$K^+$ outward currents in the outer hair cells (OHCs) of circling mice (homozygous (cir/cir) mice), an animal model for human deafness (DFNB6 type), were investigated using a whole cell patch clamp technique. Littermate heterozygous (+/cir) mice of the same age (postnatal day (P) 0-P6) were used as controls. Similar slow rising $K^+$ currents were observed in both genotypes, but their biophysical and pharmacological properties were quite different. The values of Vhalf for activation were significantly different in the heterozygous (+/cir) and homozygous (cir/cir) mice ($-8.1{\pm}2.2mV$, heterozygous (+/cir) mice (n=7) and $-17.2{\pm}4.2mV$, homozygous (cir/cir) mice (n=5)). The inactivation curve was expressed by a single first order Boltzmann equation in the homozygous (cir/cir) mice, while it was expressed by a sum of two first order Boltzmann equations in the heterozygous (+/cir) mice. The $K^+$ current of homozygous (cir/cir) mice was more sensitive to TEA in the 1 to 10 mM range, while the 4-AP sensitivities were not different between the two genotypes. Removal of external $Ca^{2+}$ did not affect the $K^+$ currents in either genotype, indicating that the higher sensitivity of $K^+$ current to TEA in the homozygous (cir/cir) mice was not due to an early expression of $Ca^{2+}$ activated $K^+$ channels. Our results suggest that the $K^+$ outward current of developing homozygous (cir/cir) mice OHCs is different in both biophysical and pharmacological aspects than that of heterozygous (+/cir) mice.
Shilajit, a medicine herb commonly used in Ayurveda, has been reported to contain at least 85 minerals in ionic form that act on a variety of chemical, biological, and physical stressors. The substantia gelatinosa (SG) neurons of the trigeminal subnucleus caudalis (Vc) are involved in orofacial nociceptive processing. Shilajit has been reported to be an injury and muscular pain reliever but there have been few functional studies of the effect of Shilajit on the SG neurons of the Vc. Therefore, whole cell and gramicidin-perfotrated patch clamp studies were performed to examine the action mechanism of Shilajit on the SG neurons of Vc from mouse brainstem slices. In the whole cell patch clamp mode, Shilajit induced short-lived and repeatable inward currents under the condition of a high chloride pipette solution on all the SG neurons tested. The Shilajit-induced inward currents were concentration dependent and maintained in the presence of tetrodotoxin (TTX), a voltage gated $Na^+$ channel blocker, CNQX, a non-NMDA glutamate receptor antagonist, and AP5, an NMDA receptor antagonist. The Shilajit-induced responses were partially suppressed by picrotoxin, a $GABA_A$ receptor antagonist, and totally blocked in the presence of strychnine, a glycine receptor antagonist, however not affected by mecamylamine hydrochloride (MCH), a nicotinic acetylcholine receptor antagonist. Under the potassium gluconate pipette solution at holding potential 0 mV, Shilajit induced repeatable outward current. These results show that Shilajit has inhibitory effects on the SG neurons of Vc through chloride ion channels by activation of the glycine receptor and $GABA_A$ receptor, indicating that Shilajit contains sedating ingredients for the central nervous system. These results also suggest that Shilajit may be a potential target for modulating orofacial pain processing.

ClC-1 is a member of a large family of voltage-gated chloride channels, abundantly expressed in human skeletal muscle. Mutations in ClC-1 are associated with myotonia congenita (MC) and result in loss of regulation of membrane excitability in skeletal muscle. We studied the electrophysiological characteristics of six mutants found among Korean MC patients, using patch clamp methods in HEK293 cells. Here, we found that the autosomal dominant mutants S189C and P480S displayed reduced chloride conductances compared to WT. Autosomal recessive mutant M128I did not show a typical rapid deactivation of Cl- currents. While sporadic mutant G523D displayed sustained activation of $Cl^-$ currents in the whole cell traces, the other sporadic mutants, M373L and M609K, demonstrated rapid deactivations. $V_{1/2}$ of these mutants was shifted to more depolarizing potentials. In order to identify potential effects on gating processes, slow and fast gating was analyzed for each mutant. We show that slow gating of the mutants tends to be shifted toward more positive potentials in comparison to WT. Collectively, these six mutants found among Korean patients demonstrated modifications of channel gating behaviors and reduced chloride conductances that likely contribute to the physiologic changes of MC.

