Effect of Iron Supplementation on Iron-Deficiency-Related Indices, Oxidative Stress and Antioxidative Enzyme Activity in Female Marathoners

  • This study was performed to evaluate the effect of iron supplementation on iron-deficiency-related indices, oxidative stress and antioxidative enzyme activity in female marathoners. Fourteen teenage female marathoners participated in the study. Subjects were divided into two groups: mild anemic and control, depending on their hemoglobin (Hb) level. The mild anemic group had significantly lower RBC count and hematocrit (Hct) and Hb levels compared to the control group. The mild anemic group (〈12.5g Hb/dI, n=7) was given iron supplements (60mg Fe/day) for four weeks during the summer training period. RBC count, Hct and Hb levels showed an increasing tendency through iron supplementation, and significant differences in these variables between the anemic and control groups disappeared in the post-period. There was no difference in plasma malondialdehyde (MDA) between the anemic and control groups. However, catalase (CAT) and glutathione peroxidase (GPx) activity were significantly higher in the anemic group. The significant difference in enzyme activity between the groups disappeared in the post-period. In addition, superoxide dismutase activity significantly decreased after iron supplementation. In conclusion, antioxidative enzyme activity was up-regulated in an anemic condition and mild iron supplementation decreased the antioxidant enzyme activity of female marathoners while improving their anemic condition.

Physiological Functionalities and Anti-oxidant activity of heated radish extract

  • This study proceeded an experiment that can increase such physiological properties of heated radish extract. During the extraction of radish, including the byproduct, an increase in antioxidant properties of radish according the heating temperature was performed. Upon the extracts of radish bark and radish green extract(mucheong), the physiological functionalities and antioxidant activity were investigated. As a result, the color of radish ethanol extract in dependence of heating temperature, showed light brown color at low heating temperature and black color from $150^{\circ}C$. The total polyphenol content significantly increased as a result of heat treatment; 6.7 times and 22 times higher than the control at $110^{\circ}C$ and $150^{\circ}C$, respectively. DPPH radical scavenging ability and antioxidant property increased with increasing heating temperature; in comparison to heat-treated radish at $110^{\circ}C$ and $150^{\circ}C$, $IC_{50}$ decreased by 1/22 times. $IC_{50}$ of the control was 23times higher than $150^{\circ}C$ heat treated radish (Control $IC_{50}$:130.305). According to the graph that represents ABTS activity, antioxidant activity increased in dependence of heat treatment likewise to the total polyphenol content and DDPH radical scavenging activity. Upon heat treatment at $150^{\circ}C$, antioxidant activity in consequence of ABTS assay increased 23 times higher than the control.

Study on Bromobenzene Metabolism in Rats with Liver Damage (흰쥐에 있어서 간손상 정도에 따른 Bromobenzene 대사)

  • To compare the severe liver damage with the slight one on the bromobeazene metabolism in rats, the animal group described as B7 group was induced the stage of slight liver damage with 7 times bromobenzene injection every other day (400 mg/Kg body wt. i.p.), whereas B40 group was induced that of more severe liver damage with bromobeazene 40 times injection as identified with determination of serum levels of alanine aminotransferase(ALT) activity and the histopathological findings. In the present experimental animal model, the decreasing rate of glutathione(GSH) and the increasing rate of glutathione S-transferase activity to the control group were higher in B7 group than B40 group. Furthermore the single dose of bromobenzene was injected to the two groups and sacrificed at 8hr and the hepatic aniline hydroxylase(AH) activity, GSH content and GST activity were determined. The increasing rate of AH activity to the control was lower in B40 group than B7 group and the decreasing rate of GSH to the control was also lower in B40 than B7 group. Moreover, B7 group showed the increased activity of hepatic GST to the control whereas B40 group showed the decrease activity of the enzyme. And Vmax value in GST was more decreased in B40 group than B7 group.

Antioxidative Activity and Tyrosinase Inhibition Effect of Ethanol Extract and Its Fractions from the Branch of Rhododendron schlippenbachii (철쭉 가지 에탄올 추출물 및 용매별 분획물의 항산화 활성과 Tyrosinase 저해 활성)

  • This study was conducted to investigate the antioxidative activity and tyrosinase inhibitory activity of 50% ethanol extract and its fractions from the branch of Rhododendron schlippenbachii. In DPPH radical scavenging ability, butanol and ethyl acetate fractions showed 59.98% and 55.17% of relative activity compared with positive control (ascorbic acid), but the 50% ethanol extract showed relatively low activity. In nitric oxide (NO) scavenging ability, the ethyl acetate and butanol fractions showed 141.80% and 131.55% relative activity compared with ascorbic acid as used for positive control. On the other hand, tyrosinase inhibitory activity of the ethyl acetate and butanol fractions showed about twice higher activity than positive control (arbutin). It means that the ethyl acetate and butanol fractions from the extract of R. schlippenbachii branch has ability for used as effective radical scavenger and tyrosinase inhibitor.

