The Morphological Characteristics of Anaerobic Granular Sludge (혐기성 입상슬러지의 형태학적 특성)

  • In this paper, the microscopic results of granular sludge from the two-phase UASB reactors are presented, particularly on the acidogenic phase where the distillery waste water is directly delivered. Distillery wastewater as a feed contained SS of 3.6~10.6 g/l and COD of 17.3~30.4 g/l. A 12.5 l and a 4.7 l UASB reactors were used for acidogenic and methanogenic phases, repectively. Morphological studies on the granular sludge consortia were made with both scanning and transmission electron microscopies. It was interesting enough to find the different colored and shaped granules at the acidogenic on the 90th day and at the methanogenic phase on the 120th day. The acidogenic granular sludge mainly consisted of acidogenic bacteria, such as long chains of large rods, short plump rods, and various size of cocci. Whereas the filamentous bacteria of Methanothrix spp. prevail in the methanogenic granule, with dirverse bacteria entrapped therein.

A Study on the Use of an Immobilized-Cell Acidogenic Reactor for the High Rate Digestion of a Distillery Wastewater (유기산 생산 세균을 고정화학 2상 메탄발효조에 의한 주정 폐수의 고효율 소화)

  • Anaerobic fermentative bacteria were isolated from the acidogenic reactor of a labora- tory scale 2-stage anaerobic digestor. The isolate 1-6 was selected for its ablity to produce more fatty acids from distillery wastewater than others, and was identified as a strain of Clostridium. The isolate Clostridium sp. 1-6 is a thermophilic bacterium growing at 55$\circ$c , and grew best at pH 5.5. An acidogenic reactor using immobilized cells of the isolate Clostridium sp. 1-6 removed about 15% of COD from distillery wastwater as hydrogen, producing about 50 mM butyrate and about 10 mM acetate, when the reactor was operated at the hydraulic retention time(HRT) of 0.8 hr. It is proposed that this system can be used to convert the distillery wastewater to hydrogen and butyrate. More than 90% of COD was removed from the wastewater by anaerobic digestion using a 2-stage digestor consisting of a UASB methanogenic reactor and an acidogenic reactor of the immobilized cells of isolate Clostridium sp. 1-6.

Characteristics and Roles of the Submerged Separation Membrane in Anaerobic Digestion (혐기성소화에 있어서 침지형분리막의 역학과 특성)

  • The substrate metabolism and bacterial population in an anaerobic digestion with the submerged separation membrane were investigated by using a laboratory-scale reactor at the hydraulic retention time(HRT) 1.0 and 0.5 day. The removal efficiencies of carbohydrate at the HRT 1.0 and 0.5 day were 99.8~99.9% and 98.0~99.6%, respectively. After the 58 days, the mixed liquor volatile suspended solids(MLVSS) concentration at the HRT 1.0 and 0.5 day were approximately 6,050 and 7,750 mg/L, respectively. According to the measurement by the most probable number(MPN) method, the numbers of acidogenic bacteria, $H_2$-utilizing and acetate-utilizing methc.nogenic bacteria were found to be $10^9$, $10^7{\sim}10^8$ and $10^6{\sim}10^8MPN/mL$, respectively. The composition of $CH_4$ in the produced gas was 46~50%. It is suggested that sulfate-reducing bacteria $10^7{\sim}10^8MPN/mL$ play an important role in producing $H_2$ and acetate in sulfate-depleted environment.

Effect of Phase Separation on Anaerobic Degradation of Phenol (페놀의 혐기성분해에 대한 상분리의 영향)

  • With the rapid industrialization, an ever-increasing quantity and kind of new organic compounds pose environmental problems due to their toxicity and physiological effect. However, research on the biodegradation of these compounds under anaerobic condition is very limited inspite of its efficiency and economical advantage. In this research, the pH effect on the ring cleavage of phenol under anaerobic condition was investigated, and the theory of phase separation was applied to the degradation of phenol for investigating the role of acidogenic bacteria. Results, obtained from biochemical methane potential(BMP) assay for 15.5 days of incubation, showed that acidic condition was more desirable for phenol degradation than alkaline condition. By both unacclimated methanogenic granular sludge and two mixed cultures, phenol was completely removed within six weeks of incubation with a gas conversion rate of over 86% of theoretical one. However, phenol was not degraded by unacclimated acidogenic culture, and thus it is considered as a syntrophic substrate. In case of phase separated biochemical methane potential(PSBMP) assay, in which acidogenic and methanogenic culture were seeded separately and consecutively, those that had been subjected to normal acidogens for 3 to 4 weeks showed higher gas production than those seeded with sterile or frozen culture.

