion: These results indicate that KRG has a therapeutic effect on CSDS-induced mood disorder by alleviating N-methyl-D-aspartate receptor overexpression in the hippocampus.

Dichlorvos 증기(蒸氣)의 지속적(持續的) 흡입(吸入)이 인체(人體) 및 가계(家鷄)에 미치는 영향 (Studies on the potential hazard of dichlorvos-resin vaporizing insecticide in human and domestic fowl)

  • Safety study of the continuous releasing dichlorvos-resin insecticide $(Mopari^{\circledR})$ was conducted in human volunteers and domestic fowls. For the purpose, the potential hazards in using the insecticide were observed in terms of the inhibition of plasma cholinesterase activity and the changes in the liver function (GOT, GPT, Alkaline phosphatase, Bilirubin, Thymol turbidity), the blood picture (RBC, WBC with differential count, Hemoglobin, Hematocrit and ESR) and the urine picture (sugar, albumin, pH and microscopic findings) in 40 healthy adult volunteers and 60 leghorn domestic fowls. In case of the human study the observation was continued for 2 months during the application of the insectiside ($1{\sim}3$ solid formulations/$30m^3$) in the living rooms of ordinary Korean dwelling houses or in the office. In the animal test, however, 1 to 5 solid formulations of the insecticide were applied in the fowl cage of $9.2m^3$ for 5 weeks. Any significant inhibition of the plasma cholinesterase activity was not observed in both the human volunteer and the fowl throughout the experimental period. And the liver function as well as the blood and urine pictures were also not changed after exposure to the vaporizing insecticide. It is considered from the result that the amount of dichlorvos released into the air by the continuous vaporizing dichlorvos-resin insecticide presents no significant hazardous effect on humans or animals in the present experimental condition.

Role of fumarates in adaptogenics like efficacies of traditionally used Fumaria indica extracts

  • Fumaria indica Linn. (Syn: Fumaria parviflora, Fumariaceae) is a wildly grown weed, mentioned and recommended in classical Ayurvedic texts for treatments of variety of ailments including dermatological diseases, topical diseases, cardiovascular complaints, circulatory disease, fever and headache etc. The present pilot study was designed to experimentally verify the possibility that fumarates are the major bioactive principles of Fumaria indica extracts involved in their stress response modulating activities, and to estimate pharmacologicallyactive dose ranges of fumarates and standardized methanolic extract of Fumaria indica (MFI). Effect of single, 5 and 10 daily oral doses of pure fumaric acid (FA), monomethyl fumarate (MMF), dimethyl fumarate (DMF) and MFI was quantified in well validated rodent models viz. apomorphine induced cage climbing, stress induced hyperthermia, and elevated plus-maze tests. Obtained results reveal high efficacy of MFI and pure fumarates possess qualitatively analogous activity profiles in all the three tests. There were no significant difference in the potencies of pure FA, MMF and DMF in the three tests, whereas efficacy of MFI in the elevated plus maze test for anxiolytics was higher than in the other two tests. Efficacies of all the four test agents in all the three tests increased with increasing number of days of oral treatments. Results of these pilot experiments should be helpful for more rational selections of pharmacologically interesting dose ranges and treatment regimens of fumarates and Fumaria indica extracts for further more holistic explorations of their diverse therapeutic potentials.

Effects of Eurycoma longifolia Jack on Masculine Copulatory Behaviour in Middle Aged Male Rats - A Comparison Study

  • The effects of Eurycoma longifolia Jack on masculine copulatory behaviour were studied in the middle aged male Sprague-Dawley rats, 9 months old and retired breeders after dosing them with 500 mg/kg twice daily for 10 days prior to test. The test lasted for 30 minutes after a 20 minute adaptation period, was carried out on the 11th day during the dark phase of the light-dark cycle (2000-0700 hours) and in subdued light, using a modified copulation cage but with the presence of a piece of mirror of appropriate size to facilitate observation. Results showed that the mean values of EL-1, EL-2 and EL-3 of the control middle aged male rats were 103.20 sec, 91.21 sec and 80.00 sec but were significantly (p<0.05) increased to 118.40-120.20 sec, 101.24-171.28 sec and 100.42-110.21 sec respectively in the methanol-chloroform, methanol-butanol-water and methanol-butanol treated middle aged male rats. However, further results also showed that PEI-1 and PEI-2 of the control middle aged male rats were 182.30 sec and 257.2 sec but were significantly (p<0.05) decreased to 100.42-121.31 sec and 40.21-132.31 sec respectively in the methanol-chloroform-butanol-water and methanol-butanol treated middle aged male rats. In conclusion, this study showed that although E. longifolia Jack continued to enhance the sexual activity of the middle aged male rats by extending the duration of coitus and decreasing the refractory period between the different series of copulation, but to a smaller degree as compared to sexually active, adult male rats (Ang and Sim, 1997).

