Determination of Total Arsenic in Drinking Water by Inductively Coupled Plasma-Mass Spectrometry (유도결합 플라스마 질량 분석법(ICP-MS)을 이용한 음용수 중의 전체 비소의 정량)

  • Total arsenic in drinking water such as spring, small water-supply system and mineral water was determined by inductively coupled plasma mass spectrometry. The contents of total arsenic were analyzed after acidification by nitric acid to become 1% in water samples. According to the results, total concentration of arsenic in drinking water was below 30ppb.

Validation of chlorophyll algorithm in Ulleung Basin, East/Japan Sea

  • The results of our observation in May 2000 indicated that the SeaWiFS algorithm (O'Reilley et al., 1998), which was adopted for OSMI data processing, overestimated the actual chlorophyll values. This was rather unexpected in that there were good reasons to expect that the bio-optical properties of East/Japan Sea belonged to Case 1 water and in such case, the OC2 algorithm would give unbiased estimates of actual chlorophyll a values. In November 2000, a cruise conducted bio-optical surveys in the same area. This time we added HPLC (High Performance Liquid Chromatography) method for measuring chlorophyll a concentration to the standard fluorometric method, which we hale been using during the past Fluorometric method with acidification is known to result in under/overestimation of chlorophyll values in many parts of the world oceans, while it is easier and cheaper than HPLC method. To our surprise, the comparison of HPLC chlorophyll and fluorometric chlorophyll values show that fluorometric values gave an underestimation up to 50%. This error was due to the presence of accessory pigments such as chlorophyll b. Considering this error, our precious result of May 2000(Yoo et al., 2000) might have to be reinterpreted. Calculation of reflectance at 490 and 555nm, however, indicated that this is not still enough to explain the discrepancies.

Isolation of Bacillus sp. SHL-3 from the Dry Soil and Evaluation of Plant Growth Promoting Ability

  • Excess use of chemical fertilizer causes soil acidification and accumulation of salt, and thus might bring to desertification of soil. To overcome this problem, it needs limited usage of chemical fertilizer and increased usage of natural fertilizer as an alternative. In this study, dry soil-borne Bacillus sp. SHL-3, which was isolated from arid and barren soil, with plant growth promoting activity was isolated for identification and to determine optimal culture condition. A bacterial strain SHL-3 had the IAA productivity ($5.16{\pm}0.10mg\;L^{-1}$), ACC deaminase activity ($0.36{\pm}0.09$ at 51 hours) and siderophore synthesis. It was identified as genus Bacillus sp.. Also, optimal culture condition of SHL-3 were $20^{\circ}C$ and pH 7 in LB medium. Bacillus sp. SHL-3 had up to 4% salt tolerance in the medium. We evaluated the plant growth promotion ability of SHL-3 using yam (Dioscorea japonica Thunb.). As a result, Bacillus sp. SHL-3 was effective on the increase of the shoot length (202.4% increase for 91 days). These results indicate that Bacillus sp. SHL-3 can serve as a promising microbial resource for the biofertilizers of soil.

Analysis on Fertilizer Application Uniformity of Centrifugal Fertilizer Distributor

  • Purpose: Chemical fertilizers contribute to agricultural productivity. Annually, 450,000 tons of chemical fertilizers are used in Korea, which is 268 kg per hectare (MAFRA, 2016). However, excessive use causes problems such as environmental pollution and soil acidification. This study proposes use conditions for a fertilizer distributor that can reduce excessive fertilization by analyzing distribution patterns. Methods: This study analyzed fertilizer application uniformity according to the number of blades on a centrifugal fertilizer distributor (three or four blades), orifice gate open ratio (50 or 100%), and blade rotation speed (400, 500, or 600 rpm). Results: When using four blades, the coefficient of variation (CV) was lower than when using three by 11-13% points, and the CV using the 50% open ratio was 10-30% points lower than using the 100% open ratio. The CV at 500 rpm blade rotating speed was 9-12% points lower than that for 400 and 600 rpm. Conclusions: The CV with four blades, 50% orifice gate open ratio, and 500 rpm of blade rotating speed was 18.4%, which provided the most uniform fertilization.

Lactic Acid Bacteria Mixture as Inoculants on Low Moisture Italian Ryegrass Silage Fermentation

  • The effects of lactic acid bacteria (LAB) mixtures on low moisture Italian ryegrass (IRG) silage fermentation was evaluated in field conditions. The experiment was categorized into two groups: Un-inoculated (Control) and Inoculated with LAB mixture for four storage periods (45, 90, 180, and 365 days, respectively). Silage inoculated with the LAB mixture had the lowest pH with highest lactic acid production than the control from beginning at 45-365 days at all moistures. Higher LAB counts were observed in inoculated silages than the control silages at whole experimental periods. It is a key reason for the rapid acidification and higher lactic acid production in silages during the storage periods. Overall results suggest that an adding of LAB mixture had positive effects on the increasing aerobic stability of silage and preserved its quality for an extended duration.

