Isolation and Characterization of Benzene-degrading Bacteria. (Benzene 분해 세균의 분리와 특성연구)

  • To evaluate the treatability of activated sludge induced by benzene with microorganisms, isolation and characterization of benzene-degrading microorganisms were carried out. Six bacterial isolates from the activated sludge were identified ; Pseudomonas fluorescens, Enterobacter agglomerans, Enterobacter cloacae, Klebsiella oxytoca, Citrobacter freundii and Klebsiella pneumoniae. P. fluorescens degraded 55% of benzene contained in the medium as a sole carbon source, E. cloacae 24%, E. agglomerans 41%, and K. oxytoca 32%. Optimal temperature, pH and benzene concentration for growth of P. fluorescens appeared to be 31$^{\circ}C$, pH 7.0, and 300mg benzene per liter. When the P. fluorescens was dominant in the activated sludge induced by benzene, the indicator protozoa was Aspidisca sp. When concentration of benzene was about 387mg per liter, the growths of Aspidisca sp. and Litonotus sp. were high. Protozoa, Litonotus sp. and Vorticella sp. did not grow over 1600mg of benzene per liter.

  • Biohydrogen production from organic wastewater by anaerobically activated sludge fermentation has already been extensively investigated, and it is known that hydrogen can be produced by glucose fermentation through three metabolic pathways, including the oxidative decarboxylation of pyruvic acid to acetyl-CoA, oxidation of NADH to $NAD^+$, and acetogenesis by hydrogen-producing acetogens. However, the exact or dominant pathways of hydrogen production in the anaerobically activated sludge fermentation process have not yet been identified. Thus, a continuous stirred-tank reactor (CSTR) was introduced and a specifically acclimated acidogenic fermentative microflora obtained under certain operation conditions. The hydrogen production activity and potential hydrogen-producing pathways in the acidogenic fermentative microflora were then investigated using batch cultures in Erlenmeyer flasks with a working volume of 500 ml. Based on an initial glucose concentration of 10 g/l, pH 6.0, and a biomass of 1.01 g/l of a mixed liquid volatile suspended solid (MLVSS), 247.7 ml of hydrogen was obtained after a 68 h cultivation period at $35{\pm}1^{\circ}C$. Further tests indicated that 69% of the hydrogen was produced from the oxidative decarboxylation of pyruvic acid, whereas the remaining 31% was from the oxidation of NADH to $NAD^+$. There were no hydrogen-producing acetogens or they were unable to work effectively in the anaerobically activated sludge with a hydraulic retention time (HRT) of less than 8 h.

Treatment Characteristics of Rapid filtration Process treating Secondary Clarifier Effluent for Wastewater Reuse (처리수 재이용을 위한 최종침전지 유출수의 급속여과공정 처리특성)

  • This reserch was focused upon experimental study for wastewater reuse and conducted to evaluate optimum operating conditions of rapid filtration process such as filter flow rate, filtration time and backwashing condition for reuse of secondary-treated effluent using the pilot plant installed in real wastewater treatment plant. Also, the experiment on treatment char-acteristics of coagulant-added activated sludge process was performed to compare with activated sludge succeeded to rapid filtration. As the filtration velocity was 100m/day, the filtration time of the rapid filter connected with activated sludge system was revealed to 40 hours. Backwashing of filter was conducted by water wash and air scour. The optimum backwashing time and backwash flow rate were 10min and 10LPM, respectively. The quantity of backwashing water of the rapid filter was about 2% of total treated water.

Types and Abundance of Filamentous Bacteria, Protozoa and Metazoa in Activated Sludge (활성오니에서 발견된 사상세균, 원생동물 및 후생동물의 유형과 우점도 분석)

  • Filamentous bacteria and other large microorganisms, which are useful indicators of bulking and poeration, were identified by microscopic observation. Activated sludge samples were taken from the aeration basin of 5 municipal wasterwater plants and 6 industrial ones. Among the filamentous bacteria founds, Microthrix parvicella was most frequently present, followed by type 0041, type 1701 and Nocardia. This frequency of occurrence was similar to those reorted in USA and the Neterland. The morphology of filamentous bacteria observed were generally identical with those previously reported. except type 1701 which had slender filament diameter. Among protozoa, Vorticella was most frequently present, followed by Aspidisca, Opercularia and Difflugia. Philodina was the only metazoa observed. Both filamentous bacteria and protozoa would be useful indicator organisms. The potential for these organisms as indicators were discussed.

