Saccharomyces cerevisiae 의 mother cell과 daughter cell 의 연결 부위의 원형질막 바로 안쪽에 존재하는 10-nm filament ring 은 세포형태 형성과정에 중요한 역할을 하리라 간주되나 그 명확한 생성기작과 기능은 밝혀지지 않았다. 본연구에서는 CDC12 유전자로부터 gene fusion technique 을 이용하여 CDC12 단백질을 만들고 이로부터 항체를 형성하였다. 이항체를 이용하여 10-nm filament ring 의 생성기작과 기능에 대하여 연구하였다. 그 결과 CDC12 단백질은 cell cycle 전주기동안 항상 정이되나 bud 가 나오기 바고 직전에 bud 가 나올 부위에 polymerization 되었다가 세포질분열 바로 직후에 unpolymerization 되며 cytoskeletal element 의 일종인 actin 과는 무관하게 행동하는 건이 밝혀졌다. 이러한 10-nm filament 는 bud 가 나올 부위의 올바른 선정과 세포질 분열에 중요한 역할을 하리라 간주된다.

Associations between periodontal infection and cardiovascular disease have been documented. Porphyromonas gingivalis is a well-established periodontal pathogen, and tissue factor (TF) is a key initiator of the coagulation cascade. In this context, P. gingivalis has been reported to enhance TF expression in human endothelial cells. The present study investigated the underlying mechanisms of TF induction by P. gingivalis in human umbilical vein endothelial cells. P. gingivalis increased TF expression in a dose- and time-dependent manner. Not only live bacteria but also glutaraldehyde-fixed bacteria increased TF expression to the same extent. However, sonicates of P. gingivalis did not induce TF expression. Cytochalasin D and SMIFH2, which are inhibitors of actin polymerization and actin nucleation, respectively, inhibited the TF expression induced by P. gingivalis. Finally, TF production was decreased or increased in the presence of various signaling inhibitors, including mitogen-activated protein kinases. These results suggest that P. gingivalis induces endothelial TF expression by a bacterial internalization-dependent mechanism and through diverse signal transduction mechanisms.

Ginsenoside Rg3, a saponin extracted from ginseng, has various pharmacological and biological activities; however, its effects against Brucella infection are still unclear. Herein, the inhibitory effects of ginsenoside Rg3 against intracellular parasitic Brucella infection were evaluated through bacterial infection, adherence assays, and LAMP-1 colocalization, as well as immunoblotting and FACS for detecting MAPK signaling proteins and F-actin polymerization, respectively. The internalization, intracellular growth, and adherence of Brucella abortus in Rg3-treated RAW 264.7 cells were significantly decreased compared with the Rg3-untreated control. Furthermore, an apparent reduction of F-actin content and intensity of F-actin fluorescence in Rg3-treated cells was observed compared with B. abortus-infected cells without treatment by flow cytometry analysis and confocal microscopy, respectively. In addition, treating cells with Rg3 decreased the phosphorylation of MAPK signaling proteins such as ERK 1/2 and p38 compared with untreated cells. Moreover, the colocalization of B. abortus-containing phagosomes with LAMP-1 was markedly increased in Rg3-treated cells. These findings suggest that ginsenoside Rg3 inhibits B. abortus infection in mammalian cells and can be used as an alternative approach in the treatment of brucellosis.
비정상적 액틴의 재구성은 암세포의 대표적 특성이다. Thymosin ${\beta}_{10}$ (TB10)과 Profilin-1 (PFN-1)은 액틴중합조절에 필수적인 단백질이다. 이전의 연구에서 본 연구진은 TB10이 F-actin의 구조를 파괴하여 난소 암 세포의 사멸을 일으킨다는 사실을 보고하였으나 그 기전에 대하여 보고 된 바는 아직까지 없다. 본 연구에서는 TB10에 의하여 PFN-1의 발현이 조절되며, PFN-1의 난소 암 저해 유전자로서의 새로운 기능을 보고하였다. 우선 난소암세포주인 SKOV3 세포에서 TB10에 의하여 발현이 조절되는 단백질들을 전기영동법과 liquid chromatography-mass spectroscopy (LC-MS/MS) 방법을 통하여 분석하였다. 그 결과 PFN-1이 TB10에 의하여 발현이 급격히 증가되는 단백질로 동정되었으며, 이 PFN-1을 난소 암 세포주인 SKOV3에 과발현 시켰을 때 암세포의 증식과 이동을 저해하고 암세포 사멸을 유도하였다. 또한 이 결과는 PFN-1에 의하여 Erk 신호전달기전이 저해되고 부수적으로 Elk-1과 Egr-1의 발현이 저해 됨으로써 유도될 가능성을 보여준다. 결론적으로, PFN-1이 난소암세포의 성장과 이동을 저해함과 동시에 세포사멸을 일으키므로 난소 암 치료에 유용하게 이용될 가능성이 높다.