Antioxidant Effects of Avocado Seeds and Seed Husks as a Potential Natural Preservative (아보카도 씨와 씨 껍질의 항산화 효과)

  • As the consumption of avocado fruits and avocado oils is steadily increasing, the amount of avocado seeds which are thrown away as by-products is also inceasing. Thus, the possibility of use of avocado seeds as natural preservatives was studied focused on the antioxidant effect. The extraction of avocado seeds and seed husks with 100% ethanol by maceration showed highest antioxidant activities and lowest IC50 values compared to 80% ethanol extract. Furthermore, 100% ethanol extract of avocado seeds and seed husks included significantly higher amount of polyphenols than 80% extract. However, total flavonoid content of 100% avocado seed extract was not signigicantly different from 80% seed extract, whereas that of 100% avocado seed husk extract was significantly higher than 80% seed husk extract. In case of acid values, heating of oil alone for 120 and 180 min significantly increased the acid values, whereas the treatment of oil with seed and seed husk extract signficantly decreased the acid values. These results suggest that antioxidant effects of avocado seeds and seed husks protected the oil against heat-induced acidification. Thus, avocado seeds and seed husks have a potential to be developed as an natural antioxidant and natural preservative which could be used commercially.

Effect of Different Pediococcus pentosaceus and Lactobacillus plantarum Strains on Quality Characteristics of Dry Fermented Sausage after Completion of Ripening Period

  • The aim of this study was to evaluate the effect of three different strains of lactic acid bacteria (LAB) starter cultures: Pediococcus pentosaceus (KC-13100) (PP), Lactobacillus plantarum (KCTC-21004) (LP1), and L. plantarum (KCTC-13093) (LP2) on the physicochemical and microbiological characteristics, and sensory quality of dry fermented sausages after 21 days of drying and ripening period. Treatments added with PP and LP2 strains showed a significant higher (p<0.05) LAB and total plat counts, and water activity (aw) of all three treatments was below 0.85 after the completion of the ripening process. A significant variation (p<0.05) in pH values of treatments was exhibited due to the difference in acidification capacity of the LAB strains: LP2PP>LP2. Substantial variations (p<0.05) in shear force values were detected amongst three batches (LP2>LP1>PP). In sensory attributes, PP treated samples had significantly higher (p<0.05) color and overall acceptability scores. The current findings proved how important the optimal assortment of starter culture. Inoculation with PP produced importantly beneficial effects on sensory quality improvement of dry fermented sausage.

Characteristics of Lignin Removal in Cellulosic Ethanol Production Process (셀룰로오스 에탄올 생산공정에서 리그닌의 제거특성)

  • In this study, we measured changes in the lignin content of acidified lignocellulosic biomass such as rice straw, saw dust, chestnut shell and peanut hull and analyzed the conversion property to cellulosic ethanol. It turns out that the lignin content increases in chestnut shell, rice straw, saw dust, peanut hull order and the conversion property to cellulosic ethanol is superior in the reverse order. Thus, the removal of lignin by acidification is necessary. In addition, as the concentration of sulfuric acid increases, the lignin content decreases and the yield of cellulosic ethanol increased. The optimum concentration of sulfuric acid is 20 wt%.

  • Acid mine drainage (AMD) resulting from pyrite oxidation in mining areas, subsequently leads to soil acidification accompanied by lowering pH and high concentration of metals and metalloids in its surrounding environment. Regarding to this, the microbial amelioration has been considered as a promising option for a more cost-effective and eco-friendlier countermeasure, compared to the use of alkaline chemicals. This study was aimed to evaluate influencing factors in microbially-mediated amelioration of acidic soil spiked by simulated AMD. For this, microcosm experiments were conducted by acid-neutralizing bacterial consortium (dominated by Klebsiella sp. and Raoultella sp.) under the various conditions of AMD spikes (0-2,500 mg SO42-/L), together with acidic mine soil (0-100 g) or sphagnum peat (0-5 g) in the 200 mL of nutrient medium. The employed bacterial consortium, capable of resisting to high level of sulfate concentration (up to 1,500 mg SO42-/L) in low pH, generated the ammonium while concomitantly reduced the sulfate, subsequently contributing to the effective soil stabilization with an evolution of soil pH up to neutral. Furthermore, it demonstrates that suitable condition has to be tuned for successful microbial metabolism to facilitate with neutralization during practical application.

Lysosome Inhibition Reduces Basal and Nutrient-Induced Fat Accumulation in Caenorhabditis elegans

  • A long-term energy nutritional imbalance fundamentally causes the development of obesity and associated fat accumulation. Lysosomes, as nutrient-sensing and lipophagy centers, critically control cellular lipid catabolism in response to nutrient deprivation. However, whether lysosome activity is directly involved in nutrient-induced fat accumulation remains unclear. In this study, worm fat accumulation was induced by 1 mM glucose or 0.02 mM palmitic acid supplementation. Along with the elevation of fat accumulation, lysosomal number and acidification were also increased, suggesting that lysosome activity might be correlated with nutrient-induced fat deposition in Caenorhabditis elegans. Furthermore, treatments with the lysosomal inhibitors chloroquine and leupeptin significantly reduced basal and nutrient-induced fat accumulation in C. elegans. The knockdown of hlh-30, which is a critical gene in lysosomal biogenesis, also resulted in worm fat loss. Finally, the mutation of aak-2, daf-15, and rsks-1 showed that mTORC1 (mechanistic target of rapamycin complex-1) signaling mediated the effects of lysosomes on basal and nutrient-induced fat accumulation in C. elegans. Overall, this study reveals the previously undescribed role of lysosomes in overnutrition sensing, suggesting a new strategy for controlling body fat accumulation.