We investigated the effects of trinitrobenzene sulfonic acid (TNBS), an amino-group reagent, on the human ether-a-go-go-related gene (HERG) $K^+$ channels expressed in Xenopus oocytes. TNBS neutralizes the positively charged amino-groups of peptide N-terminal and lysine residues. External application of TNBS at 10 mM for 5 min irreversibly shifted the curves for currents at the end of the pulse and tail currents of HERG to a more negative potential and decreased the maximal amplitude of the $I_{tail}$ curve $(I_{tail,max})$. TNBS had little effect on either the activated current-voltage relationship or the reversal potential of HERG current, indicating that TNBS did not change ion selectivity properties. TNBS shifted the time constant curves of both activation and deactivation of the HERG current to a more hyperpolarized potential; TNBS's effect was greater on channel opening than channel closing. External $H^+$ is known to inhibit HERG current by shifting $V_{1/2}$ to the right and decreasing $I_{tail,max}$. TNBS enhanced the blockade of external $H^+$ by exaggerating the effect of $H^+$ on $I_{tail,max}$, not on $V_{1/2}$. Our data provide evidence for the presence of essential amino-groups that are associated with the normal functioning of the HERG channel and evidence that these groups modify the blocking effect of external $H^+$ on the current.
Recently the business environment surrounding the financial investment industry is changing rapidly, and the demands of customers (diversity and the cycle of change etc.) are getting shorter. In this context, it can be said that companies are forced to adopt an agile management environment. In particular, non-face-to-face channels, including MTS, is adopting the agile system into the digital finance sector from a company-wide and strategic perspective. This study analyzed the effects of MTS services' environment quality on customer satisfaction and continuous intention to use for MTS users who are rapidly increasing under the agile management environment in the financial investment industry. This study surveyed the quality of service environment (accessibility, convenience, design, security), customer satisfaction, and continuous intention to use for 350 MTS users. First, accessibility, convenience, and security of MTS service environment quality had a positive effect on customer satisfaction, and design was rejected Second, customer satisfaction has a positive effect on continuous intention to use. Third, convenience and security of MTS service environment quality have positive effects on continuous intention to use, and accessibility and design were rejected. The results of this study, together with demographic analysis, are expected to provide useful implications for MTS activation studies and securities firms' strategies.
Background: Trigeminal neuralgia is a debilitating craniofacial pain syndrome that is characterized by paroxysms of intense, short-lived electric shock-like pains in the trigeminal nerve distribution. Recently, the presence of triggers has become one of the key diagnostic criteria in the 3rd edition of the International Classification of Headache Disorders. Light touch is the most common trigger, however other non-mechanical triggers, such as cold weather and certain foods, have been thought to provoke trigeminal neuralgia anecdotally. We aimed to characterize the prevalence and characteristics of these atypical triggers. Methods: We conducted a retrospective, cross-sectional study of atypical triggers in trigeminal neuralgia patients seen in a tertiary pain clinic in Singapore. Patients were recruited via clinic records, and study data were identified from physician documentation. Results: A total of 60 patients met the inclusion criteria. Weather triggers were observed in 12 patients (20%), of which five patients (8%) reported strong winds, 4 patients (7%) reported cold temperatures, and 3 patients (5%) reported cold winds as triggers. Fifteen patients (25%) had a specific food trigger, of which 10 patients (17%) reported hard or tough food, 5 patients (8%) reported hot/cold food, 4 patients (7%) reported spicy food, and 2 patients (3%) reported sweet food as triggers. Conclusions: Although trigeminal neuralgia is most commonly triggered by mechanical stimuli, atypical triggers such as cold temperatures and certain foods are seen in a significant proportion of patients. These atypical triggers may share a common pathway of sensory afferent Aδ fiber activation.
상이한 특성을 가진 퍼블릭 블록체인과 프라이빗 블록체인에 대한 전체적인 동작을 통합적으로 교육하는 것은 용이하지 않다. 현재 블록체인의 개념과 동작을 위한 교육은 대부분 비트코인, 이더리움과 같은 퍼블릭 블록체인을 대상으로 하고 있다. 그러나 실제 기업환경에서는 사용자에 대한 인증을 통하여 기업 데이터에 접근통제를 하기 때문에 하이퍼레저패블릭(HLF)과 같은 프라이빗 블록체인이 활용되고 있다. HLF 기반으로 교육하는 경우에는 피어, 오더러, 채널 등 퍼블릭 블록체인에 없는 다양한 구성요소에 대한 이해가 필요하다. 본 논문에서는 이와 같은 구성요소의 기능과 동작에 대한 효율적이고 체계적인 이해를 위하여 실습프레임워크를 설계하였다. 설계된 프레임워크은 HLF 네트워크제어 체인코드제어 탈중앙화소프트웨어(Decentralized Application, dApp) 제어기능으로 구성되어 있다. 프레임워크을 통하여 네트워크을 구성하고 체인코드의 배포 및 활성화 후 dApp 실행과정을 단계적으로 확인하였고 블록체인 서비스를 위한 전체 흐름을 이해하는 것이 매우 용이하였다. 또한 향후 네트워크의 확장 등에도 전체 흐름에 대한 체계적인 이해가 가능할 것으로 기대한다.