A Study on the Treatment of Swine Wastewater by Using Intermittently Aerated Activated Sludge Process (간헐폭기법에 의한 돈사 폐수 처리에 관한 연구)

  • In this study, an intermittently aerated activated sludge process, modified process from conventional activated sludge process, was developed to treat high strength swine wastewater, which has been blamed as major pollutant for stream pollution. Therefore, the optimum cycle for oxic and anoxic period, SRT, and OLR were studied as design parameters. The effects of different time interval for oxic and anoxic period on nitrification and denitrification were examined by operating two reactors with 60/60min and 60/90min as oxic/anoxic period. Although the reactor with 60/60min showed complete denitrification of $NO_x-N$ generated during oxic period, the reactor with 60/90min showed incomplete nitrification due to the inactivity of nitrifier by accumulated $NH_3-N$ toxicity during anoxic period. Therefore, it is recommended to operate same interval for oxic and anoxic period. In order to determine the optimum cycle for oxic/anoxic period, four different reactors with 30/30, 60/60, 90/90 and 120/120min were examined. The reactor operation with 90/90min was optimum to get the most stable results in this study. However, the optimum cycle for oxic and anoxic period should be changed with characteristics of influent wastewater and operating conditions. According to lie operation results of three reactors with SRT of 15, 20 and 30days. The reactor with 2Odays SRT showed best removal efficiency of T-N. The optimum OLR would be $2.5Kg\;COD/m^3/day$ which showed the most stable nitrification and denitrification. Since characteristics of influent wastewater in the real system has a severe fluctuation, so it is very difficult to determine each interval for oxic and anoxic period. Therefore, ORP curves, describing the change of oxidation/reduction potential in reactor, can be used as a control parameter for automatic control of oxic and anoxic period. In other words, bending point (Nitrate Knee) of ORP curve during anoxic period could be used as a starting point of oxic period.

Evaluation of Performance and Economical Efficiency of the Advanced Wastewater Treatment System (고도(高度) 하수처리(下水處理) 시스템의 처리성능 및 경제성 평가에 관한 연구)

  • For a high-rate fermentation and recovery of organic acid, we have developed a new organic acid fermentation reactor with membrane filter, which is the most important part in the new advanced wastewater treatment system. The recovered organic acid is to be reused as an organic carbon source at denitrification process. Some experiments were conducted to compare the performance of acid fermentation at different SRTs, such as 5, 10, and 20 days. The total organic acid concentration produced during the runs was in the range of 2,100-2,900 (mgC/L). The conversion efficiency from substrate to organic acid reached to from 43% to 59%. The recovery rate of organic acid from substrate based on TOC was from 26% to 53%. Regardless of operational conditions, it has been able to maintain the membrane flux constantly, in the range of 0.4-0.46 ($m^3/m^2/day$). The transmembrane pressure drop was 0.2-0.3 (kg/cm) for 100 day's operation. The result of simulation is as follows. Organic removal efficiency of the new advanced treatment system is 95%. 73% of Nitrogen is removed. The removal efficiency of Phosphorus is 93%. By coqulation, soluble phosphorus is able to remove from the water treatment lines, which is impossible at conventional activated sludge system. The unit construction cost is 65000 (yen/m3) and it was 1.4 times than that of the standard activated sludge system. The unit operation cast is 7.7 ($yen/m^3/day$) and it was 1.3 times than that of the standard activated sludge system.