수질 모니터링을 위한 유해 물질 유입에 따른 생물체의 행동 반응 분석 및 인식 (Analysis and Recognition of Behavioral Response of Selected Insects in Toxic Chemicals for Water Quality Monitoring)

  • 본 논문에서는 자동 추적 시스템을 이용하여 카바메이트 계열의 농약인 카보퓨란의 치명적인 투여에 대하여 반자연적인 조건에서 반응하는 깔따구의 움직임을 관찰하였다. 4령기에 있는 깔따구를 $6cm\times{7cm}\times{2.5cm}$ 크기의 서식 장소와 $18^\circ{C}$의 수온, 명기와 암기를 각각 10시간, 14시간의 조건에서 관찰을 하였다. 추적 시스템은 깔따구 몸체의 부분 점들을 탐지하여 추적하도록 하였다. 모든 실험은 반자연적인(semi-natural) 상태에서 진행되었으며 약제 카보퓨란(Carbofuran 0.1mg/l) 처리 전 후 이틀씩 모두 4일에 걸쳐서 연속적으로 진행되었다. 실험 결과 약제의 처리후에 압축된 지그제그 형태로 나타나는 "떨림 현상"과 같은 비정규적인 행동들이 종종 나타남을 알 수 있었다. 약제 처리된 종들의 행동 변화를 탐지하기 위하여, 웨이블릿 분석이 다른 움직임 패턴들을 특징화하기 위하여 사용되었다. 이산 웨이블릿에 기반하여 추출된 파라미터들은 약제처리 전후의 움직임에 대한 다른 유형의 패턴들을 표현하기 위하여 인공 신경망을 통하여 학습되었다. 이러한 웨이블릿과 인공 신경망의 통합 모델은 특징화된 움직임 패턴들의 발생 시점을 탐지할 수 있었으며, 수질 모니터링을 위한 독성 물질의 유입을 자동으로 탐지할 수 있는 도구로써 사용될 수 있음을 알 수 있었다.을 알 수 있었다.

복숭아순나방(Grapholita molests) 성페로몬 합성과 유인력 교란생물검정 (Chemical Synthesis and Orientation Disruption Bioassay of Sex Pheromone of the Oriental Fruit Moth, Grapholita molests (Busck))

  • 유기합성 기술을 통해 복숭아순나방(Grapholita molesta)의 성페로몬 세 가지 성분(cis-8-dodecenyl acetate (Z12Ac), trans-8-dodecenyl acetate (E12Ac), cis-8-dodecenol (Z12OH))을 제조하였다. 이중결합 페로몬 성분의 cis 이성질체 비율을 높이기 위해 이 부위의 삼중결합 유도후 $Pd/BaSO_{4}$$-20^{\circ}C$에서 환원시켰다. 최종산물인 아세테이트류의 광학이성체 비율은 92:8(Z:E)을 나타냈다. 이들 합성 페로몬의 생물활성을 검정하기 위해 유인력 분석과 유인력교란 분석으로 나누어 실시되었다. 유인력 분석은 실내에서는 수컷 날개행동유기를 기준으로 분석하였다. 표준구로서 상용 유인제를 이용하였다. 분석 페로몬 조제는 아세테이트류와 알코올류가 각각, 100:0, 99:1, 99:10(g/g)으로 조제하여 비교하였다. 실내 교미 유인력은 100:0에서 가장 높았고, 알코올류가 포함된 조성은 낮은 유인력을 보였다. 야외 유인력은 페로몬 트랩을 이용하였으며, 실내 분석과 같이 100:0에서 가장 많은 수컷 유인력을 나타냈다. 유인력교란 분석은 실내에서 간이모형을 이용하였다. 중앙에 상용페로몬을 끈끈이판에 놓고, 주위 네 방향으로 분석될 페로몬을 설치한 후, 12시간 노출시험을 진행하였다. 이 결과 상용페로몬과 합성페로몬(100:0)은 모두 100% 유인교란 효과를 보였다. 그러나 교란용 페로몬의 숫자가 줄어듬에 따라 유인력교란효과는 낮아지는 현상을 나타냈다. 이상의 결과는 복숭아순나방의 합성페로몬이 생물적 활성을 지니며, 교미교란에 이용될 수 있음을 제시하고 있다.