This study was conducted to investigate cytoskeleton alterations during vitrified (Open Pulled Straw method) porcine immature oocytes, to utilize Taxol/sup TM/ (polymerization of tubulin molecules) and Cytochalasin B (CB, depolymerization of actin filaments) during vitrification to stabilize microtubule and microfilaments (MT and MF), and to determine in vitro maturation, fertilization and development of cytoskeletal-stabilized and vitrified porcine immature oocytes. A low acid phosphatase 3 (lap3) mutant was identified and characterized from an Arabidopsis activation-tagged (Weigel) population. The roots of the lap3 plants showed lower acid phosphatase (APase) activity compared to wild-type ones under low-Pi conditions ($10{\mu}M\;Pi$). Plasmid rescue experiments revealed that the activation-tagging vector was inserted into the intergenic region between At4g31540 and At4g31550 in the Arabidopsis genome. The genotypic segregation of the lap3 mutation was tightly linked with the phenotypic segregation of root APase activity in the prgeny of lap3. The transcript level of the At4g31520 (SDA1: SEVERE DEPOLYMERIZATION OF ACTIN 1), located 7.4 kb from the CaMV 35S enhancers in the lap3 mutant, was significantly reduced compared to that in the wild type. It was speculated that cellular actin polymerization may be involved in Pi acquisition in higher plants.
PTPRT/RPTPρ is the most recently isolated member of the type IIB receptor-type protein tyrosine phosphatase family and its expression is restricted to the nervous system. PTPRT plays a critical role in regulation of synaptic formation and neuronal development. When PTPRT was overexpressed in hippocampal neurons, synaptic formation and dendritic arborization were induced. On the other hand, knockdown of PTPRT decreased neuronal transmission and attenuated neuronal development. PTPRT strengthened neuronal synapses by forming homophilic trans dimers with each other and heterophilic cis complexes with neuronal adhesion molecules. Fyn tyrosine kinase regulated PTPRT activity through phosphorylation of tyrosine 912 within the membrane-proximal catalytic domain of PTPRT. Phosphorylation induced homophilic cis dimerization of PTPRT and resulted in the inhibition of phosphatase activity. BCR-Rac1 GAP and Syntaxin-binding protein were found as new endogenous substrates of PTPRT in rat brain. PTPRT induced polymerization of actin cytoskeleton that determined the morphologies of dendrites and spines by inhibiting BCR-Rac1 GAP activity. Additionally, PTPRT appeared to regulate neurotransmitter release through reinforcement of interactions between Syntaxin-binding protein and Syntaxin, a SNARE protein. In conclusion, PTPRT regulates