Stage by stage design for primary, conventional activated sludge, SBR and MBBR units for residential wastewater treatment and reusing

  • To date, there is no central wastewater (WW) treatment plant in Erbil city, Kurdistan region, Iraq. Therefore, raw WW disposes to the environment and sometimes it used directly for irrigation in some areas of Erbil city. Disposal of the untreated WW to the natural environment and using for irrigation it causes problems for the people and the environment. The aims of the current work were to study the characteristics, design of primary and different secondary treatment units and reusing of produced WW. Raw WW samples from Ashty city-Erbil city were collected and analyzed for twenty three quality parameters such as Total Suspended Solids (TSS), total dissolved solids, total volatile and non-volatile solids, total acidity, total alkalinity, total hardness, five-day Biochemical Oxygen Demand (BOD5), Chemical Oxygen Demand (COD), biodegradability ratio (BOD5/COD), turbidity, etc. Results revealed that some parameters such as BOD5 and TSS were exceeded the standards for disposal of WW. Design and calculations for primary and secondary treatment (biological treatment) processes were presented. Primary treatment units such as screening, grit chamber, and flow equalization tank were designed and detailed calculation were illustrated. While, Conventional Activated Sludge (CAS), Sequencing Batch Reactor (SBR) and Moving Bed Biofilm Reactors (MBBR) were applied for the biological treatment of WW. Results revealed that MBBR was the best and economic technique for the biological treatment of WW. Treated WW is suitable for reusing and there is no restriction on use for irrigation of green areas inside Ashty city campus.

  • CaO-based by-products composed of CaO, SO3, Al2O3, etc. are generally used as raw materials for CaO compounds. When applied to the recovered water of ready-mixed concrete, the hydration reaction of the powder material is accelerated and concrete performance can be improved. In this study, activated sludge was prepared to apply to the recovered water of ready-mixed concrete by mixing CaO-based hot-rolled slag(C12A7) in the recycling water of ready-m ixed concrete. Cem ent paste setting time and mortar compressive strength performance tests confirmed the effect on the hydration reaction. Therefore, the possibility of concrete application using activated sludge was confirmed.

  • CaO-based by-product, which consist of CaO, SO3, Al2O3 and so on, has being used to raw materials of CaO compound. When It was applied to recycling water of remicon, concrete performance can be enhanced because hydration reaction of powder material is accelerated. In this study, activated-sludge, which was putted desulfurization gypsum of CaO-based in recycling water, was manufactured to verify effect of them, and then they was investigated by characteristics of redy-mixde concrete. As a result of concrete tests, it was confirmed that there is no problem of strength or drying shrinkage while ensuring workability. Therefore, the possibility of specific application using activated sludge was confirmed.

Effect of Sludge Age on the Toxicity of Cr6+, Zn2+, and Cd2+ in INT-Dehydrogenase Assay (INT-Dehydrogenase 시험법에서 슬러지 일령이 Cr6+, Zn2+ 및 Cd2+ 독성에 미치는 영향)

  • This study was initiated to elucidate the relation between the toxicity of $Cr^{6+}$, $Zn^{2+}$, and $Cd^{2+}$ and sludge age (Solids retention time, SRT). The effect of SRT on the toxicity of $Cr^{6+}$, $Zn^{2+}$, and $Cd^{2+}$ in activated sludge system was investigated with INT-dehydrogenase assay. Experimental results showed that the inhibitory effects of $Cr^{6+}$, $Zn^{2+}$, and $Cd^{2+}$ were reduced as the sludge age increased from SRT 5 d to SRT 25 d. It is noteworthy that the experimental results enabled to determine the relative toxicity of the tested metals depending on the sludge age. At the SRT of 5 and 9 days, the order of toxicity of the three metals to the activated sludge was $Cr^{6+}$ > $Zn^{2+}$ > $Cd^{2+}$ (the $IC_{50}$ value of $Cr^{6+}$, $Zn^{2+}$, and $Cd^{2+}$ was 16.15, 25.90, and 32.49 mg/L, respectively) and $Zn^{2+}$ > $Cr^{6+}$ > $Cd^{2+}$ (the $IC_{50}$ value of $Cr^{6+}$, $Zn^{2+}$, and $Cd^{2+}$ was 39.12, 27.93, and 42.31 mg/L, respectively), respectively. However, the toxicity of three metals was almost same at the SRT of 14 and 25 days (the range of $IC_{50}$ in SRT 14 and 25 days was from 49.80 mg/ L to 53.44 mg/L among three heavy metals). This results would be explained by that the quantity of biopolymer formed in short SRT was small, whereas that in long SRT was large. Consequently, it is recommended that sludge age be maintained at long SRT in order to avoid the toxicity inhibition of heavy metals such as $Cr^{6+}$, $Zn^{2+}$, and $Cd^{2+}$.
