A theoretical investigation of the solid phase mechanochemical synthesis of nano sized target product on the basis of dilution of the initial powdered reagent mixture by another product of an exchange reaction is presented. On the basis of the proposed 3 mode particle size distribution in mechanically activated mixture, optimal molar ratios of the components in mixture are calculated, providing the occurrence of impact friction contacts of reagent particles and excluding aggregation of the nanosized particles of the target reaction product. Derivation of kinetic equations for mechanochemical synthesis of nanoscale particles by the final product dilution method in the systems of exchange reactions is submitted. On the basis of obtained equations the necessary times of mechanical activation for complete course of mechanochemical reactions are designed. Kinetics of solid phase mechanosynthesis of nano TlCl by dilution of initial (2NaCl+$Tl_2SO_4$) mixture with the exchange reaction product (diluent,$zNa_2SO_4$, z=z*=11.25) was studied experimentally. Some peculiar features of the reaction mechanism were found. Parameters of the kinetic curve of nano TlCl obtained experimentally were compared with those for the model reaction KBr+TlCl+zKCl=(z+1) KCl+TlBr (z=z1*=13.5), and for the first time the value of mass transfer coefficient in a mechanochemical reactor with mobile milling balls was evaluated. Dynamics of the size change was followed for nanoparticle reaction product as a function of mechanical activation time.

Heat/smoke detectors are installed in most subway platforms in Korea to detect fire. Subway platform is divided by smoke-control zones for efficient smoke-control. Once the detectors detect heat or smoke, the smoke-control ventilation system in the platform and concourse is activated according to the smoke-control ventilation mode. Smoke-control mode during fires in Korean subway platforms is that the smoke zones operate by exhausting smoke while other zones in the platform and in the concourse which is the upper floor of the platform operate by supplying air or stopping any ventilation. This study is conducted to evaluate performance of passengers' evacuation for various smoke control modes in the subway station. Distribution of smoke and heat due to fire on the platform is analyzed by using Fire Dynamics Simulator(FDS V 4.06) of NIST. Various smoke-control ventilation modes and locations of fire are considered. Evacuation and movement of passengers within the platform is simulated by buildingEXODUS V 4.0.

Some peculiar features of the reaction mechanism were found. Parameters of the kinetic curve of nano-TlCl obtained experimentally were compared with those for the model reaction KBr + TlCl + zKCl = (z + 1) KCl + TlBr ($z=z_l^*=13.5$), and for the first time the value of mass transfer coefficient in a mechanochemical reactor with mobile milling balls was evaluated. Fire suppression using a water mist nozzle directly above an n-Heptane pool in a fire compartment with a door opening was numerically investigated using the Fire Dynamics Simulator (FDS) for the purpose of application in nuclear power plants. Input parameters for the numerical simulation were determined by experimental measurements. Water mist was activated 10 s after the fire began. The sensitivity analysis was conducted for three input parameters: total number of cubic cells of 6032-2,926,400, droplets per second of 1000-500,000, and extinguishing coefficient of 0-100. In a new simple calibration method of this study, the extinguishing coefficient yielding the fire suppression time closest to that measured by experiments was found for use as the FDS simulation input value. When the water mist jet flow made contact with the developed fire, the heat release rate instantaneously increased, and then rapidly decreased. This phenomenon occurred with a displacement of the flame near the liquid fuel pool. Changing the configuration of the door opening with different aspect ratios and opening ratios had impact on the maximum value of the heat release rate due to the flame displacement.
Kori unit #1, which is the first commercial nuclear power plant in Korea, was permanently shutdown in June 2017, and it is about to be decommissioned. Currently in Korea, researches on the decommissioning technology are actively conducted, but there are few researches on workers internal exposure to radioactive aerosol that is generated in the process of decommissioning nuclear power plants. As a result, the over-exposure of decommissioning workers is feared, and the optimal working time needs to be revised in consideration of radioactive aerosol. This study investigated the annual exposure limits of various countries, which can be used as an indicator in evaluating workers' internal exposure to radioactive aerosol during pipe cutting in the process of decommissioning nuclear power plants, and the growth and dynamics of aerosol. Also, to evaluate it, the authors compared/analyzed the cases of aerosol generated when activated pipes are cut in the process of nuclear power plants and the codes for evaluating internal exposure. The evaluation codes and analyzed data conform to ALARA, and they are believed to be used as an important indicator in deriving an optimal working time that does not excess the annual exposure limit.