synaptic function and neuronal development through interactions with neuronal adhesion molecules and the dephosphorylation of synaptic molecules. In this study, we report that one of the antimicrobial peptides scolopendrasin VII, derived from Scolopendra subspinipes mutilans, stimulates actin polymerization and the subsequent chemotactic migration of macrophages through the activation of ERK and protein kinase B (Akt) activity. The scolopendrasin VII-induced chemotactic migration of macrophages is inhibited by the formyl peptide receptor 1 (FPR1) antagonist cyclosporine H. We also found that scolopendrasin VII stimulate the chemotactic migration of FPR1-transfected RBL-2H3 cells, but not that of vector-transfected cells; moreover, scolopendrasin VII directly binds to FPR1. Our findings therefore suggest that the antimicrobial peptide scolopendrasin VII, derived from Scolopendra subspinipes mutilans, stimulates macrophages, resulting in chemotactic migration via FPR1 signaling, and the peptide can be useful in the study of FPR1-related biological responses. 목 적 : 특발성 신증후군의 연구를 위하여 병태생리 및 임상소견과 유사한 실험적 puromycin aminonucleoside(PAN) 신증을 이용하는데 본 연구는 배양한 사구체 상피세포에 대한 PAN의 영향을 통하여 신증후군의 주 원인 병태생리인 단백뇨의 기전을 밝히고자 하였다. 방 법 : 사구체 상피세포를 배양한 후 다양한 농도의 PAN과 항산화제를 투여하여 전자현미경관찰, 반응성 산소종 투과율 변화, confocal microscopy 등을 통하여 족세포성분의 변화를 관찰하였다. 결 과 : 사구체 상피세포의 초고배율소견에서 PAN에 의해 세포간극이 벌어지고 표면의 미세돌기가 단축되는 변화를 볼 수 있었다. 이러한 세포간극의 변화는 세포막부분의 ZO-1에 대한 면역형광검사에서도 확인할 수 있었다. DCF-DA로 측정한 반응성 산소종은 PAN에 의하여 농도에 따라 투여 2시간에 이미 유의한 증가를 보이나, 이러한 변화는 항산화제인 EGCG, probucol, vitamin C에 의해 감소하였다. 또한, 세포단층모델에서 투과율은 PAN에 의하여 농도에 따라 증가하나 항산화제에 의해 증가가 억제되었다. 세포골격구조인 ${\alpha}-actinin$은 사구체 상피세포의 세포질과 바깥 세포막부분으로 actin과 같이 분포하나 고농도의 PAN에 의해 세포질 바깥쪽의 일부분에 집중하는 형상으로 변하였다. 그러나 이러한 변화는 항산화제인 vitamin C의 처치에 의해 예방될 수 있었다. 세극막성분인 ZO-1는 고농도의 PAN에 의해 안쪽으로 이동하고 집중하는 형상으로 변하였으나, vitamin C의 처치에 의해 예방되었다. 이와 함께 ${\alpha}-actinin$과 ZO-1은 PAN에 의해 단백양이 감소하였으나 이는 항산화제에 의해 예방할 수 있었다. 결 론 : PAN은 사구체 상피세포의 반응성 산소종 생성을 증가시키고, 구조성분의 변화를 통하여 형태학적인 변화를 초래하며 이는 투과율의 증가로 나타났다. 이러한 변화들은 항산화제에 의해 어느 정도 억제할 수 있었음으로, PAN은 생체 외 사구체 상피세포에 산화스트레스기전을 통하여 구조적 변화와 이에 따른 단백뇨를 유발시키는 것으로 사료된다.
본 실험에서는 THP-1 세포의 PMA에 의하여 유도되는 초기 세포부착에 관한 메카니즘을 이해하기 위하여 다양한 요인(혈청, 신규 단백질의 합성, 세포 골격 저해제, 단백질 인신화 저해제)들의 효과를 조사하였다. 또한 본 실험에서는 이들 세포부착의 정도를 일반적으로 세포증식 분석에 사용되고 있는 SRB염색법을 도입하여 세포부착 분석에 간편한 방법의 조건을 확립하였다. PMA에 의한 초기 세포부착에는 배양액중의 혈청의 유무는 영향이 없었으나, 신규 단백질의 합성이 요구되는 것을 확인하였다. 또한 이들 초기 세포부착에 PMA처리에 의한 PKC의 활성화는 필수적이나, 그 하류 활성화 인자로 잘 알려진 MAP-kinase (erk1/2)의 인산화는 필요치 않음을 알 수 있었다. 흥미롭게도 액틴 중합 저해제인 cytochalasin D의 PMA와 공 처리는 오히려 세포부착을 PMA 단독 처리시 보다 증가시켰다. 또한 본 실험에서 사용된 SRB 염색법을 통한 세포부착 분석법은 최근 암 등 다양한 질환의 신약 표적 분자로 주목을 받고 있는 PKC 저해제의 초기 세포 기반 분석에 매우 유용하리라고 생각된다.