  • The feasibility of hydrogen production with a raw seed sludge through direct acclimation of feedstock was investigated at acidogenic stage, and methane was harvested at followed methanogenic stage in an anaerobic two-stage process. Hydrogen content was higher than 57% at all tested organic loading rates (OLRs) and the yield of hydrogen ranged from 1.5 to 2.4 mol H2/mol hexose consumed and peaked at 6 gVSl-1day-1. Normal butyrate and acetate were main volatile fatty acids (VFAs), whereas the concentration of propionate was insignificant. The hydrogen-producing bacteria, Clostridium thermosaccharolyticum, was detected with strong intensity at all tested organic loading rates (OLRs) by denaturing gradient gel electrophoresis (DGGE) of the polymerase chain reaction (PCR) analysis. From COD balance in the process, the fraction of the feed-COD converted to the hydrogen-COD at acidogenic stage ranged from 7.9% to 9.3% and peaked at 6 gVSl-1day-1, whereas the fraction of feed-COD converted to the methane-COD at methanogenic stage ranged from 66.2% to 72.3% and peaked at 3 gVSl-1day-1.

  • Pilot scale(2.5 ton) three-stage methane fermentation process was developed for the rapid production of methane from food wastes in our laboratory. Eleven strains responsible for the primary semianaerobic hydrolysis/acidogenic fermentation system were isolated and characterized. Among them, the number of gram positive bacteria was eight and that of gram negative bacteria was three. They were rod and showed positive reaction to catalase. The strain K5 was found to have the highest enzyme activities of amylase and protease.

  • Anaerobic digestion is an alternative method to digest food wastes and to produce methane that can be used as a renewable energy source. We investigated bacterial and archaeal community structures in a three-stage methane production process using food wastes with concomitant wastewater treatment. The three-stage methane process is composed of semianaerobic hydrolysis/acidogenic, anaerobic acidogenic, and strictly anaerobic methane production steps in which food wastes are converted methane and carbon dioxide. The microbial diversity was determined by the nucleotide sequences of 16S rRNA gene library and quantitative real-time PCR. The major eubacterial population of the three-stage methane process was belonging to VFA-oxidizing bacteria. The archaeal community consisted mainly of two species of hydrogenotrophic methanogen (Methanoculleus). Family Picrophilaceae (Order Thermoplasmatales) was also observed as a minor population. The predominance of hydrogenotrophic methanogen suggests that the main degradation pathway of this process is different from the classical methane production systems that have the pathway based on acetogenesis. The domination of hydrogenotrophic methanogen (Methanoculleus) may be caused by mesophilic digestion, neutral pH, high concentration of ammonia, short HRT, and interaction with VFA-oxidizing bacteria (Tepidanaerobacter etc.).

Two-Phase Anaerobic Digestion of Food and Livestock Wastewater and Hygienic Aspects of the Digested Water (음식물폐기물과 축산분뇨 혼합폐수의 이상혐기소화에 따른 소화액의 위생성 연구)

  • Temperature phase anaerobic co-digestion process was conducted with the one to one mixture of food wastewater with livestock wastewater, and the presence and the dynamics of various pathogenic microorganisms was analyzed. The mixture contained various enteric and pathogenic bacteria, such as Escherichia coli. Enterobacteriaceae, Coliform bacteria. Staphylococcus aureus, Salmonella, Shigella, Listeria, and Yeast. Anaerobic digestion has become stabilized around 21 days after the reaction started, showing about 80% to 90% of remarkable reduction rates of microorganisms until this period in acidogenic reactor (AR) and methanogenic reactor (MR), respect ively. After stabilization, the average reduction rate of organic matter was recorded as around 60% in MR. Most microorganisms in the effluent were not detected at around the last period of the reaction, except Listeria and S. aureus, which showed the growth even at the last day of the reaction.

Study of Bacteria Associated with Dental Caries Using a 3 Tone Disclosing Agent (세가지 색상차이를 보이는 착색제를 이용한 치아 우식 관련 균에 관한 연구)

  • The aim of this study was to determine the efficacy of a 3 tone plaque disclosing gel in assessing the risk of caries related to the population of Streptococcus mutans, Streptococcus sobrinus, and Lactobacillus spp. quantified using a quantitative real-time polymerase chain reaction (qRT-PCR). 15 healthy children of ages 9 - 12 years were randomly examined. The 3 tone plaque disclosing gel was applied on teeth surfaces, which changed the color to pink or red, blue or purple and light blue. Plaque was divided into 3 groups based on staining. Genomic DNA from each sample was subjected to a qRT-PCR assay for quantitative detection of target bacteria. The Kruskal-Wallis test was conducted for correlation between the color of plaque and the number of bacterial species. The levels of S. mutans, S. sobrinus, and Lactobacillus spp. were significantly different in the plaque samples of the 3 groups (p < 0.05). The proportion of S. sobrinus to S. mutans showed correlation to the color of plaque. The different color-dyed plaque was related to the number of acidogenic bacteria. The 3 tone plaque disclosing gel could be used as one of the indicators to assess the clinical risk of caries associated with the population of S. mutans, S. sobrinus, and Lactobacillus spp.