  • In this paper, we examine the effect of production uncertainty to production control policies. First, we examine two famous production control policies, namely, MRP and JIT from the view point of shop floor control perspective, and analyze the differences between them due to demand fluctuations and activity time variations. Second, we conduct simulation studies on MRP and JIT to draw out the effects of demand fluctuations and activity time variations. Demand fluctuations are further classified into demand lumpiness and demand irregularity. And, activity time variations are further classified into stationary time variations and non-stationary time variations. Experimental results show that, in terms of demand fluctuations, MRP is affected by demand lumpiness, but JIT by demand irregularity. And we also see that both MRP and JIT are influenced by stationary time variation with respect to activity time variations.

  • This study was carried out effect of red ginseng extract(1.0g/kg) on organotin compounds (10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kindey, testis, ovary, serum immuinty and sex hormone activity of rats were examinned by gastric tubing for 3 weeks. The weight of each organ in TBTO treated group were significantly increased other organs which excepted kinedy in males and only liver in females.(p<0.05, p<0.01). In case of Immunity activity of each sex, IgM level was small change comparsion with that of control group in all sex. but IgG level was significantly decreased females rather than males comparsion with that of control group.(p<0.05, p<0.01) In case of sex hormone activity, the testosterone activity of males and the estradiol activity of females were significantly decreased rather than the control group. on the other hand, red singsong treated group was only significantly increased estradiol activity.( p<0.05, p<0.01)

  • This paper proposed resource allocation algorithm for the minimum switching activity of functional unit in high level synthesis process as like DSP which is circuit to give many functional unit. The resource allocation method after scheduling use the power function calculating average hamming distance and switching activity of the between two input. First of all, the switching activity is calculated by the input value after calculating the average hamming distance between operation. In this paper, the proposed method though high If level simulation find switching activity in circuit each functional unit exchange for binary sequence length and value bit are logic one value. To use the switching activity find the allocation with minimal power consumption, the proposed method visits all control steps one by one and determines the allocation with minimal power consumption at each control step. As the existing method, the execution time can be fast according to use the number of operator and max control step. And it is the reduction effect from 6% to 8%.

Effects of Water Extracts of Camelia sinensis L on Blood Alcohol Concentration and Activities of Sub-acute Alcohol Metabolic Enzymes in ICR Mouse (ICR Mouse의 아급성 알코올 대사에 보이차(Camelia sinensis L) 추출물이 미치는 효과)

  • An eight-week-old male ICR mouse, which was induced with acute alcohol and sub-acute alcohol poisoning condition, was administered with bohee tea(Camelia sinensis L) extract. Under the inducement of the sub-acute alcohol poisoning condition, no considerable differences could be found in the blood alcohol concentration of the positive control group and the bohee tea group(p<0.05). The GOT activity of the three groups: bohee tea, Drink, and Alcodex decreased than that of the normal control group(9.064±4.687 unit)(p<0.05). In addition, the blood GOT activity of the dark green tea group dropped by 81.44% compared with that of the positive control group. On the other hand, the blood GTP activity of the bohee tea group decreased by 5.2% as opposed to that of the positive control and the Drink that decreased by 7.5% as opposed to that of the positive control. The hepatic ADH activity of the bohee tea increased by 22.7%, as opposed to that of the positive control group. The Drink, however, had an increase rate of 33.6%. In the case of the hepatic ALDH activity of the liver, no significant differences were ever recorded among all groups, except for the positive control group. Due to an intake of bohee tea extract, the hepatic ALDH activity decreased by 77.27% which could not be seen in the positive control group. However, Drink and A1codex had a decrease could be seen(p<0.05).