Janus kinase 2 (JAK2), a non-receptor tyrosine kinase, is a critical component of cytokine and growth factor signaling pathways regulating hematopoietic cell proliferation. JAK2 mutations are associated with multiple myeloproliferative neoplasms. Although physiological and pathological functions of JAK2 in hematopoietic tissues are well-known, such functions of JAK2 in the nervous system are not well studied yet. The present study demonstrated that JAK2 could negatively regulate neuronal differentiation of mouse embryonic stem cells (ESCs). Depletion of JAK2 stimulated neuronal differentiation of mouse ESCs and activated glycogen synthase kinase 3β, Fyn, and cyclin-dependent kinase 5. Knockdown of JAK2 resulted in accumulation of GTP-bound Rac1, a Rho GTPase implicated in the regulation of cytoskeletal dynamics. These findings suggest that JAK2 might negatively regulate neuronal differentiation by suppressing the GSK-3β/Fyn/CDK5 signaling pathway responsible for morphological maturation.
A family of signal transduction pathways known as wingless type (Wnt) signaling pathways is essential to developmental processes like cell division and proliferation. Mutation in Wnt signaling results in a variety of diseases, including cancers of the breast, colon, and skin, metabolic disease, and neurodegenerative disease; thus, the Wnt signaling pathways have been attractive targets for disease treatment. However, the complicatedness and large involveness of the pathway often hampers pinpointing the specific targets of the metabolic process. In our current study, we investigated the differential metabolic regulation by the overexpression of the Wnt signaling pathway in a timely-resolved manner by applying high-throughput and un-targeted metabolite profiling. We have detected and annotated 321 metabolite peaks from a total of 36 human embryonic kidney (HEK) 293 cells using GC-TOF MS and LC-Orbitrap MS. The un-targeted metabolomic analysis identified the radical reprogramming of a range of central carbon/nitrogen metabolism pathways, including glycolysis, TCA cycle, and glutaminolysis, and fatty acid pathways. The investigation, combined with targeted mRNA profiles, elucidated an explicit understanding of activated fatty acid metabolism (β-oxidation and biosynthesis). The findings proposed detailed mechanistic biochemical dynamics in response to Wnt-driven metabolic changes, which may help design precise therapeutic targets for Wnt-related diseases.
For the efficient design of baffled Powdered activated carbon(PAC) contractor, which has been widely used in water treatment plant(WTP) against the algae-related odor problems, a CFD(computational fluid dynamics) program was applied. In order to verify the performance of FLOW-3D program, the previously reported results of tracer tests from a pilot-scale PAC contractor(working volume of 288 liters) were compared to those from FLOW 3D. The results of FLOW-3D simulation were very similar to those from tracer tests conducted with the Pilot-scale PAC contactor. On the other hand, the hydrodynamic characteristics of baffled contractor in the P-WTP were simulated by using FLOW-3D. Simulation results on the distribution of PAC particles showed that there are some stagnant parts in the back side of baffles in which PAC Particles are not present. These stagnant parts might decrease the adsorption capacity of PAC particles. When the baffles were changed to maze-type intra-basin baffling, PAC particles were evenly distributed and the amount of stagnant parts reduced. In conclusion, it is anticipated that FLOW-3D simulation could be a viab1e tool for analyzing the hydrodynamic characteristics of structures used in drinking water treatment plant.

The Hippo kinase cascade functions as a central hub that relays input from the "outside world" of the cell and translates it into specific cellular responses by regulating the activity of Yes-associated protein 1 (YAP1). How Hippo translates input from the extracellular signals into specific intracellular responses remains unclear. Here, we show that transforming growth factor β (TGFβ)-activated TAK1 activates LATS1/2, which then phosphorylates YAP1. Phosphorylated YAP1 (p-YAP1) associates with RUNX3, but not with TEAD4, to form a TGFβ-stimulated restriction (R)-point-associated complex which activates target chromatin loci in the nucleus. Soon after, p-YAP1 is exported to the cytoplasm. Attenuation of TGFβ signaling results in re-localization of unphosphorylated YAP1 to the nucleus, where it forms a YAP1/TEAD4/SMAD3/AP1/p300 complex. The TGFβ-stimulated spatiotemporal dynamics of YAP1 are abrogated in many cancer cells. These results identify a new pathway that integrates TGFβ signals and the Hippo pathway (TGFβ→TAK1→LATS1/2→YAP1 cascade) with a novel dynamic nuclear role for p-YAP1.
Kinesin is an ATP-driven microtubule motor protein that plays important roles in control of microtubule dynamics, intracellular transport, cell division and signal transduction. The kinesin superfamily is composed of numerous members that are classified into 14 subfamilies. Animal kinesins have been well characterized. In contrast, plant kinesins have not yet to be characterized adequately. Here, a novel plant-specific kinesin gene, GhKCH2, has been cloned from cotton (Gossypium hirsutum) fibers and biochemically identified by prokaryotic expression, affinity purification, ATPase activity assay and microtubule-binding analysis. The putative motor domain of GhKCH2, $M_{396-734}$ corresponding to amino acids Q396-N734 was fused with 6$\times$His-tag, soluble-expressed in E. coli and affinity-purified in a large amount. The biochemical analysis demonstrated that the basal ATPase activity of $M_{396-734}$ is not activated by $Ca^{2+}$, but stimulated 30-fold max by microtubules. The enzymatic activation is microtubule-concentration-dependent, and the concentration of microtubules that corresponds to half-maximum activation was about 11 ${\mu}M$, much higher than that of other kinesins reported. The cosedimentation assay indicated that $M_{396-734}$ could bind to microtubules in vitro whenever the nucleotide AMP-PNP is present or absent. As a plant-specific microtubule-dependent kinesin with a lower microtubule-affinity and a nucleotide-independent microtubule-binding ability, cotton GhKCH2 might be involved in the function of microtubules during the deposition of cellulose microfibrils in fibers or the formation of cell wall.