Objective: This study aims to verify the front squat motions using by two different surfaces, thereby elucidating the grounds for effective training environment that can minimize the risk of injury. Method: Total of 10 healthy male crossfit athletes were recruited for this study (age: 32.30 ± 3.05 yrs., height: 173.70 ± 5.12 cm, body mass: 82.40 ± 6.31 kg, 1RM: 160 ± 13.80 kg). All participants are those who know how to do front squats well with more than five years of crossfit athlete experience. All participants have sufficient experience in front squats on two types of surface which are soft surface (SS) and hard surface (HS). In each surface, participant perform 10reps of the front squat with 80% of the pre-measured 1RM. A 3-dimensional motion analysis with 8 infrared cameras and 2 channels of EMG was performed in this study. Paired sample t-test was used for statistical verification between two surfaces. The significant level was set at α=.05. Results: The significantly decreased rectus femoris muscle activation was found in SS compared with that of HS on phase 1 (p<.05). Also, ROM of ankle joint was significantly increased in the SS compare with that of HS on phase 1 (p<.05). The muscle activity ratio of gluteus maximus/rectus femoris showed a significant difference only in SS compared with that of HS on phase 1 (p<.05). Conclusion: In conclusion, Korean crossfit boxes should consider the use of hard surface, which has a relatively less risk of injury than soft surface, in selecting flooring materials. For the Crossfit athletes, they are also considered appropriate to train on a relatively hard surface.

The purpose of this study is to investigate the recyclability of Shenlong in China using SWOT. The main analysis results are as follows. First, provided that the company's current capacity utilization rate is seriously insufficient, reducing staff is one among the effective ways. Second, Shenlong should open a web store to cater to young people's online shopping behavior, and further expand the brand visibility using national mainstream media and online shopping platforms like Taobao and JingDong to market Dongfeng Peugeot and Dongfeng Citroen on the whole network. Third, under the premise of maintaining the present best-selling models, Shenlong should appropriately reduce the amount of models, adjust the assembly capacity ratio of every model and every displacement in real time per the newest market trends, increase the agility of auto companies' production, and timely meet the wants of domestic consumers. Fourth, dual-brand coordination and channel integration are very necessary, and also the profitability and profitability of dealers are going to be further improved, thereby increasing sales. Fifth, target building new energy leading products of Shenlong, strive to attain the forefront of the industry within the sales of recent energy vehicles within 5 years, and gradually expand new energy vehicle products from passenger vehicles to passenger vehicles and commercial vehicles. In this study, pneumonia identification networks with the small number of layers were constructed by using chest X-ray images. The networks had similar trainable-parameters, and the performance of the trained models was quantitatively evaluated with the modification of the network architectures. A total of 6 networks were constructed: convolutional neural network (CNN), VGGNet, GoogleNet, residual network with identity blocks, ResNet with bottleneck blocks and ResNet with identity and bottleneck blocks. Trainable parameters for the 6 networks were set in a range of 273,921-294,817 by adjusting the output channels of convolution layers. The network training was implemented with binary cross entropy (BCE) loss function, sigmoid activation function, adaptive moment estimation (Adam) optimizer and 100 epochs. The performance of the trained models was evaluated in terms of training time, accuracy, precision, recall, specificity and F1-score. The results showed that the trained models with the small number of layers precisely detect pneumonia from chest X-ray images. In particular, the overall quantitative performance of the trained models based on the ResNets was above 0.9, and the performance levels were similar or superior to those based on the CNN, VGGNet and GoogleNet. Also, the residual blocks affected the performance of the trained models based on the ResNets. Therefore, in this study, we demonstrated that the object detection networks with the small number of layers are suitable for detecting pneumonia using chest X-ray images. And, the trained models based on the ResNets can be optimized by applying appropriate residual-blocks.