Purification, Characterization, and Cloning of Fibrinolytic Metalloprotease from Pleurotus ostreatus Mycelia

  • A fibrinolytic protease (PoFE) was purified from the cultured mycelia of the edible oyster mushroom Pleurotus ostreatus, using a combination of various chromatographies. The purification protocol resulted in an 876-fold purification of the enzyme, with a final yield of 6.5%. The apparent molecular mass of the purified enzyme was estimated to be 32 kDa by SDS-PAGE, fibrin-zymography, and size exclusion using FPLC. The optimal reaction pH value and temperature were pH 6.5 and $35^{\circ}C$, respectively. PoFE effectively hydrolyzed fibrinogen, preferentially digesting the $A{\alpha}$-chain and the $B{\beta}$-chain over the ${\gamma}$-chain. Enzyme activity was enhanced by the addition of $Ca^{2+},\;Zn^{2+},\;and\;Mg^{2+}$ ions. Furthermore, PoFE activity was potently inhibited by EDTA, and it was found to exhibit a higher specificity for the chromogenic substrate S-2586 for chymotrypsin, indicating that the enzyme is a chymotrypsin-like metalloprotease. The first 19 amino acid residues of the N-terminal sequence were ALRKGGAAALNIYSVGFTS, which is extremely similar to the metalloprotease purified from the fruiting body of P. ostreatus. In addition, we cloned the PoFE protein, encoding gene, and its nucleotide sequence was determined. The cDNA of cloned PoFE is 867 nucleotides long and consists of an open reading frame encoding 288 amino acid residues. Its cDNA showed a high degree of homology with PoMEP from P. ostreatus fruiting body. The mycelia of P. ostreatus may thus represent a potential source of new therapeutic agents to treat thrombosis.

A study on the Effects of Rehmannia Radix and Eukmigihwangtang (EMGHT) on Antioxidation Activity in Aging Rats (숙지황(熟地黃)과 육미지황탕(六味地黃湯)이 노화과정(老化過程) 흰쥐에서의 항산화(抗酸化) 기전(機轉)에 미치는 영향(影響))

  • This experimental study was designed to verify the anti-aging efficacy of Eukmigihwangtang (EMGHT) and Rehmannia Radix, and determine the specific role and actions of Rehmannia Radix. Normal rat (2 months old), aging rat (8 months old), and pathologically induced rat (2 months old, injected 30mg/kg of streptozotocin) are observed to study the aging eliciting factors such as peroxide contents and enzyme activities. The following results were obtained in this study: 1. For the body weight changes, normal group given Rehmannia Radix showed decrease in the body weight compared to the control group, aging group given EMGHT and Rehmannia Radix showed significant decrease in the body weight, and STZ injected group showed suppression to the body weight loss when given EMGHT and Rehmannia Radix. 2. For the content changes in serum lipid peroxide, normal group showed increasing level as the rat gets older. Aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the lipid peroxide level compared to the control group. Decrease was more prominant in the group given EMGHT. 3. For the changes in serum hydroxyl radical, normal group did not show significant changes, but aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the hydroxyl radical level compared to the control group. Decrease was more prominant in the group given EMGHT. 4. For the changes in serum superoxide dismutase (SOD) activity, normal group did not show significant changes, but aging group given EMGHT and Rehmannia Radix showed significant increase in the SOD activity compared to the control group. STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the SOD activity compared to the control group. 5. For the content changes in hepatic lipid peroxide, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the lipid peroxide level compared to the control group. 6. For the changes in hepatic cytochrome P-450 activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease compared to the control group. Cytochrome b5 activity was significantly decreased only in the STZ injected group given EMGHT and Rehmannia Radix. 7. For the changes in hepatic aminopyrine demethylase and aniline hydroxylase activity, aging group given EMGHT and Rehmannia Radix showed significant decrease compared to the control group. STZ injected group given EMGHT and Rehmannia Radix showed significant increase in the aminopyrine demethylase activity, and showed significant decrease in the aniline hydroxylase activity compared to the control group. 8. For the content changes in hepatic protein bound-SH and nonprotein bound-SH, againg group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase compared to the control group. 9. For the content changes in hepatic glutathione level, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase compared to the control group. 10. For the changes in hepatic glutathione S-transferase activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase and decrease, respectively, compared to the control group. 11. For the changes in hepatic glutathione reductase activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase compared to the control group, while $\gamma$-Glutamylcystein synthetase activity did not show significant changes. 12. For the changes in hepatic superoxide dismutase activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease compared to the control group. From the above results, the antioxidant effects of EMGHT and Rehmannia Radix were proved, as well as the role of Rehmannia Radix, a chief of EMGHT, was examined. In addition, since no change was reconized as the quantity of Rehmannia Radix and the order herbs increased, the reasonableness on EMGHT was proven with respect to its composition and quantity. Thus, the significance of EMGHT could be objectively exmined in terms of its composition and quantity. Considering animals used in the experiment, there were obvious changes in aging rats and pathologically induced rats than in normal rats. Consequently, it was noticeable that EMGHT and Rehmannia Radix were working selectively on the subjects.
